Stachybotrys atra에서 추출한 섬유소분해효소에 관한 연구 II (STudies on the Cellulolytic Enzymes of Stachybotrys atra)

  • A cellulase fraction (F IV-1) purified to about 8-folds was obtained from crude cellulase prepared from the wheat bran culture of S.atra. The partial purification of the enzyme was made by DEAE Sephadex and Sephdex cloumn chromatography in conjuction with ammonium sulfate precipitation. After stading at various pH's for 22 hours at $20^{\circ}C$, F IV-1 was most stable at pH 5.0 but when the enzyme fraction was stood for 74 hours, the point of pH stability was raised to around pH 6.0-7.0. After heating at various temperatures for 1 hour, F IV-1 was most stable at $20^{\circ}C$. The optimal enzyme activities of F IV-1 were seen at pH 6.0 and $50^{\circ}C$. The optimal concentrations of $Zn^{++}\;and\;Ca^{++}$ for the activities of crude cellulase were 6 and 4 mM respectively, but $Ca^{++}$ inhibited the enzyme activity at concentrations below 2 mM and above 6mM. Both $Cu^{++}\;and\;Mn^{++}$ ions inhibited cellulase activities and a ocmplete inactivation of crude cellulase was achieved at concentratioins of 5 and 2 mM of ions respectively. When Na-CMC was used as substrate, the Km values of crude cellulase and F IV-1 were calculated to be $5{\times}10^{-4}\;and\;2{\times}10^{-5}mM$, and V values 32 and 1.35 mmoles/hour, respectively. The Ki values of $Mn^{++}$ for crude cellulase and F IV-1 were found to be $8{\times}10^{-2}\;and\;3{\times}10^{-2}\;mM\;while\;those\;of\;Cu^{++}\;were\;at\;2{\times}10^{-1}\;and\;1{\times}10^{-1}\;mM\;respectively.\;Both\;Mn^{++}\;and\;Cu^{++}$ showed competitive inhibition with substrate.

온라인 플레밍(flaming) 발생 요인과 해소 방안에 대한 연구 (A Study on the Triggering and Medicating Factor of Online Flaming)

  • 본 논문은 반사회적 활동으로 평가받고 있는 온라인 플레밍 현상을 설명하고 이를 해소할 수 있는 요인은 무엇인지를 살펴보고 있다. 이를 위해 본 논문은 상황에 걸쳐 일관된 행위 패턴을 보여주는 개성 특질(personality trait) 그리고 사회인구학적 요인(성별, 연령)을 통해 플레밍 행위를 설명하고자 하였다. 나아가 높은 플레밍을 보여주는 유의미한 설명변인 집단들에게 메시지 신뢰도가 미치는 영향 정도를 연구하고 있다. 이를 위해 인터넷 조사기관의 온라인 설문을 통한 자료수집이 이뤄졌다. 분석 결과에 따르면, 성별과 연령 그리고 언어적 공격(verbal aggression) 특질은 플레밍과 유의미한 관계가 있는 것으로 밝혀졌다. 특히 남성, 10대, 그리고 공격 특질이 높은 사람들은 플레밍 가능성이 높은 것으로 나타났다. 또한 플레밍 해소방안으로서 분석된 메시지의 신뢰도는 성별, 연령과는 무관한 것으로 나타난 반면, 개성 특질과는 어느 정도 유의미한 것으로 나타났다. 이러한 결과는 온라인에서 이뤄지는 커뮤니케이션의 질(quality)은 플레밍을 해소할 수 있는 하나의 방안일 수 있음을 시사하고 있다.

NDS공법의 현장시험시공을 통한 지반개량 효과에 관한 연구 (A Study on the Improvement Effects of Soft Ground through In-Situ Construction of NDS)

  • This study applied the NDS method to the creek levee and the foundation of a building, for the purpose of ground improvement and cut-off. The conclusions are shown in the following. (1) The results of the field tests in the creek levee are as follows; the N value of the standard penetration test is $2{\sim}3$ before NDS grout and $9{\sim}21$ after NDS grout; in the alluvial layer, $7{\sim}11$ before NDS grout and $14{\sim}23$ after NDS grout. This confirms increasing ground strength with consistency ranging from stiff to very stiff. (2) The result of the permeability test in the creek levee shows that the ground had a great hydraulic conductivity with complete leakage before ground improvement but that the hydraulic conductivity has significantly increased to $3.17{\times}10^{-5}{\sim}4.65{\times}10^{-5}cm/sec$ after ground improvement by the NDS method. (3) The result from the field test of the foundation of the building confirms great reinforcing effects, showing that the allowable bearing capacity has increased from Pa = 5.0 $t/m^2$ before reinforcement to Pa = 25.0 $t/m^2$ after reinforcement.

