PLGA 미립구로부터 G-CSF의 방출 거동 양상을 향상 시키기 위하여 G-CSF를 분자량 5000인 methoxy polyethylene glycol-aldehyde로 PEGylation 시켰다. 대부분의 G-CSF는 mono-PEGylation 되었으며, 이를 SDS-PAGE, HPLC, 및 펩타이드 지도 분석을 통해 확인하였다. W/O/W 방법을 사용하여 G-CSF 및 PEGylated G-CSF의 PLGA 미립구를 제조하였으며, 이때 봉입율은 높은 상태였다. 미립구내로 더 많은 G-CSF를 봉입하기 위하여 액상의 G-CSF 및 PEGylated G-CSF을 농축하였고 native gel과 gel filtration 크로마토 그래피를 통하여 단백질이 안정함을 확인하였다. 이렇게 제조한 PLGA 봉입체의 in vitro 방출 거동을 조사한 결과 PEGylated G-CSF는 native G-CSF에 비하여 더 오랜 시간 동안 방출이 지속되었고 최대 방출량도 증가하였다.
인간 과립구 성장인자(hG-CSF)는 골수에서 생산되는 단백질로 호중구의 분화 및 생성을 촉진시키는 역할을 한다. 현재 재조합 hG-CSF는 암화학요법에 의한 호중구감소증, 골수이식시 호중구 감소증, 재생불량성 빈혈에 수반되는 호중구 감소증 등으로 적응증이 확대되고 있다. 본 연구에서는 OmpA signal sequence를 삽입하여 인간 과립구 성장인자(hG-CSF)가 분비발현되도록 고안된 T7 promoter 에 의하여 발현되는 pYRCl 발현백터를 제조하였다. E. coli BL2l (pYRCl) 발현시 $37^{\circ}C$에서 배양하는 경우 많은 양의 봉입체(aggregates)를 형성한다. 이에 비하여 $10\mu$M ucose를 포함하는 변형된 MBL배지에서 10 g/$\ell$IPTG를 유도물질로 7시간동안 $25^{\circ}C$에서 배양하였을 때 전체 periplasm단백질의 15%가 soluble rhG-CSF이었다. 또한, 유가식 배양방법을 사용하여 E. coli BL2l(pYRCl)에서 soluble rhG-CSF의 생산조건을 조사하였다. 유가식 배양에서 rhG-CSF의 발현량이 비증식속도를 $0.43 h^{-1}$ 에서 0.14 $h^{-1}$ 으로, 유도 배양시간을 최적화함으로써 rhG-CSF의 발현량이 4.4mg/$\ell$에서 24mg/$\ell$ 로 증가하였다.
목 적 : G-CSF와 GM-CSF는 과립구생성에 중요한 사이토카인으로서, 각각의 수용체 G-CSFr과 GM-CSFr에 결합하여 기능을 하게 되며, 이들 수용체들은 미성숙 골수 세포로부터 성숙 된 말초 과립구까지 발현된다. 일반적으로 혈중 G-CSF와 GM-CSF의 농도 및 이들 수용체의 발현은 과립구가 증가하는 감염질환에서 변화한다고 알려져 있으나, 백혈구 증가증에서 과립구수 증가와 관련된 변화에 대해서는 아직 연구된 바가 없다. 본 연구에서는 백혈구 증가증 환아와 정상아의 혈중 G-CSF와 GM-CSF의 농도를 측정하고 말초혈액의 과립구 표면에 존재하는 이들의 수용체를 정량분석하였으며, 활동성이 있는 수용체 발현 양에는 어떤 변화가 있는 지를 분석하여 백혈구 증가증 환아의 과립구 수 증가와 G-CSF와 GM-CSF 및 이들 수용체 변화의 관련성을 규명하고자 하였다. 