Cultural Conditions for Mycelial Growth and Molecular Phylogenetic Relationship in Different Wild Strains of Schizophyllum commune

  • The common split-gilled mushroom, Schizophyllum commune is found throughout the world on woody plants. This study was initiated to evaluate conditions for favorable vegetative growth and to determine molecular phylogenetic relationship in twelve different strains of S. commune. A suitable temperature for mycelial growth was obtained at $30^{\circ}C$. This mushroom grew well in acidic conditions and pH 5 was the most favorable. Hamada, glucose peptone, Hennerberg, potato dextrose agar and yeast malt extract were favorable media for growing mycelia, while Lilly and glucose tryptone were unfavorable. Dextrin was the best and lactose was the less effective carbon source. The most suitable nitrogen sources were calcium nitrate, glycine, and potassium nitrate, whereas ammonium phosphate and histidine were the least effective for the mycelial growth of S. commune. The genetic diversity of each strain was investigated in order to identify them. The internal transcribed spacer (ITS) regions of rDNA were amplified using PCR. The size of the ITS1 and ITS2 regions of rDNA from the different strains varied from 129 to 143 bp and 241 to 243 bp, respectively. The sequence of ITS1 was more variable than that of ITS2, while the 5.8S sequences were identical. A phylogenetic tree of the ITS region sequences indicated that the selected strains were classified into three clusters. The reciprocal homologies of the ITS region sequences ranged from 99 to 100%. The strains were also analyzed by random amplification of polymorphic DNA (RAPD) with 20 arbitrary primers. Twelve primers efficiently amplified the genomic DNA. The number of amplified bands varied depending on the primers used or the strains tested. The average number of polymorphic bands observed per primer was 4.5. The size of polymorphic fragments was obtained in the range of 0.2 to 2.3 kb. These results indicate that the RAPD technique is well suited for detecting the genetic diversity in the S. commune strains tested.

축전식 탈염공정을 이용한 하수중의 용존염 제거특성 연구 (The removal characteristics of dissolved solid in wastewater during a capacitive deionization process)

  • Capacitive deionization(CDI) has many advantages over other desalination technologies due to its low energy consumption, less environmental pollution and relative low fouling potential. The objectives of this study are evaluate the performance of CDI which can be used for dissolved salts removal from sewage. To identify ion selectivity of nitrate and phosphate in multiionic solutions and adsorption/desorption performance related to applied potential, a series of laboratory scale experiments were conducted using a CDI unit cell with activated carbon electrodes. The CDI process was able to achieve more than 75 % TDS and $NO_3{^-}$, $NH_4{^+}$ removals, while phosphate removal was 60.8 % and is inversely related in initial TDS and $HCO_3{^-}$ concentration. In continuous operation, increasing the inner cell pressure and reduction of TDS removal ability were investigated which are caused by inorganic scaling and biofouling. However a relative mild cleaning solution(5 % of citric acid for calcium scaling and 500 mg/L of NaOCl for organic fouling) restored the electrochemical adsorption capacity of the CDI unit to its initial level.

한라산 지역 용천수의 수리지화학적 특성 (Hydrogeochemical Characteristics of Spring Water in Halla Mountain Region, Cheju Island)

  • 본 연구는 한라산 지역에 분포하는 대표적인 9개 용천수에 대한 고도별 수질특성과 그 진화과정을 연구하였다. 조사 지역 9개 용천수의 수질분석결과 관음사용천수는 수소이온 농도에서 비정상적인 수질특성을 보였다. 해발고도 1000m 이하에 위치하는 영실, 남국선원, 성판악, 어리목 및 관음사용천수에서는 염소이온, 황산이온, 질산성질소 및 나트륨이온 함량이 높아 주변 육상오염원들에 의해 오염이 진행 중인 용천수로 분류된다. 중탄산이온, 황산이온 및 수소이온 농도는 강수량이 많았을 때 그 농도가 증가하였으며, 염소이온, 칼슘이온 및 질산성질소 농도는 강수량이 많았을 때 그 농도가 감소하였고, 마그네슘이온, 나트륨이온 및 전기비전도도 농도는 강수량의 영향이 미미하였다. 한라산 용천수는 백록담 담수와 윗세오름용천수를 제외한 전 용천수가 나트륨 또는 칼륨형(sodium or potassium type)과 중탄산형(bicarbonate type)의 용천수군으로 구분된다.

