천연항생제로서 봉독의 기원 및 지표성분 설정 (Origin and effective ingredient standards of honeybee venom as natural antibiotic ingredients)

  • This research was performed in order to investigate the origin, standard compound, and structural and physical properties of honeybee venom which used as natural antibiotic ingredients to animal. We compared the nucleotide sequence of mitochondrial cytochrome c oxidase subunit 1 gene (COI) of honeybees were collected from Gangwon, Gyeonggi, Chungnam, Gyeongbuk, Gyeongnam province and Suwon. As major constituent of honeybee venom, melittin was assayed by liquid chromatography. X-ray, differential scanning calorimetry (DSC) and fourier transform infrared spectroscopy (FT-IR) were utilized to examine the structural and physical properties of honeybee venom. Based on the 627bp sequence of COI, Apis mellifera ligustica was determinated honeybees collected from all six regions. Melittin content varied from 50.7 to 68.6 and averaged 59.8%. According to XRD analysis, honeybee venom showed regular crystal structure peaks at $2{\Theta}=8.5^{\circ}$ and $21.5^{\circ}$. DSC showed that the maximum degration temperature of powder was around $230^{\circ}C$. Through FT-IR analysis, we could identify cross-linking by the presence of peptide peak at 1,500~1,600 $cm^{-1}$. In conclusion, the origin of honeybee venom was Apis mellifera ligustica and effective ingredient standards was melittin content varied from 50.7 to 68.6 as natural antibiotic ingredients.

Developmental characteristics and genetic diversity of the two-spotted cricket Gryllus bimaculatus De Geer, 1773 (Orthoptera: Gryllidae) in South Korea

  • In this study, we investigated the developmental characteristics and genetic diversity of seven populations of two-spotted crickets (Gryllus bimaculatus De Geer, 1773 (Orthoptera: Gryllidae)) raised in South Korea. Regarding the developmental characteristics of the species, we observed no statistically significant difference in the weight of the nymphs in the six populations we tested. After molting, although weight differences were observed between the populations in each stage of the developmental period, the average weight for each developmental stage was constant. We also analyzed mitochondrial COI gene sequences (DNA barcoding region) of the reared crickets collected from five insect farms and two national insect rearing facilities and the resultant sequences were analyzed together with the 12 sequences from foreign countries specimens obtained from public data. We detected six haplotypes from 111 specimens, indicating a low intraspecific genetic distance (~1.8%). The most dominant haplotype was overwhelmingly haplotype 1, which was found in all South Korean specimens and four specimens from China, Indonesia, and Germany. These findings indicate that the low genetic diversity of South Korean specimens can be explained by the fact that the G. bimaculatus population imported for feed from Japan in the early 2000s became a maternal group that spread throughout cricket farms in South Korea. In order to breed healthy cricket strains, it is necessary to increase genetic diversity by importing them from other countries through appropriate quarantine procedures.

  • 과실파리, 굴파리의 유충에 기생하는 Areotetes van Achterberg & Li, 2013(벌목: 고치벌과: 꽃파리고치벌아과)는 중국에서 처음으로 보고된 바 있다. 현재까지 Areotetes속에는 4종이 보고되어 있다. 금번 연구에서 Areotetes속의 1종, Areotetes carinuliferus van Achterberg & Li, 2013를 한국에서 처음으로 보고하며, 본 종의 형태 진단, 분포, 도해도를 작성하였고, 추가적으로 미토콘드리아 COI 데이터를 제공한다.

Analysis of genetic differentiation and population structure of the Korean-peninsula-endemic genus, Semisulcospira, using mitochondrial markers

  • The genus Semisulcospira is an economically and ecologically valuable freshwater resource. Among the species, Semisulcospira coreana, Semisulcospira forticosta and Semisulcospira tegulata are endemic to the Korean peninsula and Semisulcospira gottschei is widespread in Asia. Therefore, maintenance and conservation of wild populations of these snails are important. We investigated the genetic diversity and population structure of Semisulcospira based on the mitochondrial cytochrome c oxidase subunit I (COI), NADH dehydrogenase subunit 4 (ND4), and combined mitochondrial DNA (COI + ND4) sequences. All four species and various genetic makers showed a high level of haplotype diversity and a low level of nucleotide diversity. In addition, Fu's Fs and Tajima's D neutrality tests were performed to assess the variation in size among populations. Neutrality tests of the four species yielded negative Fu's Fs and Tajima's D values, except for populations with one haplotype. The minimum spanning network indicated a common haplotype for populations of S. coreana, S. tegulata and S. gottschei, whereas S. forticosta had a rare haplotype. Also, genetic differences and gene flows between populations were assessed by analysis of molecular variance and using the pairwise fixation index. Our findings provided insight into the degree of preservation of the species' genetic diversity and could be utilized to enhance the management of endemic species.

  • 본 연구를 통해 한국의 양식 가숭어에서 최초로 분리된 등각류는 Nerocila japonica와 가장 근연한 종으로 확인되었으나, 향후 형태학적 분류 등 명확한 종 동정이 추가로 필요할 것으로 판단되며, 약물의 직접 노출에 의한 구충 효과를 확인한 결과 trichlorfon이 가장 효과가 있는 것으로 확인되었다.

