Zinc Deficiency Decreased Alkaline Phosphatase Expression and Bone Matrix Ca Deposits in Osteoblast-like MC3T3-E1 Cells

  • It is well established that zinc plays an important role in bone metabolism and mineralization. The role of zinc in bone formation is well documented in animal models, but not much reported in cell models. In the present study, we evaluated zinc deficiency effects on osteoblastic cell proliferation, alkaline phosphatase activity and expression, and extracellular matrix bone nodule formation and bone-related gene expression in osteoblastic MC3T3-E1 cells. To deplete cellular zinc, chelexed-FBS and interpermeable zinc chelator TPEN were used. MC3T3-E1 cells were cultured in zinc concentration-dependent (0-15 ${\mu}M\;ZnCl_2$) and time-dependent (0-20 days) manners. MC3T3-E1 cell proliferation by MTT assay was increased as medium zinc level increased (p<0.05). Cellular Ca level and alkaline phosphatase activity were increased as medium zinc level increased (p<0.05). Alkaline phosphatase expression, a marker of commitment to the osteoblast lineage, measured by alkaline phosphatase staining was increased as medium zinc level increased. Extracellular calcium deposits measured by von Kossa staining for nodule formation also appeared higher in Zn+(15 ${\mu}M\;ZnCl_2$) than in Zn-(0 ${\mu}M\;ZnCl_2$). Bone formation marker genes, alkaline phosphatase and osteocalcin, were also expressed higher in Zn+ than in Zn-. The current work supports the beneficial effect of zinc on bone mineralization and bone-related gene expression. The results also promote further study as to the molecular mechanism of zinc deficiency for bone formation and thus facilitate to design preventive strategies for zinc-deficient bone diseases.

난소적출 수술을 받은 흰쥐에서의 골밀도, 골강도 및 골대사의 생화학적 지표에 대한 한방요법의 효과 (Effects of Alternative Medicine Extract on Bone Mineral Density, Bone Strength and Biochemical Markers of Bone Metabolism in Ovariectomized Rats)

  • 본 실험은 칼슘 결핍과 난소 제거로 유발한 골다공증에 대한 한방요법의 효과에 대하여 고찰하였다. 1) 난소 제거로 인한 에스트로겐의 결핍 및 칼슘 결핍은 폐경여성에서 관찰되어지는 것처럼 골강도 및 골밀도의 손실을 일으켰다. 2) 한방 추출물의 투여는 에스트로겐 결핍 및 칼슘 결핍으로 유도된 골강도 및 골밀도 손실에 대한 예방효과가 관찰되었다 3) 난소제거로 인한 에스트로겐의 결핍 및 칼슘 결핍은 골대사의 생화학적 지표들(혈청 중 alkaline phosphatase, acid phosphatase, osteocalcin, 그리고 뇨중 deoxyopyridinoline 농도)에 영향을 미치지 않았다. 4) 한방 추출물의 투여 또한 골 대사의 생화학적 지표들에 영향을 미치지 않았다. 결론적으로, 한방 추출물의 골손실에 대한 예방효과는 한방 추출물의 골다공증 치료제로서의 가능성을 제시하며, 새로운 골다공증 치료제 개발 일환으로 한방추출물의 적용기간 및 농도 변화를 통한 계속적인 연구가 요구되어진다.

폐경 전$\cdot$후 여성의 신체 계측치와 영양소 섭취량이 골밀도 및 골밀대사 지표에 미치는 영향 (Effects of Anthropometric Indices and Nutrients Intake on Bone Mineral Density and Biochemical Markers of Bone Metabolism in Korean Premenopausal and Postmenopausal Women)

  • This study investigated the influence of anthropometric data and nutrient intake on bone mineral density(BMD) and biochemical markers of bone metabolism The mean age of 21 premenopausal women were 47.0 years and that of 41 postmenopausal women whose menopausal age was 49.46 years were 60.56 years. The waist and WHR of postmenopausal women were significantly higher than those of premenopausal ones. The animal protein intake of premenopausal and postmenopausal women were 38.5 and 21.03 g which comprised 54.35 and $31.84\%$ of total protein intake, respectively. The calcium intake of premenopausal and postmenopausal women were 446.45 and 546.97mg which was 63.78 and $78.14\%$ of Korean RDA, respectively. The ALP(Alkaline phosphatase) of premenopausal women was 65.81 U/L, which was significantly lower than that(90.24 U/L) of postmenopausal women (p<0.01). BMD of lumbar spine of premenopausal women was correlated significantly with body weight(r=0.690, p<0.01), waist(r=0.682, p<0.01), WHR(r=0.672, p<0.01), BMI(r=0.559, p<0.01), and body fat(r=0.457, p<0.01). Urinary Ca/creatinine ratio of the premenopausal women was negatively correlated with plant protein(r=-0.529, p<0.05) and plant calcium(r=-0.579, p<0.05). BMD of lumbar spine of postmenopausal women showed positive correlation with lean body mass(r=0.469, p<0.01) and body weight(r=0.383, p<0.05). Urinary Ca/creatinine ratio for the postmenopausal women was positively correlated with ALP(r=0.404, p<0.01) and urinary Na/creatinine ratio(r=0.389, p<0.05). In conclusion, it is necessary to maintain adequate body weight and to increase calcium intake for the premenopausal women. It is also important to increase muscle mass and reduce salt intake for the postmenopausal women.

