Background: A previous study reported that hyperlipidemia increases the incidence of tears in the rotator cuff tendon and affects healing after repair. The aim of our study was to compare the gene and protein expression of torn rotator cuff tendons in patients both with and without hypercholesterolemia. Methods: Thirty patients who provided rotator cuff tendon samples were classified into either a non-hypercholesterolemia group (n=19, serum total cholesterol [TC] <200 mg/dL) and hypercholesterolemia group (n=11, serum TC ≥240 mg/dL) based on their concentrations of serum TC. The expression of various genes of interest, including COL1A1, IGF1, IL-6, MMP2, MMP3, MMP9, MMP13, TNMD, and TP53, was analyzed by real-time quantitative reverse transcription polymerase chain reaction (qRT-PCR). In addition, Western blot analysis was performed on the proteins encoded by interleukin (IL)-6 and TP53 that showed significantly different expression levels in real-time qRT-PCR. Results: Except for IGF1, the gene expression levels of IL-6, MMP2, MMP9, and TP53 were significantly higher in the hypercholesterolemic group than in the non-hypercholesterolemia group. Western blot analysis confirmed significantly higher protein levels of IL-6 and TP53 in the hypercholesterolemic group (p<0.05). Conclusions: We observed an increase in inflammatory cytokine and matrix metalloproteinase (MMP) levels in hypercholesterolemic patients with rotator cuff tears. Increased levels of IL-6 and TP53 were observed at both the mRNA and protein levels. We suggest that the overexpression of IL-6 and TP53 may be a specific feature in rotator cuff disease patients with hypercholesterolemia.
This study examined whether the oral administration of low-molecular-weight collagen peptide (LMCP) containing 3% Gly-Pro-Hyp with >15% tripeptide (Gly-X-Y) content could ameliorate osteoarthritis (OA) progression using a rabbit anterior cruciate ligament transection (ACLT) model of induced OA and chondrocytes isolated from a patient with OA. Oral LMCP administration (100 or 200 mg/kg/day) for 12 weeks ameliorated cartilage damage and reduced the loss of proteoglycan compared to the findings in the ACLT control group, resulting in dose-dependent (p < 0.05) improvements of the OARSI score in hematoxylin & eosin (H&E) and Safranin O staining. In micro-computed tomography analysis, LMCP also significantly (p < 0.05) suppressed the deterioration of the microstructure in tibial subchondral bone during OA progression. The elevation of IL-1β and IL-6 concentrations in synovial fluid following OA induction was dose-dependently (p < 0.05) reduced by LMCP treatment. Furthermore, immunohistochemistry illustrated that LMCP significantly (p < 0.05) upregulated type II collagen and downregulated matrix metalloproteinase-13 in cartilage tissue. Consistent with the in vivo results, LMCP significantly (p < 0.05) increased the mRNA expression of COL2A1 and ACAN in chondrocytes isolated from a patient with OA regardless of the conditions for IL-1β induction. These findings suggest that LMCP has potential as a therapeutic treatment for OA that stimulates cartilage regeneration.
우리나라에서 진주양식의 대상패류인 진주조개 (Pinctada fucata martensii) 담륜자를 냉동보존하기 위하여 4가지 동해방지제를 사용하여 냉동실험을 실시하고, 적합한 동해방지제를 찾고자 하였다. 동해방지제인 dimethyl sulfoxide (DMSO), ethylene glycol, glycerol과 1, 2-propanediol을 희석액인 0.2 M sucrose에 각각 0.01, 0.1, 1.0과 2.0 M이 되도록 혼합한 다음, 10분간 평형상태를 유지한 후 냉동하였다. 냉동후 해동시킨 담륜자는 모든 동해방지제에서 농도가 증가할수록 생존율이 높아졌으나, 일부 세포내 함유물이 세포외로 유출되어 손상을 입은 개체도 보였다. 진주조개의 담륜자 냉동보존에 적합한 동해방지제는 1.0∼2.0 M의 DMSO와 ethylene gly-col로서 해동후 생존율이 82.8∼97.4%로 가장 높았다.
