N6 배지에서 오차드그래스 캘러스로부터 빠른 재분화 (Rapid Regeneration of Plants on N6 Medium from Orchardgrass (Dactylis Glomerate L.) Calli)

  • 5품종의 오차드그래스 종자로부터 직접 캘러스를 유도하고, 형성된 캘러스로부터 식물체를 재분화하는 조건을 확립하였다. 공시품종증 합성 19호가 캘러스 유도 및 재분화에서 가장 우수한 것으로 판명되었으며, SH, MS, N6 배지주에서 캘러스 유도시에는 MS 배지가, 재분화시에는 N6 배지가 유리한 것으로 나타났다. 각 단계별 배지중 호르몬 첨가는 캘러스 유도 및 증식시에 dicamba $3\;mg/\;{\ell}$, 뿌리와 잎의 유도시에 NAA $1\;mg/\;{\ell}$와 kinetin $5\;mg/\;{\ell}$, 재분화 완성기에 무첨가한 조건이 가장 효율이 좋았으며, 캘러스로부터 완전한 식물체로 재분화되는데 필요한 시간은 약 50~80일이었다.

Multiple Shoots Regeneration and In vitro Bulblet Formation from Garlic Callus

  • The leaf segments of garlic (Allium sativum L.) were cultured in vitro and determined optimal concentration of plant growth regulators and sugars for callus induction, multiple shoots regeneration and in vitro bulblet formation. Highest yield of callus was observed in the leaf segment culture on Murashige and Skoog (MS) medium supplemented with 1.0 mg/L 2,4-D, 30 g/L sucrose and 8 g/L agar. Regeneration rate of multiple shoots from callus was high in the MS medium supplemented with kinetin 3.0 + NAA 3.0 mg/L or SA 1.0 + NAA 3.0 mg/L, containing 30 g/L sucrose. High rate of bulblet formation was observed as the concentration of jasmonic acid increased from 0.5 to 2.0 mg/L in medium, whereas addition of gibberellic acid significantly suppressed bulblet formation. The rate of in vitro bulbing was as high as $96\%$ in MS medium supplemented with 2.0 mg/L jasmonic acid and 120 g/l sucrose after two month culture at $25{\pm}1^{\circ}C$ under 16 hours day light.

벼 뿌리조직 유래의 캘러스로부터 체세포배 형성과 식물체 재분화 (Plant Regeneration and Somatic Embryo Formation from Root-Derived Callus of Rice)

  • 벼의 뿌리조직을 기내배양하여 배발생캘러스의 형성과 식물체 재분화율의 품종간 차이 및 완숙배와 뿌리조직에서 유래된 캘러스의 현탁배양 기간별 부정배 형성정도와 식물체 재분화율 등에 대한 몇가지 실험을 수행하여 얻어진 결과를 요약하면 다음과 같다. 벼 뿌리조직으로부터 캘러스 형성 및 식물체 재분화 능력의 품종간 차이는 뚜렷하게 나타났으며, 자포니카형 품종들이 통일형 품종에 비해 캘러스의 생장량도 많고 식물체 재분화율도 높은 경향이었고, 공시품종 중 "영덕벼"의 식물체 재분화율이 13%로 가장 높았다. 뿌리 및 현미배양에서 형성된 캘러스를 고체배지에서 2 주 간격으로 7회 계대배양한 바, 계대배양 횟수가 증가됨에 따라 식물체 재분화율이 증가되었다가 5회째부터는 점차 감소하였다. 뿌리조직에서 형성된 캘러스의 현탁배양에서 모양이 둥근세포와 그들의 세포괴는 배양 24일 후에 최대치를 나타내었고 그 이후는 감소하였다. 현탁배양 기간별 뿌리조직에서 형성된 캘러스의 체세포배 형성률은 배양 90일까지는 배양기간이 길어질 수록 증가하였으나, 그 이후는 감소하였고, 식물체 재분화율도 배양기간이 길어질수록 감소하는 경향이었다.소하는 경향이었다.

  • Callus formation and plant regeneration from the seeds of zoysiagrass cv. Zenith was tested on MS basal medium supplemented with various concentrations of 2,4-dichlorophenoxyacetic acid(2,4-D) and of several cytokinins. A concentration of 1mg/L 2,4-D on medium stimulated callus formation. In the presence of 5mg/L 2,4-D, addition of 1mg/L kinetin significantly enhanced callus formation and plant regeneration over 2,4-D alone. To transfer foreign DNA into turfgrass, parameters for the bombardment of embryogenic callus with the particle bombardment were partially optimized using transient expression assay of a $chimeric \beta$-glucuronidase(GUS) gene driven by the CaMV 35S promoter. GUS gene was strongly expressed at helium pressure 1,100 psi and 6~9cm target distance.

An Improved Method of Organogenesis from Cotyledon Callus of Acacia sinuata (Lour.) Merr. using Thidiazuron

  • An efficient protocol for in vitro multiple shoot bud induction and plant regeneration from mature green cotyledon derived callus tissues of Acacia sinuata has been developed. Callus formation occurs at all the concentrations of thidiazuron (TDZ) in Murashige and Skoog's (MS) medium, but 0.6 ${\mu}M$ proved to be the best with maximum callus formation frequency. Supplementation of TDZ in combination with indole-acetic acid (IAA) in MS media accelerates shoot bud organogenesis in differentiating callus tissues with 60-70% conversion of shoot buds into shoot Most efficient shoot organogenesis was recorded when TDZ induced calli were subcultured at different concentrations of 6-benzyla-denine (BA). Optimum shoot bud induction and plant regeneration from callus was achieved when 0.6 ${\mu}M$ (TDZ) induced calli were subcultured at 3.0 ${\mu}M$ (BA) where $16.6{\pm}0.74$ shoots/unit callus on obtained. Rooting in in vitro differentiated shoots was achieved when transferred to medium containing different concentration of indole-3-butyric acid (IBA) in full & half strength MS medium. The well rooted plantlets were hardened and transferred to net house with 90% survival rate.