방 법 : 정상 대조군으로서 비감염성 질환으로 입원한 같은 연령대의 아동 중 말초혈액 백혈구 수 및 중성구 수가 정상인 아동 13명과 급성기 초기의 백혈구 증가증 환아 14명, 총 27명의 혈중 G-CSF와 GM-CSF의 농도를 측정하였고 세포표면 G-CSFr와 GM-CSFr의 발현양은 각각 항 G-CSF 수용체 단 클론항체, 항 GM-CSF 수용체 단클론항체와 혼합 후 유세포 분석기를 이용하여 정량적으로 분석하였으며, 활동성 G-CSFr, GM-CSFr의 양적변화를 유세포 분석기를 이용하여 측정하였다. 결 과 : 백혈구 증가증 환아의 총 백혈구 수는 $24,100{\pm}6960/{\mu}L$로 정상 대조군 $8,680{\pm}378/{\mu}L$에 비해 유의하게 증가되었으며 중성구 수는 백혈구 증가증 환아에서 유의하게 증가되어 있었으나($20,800{\pm}2120/{\mu}L$ vs $3,360{\pm}2,120/{\mu}L$) 단구수는 차이가 없었다. 혈중 G-CSF의 농도 $164.0{\pm}187.5pg/mL$는 정상 대조군 $71.9{\pm}94.1pg/mL$과 감소된 경향을 보였으나 통계적으로 유의한 차이가 없었고(P=0.217) 혈중 GM-CSF의 농도도 유의한 차이가 없었다(P=0.968). 백혈구 증가증 환아의 중성구의 G-CSFr 발현양 $963.6{\pm}575.7$은 정상대조군 $1711.1{\pm}452.6$에 비해 유의한 감소를 보였으며(P=0.012), 백혈구 증가증 환아의 중성구의 GM-CSFr의 발현양 $471.7{\pm}217.0$은 정상 대조군의 $854.8{\pm}383.0$과 통계적으로 유의한 차이가 없었다(P=0.220). 항 G-CSFr항체는 수용체가 G-CSF와 결합하고 나면 수용체에 결합하지 못하는 epitope에 대한 항체로 보여지며, 수용체를 포화시키기에 충분한 과량의 CSF와 배양시킨 후 포화농도에도 결합하지 않은 수용체의 양을 측정하면 남은 수용체 즉, 활동성 수용체 수는 감소하게 되는데, 백혈구 증가증 환아에서 과량의 G-CSF에 배양한 후 감소된 중성구 G-CSFr의 발현양은 $407.8{\pm}405.1$로 정상 대조군의 $1,012.2{\pm}488.5$에 비해 유의한 감소를 보였고(P=0.050), 혈중 G-CSF 농도 및 혈중 GM-CSF 농도와 무관하였다(P=0.735, P=0.087). 백혈구 증가증 환아와 정상대조군에서 수용체를 포화시키기에 충분한 과량의 GM-CSF에 배양한 후 중성구의 GM-CSFr의 발현양을 분석한 결과는 발현이 증가된 소견을 보였다. 증가된 중성구의 GM-CSFr의 발현 양은 정상아 및 백혈구 증가증 환아에서 유의한 차이가 없었다(P=0.828). 결 론 : 백혈구 증가증에서는 중성구 수 증가에 의하여 총 백혈구 수가 증가되고 혈중 G-CSF의 농도는 중성구 증가의 원인으로 생각되며 G-CSFr과 결합하여 백혈구 증가증을 일으키는 것으로 보인다. GM-CSF 농도 및 GM-CSFr은 백혈구 증가에 영향을 주지 않음을 알 수 있었다.
