An Experimental study on the effects of Hyperbaric oxygen therapy on the healing process of mandibular fracture of normal rats and streptozotocin-prduced diabetic rats (일반백서와 당뇨백서에서의 하악골절 치유시 고압산소요법이 미치는 영향에 관한 실험적 연구)

  • The main objectives of this study was to observe the effects of Hyperbaric oxygen therapy on the healing processes of mandibuar fracture of normal rats & streptozotocin - induced diabetic rats. Author used 120 rats (Sprague - Dawley Strain) dividing into controf (60) & experimental group (60) of normal & diabetic rats. Complete fracture was produced on the left mandibular body of 120 rats, rendered hyperbaric oxygen therapy (2 hrs, daily at 2.5 atm) on experimental group and observed effects of hyperbaric oxygen therapy microscopically. The obtained results were as follows : 1. The experimental group of HBO in normal rats had the good effect until 6th week, especially the better effect at 3rd week, because of decrease of inflammatory cell infiltration, heavy proliferation of fibroblast & capillary and active callus formation. 2. The hyperbaric oxygen therapy in mandibular fracture of diabetic rats influenced especially on the healing process at 5th week, because there were much decrease of inflammatory cell infiltrations, heavy proliferation of fibroblast, capillary, osteoblasts, moderate fibrous callus formation, osteocastic activity and mild bony callus formation.

High frequency plant proliferation via direct fronds regeneration of Korean endemic duckweed species (국내 자생 좀개구리밥 (Lemna paucicostata) 및 개구리밥(Spirodela polyrhiza)의 엽상체 증식을 통한 기내 식물체 대량 증식체계 확립)

  • High frequency plant proliferation system via direct frond regeneration of endemic duckweed plants Lemna paucicostata and Spirodela polyrhiza was established. Fronds of L. paucicostata and S. polyrhiza were able to multiply half-strength MS basal medium without plant growth regulators. However, addition of BA at a range of 1 to 3 mg/L was more effective than high concentration of BA treatments for fronds proliferation. Also half-strength MS salts was suitable for the fronds proliferation. Increase of salts concentration had inhibitory effect on fronds proliferation. Also the frequency of callus formation from fronds of L. paucicostata was 3.3%, when they cultured onto 1/2 MS medium supplemented with 1 mg/L of BA. Similarly the frequency of callus formation from S. polyrhiza was very low. After subculture of white globular structures derived from fronds of L. paucicostata, numerous globular somatic embryos and calluses were developed onto the surface of fronds. However these somatic embryos did not fully develop into normal plants when transferred to 1/2 MS basal medium. Therefore direct frond regeneration system was more efficient for mass proliferation of L. paucicostata and S. polyrhiza. The plant regeneration system of L. paucicostata and S. polyrhiza established in this study, might be applied to mass proliferation and genetic transformation for molecular breeding.

Isoflavones and biotransformed dihydrodaidzein production with in vitro cultured callus of Korean wild arrowroot Pueraria lobata (한국산 야생칡 캘러스에서의 이소플라본 및 생물전환에 의한 디하이드로다이드제인 분석)

  • Pueraria lobata, a medicinally important leguminous plant produces various isoflavones including puerarin, daidzin and daidzein which are metabolized to equol via dihydrodaidzein and tetrahydrodaidzein by the bacterial fermentation of natural isoflavone sources in human intestines. In this study, we described callus proliferation and isoflavone production in callus of Korean wild arrowroot and dihydrodaidzein biosynthesis in callus extract fermented with Pediococcus pentosaceus. Proliferation was the best at callus cultured in the medium containing 1.0 mg/L TDZ and 1.0 mg/L NAA at light condition for 12 days. Puerarin was significantly more produced at callus cultured in the medium containing 2.0 mg/L kinetin and 1.0 mg/L NAA at dark condition for 16 days, but daidzin and daidzein were not significant. Callus extract was successfully fermented with P. pentosaceus and dihydrodaidzein, which is one of equol precursors formed by biotransformation, was confirmed to be produced. These results will facilitate mass production of callus and isoflavones as equol precursors from Korean wild arrowroot and can be applied for the production of equol by biotransformation in vitro.

