Functionality of Extracted Proteins by Additives and Ionic Strength (첨가제의 종류와 이온강도에 따라 추출되는 육단백질의 기능적 특성)

  • This study was investigated to determine the effect of additives and ionic strength on the functionality of extracted proteins in preblends in order to use less additive in restructured meat products. Preblends contained the combinations of sodium chloride (NaCl; 0, 4.5, 9.0%), sodium tripolyphosphate (STPP; 0, 2.5, 5.0%), and tetrasodium pyrophosphate (PP; 0, 2.44, 4.88%). The pH values increased linearly with increasing STPP and PP concentrations (p<0.01). In the equivalent ionic strengths, PP was more effective than STPP in increasing pH. Phosphate ions were more effective on total extractable protein (used 1 M NaCl buffer) than chloride ion at equivalent ionic strengths. Solubility was decreased by adding NaCl and increasing total extractable proteins. Meat sulfhydryl contents were high with increasing total extractable proteins. When protein extracts were heated at $65^{\circ}C$, 7 min, meat sulfhydryl contents decreased and surface hydrophobicity increased (p<0.01). However, all protein extracts showed no differences in SDS-PAGE pattern. In conclusion, PP is more effective than STPP in order to use less additive but there was no linear relationship between functionnal improvement and ionic strength.

Spectrophotometric Determination of Traces of Phosphorus in Semiconductor-grade Trichlorosilane (반도체급 삼염화실란 중의 극미량 인의 분광광도법적 정량)

  • A procedure for spectrophotometric determination of traces of phosphorus(P) in high-purity trichlorosilane(TCS) is proposed using an adsorptive separation. $PCl_3$, which is a dominant P impurity within TCS, is first oxidized by oxygen to a stable form as $POCl_3$. $AlCl_3$ is selected as an adsorbent which forms a thermally stable complex with $POCl_3$ in TCS and can be well dissolved in aqueous ethanol solution. The proposed adsorptive separation method is free from the formation of silica gel and gas bubbles during the colorimetric analysis of TCS. The method reveals that the P concentration in a semiconductor-grade TCS is 5.32 ${\mi}g/l$ within the standard deviation of ${\pm}$ 17%. On the other hand, the P concentration of the purified TCS which is separated from the $AlCl_3$${\cdot}$$POCl_3$ complex is reduced to be less than 0.15 ${\mi}g/l$, showing the efficient applicability of $AlCl_3$ to the wet chemical analysis. The proposed method is also tested to verify the effectiveness of other well-known adsorbents.

Solution Properies of Algin Affected by Various Food Additives (여러 가지 식품처가제에 의한 Algin 용액의 유동특성)

  • In order to obtain data for use of algin in drink making process solution properties of algin have been investigated at various condition of algin concentration temperature pH and various food additives. At same revolution velosity viscosities of algin were increased as algin concentration raised. Algin solution showed dilatnat type flow in concentration of 0.25% to 0.4% but pseudoplastic type flow in above 0.5% showed dilatant type flow in concentration of 0.25% to 0.4% but pseudoplastic type folw in above 0.5% A maximum viscosity of algin was observed at pH 5.5 and its viscosities were also decreased as the tem-perature increased and heating at 8$0^{\circ}C$ above. Organic acids affected on the viscosity of algin with pH dependently and gel formed in pH below 3.0 Sweetners have no effect to the viscosity of algin. How-ever addition of NaCl and KCL upto 1.0% decreased a little its viscosity and CACl2 MgCl2 and FeCl3 increased the viscosity of algin Glutamic acid decreased the viscosity of algin.

  • $SrTiO_3$ reaction mechanism formed from $TiCl_4$ and $SrCl_2$ solution by direct-wet-process was studied. Through this study it is identified that crystalline $SrTiO_3$ is formed above pH 13.8 amorphous $SrTiO_3$ above pH7 and crystallization temperature of amorphous $SrTiO_3$ is at 37$0^{\circ}C$. the final products are composed of 60% crystalline $SrTiO_3$ and 40% amorphous $SrTiO_3$ The amorphous $SrTiO_3$ is identified with the IR absorption spectrum of Sr-Ti-O by FT-IR spectrometer. Under pH 7 gelationus metatitanate (H2TiO3) is formed from TiCl4 but above pH7 the activity by the formation of metatitnic acid ion $[TiO_3]^{2-}$ is so high that $SrTiO_3$is formed more easily through the reaction with $Sr^{2+}$.

Chemical Equilibria of Nickel Chloride in HCl Solution at 25˚C

  • Chemical equilbria of nickel chloride in HCl solution at $25\;{^{\circ}C}$ were analyzed by considering chemical equilibria, mass and charge balance equations. The activity coefficients of solutes were calculated by using Bromley equation. The necessary thermodynamic properties, such as the equilibrium constant for the formation of Ni$Cl^+$ at zero ionic strength and interaction parameter, were evaluated by applying Bromley equation to the stability constant data reported in the literature. It was found that most nickel exists as $Ni^{2+}$ in HCl solution up to 5 molality HCl. The pH values of Ni$Cl_2-HCl-H_2O$ system at $25\;{^{\circ}C}$ calculated in this study agreed well with the pH values measured by employing pH meter.

  • Analyses of $_1$H-NMR, infrared and electronic spectral data for $[Ir(CH_2 = CHCN)(CO)(P(C_6H_5)_3)_2]ClO_4 (1)$prepared by the reaction of $Ir(OClO_3)(CO)(P(C_6H_5)_3)_2$ with $CH_2 = CHCN$, agree with the suggestion that 1 is a mixture of the nitrogen-bonded acrylonitrile complex, $[(CO)(P(C_6H_5)_3)_2Ir-NCCH = CH_2]ClO_4$ and other compound which may be the C = C ${\Pi}$ -system-bonded acrylonitrile complex, "[(CO)(P(C6H5)3)2Ir-CHCN = CH2]ClO4.