빛은 식물에서 성장과 발달을 비롯한 다양한 생리화학적인 역할은 지닌다. 본 연구는 hGM-CSF 유전자가 도입된 형질전한 담배의 callus를 현탁배양하여 hGM-CSF를 생산할 때에 빛을 조사하는 시간에 따른 hGM-CSF의 생산에 미치는 영향을 확인하고자 실시하였다. 24시간 명배양, 18시간 명배양과 암배양을 실시하여 세포성장과 분비된 총단백질, hGM-CSF 생산량을 비교 관찰하였다. 세포의 성장은 24시간 명배양일 때 건조중량이 14.4 g/L로 가장 높았다. 분비된 총단백질의 양은 세가지 경우에서 큰 차이를 관찰할 수가 없었지만, 단위 세포당 분비된 총단백질의 양은 암배양이 다른 것에 비해 1.5배 가량 높았다. hGM-CSF의 생산은 18시간 명배양 조건이 가장 좋았으며 최대생산량이 495.5ug/L에 이르렀다. 또한 분비된 총단백질에서 hGM-CSF가 차지하는 비율은 18시간 명배양이 24시간 명배양에 비해 최대 1.8배 높았다.
Background: Granulocyte colony stimulating factor (G-CSF) is known as a cytokine central to the hematopoiesis of blood cells and to modulate their cellular functions. Besides granulocytes and their precursors, monocytes/macrophages and endothelial cells are direct target cells of G-CSF action. G-CSF influences immune cells in an anti inflammatory way. Methods: To evaluate whether G-CSF has a potential for preventing or ameliorating diseases characterized by mucosal inflammation, we used a mouse model with trinitrobenzene sulfonic acid (TNBS)-induced inflammatory colitis. To the mice model G-CSF was administrated daily by intraperitoneal injection. Macroscopic evaluation and immunohistochemical analysis of colonic tissues were performed. Results: Re combinant human G-CSF significantly inhibited LPS-induced TNF-${\alpha}$ mRNA expression in THP-1 cells. As for in vivo relevance, G-CSF dramatically reduced the weight loss of mice, colonic damage, and mucosal ulceration that characterize TNBS colitis. Moreover, G-CSF suppressed the expression of tumor necrosis factor-${\alpha}$, interleukin-$1{\beta}$, and intercellular adhesion molecule-1 in TNBS colitis. Conclusion: Current results demonstrate that G-CSF may be an effective agent for the treatment of diseases characterized by mucosal inflammation.

Macrophage colony-stimulating factor (M-CSF) is a growth factor required for growth and differentiation of mononuclear phagocyte lineage. Total and 16 poly (A) mRNA of bovine M-CSF were isolated from healthy bovine peripheral mononuclear cells stimulated by phobol 12-myristste 13-acetate (TPA). The more compatible cultured mononuclear cells were 5${\times}$10/ml for RNA isolation. TPA-activated mononuclear cells increased the level of M-CSF-mRNA more than concanavalin A (Con A) and lipopolysaccharide (LPS). The optimal analysis of reverse transcriptase-polymerase chain reaction (RT-PCR) for14 Macrophage colonystimulating factor (M-CSF) as a growth factor required for bovine M-CSF was denaturation at 94$^{\circ}C$ for 1 minute, annealing at 57$^{\circ}C$ for 1 minute, extension at 72$^{\circ}C$ for 1 minute for 30 cycles. The size of cDNA of bovine M-CSF by RT-PCR was 774 base pairs. A 774 base pairs cDNA encoding bovine M-CSF was synthesized by reverse transcriptase polymerase chain reaction (RT-PCR). Ligated cDNA was transformed to competent cells and then plasmid isolation and digestion was performed. Molecular cloning and sequencing were performed for cDNA of bovine M-CSF. The size of cloned cDNA of bovine M-CSF was 774base pairs. The homology of base sequence and amino acid sequence was 88% and 86% compared with known human M-CSF, respectively. From a high degree of sequence similarity, the obtained cDNA of bovine M-CSF is thought be a specific gene of bovine M-CSF.
INI1/hSNF5/BAF47 is a core component of the hSWI/ SNF ATP-dependent chromatin remodeling complex, and it has been implicated in regulating gene expression, cell division and tumorigenesis. We investigated whether INI1/hSNF5/BAF47 functions in activation of the colony stimulating factor 1 (CSF1) promoter in HeLa cells. Overexpression of INI1/hSNF5/BAF47 promoted CSF1 transcription, and siRNA targeting INI1/hSNF5/ BAF47 (siINI1) strongly inhibited the activity of the CSF1 promoter. We demonstrated that all conserved domains of INI1/hSNF5/BAF47 are needed for CSF1 transcription. ChIP experiment showed that INI1/ hSNF5/BAF47 is recruited to the region of the CSF1 promoter. Taken together, these results indicate that INI1/hSNF5/BAF47 is involved in activation of the CSF1 promoter.