연구목적: 이 연구에서는 mineral trioxide aggregate 제재인 white ProRoot MTA (wMTA)와 수산화칼슘 제재인 Dycal을 인간치수세포에 적용한 후 치수세포의 분화와 증식, 석회화, 신생혈관형성(angiogenesis) 그리고 염증에 관여하는 유전자들의 발현 변화를 비교하였다. 연구 재료 및 방법: 실험군은 wMTA와 Dycal을 테플론 튜브(내경 10 mm, 길이 1 mm)에 담아 4시간 경화시킨 후 일차세포배양한 인간치수세포에 적용하였고, 대조군은 빈 튜브만을 적용하였다. 3시간, 6시간, 9시간, 24시간 후 total RNA를 추출하고 oligonucleotide microarray 방법을 통하여 유전자 발현 양상을 분석하였다. 위의 결과를 역전사 중합효소 연쇄반응(reverse transcriptase polymerase chain reaction)으로 재확인하였다. 결과: wMTA를 적용한 실험군에서 24,546개의 유전자 중 43개 유전자의 발현이 2배 이상 증가하였으며(예. BMP2, FOSB, THBS1, EDN1, IL11, COL10A1, TUFT1, HMOX1) 25개 유전자의 발현이 50% 이하로 감소하였다(예. SMAD6, TIMP2, DCN, SOCS2, CEBPD, KIAA1199). Dycal을 적용한 실험군에서 239개 유전자의 발현이 2배 이상 증가하였으며(예. BMP2, BMP6, SMAD6, IL11, FOS, VEGFA, PlGF, HMOX1, SOCS2, CEBPD, KIAA1199) 358개 유전자의 발현이 50% 이하로 감소하였다(예. EDN1, FGF). 결론: wMTA를 적용한 치수세포에서는 분화와 증식 그리고 석회화에 관여하는 유전자들의 변화가 관찰되었다. Dycal을 적용한 치수세포에서는 분화와 증식 그리고 신생혈관형성에 관여하는 유전자들의 변화가 관찰되었다. 또 Dycal이 염증에 관여하는 유전자들을 더 많이 발현시키는 양상을 보였다.
Bacillus thuringiensis subsp. kurstaki HD-1으로부터 생산된 살충성 내독소 단백질을 coding하는 CryIIA 유전자를 클로닝하고 염기서열을 조사하였다. HD-1 균주의 12개 plasmid 중 225kb plasmid를 분리하여 CryIIA 유전자를 포함하는 5kb HindIII 절편을 hybridization하여 찾아냈다. 이 절편을 plasmid pUC19에 ligation하여 E. coli에 형질 전환하였다. 이 독소 유전자를 포함하는 4kb BamHI-HindIII 절편은 vector pT7-5에 ligation하여 pSKIIA라하였다. pSKIIA는 3개의 open reading frames(orf1, orf2, orf3)로 구성되어 있으며 염기서열은 3,952base로 되어 있었다. 이러한 3개의 orf 각각의 발현 여부를 확인하기 위하여 생물검정을 하였다. 그러나 orf1 또는 orf2에 의한 형질 전환체는 독성이 없는 것으로 나타났다. orf3를 포함하는 형질 전환체는 3종의 나비목 곤충(배추점나방, 담배거세미나방, 담배나방) 및 1종의 파리목 곤충(집모기) 유충에 대하여 독성을 나타내었다.

Resveratrol (3,4',5,-trihydroxystilbene), a phytoalexin present in grapes, exerts a variety of actions to reduce superoxides, prevents diabetes mellitus, and inhibits inflammation. Resveratrol acts as a chemo-preventive agent and induces apoptotic cell death in various cancer cells. However, the role of resveratrol in odontoblastic cell differentiation is unclear. In this study, the effect of resveratrol on regulating odontoblast differentiation was examined in MDPC-23 mouse odontoblastic cells derived from mouse dental papilla cells. Resveratrol significantly accelerated mineralization as compared with the control culture in differentiation of MDPC-23 cells. Resveratrol significantly increased expression of ALP mRNA as compared with the control in differentiation of MDPC-23 cells. Resveratrol significantly accelerated expression of Col I mRNA as compared with the control in differentiation of MDPC-23 cells. Resveratrol significantly increased expressions of DSPP and DMP-1 mRNAs as compared with the control in differentiation of MDPC-23 cells. Treatment of resveratrol did not significantly affect cell proliferation in MDPC-23 cells. Results suggest resveratrol facilitates odontoblast differentiation and mineralization in differentiation of MDPC-23 cells, and may have potential properties for development and clinical application of dentin regeneration materials.