충남지역 일부 여대생의 칼슘 섭취상태와 혈청 골대사지표 , 단백질 및 지질 수준과의 관계에 대한 연구 (Relation among Calcium Intake , Bone Metabolism Parameters , Serum Protein and Lipids of Female College Students in Chungnam)

  • The purpose of this study was to investigate the relationship among calcium intake, blood parameters related with bone metabolism, and serum lipids in healthy adults on self-selected diet. Subjects were consisted of 40 female college students residing in Chungnam. Anthropometric measurements, dietary intake measurements and blood collection were conducted. Serum concentrations of total protein, albumin, alkaline phosphates, leucine amino peptidase, BUN, calcium, inorganic phosphorus, and lipids were measured by biochemical analyzer and ICP spectrometer. The results were as follows. The mean age of subjects was 22.34 years and weight, height and BMI were 52.89kg, 161.29cm and 20.34, respectively. The daily mean energy and calcium intakes were 81.75% and 64.38% of RDA. The mean animal 1:2. The mean serum concentrations were 6.54g/dl(total protein), 4.12g/dl(albumin), 123.24U/(alkaline phosphates), 36.59U/l(leucine amino peptidase), 8.26mg/dl(calcium), 3.29mg/dl(inorganic phosphorus), 60.73mg/dl(triglyceride), 138.49mg/dl(total cholesterol), 65.95mg/dl(HDL-cholesterol), and 60.39mg/dl(LDL-cholesterol). There were no significant differences among calcium intake, bone metabolism parameters, and serum lipids when analyzed by Pearson's correlation coefficient. More systematic studies are required to investigate the roles of calcium in healthy persons on self-selected diets containing different levels of calcium.

난소절제 흰쥐의 골격대사에 대한 식이 칼슘과 대두 이소플라본의 섭취효과 (Effects of Dietary Calcium and Soy Isoflavones Supplementation on Bone Metabolism in the Ovariectomized Rats)

  • 본 연구는 난소절제 휜쥐를 이용하여 저$(0.1\%)$ 및 정상 $(0.5\%)$ 칼슘 식 이 에 대두 이소플라본 첨가수준을 달리 하여 급여했을 때 뼈 무기질 함량에 미치는 영향을 조사했으며 그 결과는 다음과 같다. 체중은 난소절제에 의해 증가하였고 식이 칼슘 및 대두 이소플라본의 첨가에 따른 효과는 없었다. 혈청 칼슘 농도는 난소절제, 식이 칼슘 및 대두 이소플라본에 의해 변화되지 않았으며 혈청 인의 농도는 정상 칼슘 식이 급여시 감소하였으나 정상 범위였고, 대두 이소플라본은 이에 영향을 미치지 않았다. 혈청 ALP활성은 난소절제에 의해 증가하였는데,그 정도는 저 칼슘 식이군에서 더 높았고 대두 이소플라본은 이를 변화시키지 못하였다. 혈청 TrACP활성은 난소절제, 식이 칼슘 및 대두 이소플라본에 따른 차이를 나타내지 않았다. 대퇴골 및 요추골의 무게 및 크기는 난소절제, 식이 칼슘, 대두 이소플라본에 의해 영향을 받지 않았다. 대퇴골의 파단력은 난소절제 에 의해 변화되지 않았으나 식이 칼슘은 이를 증가시켰다. 대퇴골 및 요추 골의 칼슘과 인의 함량은 난소절제에 의해 감소하였고 대두 이소플라본 80 ppm 첨가는 이를 증가시켰다. 식이 칼슘의 영향은 요추골에서 나타나 정상 칼슘식이를 급여했을 때 요추골의 칼슘과 인의 함량을 증가시켰다. 대두 이소플라본 160 ppm첨가는 뼈의 칼슘과 인의 함량을 증가시키지 않았다 이상의 결과로부터 난소절제한 흰쥐에서 대두 이소플라본 80 ppm을 첨가한 식이를 급여하면 뼈의 무기질 함량을 증가시킬 수 있으며 이런 효과는 식이 칼슘이 정상일 때 더 높은 것으로 나타났다. 본 연구의 결과는 대두 이소플라본의 첨가는 폐경성 골다공증을 예방하는데 유효하며 섭취하는 칼슘이 결핍되지 않을 때 그 효과가 더 높을 가능성을 제시하였다.

