Effects of Dietary Modification on Plasma Glucose and Insulin Sensitivity in Streptozotocin-induced Diabetic Rats

  • There is substantial evidence that insulin sensitivity can be enhanced through appropriate dietary management . In this study, insulin sensitivity was evaluated using and insulin suppression test. Male Sprague-Dawley rats, were caused to be in a diabetic condition by the injection of streptozotocin, and divided into four groups. They were fed one of the following diets for 2 weeks : (group 1) a high-carbohydrate(CHO) low-fat low-fiber diet, (group 2) a high CHO low-fat and high-fiber diet, (group 3) a low-CHO high-fat and low-fiber diet, and (group 4) a low-CHO low-fat diet groups (as comparison between group 1 and group 2 shows). In the low-CHO high-fat diet groups, dietary fiber tended to decrease plasma glucose levels at the end of the experiment, but not significantly (as comparison between group 3 and group 4 shows). The average steady state plasma glucose level in rats on the group 3 diet was the highest among all four groups(p<0.05), indicating the poorest insulin sensitivity . However, high fiber increased insulin sensitivity in rats on the low-CHO high-fat diets(as shows by a comparison between group 3 and group 4). On the other hand , the high-CHO low-fat enhanced insulin sensitivity in rats on the low fiber diet(group 1 and group 3). The degree of enhancement of insulin sensitivity depends on the combination of CHO, fat , and fiber in the diet. In conclusion, this study demonstrates that a low-CHO high-fat low -fiber diet may be deleterious to diabetic rats. In view of insulin sensitivity enhancement , dietary fiber level is irrelevant, as long as the diet has a high-CHO and low-fat level.

대리모의 준비 조건 변화를 통한 복제미니돼지의 생산 (Production of Cloned Miniature Pig by Surrogate Mother Conditions)

  • Somatic cell nuclear transfer (SCNT) for miniature pig has been developed for xenotransplantation and many other biomedical experiments. However, the efficiency of SCNT is still very low due to many factors. To optimize the surrogate mother condition for improvement of cloned miniature pigs efficiency, we investigated the effect of the status of surrogate mother on pregnancy, farrowed rate in SCNT pigs. After SCNT with mesenchymal stem cells as donor cells, the SCNT embryos were surgically transferred into the oviduct of surrogated pigs. To compare the effects of status of surrogate pigs on pregnancy, surrogate pigs were prepared by artificial abortion at day 20~29 (Group 1), 30~39 (Group 2), and 40~45 (Group 3) of gestation. After SCNT embryos transfer in three different status of surrogate pigs, Group 2 (56.3%) and 3 (55.6%) had significantly ($p$ <0.05) higher the pregnancy rate than group 1 (0%) at day 30 of gestation. The status of ovulation in surrogate pig also was investigated. Post-ovulation status (54.8%) had higher proportion than pre-ovulation status (38.7%) and ovulation status (6.5%). We obtained 19 cloned miniature piglets from seven surrogate gilts and five piglets are living healthy but fourteen piglets died soon after birth or stillbirth. The weights of piglets greatly differ from 254 to 1,296 g. Microsatellite analysis showed that cloned piglets were genetically different from the surrogate mother and cloned piglets were genetically equal to the donor cell. In conclusion, the present result indicates that artificially abortion method can improve the efficiency of pregnancy after SCNT in pigs. This study will provide available method for the further study and application in the field of xenotransplantation.

인삼사포닌 (ginsenoside) 저밀도 지방단백질 수용체 생합성에 미치는 영향 (Effect of Ginsenosides on .the Biosynthesis of Low density Lipoprotein Receptor in Cultured Chinese Hamster Ovary(CHO) Cell)

  • 여러가지 농도의 cholesterol을 함유한 배지에서 배양한 CHO 세포내로 흡입된 cholesterol 양을 조사한 결과 흡입량이 배지의 cholesterol 농도에 비례하였으므로 cholesterol 흡입은 확산에 의한 것으로 생각된다. $^{125}I$으로 표지된 저밀도 지방단백질($^{125}I$-LDL)을 이용하여 여러가지 농도의 cholesterol을 함유한 배지에서 배양한 CHO 세포와 cholesterol이 없는 정상배지에서 배양한 CHO 세포에서의 LDL 수용체의 합성양상을 조사한 결과 배지에 가해준 cholesterol이 LDL 수용체 합성을 억제함을 확인하였다. Cholesterol의 LDL 수용체 생합성 억제작용에 미치는 ginsenoside의 영향을 조사하기 위해 ginsenoside와 cholesterol을 함께 함유한 배지에서 배양한 CHO 세포(시험군)과 cholesterol만을 포함한 배지에서 배양한 CHO 세포(대조군)에서의 LDL 수용체의 활성양상을 분석한 결과 대조군에 비해 시험군에서의 LDL 수용체 활성이 크게 증가하였다. RNA 및 단백질 합성도 시험군이 대조군보다 증가하였음을 관찰하였다. 그러나 이와 같은 실험조건하에서의 대조군과 시험군의 cholesterol농도를 측정한 결과 시험군의 cholesterol 농도가 대조군보다 훨씬 저하되고 있었다. Ginsenoside의 cholesterol농도 저하작용을 관찰하기 위해 CHO세포에서의 cholesterol의 steroid hormone(estradiol, progesterone)으로의 전환에 미치는 ginsenoside의 영향을 조사한 결과 ginsenoside는 cholesterol의 hormone으로의 전환을 촉진하였음이 확인되었다. 위와 같은 실험결과로 볼 때 ginsenoside는 CHO 세포내부에서의 cholesterol의 LDL 수용체 합성억제를 완화시켜 주는 것이라고 생각된다.