Evaluation of Some Biochemical Parameters and Brain Oxidative Stress in Experimental Rats Exposed Chronically to Silver Nitrate and the Protective Role of Vitamin E and Selenium

  • Due to undesirable hazardous interactions with biological systems, this investigation was undertaken to evaluate the effect of chronic exposure to silver on certain biochemical and some oxidative stress parameters with histopathological examination of brain, as well as the possible protective role of selenium and/or vitamin E as nutritional supplements. Thirty six male rats were divided into six groups of six each: the first group used as a control group. Group II given both vitamin E (400 mg/kg) of diet and selenium (Se) (1 mg/L) in their drinking water. Group III given silver as silver nitrate ($AgNO_3$) (20 mg/L). Group IV given vitamin E and $AgNO_3$. Group V given both $AgNO_3$ and selenium. Group VI given $AgNO_3$, vitamin E and Se. The animals were in the same exposure conditions for 3 months. According to the results which have been obtained; there was an increase in serum lactate dehydrogenase (LDH), lipase activities and cholesterol level, a decrease in serum total protein, calcium and alkaline phosphatase (ALP) activity in Ag-intoxicated rats. Moreover, the findings showed that $Ag^+$ ions affected antioxidant defense system by decreasing superoxide dismutase (SOD) activity and increasing vitamin E concentration with a high level of malondialdehyde (MDA) in brain tissue. The histological examination also exhibited some nervous tissue alterations including hemorrhage and cytoplasm vacuolization. However, the co-administration of selenium and/or vitamin E ameliorated the biochemical parameters and restored the histological alterations. In conclusion, this study indicated that silver could cause harmful effects in animal body and these effects can be more toxic in high concentrations or prolonged time exposure to this metal. However, selenium and vitamin E act as powerful antioxidants which may exercise adverse effect against the toxicity of this metal.

  • 방사성(放射性) 물질(物質)을 오염(汚染)시킨 토양(土壤)에 무우와 배추를 pot 재배하면서 질소와 인산시비(燐酸施肥)에 따른 $^{90}Sr$ 흡수양상(吸收樣相)을 조사하였다. 질소와 인산시비(燐酸施肥)는 무와 배추의 수량증수(收量增收) 효과가 있었으며 무우는 질산태질소가, 배추는 암모니아태질소가 더욱 효과적이었고 인산시비(燐酸施肥)는 식물체의 K 함량을, 질소 시비(施肥)는 Ca함량을 증가시켰다. 또한 배추에 비해 무가 $^{90}Sr$ 흡수량(吸收量)이 높았고 질소와 인산시비(燐酸施肥)는 $^{90}Sr$ 흡수(吸收) 억제효과(抑制效果)가 있었으며 암모니아태 질소보다는 질산태질소가 더욱 효과적(效果的)이었다.

물유리를 이용한 실리카계 박막의 광학적 및 기계적 특성 (Optical and mechanical properties of silicate film using a water glass)

  • 본 연구에서는 물 유리의 경제성을 바탕으로 광학적 및 기계적인 특성을 조사하여 투명하고 높은 경도를 가진 표면 보호막의 기능을 검토하기 위하여 $SiO_2-Na_2O-R_mO_n$계 박막을 제조하였다. 물 유리에 CaO와 $Al_2O_3$를 소량의 1 N HCl 1N $NH_4OH$와 함께 각각 첨가하여 코팅용 졸을 준비하였다. Stainless steel. Si wafer. soda-lime-silica glass등 다양한 기판 위에 spin-coating 한 후 질소 분위기 하에서 500, 750 및 $900^{\circ}C$로 최종 열처리를 행했다. 제조된 막은 Knoop 경도계로 micro-hardness를 측정하였다. 막의 표면 질소 함유량을 알아보기 위하여 EDX 분석을 행하였다. 그리고 Field Emission Scanning Electron Microscope(FE-SEM)을 이용하여 막의 표면구조를 관찰하였으며, UV-VIS 스펙트라 측정을 통하여 막의 두께와 반사 특성을 조사하였다.

2010년 서울에서 관측한 황사와 연무사례의 물리, 화학, 광학적 특성비교 (Physical, Chemical and Optical Properties of an Asian Dust and Haze Episodes Observed at Seoul in 2010)

  • This study investigated physicochemical and optical characteristics for three episodes of Asian dust, stagnant haze and long-range transport haze and for one clean day. $PM_{10}$ mass concentration during Asian dust and two haze days was increased by 2~9 times compared to that of clean episode. During Asian dust episode, coarse particle concentration was increased and the mass concentration of calcium in a coarse mode ($1.8{\sim}10{\mu}m$) was $5.4{\mu}g/m^3$ which was 7 times higher than that of clean episode. The calcium was presented as a form of $CaCO_3$ in a coarse mode. During the two haze episodes, fine particle (< $1.8{\mu}m$) concentration was increased and secondary inorganic pollutants such as sulfate, ammonium and nitrate composed of 90% of the total ions. $(NH_4)_2SO_4$ and $NH_4NO_3$ were dominant in a fine mode for stagnant haze episode. But they were the most dominant form in both fine mode and coarse mode for long-range transport haze episode. According to the optical properties for each episode (Asian dust, stagnant haze and long-range transport haze) were classified as dust, black carbon and mixture, respectively.

