Selection of Cryoprotectants for Cryopreservation of Pearl Oyster, Pinctada fucata martensii Trochophore (진주조개, Pinctada fucata martensii 담륜자의 냉동보존을 위한 동해방지제의 선택)

  • To find out the desirable cryoprotectant for cryopreservation of bivalve trochophores, four types of cryoprotectant were tested with trochophores of the pearl oyster (pinctada fucata martensii) generally used in the pearl production in Korea. Each cryoprotectant (dimethyl sulfoxide, ethylene glycol, glycerol and 1,2-propanediol) was mixed with 0.2 M sucrose to make final concentrations of 0.01, 0.1, 1.0 and 2.0 M. The trochophores were immersed in each preparation, waiting for 10 minutes to reach equilibration and cryopreserved in the liquid nitrogen (-l96$^{\circ}C$). Survival rate of trochophores thawed after cryopreservation increased as the media concentration increase. However, a few number of the trochophores seemed to be damaged with the efflux of cell inclusions. Our study rsults indicate that desirable cryoprotectants for cryopreservation of pearl oyster trochophores are 1.0~2.0 M dimethyl sulfoxide and ethylene glycol : 82.8~97.4% of the trochophores cryopreserved with these media survived after thawing.

  • An preliminary study was made with special references on the insect fauna of Mt. Gyeryong during the period from mid-May to mid-September 1981. The results were obtained as follows; 1. 673 species of insects within 156 families of 20 orders were identified and listed herewith. 2. The 16 species are first records from Korea : Phaneraptera nigro-antennata (Tettigoniidae : Orthop.), Phraraortes kumamotoensis(Phasmidae : Phasm.), Nesogaster lewisi (Nesogasteridae : Dermap), Polymerus palustris(Miridae : Hemip.), Agrosteomela indica (Chrysomelidae:Col.), Monochimus sparsutus (Cermbicidae : Col.), Oberthiiria ialcigera (Bombycidae: Lep.), Alcis albiiera (Geometridae : Lep.), Proplepsis diazama (G eometridae : Lep.) Pandemis cinnamomeana (Tortricidae : Lep.), Hypenodes squalida (Noctuidae : Lep.), Hypolimnas bolina (Nymphalidae : Lep.), Ctenophora nohirae (Tipulidae: Dip.), Ortalotrypeta isshiki (Trypetidaedae : Dip.), Trypeta artemisicola (Trypetidae : Dip.), Ichneumom 8-guttatus ( Ichneumonidae : Hymenop.). 3. We could find a considerable number of Oriental fauna species, Lepismachilis nipponica, Nezara antennata, Parapolybia varia, Anthophora zonata, Parnara guttata, Byasa alcinous, Eurema laeta, Pieris canidia, Eizera maha, Kaniska canace, Danaus sita including two newly recorded species, Monochimus sparstus and Hypolimnas bolina.

  • This paper deals with the development of Laser Doppler Vibrometer (LDV) that can mea~ure the tlequency and amphlude of the ultrasonic vibralion. Hc-Ne laser (632.8 om) is used as a light source, and Michelson interferometer in winch frequency of the objective beam is shIfted by Bragg cell IS adopted The frequency modulated signal centered at 40 MHz flom the PIN diode IS amplified. down-col1vel1ed to 2.5 MHz, filtered and digiLized. The voltage output that is proportional to the velocity of the vibratwg surface is obtawed using digItal PLL. A microprocessor is used to extract the frequcncy aud amplitude of the vibratIOn from the voltage output. It is found that the developed LDV can measure up to 300 kHz vibratIOn and the mlillmUITI measurable amplitude is I nm at 300 kHz. We believe thatlhis LDV can be used to measure the vibratIOn of the heavy electric maclllnery and micro-size structures. tures.

Induction of Effective Osteogenesis by Mesenchymal Stem Cells from the Human Subchondral Bone (사람 연골하골 중간엽 줄기세포의 효율적인 골형성 유도)