석면에 의한 CHO 세포의 염색체 이상 유발 기전에 관한 연구 (Mechanism of Asbestos Induced Chromosome Aberration in CHO Cells)

  • In order to examine the mechanism of asbestos clastogenicity, CHO cells were treated with chrysotile and crocidolite. Crocidolite and chrysotile were able to induce lipid peroxidation in a dose dependent manner. Ultrafiltrate of culture media from CHO cells treated with chrysotile/crocidolite induced sister chromatid exchange in CHO cells. Ultrafiltrate of culture media from CHO cells treated with chrysotile induced chromosome aberration but it was not statistically significant. Simultaneous treatment of 3-Aminobenzamide (3-AB) or cytosine arabinoside (Ara C) with crocidolite had no effect on the frequency of chromosome aberration by crocidolite whetease posttreatment of caffeine significantly increased the chromosomel aberration by crocidolite. This indicated that DNA damage by asbestos took place at late stage of cell cycle. The results suggested that the ultrafiltrate of media contained clastogenic factor (CF) and lipid peroxidation might be involved in the formation of CF.

  • Photovoltaic system has very low energy conversion efficiency and the output characteristics of solar cell is varied by the Insolation quantity and the temperature. In order to improve the efficiency of photovoltaic system, the energy which has got from solar cell must be use maximum. In this paper, it was stimultaneous executed both MPPT control and instrumentation in order that the operating point of solar cell is located maximum power point, using the PWM inverter and micro-computer, which is for the purpose of acquiring maximum power from the solar cell. As a result, maximum power point tracking had carried out and the efficiency of photovoltaic system improved, even if insolation quantity and the temperature are varied.

익모초의 생약학적 연구 (Pharmacognostical Studies on the Ig Mo Cho)

  • Ig Mo Cho (益母草) has been used as remedy for invigorating blood circulation to remove blood stasis and relieving edema by inducing diuresis. With regard to the botanical origin of Ig Mo Cho, it has been considered to be Leonurus spiecies of Labiatae, but there has never been pharmacognostical confirmation on it. To clarify the botanical origin of Ig Mo Cho from Korea, the anatomical characteristics of Leonurus japonicus and L. sibiricus were studied. As a result, the botanical origin of Ig Mo Cho from Korea was proved the herba of Leonurus japonicus.

초오의 수치방법에 따른 알칼로이드 함량 (Effects of Various Processing Methods on the Contents of Alkaloids in the Cho O(Aconiti Ciliare Tuber))

  • The contents of alkaloids in raw Cho O(Aconiti Ciliare Tuber) and its processed products were determined by high performance liquid chromatography. Processing methods 1 and 2 lowered the contents of alkaloids more than the method 3. Cho O processed with method 1 or 2 showed even lower alkaloid contents than commercially available Processed Aconiti Tuber Powder(PATP). The contents of aconitine and hypaconitine in MeOH extracts of Cho O treated with method 3(10 min irradiation) and mesaconitine of Cho O treated with method 3(5 min) were comparable to those of PATP.

민간약 "진해초"의 생약학적 연구 (Pharmacognostical Studies on the Korean Folk Medicine 'Jin Hae Cho')

  • Korean folk medicine 'Jin Hae Cho' has been used as a remedy for neuralgia and an invigorating drug after a childbirth in Korea. The botanical origin of the crude drug has been no pharmacognostical confirmation on it. To clarify the botanical origin of 'Jin Hae Cho', studied on the morphological and anatomical characteristics of the roots of Potentilla species growing wild in Korea i.e. Potentilla chinensis Ser., P. cryptotaeniae Maxim., P. dickinsii Fr. et Sav., P. discolor Bunge, P. fragarioides L. var. major Maxim., P. freyniana Bornm., P. kleiniana Wight et Arnott, P. paradoxa Nutt., P. yokusaiana Makino and 'Jin Hae Cho' from Korea on Korean market. As a result, it was made clear that 'Jin Hae Cho' from Korea was derived from the roots of Potentilla chinensis Ser. and Potentilla discolor Bunge.

민간약 "암초"의 생약학적 연구 (Pharmacognostical Studies on the Folk Medicine 'Am Cho')

  • The Korean folk medicine 'Am Cho' has been used as the folk remedy for diuretic, stomach and liver cancer. The botanical origin of the crude drug has not been studied pharmacognostically. To clarify the botanical origin of 'Am Cho', the comparative morphological and anatomical characteristics have been studied of Lepisorus species growing wild in Korea, i.e. L. annuifrons, L. onoei, L. thunbergianus, L. ussuriensis and 'Am Cho' form Korea. As a result, it was clarified that 'Am Cho' from Korea was derived from Lepisorus onoei.