토양에서 분리한 Aspergillus sp. PS-104 균주에 의한 난용성 인산염 분해 (Solubilization of Insoluble Phosphates by Aspergillus sp. PS-104 Isolated from Soil)

  • PDA-calcium phosphate 평판배지를 이용하여 경상남북도 일대의 토양으로부터 인산가용화능이 우수한 사상균을 분리하고 배양특성을 조사하였다. 최종적으로 분리한 인산가용화능이 우수한 사상균 PS-104 균주를 선발하여 형태적 특성을 조사한 결과 이 균주의 분생자는 비가지형으로 끝이 돌출되어 있으며, 균사가 격막으로 형성되는 등의 특징을 갖는 Aspergillus sp. PS-104 균주로 동정되었다. 이 균주의 인광석 분해능이 최대가 되는 최적 배양온도와 pH는 각각 $30^{\circ}C$와 pH 7.0이었다. 이와 같은 배양조건에서 tricalcium phosphate, aluminium phosphate, hydroxyapatite, 인광석 등의 난용성 인산염에 대한 이 균주의 인산가용화능을 살펴보면 최대 인산가용화능은 tricalcium-phosphate 처리구(1,900 ppm)에서, 최소 인산가용화능은 hydroxyapatite 처리구(320 ppm)에서 확인되었다. 또한 이 균주의 인산가용화능은 질소원으로 첨가한 ammonium 염보다 nitrate 염에서 높게 나타났다.

  • 김의 성장과 품질에 대한 환경수질의 중요성은 다시 말할 필요가 없다 김 양식기에 있어서의 환경수질의 변동을 보기 위해 저자의 한 사람은 섬진강 및 낙동강 하구 김밭에 대해 이미 조사한 바 있거니와 이번에는 우리나라 유수의 김 생산지인 완도군하 김밭의 수질을 전보와 같은 방법으로 조사하였다. 조사지점으로서는 완도군 에서대체로 대표적 김밭이라 볼 수 있는 두 지점을 선정하여 1968년 10월부터 1969년 2월까지 매월 대조일에서의 수질변동을 조수의 일조기에 걸쳐 1시간마다 각 성분량을 측정하므로써 조사하였다.

Hydrogeochemical study of a watershed in Pocheon area: controls of water chemistry

  • The groundwater in the Pocheon area occurs from both a fractured bedrock aquifer in igneous and metamorphic rocks and an alluvial aquifer with a thickness of <50 m, and forms a major source of domestic and agricultural water supply. In this study, we performed a hydrogeochemical study in order to identify the control of geochemical processes on groundwater quality. For this study, groundwater level and physicochemical parameters (EC, Eh, pH, alkalinity) were monitored once a month from a total of 150 groundwater wells between June 2003 to August 2004. A total of 153 water samples (13 surface water, 66 alluvial groundwater, 74 bedrock groundwater) were also collected and analyzed in February 2004. Groundwater chemistry in the study area is very complex, depending on a number of major factors such as geology, degree of chemical weathering, and quality of recharge water. Hydrochemical reactions such as the leaching of surficial and near-solace soil salts, dissolution of calcite, cation exchange, and weathering of silicate minerals are proposed to explain the chemistry of natural groundwater. Alluvial groundwaters locally have very high TDS concentrations, which are characterized by their chloride(nitrate)-sulfate-bicabonate facies and low Na/Cl ratio. Their grondwater levels are highly fluctuated according to rainfall event. We suggest that high nitrate content and salinity in such alluvial groundwaters originates from the local recharge of sewage effluents and/or fertilizers. Likewise, high concentrations of nitrate were also locally observed in some bedrock groundwaters, suggesting their effect of anthropogenic contamination. This is possibly due to the bypass flow taking place through macropores. Tile degree of the weathering of silicate minerals seems to be a major control of the distribution of major cations (sodium, calcium, magnesium, potassium) in bedrock groundwaters, which show a general increase with increasing depth of wells. Thermodynamic interpretation of groundwater chemistry shows that the groundwater in the study area is in chemical equilibrium with kaolinite and Na-montmorillonite, which indicates that weathering of plagioclase to those minerals is a major control of hydrochemistry of bedrock groundwater. The interpretation of the molar ratios among major ions, as well as the mass balance calculation, also indicates the role of both dissolution/precipitation of calcite and Ca-Na cationic exchange as bedrock groundwaters evolves progressively.