Effect of Temperature and Sodium Chloride on Seed Germination of Thuja orientalis (온도 및 염화나트륨이 측백나무 종자 발아에 미치는 영향)

  • This study was conducted to investigate effect of temperature and NaCl on the seed germination and water absorption of Thuja orientalis. Seeds were treated with 0, 500, 1,000, 2,000 and 4,000ppm of NaCl and placed in different chambers at 15, 20, 25 and $30^{\circ}C$, respectively. And seed properties and relative water absorptions were analyzed. Germination decreased with the increase of both temperature and NaCl concentration, and especially the difference was obvious at $30^{\circ}C$. Dormancy and mortality increased with the increase of temperature in non-NaCl treatment. Two-way analysis of variance showed significant effects of temperature, NaCl concentration and interaction between temperature and NaCl concentration (p<0.001). Mean germination time increased with the increase of NaCl concentration at 15, 20 and $25^{\circ}C$ but decreased at $30^{\circ}C$ because the seeds were mortal by NaCl high concentration, Germination speed and germination performance index decreased with the increase of NaCl concentration. Those represented decreasing tendency with NaCl concentration but high positive correlation with germination. Relative water absorption decreased with the increase of NaCl concentration and represented high values at $15^{\circ}C$, and showed high positive correlations with germination, germination speed and germination performance index. It was reported that the high temperature and salinity were inhibitive factors of seed germination of Thuja orientalis.

Roles of MicroRNA-21 and MicroRNA-29a in Regulating Cell Adhesion Related Genes in Bone Metastasis Secondary to Prostate Cancer

  • Background: There is an increasing concern in the role of microRNA (miRNA) in the pathogenesis of bone metastasis (BM) secondary to prostate cancer (CaP). In this exploratory study, we hypothesized that the expression of vinculin (VCL) and chemokine X3C ligand 1 (CX3CL1) might be down-regulated in clinical samples, most likely due to the post-transcriptional modification by microRNAs. Targeted genes would be up-regulated upon transfection of the bone metastatic prostate cancer cell line, PC3, with specific microRNA inhibitors. Materials and Methods: MicroRNA software predicted that miR-21 targets VCL while miR-29a targets CX3CL1. Twenty benign prostatic hyperplasia (BPH) and 16 high grade CaP formalin-fixed paraffin embedded (FFPE) specimens were analysed. From the bone scan results, high grade CaP samples were further classified into CaP with no BM and CaP with BM. Transient transfection with respective microRNA inhibitors was done in both RWPE-1 (normal) and PC3 cell lines. QPCR was performed in all FFPE samples and transfected cell lines to measure VCL and CX3CL1 levels. Results: QPCR confirmed that VCL messenger RNA (mRNA) was significantly down-regulated while CX3CL1 was up-regulated in all FFPE specimens. Transient transfection with microRNA inhibitors in PC3 cells followed by qPCR of the targeted genes showed that VCL mRNA was significantly upregulated while CX3CL1 mRNA was significantly down-regulated compared to the RWPE-1 case. Conclusions: The down-regulation of VCL in FFPE specimens is most likely regulated by miR-21 based on the in vitro evidence but the exact mechanism of how miR-21 can regulate VCL is unclear. Up-regulated in CaP, CX3CL1 was found not regulated by miR-29a. More microRNA screening is required to understand the regulation of this chemokine in CaP with bone metastasis. Understanding miRNA-mRNA interactions may provide additional knowledge for individualized study of cancers.

  • The separation of trypsin inhibitir using reverse micellar system was invertigated. Among the biffer system tested, 1.0M $CaCl_2$ solution (pH 3.0) and 1.0M NaCl soluation (pH 11.5) were most effective for solubilization and de-solubilization of protein, respectively. When these conditions were applied to two model sampeles, one of which was composed of the same amount of 7S protein and trpsin inhibitor, and the other of which was composed of the same amount of soluble soybean protein isolates and trypsin inhibitor, highly pure trypsin inhibitor was obtained. And in the real soybean, Kwng Gyo, pure trypsin inhibior was also obtained.

Desalinization Characteristics after Reclamation of Tidal Flat on the Western Coast of Korea (서해안 간척지 토양의 탈염특성)

  • Vertical and temporal characteristics of desalinized reclaimed soil were analyzed from reclaimed coastal land on the western coast of Korea. Of the vertical changes during desalting, pH valuse were the lowest at the topsoil without regard to reclaimation time. The content of C1 were designated as the early period (the first 2-4 years) which decreased exponentially and the later period(the last 5-7 years) which was almost constant, from top to down. In temporal changes of the soil attributes, pH values increased for 5 years and decreased at 6 year after reclamation. Chlorine leaches more rapidly than Na does, K and Ca are constant but Mg increases as time elapsed after reclamation. Sometimes the content of Ca and K in the reclaimed soil are of higher concentration than that of the seawater after reclamation. During desallinization as exemplified by decreasing EC of the soil, Cl and Na are rapidly leached, but K, Ca and Mg are somewhat enhanced. The ration of Na/Cl in the soil equals 1 when the EC registers 5 mmho and then increases dramatically as the EC decreases. Rapid leaching of $Cl^{-}$ elicits an increasing pH valus. The electrostatic balance of the soil is achived by replacement of $Cl^{-}$ with $OH^{-}$ until stationary or until a decreasing pH value is reached again.