  • This experiment was carried out to investigate the effects of plant growth regulators on the organogenesis from the tissue culture of Arabidopsis thaliana stem, and the origin of the callus development. When the stem segments were cultured on medium with 2mg/L IAA or NAA, adventitious roots and trichomes were differentiated after 11 days of culture. Callus vigorously formed on medium with 2/L2,4 after 7 days of culture, but adventitious roots and trichomes were not differentiated from callus after 10 days of culture. This results suggesting that picloram is very effective auxin for the callus formation and organogenesis. Callus weakly formed on 0.05mg/L kinetin, and formed on combination of auxins(2mg/L) with 0.05mg/L kinetin. But the effect of combination of auxins and kinetin the callus formation was less than 2,4-D or picloram alone. A histological examination of callus formed on picloram showed that phloram showed that phloem parenchyma cells were divided and enlarged after 2 days of culture. Cortex parenchyma cells were divided and meristematic nodules were developed from these cells after 4 days of culture. Finally, callus formed on outside of cortex and epidermis by division of meristematic nodules after 7 days of culture.

  • 'Ggoma' is a new Hibiscus dwarf type variety developed by gamma irradiation at the Korea Atomic Energy Research Institute (KAERI). This study was conducted to determine the best optimum cultural callus formation and shoot regeneration condition. Sterilized leaf tissues were cultured on MS (Marashige and skoog's) medium containing 3% sucrose, 0.8% agar with different concentration and combination of TDZ, 2, 4-D, KT, BA, and 2iP for 4 weeks in vitro culture. MS medium containing 2,4-D $0.1mg\;l^{-1}$ and BA $0.5mg\;l^{-1}$ were most effective on callus formation and growth. After 4 weeks, callus was transferred on BA (0.5, 1, $1.5mg\;l^{-1}$) and TDZ (0.1, 0.2, $0.3mg\;l^{-1}$) for shoot formation. The best condition for inducing the shoot from callus was BA $1.5mg\;l^{-1}$ and TDZ $0.3mg\;l^{-1}$. This result will be useful for the rapid multiplication of Hibiscus syriacus L.var. Ggoma.

강남콩(Phaseolus vulgaris L.) Callus의 원형질체 유이 및 배양 (Isolation and Culture of Phaseolus vulgaris L. Callus Protoplasts)

  • The isolatin and culture of protoplasts from hypocotyl originated callus of Phaseolus vulgaris cv. Damyang were carried out. The maximum protoplast yield of 4.6$\times$105 per gram fresh callus, using the 13-day-old callus, was obtained by digeston for 6 hours in the enzyme solution. After 10 day-culture of the isolated callus protoplsts, plating efficiency was 50%. Thereafter, cell cluster medium, and followed by leading to callus formation on an agar medium after 3 weeks of the liquid culture.

  • 배지, 절편체의 종류 그리고 생장조절제가 약용작물인 구기자나무 (Lycium chinese Mill. 'Cheongyang')의 캘러스 유도 및 신초 분화조건에 미치는 영향을 조사하였다. 배지 종류별 기관분화는 신초 분화율, 신초 발생수 및 생체중이 MS, B5, WPM 배지 순으로 양호하였다. 구기자나무의 캘러스 유도는 절간 절편체보다 잎 절편체가 좋았고, 캘러스는 0.5mg/L NAA와 0.2 mg/L BA가 첨가된 MS배지에서 효율적으로 유도되었다. 신초 분화에 관여하는 cytokinin으로는 TDZ보다는 BA가 좋았으며 BA 단일처리보다는 NAA와의 혼용처리에서 더 양호하였다. 신초분화는 0.01 mg/L NAA와 0.2 mg/L BA가 첨가된 MS 배지에서 가장 좋았다

국내육성 오차드그라스 품종들의 캘러스 형성율 및 식물체 재분화 효율 (Callus formation Ratio and Regeneration Efficiency of Orchardgrass Varieties Developed in Korea)

  • 국내육생 품종인 Hapsung 2, Jangbeol 101, Jangbeol 102와 대비품종인 Potomac의 종자로부터의 캘러스 형성율, 캘러스 크기, 식물체 재 분화율 및 재분화 효율을 4주령 및 6주령의 캘러스에 대하여 조사한 결과는 다음과 같다. 캘러스 형성율은 4주령 캘려스에서는 Jangbeol 102가 가장 높았고, 6주령 캘러스에서는 대비 품종인 Potomac아 가장 높았다. 캘러스 크기는 4주령 캘러스에서 대비품종인 Potomac (3.93cm)이 가장 컸으며, 6주령 캘러스에서는 Jangbeol 101 (4.32cm)이 가장 컸지만, 품종간의 차이는 적은 편이었다. 식물체 재분화율운은4주령 캘러스에서 10.3~17.7%의 넓은 범위의 차이를 보였는데, Jangbeol 101이 가장 높았으며 6주령 캘러스에서도 역시 11.2~37.4%로서 품종간에 큰편차를 보였으며, 특히 대비품종인 Potomac은 월등하게 재분화율이 높았다. 식물체 재분화 효율은 4주령 캘러스에서 그 범위가 6.8~11.5%로서 Jangbeol 102가 가장 높았으며, 6주령 캘러스에서는 범위가 7.2~25.6%로서 Potomac이 가장 높았다.