Trace mineral studies involving metal ion chelators have been conducted in investigating the response of gene and protein expressions of certain cell lines but a few had really focused on how these metal ion chelators could affect the availability of important trace minerals such as Zn, Mn, Fe and Cu. The aim of the present study was to investigate the availability of Zn for the treatment of MC3T3-E1 osteoblast-like cells and the availability of some trace minerals in the cell culture media components after using chelexing resin in the FBS and the addition of N,N,N',N'-tetrakis-(2-pyridylmethyl)ethylenediamine (TPEN, membrane-permeable chelator) and diethylenetriaminepentaacetic acid (DTPA, membrane-impermeable chelator) in the treatment medium. Components for the preparation of cell culture medium and Zn-treated medium have been tested for Zn, Mn, Fe and Cu contents by atomic absorption spectrophotometer or inductively coupled plasma spectrophotometer. Also, the expression of bone-related genes (ALP, Runx2, PTH-R, ProCOL I, OPN and OC) was measured on the cellular Zn depletion such as chelexing or TPEN treatment. Results have shown that using the chelexing resin in FBS would significantly decrease the available Zn (p<0.05) $(39.4{\pm}1.5{\mu}M\;vs\;0.61{\pm}10.15{\mu}M)$ and Mn (p<0.05) $(0.74{\pm}0.01{\mu}M\;vs\;0.12{\pm}0.04{\mu}M)$. However, levels of Fe and Cu in FBS were not changed by chelexing FBS. The use of TPEN and DTPA as Zn-chelators did not show significant difference on the final concentration of Zn in the treatment medium (0, 3, 6, 9, $12{\mu}M$) except for in the addition of higher $15{\mu}M\;ZnCl_2$ which showed a significant increase of Zn level in DTPA-chelated treatment medium. Results have shown that both chelators gave the same pattern for the expression of the five bone-related genes between Zn and Zn+, and TPEN-treated experiments, compared to chelex-treated experiment, showed lower bone-related gene expression, which may imply that TPEN would be a stronger chelator than chelex resin. This study showed that TPEN would be a stronger chelator compared to DTPA or chelex resin and TPEN and chelex resin exerted cellular zinc depletion to be enough for cell study for Zn depletion.
This study was conducted to investigate the physical characteristics and risk factors for hyperchol-esterolemia (HC) in Korean. 344 adult men who took the annual health check-ups at D or J hospitals were participated in this cross-sectional study. The subjects were grouped by plasma total cholesterol level in to three groups: normal cholesterolemic (n=139) borderline hypercholesterolemic(n=93) and hypercholesterolemic (n=112) groups. The data of height weight and plasma cholesterol level were col-lected from medical records. Body circumferences(midarm, waist, hip, and thight) skinfold thicknesses (biceps, triceps, subcostal, abdomen, and suprailic), and body composition (fat mass and fat free mass) were measured. Body mass index (BMI) height/weight ratio (HWR) waist/hip circumference ratio (WHR) waist/ thigh circumference ratio (WTR) central skinfold thickenss (CSF) and peripheral skin-fold thickness were calculated. The subjects with HC had significantly higher weight BMI waist cir-cumference skinfold thickness and body fat mass than those of the normal subjects. The relative and attributable risks on HC were 1.61 and 0.17 for obesity (BMI$\geq$25) 1,30 and 0.11 for upper body obesity (WTR$\geq$1.30) and 1.54 and 0.18 for central body obesity (CSF$\geq$95.7). Plasma total cholesterol level was positively correlated with several antropometric parameters: BMI (p<0.001) weight(p<0.001) waist circumference(p<0.001) and skinfold thickness of abdomen (p<0.001) spraillic (p<0.01) triceps(p<0.01) subcostal (p<0.01) and biceps (p<0.05) In conclusion the major influencing factors to plasma cholesterol level was BMI. Among the each physical parameters the circumference of waist the skinfol-d thickness of abdomen and the percentage of body fat were closely related to plasma cholesterol level. The important risk factor for hypercholesterolemia was obesity specially upper body obesity and central body obesity.