  • Global climate change, followed by an increase in anthropogenic activities in aquatic ecosystems, and species invasions, has resulted in a decline in aquatic organism biodiversity. The Batanghari River, Sumatra's longest river, is polluted by mercury-containing illegal gold mining waste (PETI), industrial pollution, and domestic waste. Several studies have provided evidence suggesting a decline in fish biodiversity within the Batanghari River. However, a comprehensive evaluation of the present status of biodiversity in this river is currently lacking. The species under investigation were identified through various molecular-based identification methods, as well as morphological identification, which involved the use of neighbor-joining (NJ) trees. All collected specimens were initially identified using morphological techniques and subsequently confirmed with molecular barcoding analysis. Morphological and DNA barcoding identification categorized all specimens (1,692) into 36 species, 30 genera and 16 families, representing five orders. A total of 36 DNA barcodes were generated from 30 genera using a 650-bp-long fragment of the mitochondrial cytochrome oxidase subunit I (COI) gene. Based on the Kimura two-parameter model (K2P), The minimum and maximum genetic divergences based on K2P distance were 0.003 and 0.331, respectively, and the average genetic divergence within genera, families, and orders was 0.05, 0.12, 0.16 respectively. In addition, the average interspecific distance was approximately 2.17 times higher than the mean intraspecific distance. Our results showed that the COI barcode enabled accurate fish species identification in the Batanghari River. Furthermore, the present work will establish a comprehensive DNA barcode library for freshwater fishes along Batanghari River and be significantly useful in future efforts to monitor, conserve, and manage fisheries in Indonesia.

The Arabidopsis AtLEC Gene Encoding a Lectin-like Protein Is Up-Regulated by Multiple Stimuli Including Developmental Signal, Wounding, Jasmonate, Ethylene, and Chitin Elicitor

  • The Arabidopsis gene AtLEC (At3g15356) gene encodes a putative 30-kDa protein with a legume lectin-like domain. Likely to classic legume lectin family of genes, AtLEC is expressed in rosette leaves, primary inflorescences, and roots, as observed in Northern blot analysis. The accumulation of AtLEC transcript is induced very rapidly, within 30 min, by chitin, a fungal wall-derived oligosaccharide elictor of the plant defense response. Transgenic Arabidopsis carrying an AtLEC promoter-driven ${\beta}$-glucuronidase (GUS) construct exhibited GUS activity in the leaf veins, secondary inflorescences, carpel heads, and silique receptacles, in which no expression could be seen in Northern blot analysis. This observation suggests that AtLEC expression is induced transiently and locally during developmental processes in the absence of an external signal such as chitin. In addition, mechanically wounded sites showed strong GUS activity, indicating that the AtLEC promoter responds to jasmonate. Indeed, methyl jasmonate and ethylene exposure induced AtLEC expression within 3-6 h. Thus, the gene appears to play a role in the jasmonate-/ethylene-responsive, in addition to the chitin-elicited, defense responses. However, chitin-induced AtLEC expression was also observed in jasmonate-insensitive (coi1) and ethylene-insensitive (etr1-1) Arabidopsis mutants. Thus, it appears that chitin promotes AtLEC expression via a jasmonate- and/or ethylene-independent pathway.

  • The venus clam, Ruditapes philippinarum, is an aquaculture shellfish mainly distributed in an intertidal zone of East Asia including Korea, China and Japan. The morphological variation of this species is great. In fact, two of the most popular markers used in molecular evolution, mitochondrial DNA (mtDNA) and nuclear ribosomal DNA (rDNA), have quite different properties, which could translate into different consequences of mutation, drift, migration and selection on patterns of geographical variation and molecular divergence. Survey of Busan Oligochaeta earthworms supported by DNA barcodes

  • An earthworm survey of Busan metropolitan area unearthed a dozen taxa in four families (including Enchytraeidae). Members of mostly common, cosmopolitan earthworm species-complexes were: Drawida cf. koreana Kobayashi, 1938, Amynthas cf. corticis (Kinberg, 1867), Aporrectodea trapezoides (Dug$\grave{e}$s, 1828) and Eisenia fetida (Savigny, 1826). Also found were Amynthas hupeiensis (Michaelsen, 1895), A. masatakae (Beddard, 1892) and Metaphire ryunome Blakemore, 2012 - the latter a new Korean record. New taxa are: moniligastrid Drawida songae yeongdo subsp. n.; megascolecid Amynthas carnosus roki subsp. n. which is compared to nominal taxon A. carnosus (Goto and Hatai, 1899) from Japan, to A. carnosus monstriferus (Kobayashi, 1936) stat. n. from Korea and to A. lichuanensis Wang and Qiu, 2005 stat. n. from China; plus lumbricid Eisenia japonica vaga subsp. n. deemed an objectively-based molecular taxon on its unique DNA COI gene barcode. Restoration of Eisenia xanthurus (Templeton, 1836) for E. andrei is mooted (in Appendix).