디젤오염토양 복원 효율 증진을 위한 음이온/비이온 계면활성제 토양세척공정에 초음파 적용 영향 (The Effect of Ultrasound Application to Anionic/Non-ionic Surfactant Aided Soil-washing Process for Enhancing Diesel Contaminated Soils Remediation)

  • Ultrasound and Surfactant aided soil washing process has been shown to be an effective method to remove diesel from soils. The use of surfactants can improve the mobility of diesel in soil-water systems by increasing solubility of adsorbed diesel into surfactant micelles. However, a large amount of surfactant is required for treatment. In addition, synthetic surfactants, specially anionic, are more toxic and the surfactant wastewater is hard to treat by conventional wastewater treatments even by AOPs. Ultrasound improves desorption of the diesel adsorbed on to soil. The mechanisms are based on physical breakage of bonds by hot spot, directly impact onto soil particle surface, the fragmentation of long-chain hydrocarbons by micro-jet and microstreaming in the soil pores. The use of ultrasound as an enhancement method in both anionic and nonionic surfactant aided soil-washing processes were studied. And all experiments were examined proceeded under CMC surfactant concentration, frequency 35 khz, power 400 W, Soil-water ratio 1:3(wt%), particle size 0.24 ~ 2mm and initial diesel concentration. 20,000 mg/kg. Combination with ultrasound showed significant enhancements on all the processes. Especially, nonionic surfactant Triton-X100 with ultrasound showed remarkable enhancements and diesel removal rate enhanced by ultrasound helps desorpting of surfactant adsorbed onto soils which prevented decreasing surfactant activity.

가미통규탕(加味通竅湯)의 항염(抗炎).진통(鎭痛).항(抗)알러지.항균(抗菌)에 대(對)한 실험적(實驗的) 연구(硏究) (Effects of Gamitonggyutang on the Experimental Animals on Anti-inflammatory, Analgesic, Anti-Allergic Effect and Antibacterial Activity)

  • Gamitonggyutang has been used for treatment of rhinitis in oriental medical science. It is reported that Gamitonggyutang has a good effect on sinusitis in clinical medicine. So this study analysed the effect of Gamitonggyutang on anti-inflammatory, analgenic, anti-Allergic effect and Antibacterial Activity. The result were summerised as follows; 1. Gamitonggyutang extract decreased the edema indused by Carregennin at 200mg/kg and 400mg/kg. 2. Gamitonggyutang extract decreased the protein exudation indused by CMC-pouch at 200mg/kg and 400mg/kg. 3. Gamitonggyutang extract showed the ataralgesia at 800mg/kg by measurement of writhing syndrome, paw licking time and escape time induced by the i.p. infection of acetic acid and hot plate. 4. Gamitonggyutang extract decreased the effluent of vascular permeability indused by Evans blue at 600mg/kg and 800mg/kg. 5. Gamitonggyutang extract decreased the acute edema indused by Carregennin about 2 and 4 hours but didn't show useful effect. 6. Gamitonggyutang extract decreased the death rate, resulted from the effect of active systemic anaphylaxis reaction indused by CGG, but didn't show useful effect.. 7. Gamitonggyutang extract showed the growth inhibitory effect of each bacterias at $52mg/m{\ell}$. 8. Gamitonggyutang extract suppressed the growth of Streptococcus mutans 10449, and showed the supression of acid fabrication in case of 1:10 more than 1:100.

삼비탕(三痺湯)의 항염(抗炎), 항응고(抗凝固) 및 진통효과(鎭痛效果)에 대(對)한 연구(硏究) (Study on the Antiinflammatory, Anticoagulative and Analgesic effects of Sambitang in the experimental animal model)

  • This study was designed to elucidate the antiinflammatory, cardiovascular, antithrombotic, and analgesic effect of Sambitang. The antiinflammatory effects was measured by the method of carrageenin induced edema, protein leakage test using CMC-pouch, and the effect of Sambitang on the cardiovascular system was observed by the change of flow rate of Ringer solution in the vascular system in the ear of rabbit, and the contraction and dilatation of rat tail artery. Death rate, platelet aggregation, plasma coagulation activity was observed for the measurement of the anticoagurative effect of Sambitang, and the analgesic effect was measured by the acetic acid method and hot plate method. The result was as follows: 1. Sambitang administration, edema and protein leakage was significantly decreased. 2. The drug increased the auricular blood flow in rabbit. 3. The drug relaxed the artery contraction by pretreated norepinephrine in rat. 4. The drug inhibited the death rate of mouse which was led to thromboembo- lism by serotonin and collagen. 5. The drug inhibited the platelet aggregation in rat. 6. The drug prolonged the prothrombin time and activated partial thromboplastin time on the test of plasma coagulation factor activity in rat, but was not valuable. 7. The slight anagesic effect of Sambitang extract was confirmed by the observation of writhing syndrome, paw licking time, and escape time.