ACP (annealing control primer)를 사용하여 DDRT (differential display reverse transcription)-PCR 방법으로 볼락의 성장단계에 따라 발현량 차이를 나타내는 DEG (differentially expressed gene)를 확보하였다. ACP 120개를 분석하여 18개월령 근육조직에서보다 6개월령 근육조직에서 발현량이 많은 DEG 16개와 6개월령 근육조직에서보다 18개월령 근육조직에서 발현량이 더 많은 DEG22개의 염기서열을 분석하였다. DEG 염기서열을 BLAST 검색한 결과, parvalbumin (PVALB) 등 18개의 유전자(PVALB, NDKB, TPM, TnI, GAPDH, CKM2, factor 2 SERF2, AMPD, TRICA, ARHGAP15, ESD, hsp70, COL1A2, GST, Midllip1, MYL1, SERCA1B, FTH1)와 69~95%의 상동성을 나타냈다. Real time PCR 분석법으로 6개월령 근육조직에서 발현량이 많은 DEG14와 PVALB 유전자의 성장단계별 발현양상을 조사한 결과, 볼락이 성장함에 따라 발현량이 감소하였으며, 특히 PVALB 유전자는 6개월령 이후에는 발현량이 극히 적었다. 6개월령 근육조직에서보다 18 개월령 근육조직에서 발현량에서 많았던 CKM2 유전자는 성장함에 따라 발현량이 계속 증가하였고, 4세 이후에는 발현량이 감소하였다. DEG의 조직특이적 발현양상을 분석한 결과, DEG14는 근육, 간, 신장, 및 비장조직에서 발현되었으며, PVALB 유전자는 근육과 신장조직에서 발현되었고, 간과 비장조직에서는 발현되지 않았다. CKM2 유전자는 근육, 신장 및 비장조직에서 발현되었고, 간 조직에서는 발현되지 않았다. PVALB 유전자의 mRNA 크기는 659 bp 이며, 110개의 아미노산으로 구성되어 있다. Parvalbumin과 CKM2 유전자는 성장속도가 빠른 어류 선발에 이용할 수 있는 분자마커 개발에 활용하고자한다.
식물생육을 촉진하고 고추역병균을 방제하는 다기능 PGPR 균주 Bacillus subtilis AH18 항진균성 cellulase 유전자를 PCR을 이용해 pUC18과 재조합 후 E. coli DH5$\alpha$에 cloning하여 E. coli내에 발현시켰으며, 그 형질전환 균주를 E. coli DH5$\alpha$(pCM 41)이라 명명하였고, 발현된 cellulase를 ce/H라 하였다. E. coli DH5$\alpha$(pCM 41)의 inset 부위는 B. subtilis AH18의 1,582 bp 유전자를 포함하며 cellulase의 유전자는 1,524 bp로 508개의 amino acid가 암호화된 것으로 추정되었고, CMC를 함유한 SDS-PAGE의 방법으로 약 55 kDa의 분자량을 확인하였다. B. subtilis AH18이 가지는 ce/H는 3종의 대표적인 Bacillus spp.들의 cellulase 유전자의 DNA와 아미노산 배열이 98% 이상 유사하였으며, CMC(carboxymethyl-cellulose) 뿐만 아니라, 불용성 섬유소인 Avicel, filter paper(Whatman No. 1) 특히 고추역병균인 Phytophthora capsici의 건조 cell wall도 분해하였다. 또한 colH의 cellulase는 $50^{\circ}C$에서 효소활성이 가장 높았으며, 최적 pH는 pH 6.0이었다. 그리고 $AgNO_3$ 또는 $CoCl_2$ 첨가시 활성이 1.7배, 2배 정도 증가하였고 $HgC1_2$ 첨가시는 활성이 20%까지 떨어졌다. 또한 여러 화학 저해제들 중 Sodium azide 또는 Hydroxy urea는 효소 활성을 증가시켰으며, CDTA 또는 EDIA는 섬유소분해능을 감소시켰다. 이들의 결과는 고추역병균 P. capsici의 생육을 억제하는 B. subtilis AM18의 진균세포벽 용해성 cellula의 효소학적 특성을 구명한 것이라고 할 수 있다.

제 15 조 (계약 해지)
회원이 이용계약을 해지하고자 하는 때에는 [가입해지] 메뉴를 이용해 직접 해지해야 합니다.
제 16 조 (서비스 이용제한)
① 당 사이트는 회원이 서비스 이용내용에 있어서 본 약관 제 11조 내용을 위반하거나, 다음 각 호에 해당하는
경우 서비스 이용을 제한할 수 있습니다.
- 2년 이상 서비스를 이용한 적이 없는 경우
- 기타 정상적인 서비스 운영에 방해가 될 경우
② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