  • Background : Mesenchymal stem cells (MSCs) are present in most of the tissue matrix, taking part in their regeneration when injury or damage occurs. The aim of this study was to investigate the presence of cells with pluripotential characteristics in human subchondral bone and the capacity of these cells to differentiate to osteoblast. Methods : Human subchondral bone were digested with collagenase. Isolated cells were cultured with a-MEM, 15% FBS, 10-8M dexamethasone and 50 ng/mL ascoric acid. Cells from 0 day(isolated cells), 7 day (first subculture) and 14 days (third subculture) were used to carry out phenotypic characterization experiments flowcytometry analysis with 11 monoclonal antibodies) and osteogenic differentiation experiments. Osteogenic differentiation of cells was assessment by quantification of bone extracellular matrix components by following analysis: alkaline phosphatase(ALP) stains to detect ALP activity, RT-PCR and western blot to detect osteocalcin (OCN), osteopontin (OPN) and type I collagen(Col I), and Alizarin red stains to detect calcium deposition. Results : Flowcytometry analyses showed that in our population more than 98% of cells were positive for MSC markers: SH-2(CD105, 99%), CD29 (95%), CD73 (95%). Cells were negative for hematopoietic markers (CD11b, CD34, and CD45). Furthermore, cells showed positive stain to multipotent markers such as CDl17 (c-kit) (15.1%), and CD166 (74.9%), and cell adhesion molecules such as CD54 (78.1%) and CD106 (63.5%). The osteogenic specific marker analyses showed that the culture of these cells for 7 and 14 days stimulates ALP, OCN, OPN and Col I synthesis by RT-PCR and Western blot analysis. Also, after 14 days in the culture of MSCs induces mineralization by Arizarin red stain. Conclusion : In this work, we demonstrated a new and efficient method for osteoblastic differentiation of human subchondral bone stem cells. As MSCs takes part in reparative processes of adult tissues, these cells could play an important role in osteogenesis.

Identification of Genomic Aberrations by Array Comparative Genomic Hybridization in Patients with Aortic Dissections

  • Background: The aim of the present study was to identify chromosomal loci that contribute to the pathogenesis of aortic dissection (AD) in a Korean population using array comparative genomic hybridization (CGH) and to confirm the results using real-time polymerase chain reaction (PCR). Materials and Methods: Eighteen patients with ADs were enrolled in this study. Genomic DNA was extracted from individual blood samples, and array CGH analyses were performed. Four corresponding genes with obvious genomic changes were analyzed using real-time PCR in order to assess the level of genomic imbalance identified by array CGH. Results: Genomic gains were most frequently detected at 8q24.3 (56%), followed by regions 7q35, 11q12.2, and 15q25.2 (50%). Genomic losses were most frequently observed at 4q35.2 (56%). Real-time PCR confirmed the results of the array CGH studies of the COL6A2, DGCR14, PCSK6, and SDHA genes. Conclusion: This is the first study to identify candidate regions by array CGH in patients with ADs. The identification of genes that may predispose an individual to AD may lead to a better understanding of the mechanism of AD formation. Further multicenter studies comparing cohorts of patients of different ethnicities are warranted.

Inhibitory Effects of Boesenbergia pandurata on Age-Related Periodontal Inflammation and Alveolar Bone Loss in Fischer 344 Rats

  • Periodontitis, an infective disease caused by oral pathogens and the intrinsic aging process, results in the destruction of periodontal tissues and the loss of alveolar bone. This study investigated whether Boesenbergia pandurata extract (BPE) standardized with panduratin A exerted anti-periodontitis effects, using an aging model representative of naturally occurring periodontitis. In aged rats, the oral administration of BPE ($200mg{\cdot}kg^{-1}{\cdot}day^{-1}$) for 8 weeks significantly reduced the mRNA and protein expression of $interleukin-1{\beta}$, nuclear factor-kappa B, matrix metalloproteinase (MMP)-2, and MMP-8 in gingival tissues (p < 0.01). In alveolar bone, histological analysis with staining and micro-computed tomography revealed the attenuation of alveolar bone resorption in the BPE-treated aged group, which led to a significant reduction in the mRNA and protein expression of nuclear factor of activated T-cells c1 (NFATc1), c-Fos, tartrate-resistant acid phosphatase, and cathepsin K (p < 0.01). BPE not only increased the expression of osteoblast differentiation markers, such as alkaline phosphate, and collagen type I (COL1A1), but also increased the ratio of osteoprotegerin to RANKL. Collectively, the results strongly suggested that BPE is a natural resource for the prevention or treatment of periodontal diseases.

Synthesis of PSZ-seeding Mullite Composite from Metal Alkoxides and Its Characteristics of Sintered Body (금속 알콕사이드로부터 PSZ-seeding Mullite 복합체의 합성 및 소결체의 특성)

  • Mullite-PSZ composite was prepared by sol-gel method using $Al(sec-OC_4H_9)_3,\;Si(OC_2H_5)_4,\;ZrOCl_2\;8H_2O\;and\;Y_2O_3$. The sinterability ana mechanical properties of powder compacts sintered at $1,650^{\circ}C$ for 4 hrs were investigated for various PSZ contents. In result Al-Si spinel formed at $980^{\circ}C$ from amorphous dried gel, and zirconia as well as mullite crystal formed above $1,200^{\circ}C$. The sintered body was densified to $97{\sim}98%$ except the specimen containing 25vol% PSZ which showed the relative density of about 95% obtained by sintering at $1,650^{\circ}C$ for 4 h. The flexural strength of the sintered body was a maximum value of 290 MPa in 20 vol% PSZ, which was also considerably larger than the value of 200 MPa without PSZ. The value of the fracture toughness increased linearly with increase of PSZ content and showed a maximum value of $4.3MPam^{1/2}$ in 25 vol% PSZ, Namely this value was remarkably larger than the $value(2.6MPam^{1/2})$ of pure mullite without PSZ.