The purpose of this study is to assess and develop effective MIS planning method by applying the CSF method. With the comparative analysis of the Rockart's basic concept of CSF and the examples of the corporations which employed CSF methodology, a new CSF method more suitable to Korean business environment is proposed and applied to another company for evaluation. The new CSF methodology consists of three steps. The first step is to preexamine the business conditions. The second step contains an Introductory Workshop, interview with the managers, survey questionnaire, Focusing Workshop, and decision of the priorities for system development. The third step is a real development of an information system out of the first and second steps. As a result of this study, it is found that revised CSF method can induce top management to participate more in the activities of information system development and that it can make top management to focus on the critical area. The fulfillment of the information requirement out of the enhanced MIS planning will lead to more successful management activities.

Background: Ultraviolet (UV) goes through the epidermis and promotes release of inflammatory cytokines in keratinocytes. Granulocyte-macrophage colony-stimulating factor (GM-CSF), one of the keratinocyte-derived cytokines, regulates proliferation and differentiation of melanocytes. Extracellular signal-regulated kinase (ERK1/2) and protein kinase C (PKC) signaling pathways regulate expression of GM-CSF. Based on these results, we found that ginsenoside Rh3 prevented GM-CSF production and release in UV-B-exposed SP-1 keratinocytes and that this inhibitory effect resulted from the reduction of PKCδ and ERK phosphorylation. Methods: We investigated the mechanism by which ginsenoside Rh3 from Panax ginseng inhibited GM-CSF release from UV-B-irradiated keratinocytes. Results: Treatment with 12-O-tetradecanoylphorbol-13-acetate (TPA) or UV-B induced release of GM-CSF in the SP-1 keratinocytes. To elucidate whether the change in GM-CSF expression could be related to PKC signaling, the cells were pretreated with H7, an inhibitor of PKC, and irradiated with UV-B. GM-CSF was decreased by H7 in a dose-dependent manner. When we analyzed which ginsenosides repressed GM-CSF expression among 15 ginsenosides, ginsenoside Rh3 showed the largest decline to 40% of GM-CSF expression in enzyme-linked immunosorbent assay. Western blot analysis showed that TPA enhanced the phosphorylation of PKCδ and ERK in the keratinocytes. When we examined the effect of ginsenoside Rh3, we identified that ginsenoside Rh3 inhibited the TPA-induced phosphorylation levels of PKCδ and ERK. Conclusion: In summary, we found that ginsenoside Rh3 impeded UV-B-induced GM-CSF production through repression of PKCδ and ERK phosphorylation in SP-1 keratinocytes.
Members of the colony stimulating factor cytokine family play important roles in macrophage activation and recruitment to inflammatory lesions. Among them, granulocyte-macrophage colony stimulating factor (GM-CSF) is known to be associated with immune response to mycobacterial infection. However, the mechanism through which Mycobacterium tuberculosis (MTB) affects the expression of GM-CSF is poorly understood. Using PMA-differentiated THP-1 cells, we found that MTB infection increased GM-CSF mRNA expression in a dose-dependent manner. Induction of GM-CSF mRNA expression peaked 6 h after infection, declining gradually thereafter and returning to its basal levels at 72 h. Secretion of GM-CSF protein was also elevated by MTB infection. The increase in mRNA expression and protein secretion of GM-CSF caused by MTB was inhibited in cells treated with inhibitors of p38 MAPK, mitogen-activated protein kinase kinase (MEK-1), and PI3-K. These results suggest that up-regulation of GM-CSF by MTB is mediated via the PI3-K/MEK1/p38 MAPK-associated signaling pathway.