안쪽축이 회전하는 환형관내 비뉴튼 유체의 천이 유동 연구 (Transitional Flow study on non-newtonian fluid in a Concentric Annulus with Rotating Inner Cylinder)

  • This experimental study concerns the characteristics of a transitional flow in a concentric annulus with a diameter ratio of 0.52, whose outer cylinder is stationary and inner one rotating. The pressure losses and skin-friction coefficients have been measured for the fully developed flow of $0.1\sim0.4%$ aqueous solution of sodium carbomethyl cellulose (CMC), respectively at inner cylinder rotational speed of $0\sim600rpm$. The transitional flow has been examined by the measurement of pressure losses to reveal the relation of the Reynolds and Rossby numbers with the skin-friction coefficients. The present results show that the skin-friction coefficients have the significant relation with the Rossby numbers, only for laminar regime. The occurrence of transition has been checked by the gradient changes of pressure losses and skin-friction coefficients with respect to the Reynolds numbers. The increasing rate of skin-friction coefficients due to the rotation in uniform for laminar flow regime, whereas it is suddenly reduced for transitional flow regime and, then, is gradually declined for turbulent flow regime. Consequently, the critical(axial-flow) Reynolds number decrease as the rotational speed increases. Thus, the rotation of inner cylinder promotes the early occurrence of transition due to the onset of taylor vortices.

고농축에멀션중합방법을 이용한 분리막제조와 선택적 흡수성 연구 (Preparation of Hydrophilic-Hydrophobic Composites by A Concentrated Emulsion Polymerization Method and Their Permselectivity to Mixture of Water-Ethanol)

  • 고농축에멀션중합방법을 이용하여 친수성-친유성 고분자복합소재를 합성하였다. w/o 형태의 고농축에멀션의 연속상은 styrene이며, 분산상은 수용성 acrylamide용액으로 분산상의 부피분율이 0.74 이상이다. 중합온도에서 에멀션의 안정성에 미치는 주요 요인들로는 친유성액체와 친수성액체의 물리화학적 특성인 극성, pH, 이온세기 및 계면활성제의 종류와 농도가 있다. 친유성액체와 친수성액체 사이의 계면장력이나 친수성액체와 계면활성제 수용액사이의 계면장력에 있어 계면장력의 값이 큰 경우는 높은 에멀션의 안정성을 나타낸다. 계면활성제의 HLB가 3.0~9.0 경우와 계면활성제의 농도가 CMC가 이상에서 에멀션이 높은 안정성을 나타낸다. 전자현미경을 이용한 조사에 의하며 복합재료는 분산상의 Poly acrylamide입자가 망상구조의 얇은 polystyrene필림에 의해 분리된 구조로 이루어져 있음을 보여주고 있다. 고농축에멀션에서부터합성된 고분자복합재료막은 물-에탄온 혼합물에서 물에 대해서 높은 선택성을 나타내었으며, 또 에탄올의 농도가 증가할수록 막의 선택도는 증가하였으나 투과속도는 감소함을 나타내었다.

  • 근래의 전기 전자 제품은 기능은 향상되는 반면 크기는 소형화되는 추세에 있다. 기존 입체 형태의 음향 신호 재생 장치로서의 엑츄에이터 대신 압전 소자를 이용한 스피커가 개발되기 시작하였다. 압전 소자는 압전 특성을 갖는 물질로서 물체의 외부에서 전압을 가할 때 기계적인 변형을 일으키는 특징(즉 피에조 특징)을 이용한다. 초기의 필름 스피커는 화학적 성질의 한계로 인하여 전도성 고분자 막을 형성하기 힘들다는 한계가 있었고, 도포한 두께를 균일하게 유지하기 힘들었으며, 음향 신호의 출력과 동작 주파수의 한계가 있다는 등의 종래 기술의 문제점을 해결하기 위해, 전도 물질과 피에조 물질(필름)의 결합력을 증가시킴으로써 전도체의 저항을 줄이고 주파수 영역을 확장하고자 한다. 피에조 필름의 표면 접착력을 향상시키고, 고분자 전도체 막의 흡착력을 증가시킴으로써 음향시스템의 출력 향상을 위해 코로나 표면처리방식에 의한 표면 특징을 변화시킴으로써 출력개선을 하였다.

구조적 분석에 의한 e-BBS 토의 활동 및 참여자의 행동 분석 (The Analysis of Discussion Activities and Participants' Behavior in the e-BBS based on a Structural Analysis)

  • 비동기 통신 수단으로써의 e-BBS (electronic-Bulletin Board System, 전자게시판)는 협력 학습을 위한 유용한 매체로 알려져 왔다. 그러나 e-BBS의 구조적 분석을 이해하기 위한 이론적 결과는 이직 충분하지 못하다. 그 결과, 복잡한 의사소통 현상을 이해하고 교수자를 도와 학생들을 교육 목적으로 이끌 수 있도록 보조하는 적절한 도구의 부재를 초래하게 되었다. 그로 인해 BBS를 활용하는 교수자는 토의 상황의 구조적, 종합적 이해가 부재한 상태에서 거의 방임적으로 운영하게 된다. 이러한 문제점에 대처하기 위하여 본 논문에서는 BBS의 구조적 이해를 돕기 위한 이론적 토대를 제시하고 그를 바탕으로 토의 활동 참여자의 행동 분석을 위한 계산 모델을 제시하고자 한다. 쌍방향 토의 활동의 형식 표현은 응답 그래프와 연결 그래프의 두 그래프로 이루어지며 분석을 위한 여러 측정치와 통계량이 제시된다. 분석의 결과를 명확하게 제시하기 위하여 유형별로 분류한다.