가임기 동안 칼슘섭취 부족 흰쥐에서 임신기의 칼슘보충 수준이 무기질대사에 미치는 영향 (Effects of Calcium Supplementations on Mineral Metabolism during Pregnancy with Calcium-Deficient Young Adult Rats)

  • We examined the effects of calcium intake levels on mineral metabolism during pregnancy using calcium-deficient young adult rats. Five week-old female Sprague-Dawley rats were fed normal Ca (NCa, 0.5%) and low Ca (LCa, 0.15%) diets for five weeks (pre-pregnancy). The low Ca intake group was then divided into three groups and fed low Ca (0.15%), normal Ca (0.5%) and high Ca (1.5%) diets for 3 weeks (pregnancy). All of the rats were mated with normal male rats. The control group was fed a consistently normal Ca (0.5%) diet during pre-pregnancy and pregnancy. On the day after delivery, dams and their pups were sacrificed. We measured total protein, albumin, alkaline phosphatase and mineral content in serum and weight, length, breaking force, ash and mineral content in the femur and lumbar (L2-L4) of the dams. Whole body mineral content was measured in the pups. There was no difference in weight gain and food intake among the groups. Serum total protein and albumin were in the normal range but a little lower during pregnancy. High Ca supplementation decreased serum Mg and Fe during pregnancy. Weight, ash and Ca of the femur and lumbar significantly decreased in rats fed a chronically low Ca diet during pre-pregnancy and pregnancy. Calcium supplementation levels were above normal during pregnancy and increased the bone weight and breaking force of rats fed the low Ca diet during pre-pregnancy. However, Ca supplementation did not increase the ash and Ca contents in the bones. High Ca supplementation during pregnancy significantly decreased Mg in the bones and increased Ca and P in the kidneys. Ash content of pups from dams fed the chronically low Ca diet decreased but there was no difference in whole body Ca among the groups. Mg and Fe in the whole body decreased in pups from dams fed the high Ca diet. Pregnancy performance was reduced in dams fed the low Ca diet. These results suggest that above normal Ca supplementation levels during pregnancy restored maternal bone status to some extent in rats fed the chronically low Ca diet. The same could not be said for mineral content. Also, high Ca supplementation during pregnancy may deteriorate mineral composition in bones and other tissues. Therefore, more detailed research is needed to facilitate sound recommendations on appropriate calcium intake during pregnancy. (Korean J Nutrition 36(5): 459∼469, 2003)

식습관과 노년기 골격상태와의 관계 연구 (The Study of the Relationship between Food Habits and Bone State in the Elderly)

  • To investigate the effect of food habits on the bone state of the senior citizens, two groups were tested: one(111 senior citizens) was healthy ordinary senior citizens over 65 years old and the other(51 senior citizens) was patients distinguished as having osteoporosis. The present dietary intake was estimated by a 24-hr recall method, and individual history. For the data analysis, percentages and frequencies were calculated and χ²-test was undertaken to test the relation among values. The following results were obtained: patient group with osteoporosis was less in height and weight than the group of ordinary senior citizens(160.33cm, 59.99kg). It was much less than the average Korean senior citizens(158cm, 54.9kg). Food appetite in the group of patient was worse than that of ordinary senior citizens group. According to their dietary history(58.8%), the food intake pattern was most of vegetables(62.0%). Eventhough they haven't been intaken milk after recognizing of their osteoporosis(74.5%). Most of them didn't improve their food habits to help Ca metabolism. Also they have depress of their life(50%). All subjects certainly took insufficient energy, Ca, protein from their diets. Moreover the major source of Ca were vegetables, seaweeds and legumes.