  • Colmap is one of the innovative artificial intelligence technologies, highly effective as a tool in 3D reconstruction tasks. Moreover, it excels at constructing intricate 3D models by utilizing images and corresponding metadata. Colmap generates 3D models by merging 2D images, camera position data, depth information, and so on. Through this, it achieves detailed and precise 3D reconstructions, inclusive of objects from the real world. Additionally, Colmap provides rapid processing by leveraging GPUs, allowing for efficient operation even within large data sets. In this paper, we have presented a method of collecting 2D images of traditional Korean towers and reconstructing them into 3D models using Colmap. This study applied this technology in the restoration process of traditional stone towers in South Korea. As a result, we confirmed the potential applicability of Colmap in the field of cultural heritage restoration.

The Effects of Some Halitosis Removal Methods on the Reduction of Intraoral Volatile Methyl Mercaptan Concentrations (수종 구취제거법이 구강내 휘발성 메틸머캅탄 감소에 미치는 영향)

  • In order to evaluate the effectiveness of tooth brushing, mouth gargling and gum chewing in reducing halitosis, 84 individuals ranging in age from 22 59 28 years old were examined. These individuals had no gross oral abnormalities, other than mild gingival inflammation, dental caries, nasopharyngeal disorder, or systemic diseases that were associated with halitosis. They were divided into a tooth brushing group, a mouth garging group, a gum chewing group and a control group that did not use any halitosis removing method. Each of the groups included 21 persons, B.B. Checker (Tokuyama Soda Col, LTDl, Japan) was used to measure the concentrations of intraoral volatile methyl mercaptan of each group. The concentrations of intraoral volatile methyl mercaptan were measured before and after lunch, and after removing halitosis by toothe brushing, mouth gargling and gum chewing. The obtained results were as follows : 1. The average concentration of intraoral volatile methyl mercaptan before lunch was 1.79ppm and after lunch it was 2.02ppm, an increase of 12.9%. 2. In the tooth brushing group the average concentration of intraoral volatile methyl mercaptan was 0.61ppm, in the mouth gargling group it was 1.15ppm, in the gum chewing group it was 1.64ppm and in the control group it was 1.92ppm. It decreased 69.5% in the tooth brushing group, 43.8% in the mouth gargling group, 18.4% in the gum chewing group and 5.4% in the control grop (p<0.05). 3. There were significant differences between the tooth brushing and control group, tooth brushing and gum chewing group and between mouth gargling and control group in concentrations of intraoral volatile methyl mercaptan after using the halitosis removing methods (p<0.05). According to the above results, tooth brushig and mouth gargling are effective ways to reduce halitosis.

The Role of Autonomous Wntless in Odontoblastic Differentiation of Mouse Dental Pulp Cells

  • Purpose: Wnt signaling plays an essential role in the dental epithelium and mesenchyme during tooth morphogenesis. Deletion of the Wntless (Wls) gene in odontoblasts appears to reduce canonical Wnt activity, leading to inhibition of odontoblast maturation. However, it remains unclear if autonomous Wnt ligands are necessary for differentiation of dental pulp cells into odontoblast-like cells to induce reparative dentinogenesis, one of well-known feature of pulp repair to form tertiary dentin. Materials and Methods: To analyze the autonomous role of Wls for differentiation of dental pulp cells into odontoblast-like cells, we used primary dental pulp cells from unerupted molars of Wls-floxed allele mouse after infection with adenovirus for Cre recombinase expression to knockout the floxed Wls gene or control GFP expression. The differentiation of dental pulp cells into odontoblast-like cells was analyzed by quantitative real-time polymerase chain reaction. Result: Proliferation rate was significantly decreased in dental pulp cells with Cre expression for Wls knockout. The expression levels of Osterix (Osx), runt-related transcription factor 2 (Runx2), and nuclear factor I-C (Nfic) were all significantly decreased by 0.3-fold, 0.2-fold, and 0.3-fold respectively in dental pulp cells with Wls knockout. In addition, the expression levels of Bsp, Col1a1, Opn, and Alpl were significantly decreased by 0.7-fold, 0.3-fold, 0.8-fold, and 0.6-fold respectively in dental pulp cells with Wls knockout. Conclusion: Wnt ligands produced autonomously are necessary for proper proliferation and odontoblastic differentiation of mouse dental pulp cells toward further tertiary dentinogenesis.