Embryogenic callus culture of Tribulus terrestris L. a potential source of harmaline, harmine and diosgenin

  • In the present study, a simple one medium formulation protocol for callus culture, somatic embryogenesis and in vitro production of ${\beta}-carboline$ alkaloids and diosgenin in Tribulus terrestris L. was developed. Extensive callus induction and proliferation was obtained in leaf explant on Murashige and Skoog (MS) medium supplemented with $5.0{\mu}M$ 6 benzyl adenine (BA) and $2.5{\mu}M$ ${\alpha}-naphthaleneacetic$ acid (NAA). The embryogenic callus was maintained on subculture to fresh parental medium at 4-week intervals over a period of 28 months. The frequency of embryo formation was at a maximum ($18.1{\pm}0.9$ per g of callus) on MS medium containing $5.0{\mu}M$ BA and $2.5{\mu}M$ NAA together with $75mg\;1^{-1}$ casein hydrolysate. Globular embryo developed into torpedo stage embryo under the influence of starvation. The accumulation of ${\beta}-carboline$ alkaloids (harmaline and harmine) and steroidal saponin (diosgenin) in non-embryogenic and embryogenic callus culture derived from leaf explant was compared with root, leaf, stem, and fruit of the mother plant. The embryogenic callus accumulated equivalent amounts of harmaline ($66.4{\pm}0.5{\mu}g/g$ dry weight), harmine ($82.7{\pm}0.6{\mu}g/g$ dry weight), and diosgenin ($170.7{\pm}1.0{\mu}g/g$ dry weight) to that of the fruit of T. terrestris. The embryogenic callus culture of this species might offer a potential source for production of important pharmaceuticals.

Functional Characterization of Callus Extracts of Apple 'Hirosaki' for Cosmetic Materials (사과 '히로사키' 캘러스 추출물의 기능성 화장품 소재로서의 특성)

  • In order to investigate functional characterization of callus extracts of apple 'Hirosaki' for cosmetic materials, biological activities of its extracts including wrinkle improvement, hair growth, and anti-inflammatory effect were investigated. The callus extract showed similar activity with TGF-${\beta}$ used as positive control at 50 ${\mu}g/mL$ in the test of collagen synthesis, and increased 40% of proliferation of hair follicle dermal papilla cells. Especially, in case of anti-inflammatory effect, callus extract inhibited about 50% of COX-2 expression which was known as response for intermediating inflammation, and about 70% of eotaxin-1 production which was increased by atopy dermatitis.

Improvement of Catechin Productivity in Callus Cultures of Camellia sinensis Leaves (차나무 잎의 캘러스 배양을 통한 카테킨류의 생산성 개선)

  • The effects of thiamine-HCl or polyvinylpyrrolidone (PVP) on growth and catechin contents of calli from Camellia sinensis leaves were investigated to improve catechin productivity in callus cultures on the selective medium. The growth of calli was great on the proliferation medium (the MS medium with 0.2 mg/L 2,4-D and 1.0 mg/L TDZ) supplemented with $20{\sim}30\;mg/L$ thiamine-HCl. Although the caffeine content was low in calli compared to young leaves, the total content of catechins was high in calli. Particularly,(-)-epicatechin (EC) which not detected in young loaves was also detected in calli on the propagation medium containing thiamine-HCl or PVP. In addition, the (-)-epicatechingallate (ECG) content was significantly higher in calli than in young leaves. In conclusion, the proliferation medium supplemented with 30 mg/L thiamine-HCl as a vitamin source seemed to be optimal condition for the growth and catechin production in callus culture.