두충(杜冲)이 난소적출(卵巢摘出) 흰쥐의 골조직(骨組織) 형태(形態)와 골대사(骨代謝)에 미치는 영향(影響) (Effects of Eucommiae Cortex (杜冲) on the Bone Histomorphometry and Metabolism in Ovariectomized Rats)

  • Objective : This study was undertaken to investigate the effects of Eucommiae Cortex on the osteoporosis in ovariectomized rats. Materials and Methods : We used Sprague-Dawley female rats in 8-week-old. They were divided into three groups. Sample group was ovariectomized and administered with 10 mg/100 g/day Eucommiae Cortex extract solution for 10 weeks. Control group was ovariectomized and sham group was conducted by sham's operation. And control group and sham group were administered with normal saline as the same way. We measured rats's body and uterus weight and also measured the serum levels of Ca, phosphorus and ALP. We stained the specimens of rat's tibial bones with Goldner's modified Masson's Trichrome and then examined bone histomorphometry with Bioquant computer program of image analysis system. We measured the thickness of osteoid and callus as static parameters and measured bone volume and mineral apposition rate as dynamic parameters. We observed the expressions of RANKL and OPG mRNA of the tibial bone by RT-PCR. Results : The body weight was significantly (p<0.05) increased in control and sample groups compared with sham group, respectively. The uterus weight was significantly (p<0.05) decreased in control and sample group compared with sham group. In the change of Ca, phosphorus and ALP there were no significant changes among three groups. There were no significant changes of trabecula cortical and osteoid bones' thickness and volume. But trabecula mineral apposition rate (MAR) was significantly (p<0.05) increased in sham and sample group compared with control group. In the expression of RANKL mRNA, sample group was decreased compared with control group, and in that of OPG mRNA, sample group was increased compared with control group. Conclusion : This study shows that Eucommiae Cortex has the beneficial effects on bone histomorphometry and metabolism in the ovariectomized rats. We suggest that Eucommiae Cortex be useful for the treatment of osteoporosis.

칼슘급원식품의 체내이용성 연구 (A Study on the Bioavailability of Dietary Calcium Sources)

  • This study was conducted to investigate the effect of various types of calcium sources on calcium metabolism. Sprague-Dawley male rate weighing approximately 82g were divided into 6 groups and fed experimental diets containing about 0.2% calcium for 4 weeks. Perilla leaves, dried sea mustard, mulberry leaves, loach, skim milk powder, and CaCO3 were used as calcium sources for this study. Food intake of experimental groups showed no significant difference from that of control group, but food efficiency ratio were higher in group fed loach as a calcium source. Apparent calcium absorption from perilla leaves, and skim milk powder groups as good as that in CaCO group. Femur length showed no significant difference among exjerimental groups with different calcium sources. The breaking force of bone was higher in loach and dried sea mustard groups. Weight, ash weight, and calcium content of the femur were higher in the loach diet group than in the others. Thus, calcium from not only skim milk powder but also perilla leaves, dried sea mustard, mulberry leaves, and loach appears readily available and all of these can be recommended as calcium sources.

한국산 홍화씨분말 보충식이의 급여가 골절된 흰쥐의 골대사지표에 미치는 영향 (The Effects of Korean Safflower(Carthamus tinctorious L.) Seed Powder Supplementation Diet on Bone Metabolism Indices in Rats during the Recovery of Rib Fracture)

  • This study was carried out to investigate the effects of Korean safflower(Carthamus. tinctorious L.) seed powder supplementation on bone metabolism during the recovery of rib-fracture induced by surgical operation in rats. Male Sprague-Dawley rats of 10 weeks old, weighing 370$\pm$5g, were divided into two groups after arrival : the control group(C group, AIN-76 semifurified diet) and safflower seed group(S group, AIN-76 semifurified diet+10% safflower seed powder). They were fed experimental diets for 12 days before the rib fracture operation and for 30 more days after the operation. A number 9 rib was fractured surgically and sham-operation was also performed. After the rib-fracture operation, rats were sacrificed on the 8th, 11th, 16th, 21st, and 30th day, respectively. The body weights were decreased on the 8th day after operation in both groups but recovered gradually thereafter. The fractured sections of the S group were more strongly adhered and more rapidly repaired than those of C group. Effects of safflower seed powder supplementation on indices of bone metabolism during the repairing of rib-fracture can be summarized as following. There were no differences between C and S group. In the levels of PTH and calcitonin, which regulate the concentration of plasma calcium. However, PTH level on the 8th day was significantly higher than that on the 16th day in control group. There were also no differences between groups or within groups in the levels of calcitonin. Osteocalcin levels, one of the bone-formation indices, were similar in two groups. Activities of total-ALP in plasma were highest at the 8th day in both groups and decreased thereafter, except on the 30th day in control group. The activities of bone-specific ALP that is directly related to bone calcification, showed higher levels in the S group than in the C group throughout the experimental period. They were significantly increased in the S group on the 11th, 16th, and 21st days. In comparisons of plasma calcium and phosphate levels, Ca levels were tended to be lower in the S group than in the C group and P levels showed the opposite trend. It seems that the fractured ribs were more rapidly repaired in rats supplemented with safflower seed powder than in control rats, possibly due to stimulation of bone calcification from increased activity of bone-specific ALP.