Callus Induction and Plant Regeneration in Angelica koreana MAX. (강활(Angelica koreana MAX.) 조직배양을 통한 캘러스 유도와 식물체 재생)

  • This study was conducted to investigate the possibility of callus induction and plant regeneration from immature inflorescence, stem and petiole of A. koreana MAX. which is worth enough to be used as food and medicine. The callus induction and its proliferation was best when immature inflorescence segments were placed on MS medium supplemented with 2, 4-D 2mg / l. The white and compact embryogenic callus on the surface of dark yellow and soft callus which was induced from immature inflorescence segments came into being only on MS medium with 2, 4-D 1mg /l and 2mg /l, but didn't come into being on the other ones. The shoot came into being effectively from callus derived from immature inflorescence on MS medium mixed 2, 4-D 0. 1mg /l with Kinetin 1mg /l, and 2, 4-D 0.5mg /1 with Kinetin 2mg /l. Immature inflorescence was most appropriate material for callus induction and plant regeneration.

Induction of Callus from the Anther Culture of Prunus persica (복숭아(Prunus persica) 약배양(葯培養)에 의(依)한 Callus 유기(誘起))

  • In order to establish the effective method of producing calluses of Prunus persica, anthers were cultured on Nitsch's medium supplemented with combinations of several growth regulators. Anthers of tetrad stage were preserved in the refrigerator at $4^{\circ}C$ for 50~60 days. Calluses were embeded on the Murashige and Skoog's medium supplemented with multiplicate and differentiate the calluses. Changes of anther color, callus formation, and proliferation of haploid callus were observed under the different medium conditions. The results obtained were summarized as fellows: 1) The cultured anthers were turned dark brown 2~6days after were explanted anthers into the medium. 2) The anthers which were not changed in color were observed more frequently in the medium not added the growth regulators. 3) Calluses were induced from anthers which were turned dark brown and liberated from the anther slit. 4) BA. was very effective to induce calluses and to form the chlorophyll. The medium supplied with BA 0.5ppm were best to induce calluses. 5) The best medium supplied with BA 0.5ppm+IAA or 2.4-D were best to proliferation of calluses. 6) The medium was adjusted to pH 4.5 and supplied with 250mg/l of $CaCl_2{\cdot}2H_2O$ were induced calluses from anthers.

  • This study was conducted to optimize phytohormones combination and concentration on resveratrol production of Vitis vinifera callus cultures. TDZ was so effective for callus proliferation and resveratrol production and can be expected as a stimulus to the cells which keep the ability of producing resveratrol. We optimized the hormone combination of NAA 0.5 mg/L and TDZ 2 mg/L to stimulate resveratrol production very effectively. And callus under MS medium with the optimized phytohormones combination was treated by fungal elicitor, Botrytis.

In vitro Callus formation and Plant Regeneration of Epimedium koreanum Nakai

  • The efficiency of regeneration of callus and explants from leaf and stem disks of Epimedium koreanum was examined on the MS media containing 2,4-D, NAA, Kinetin, BA and TDZ. Calli were formed on the 2mg/l 2,4-D media at the rate of 32% from leaf discs and 52% from stems. No callus was produced on the media which are containing BA or TDZ alone. The combination of 2,4-D and BA showed the effect on the formation of callus. The combination of 2mg/l 2,4-D and 0.lmg/l BA in the MS media had produced the highest percentage of callus formation, 50% from leaf discs and 40% from stems, respectively. The combination of 2mg/l 2,4-D and 1mg/1 BA in the MS media had affected the formation of callus in the rate of 40% from leaf discs and 25% from stems. The combined plant growth regulators of 2,4-D and BA increased the formation of calli from leaf discs, but single treatment of 2,4-D showed the highest callus formation from stems. Multiple shoots from leaf discs were formed on the media containing NAA, BA, kinetin, and TDZ. The highest number of multiple shoots were obtained 0.1mg/l NAA combined with 1mg/l kinetin. As a result, leaf discs or stems can be used for the mass propagation of Epimedium koreanum, but stem elongation of shoots from calli was not easy.
