Inactivation of Photosystem I in Cucumber Leaves Exposed to Paraquat-Induced Oxidative Stress

  • Cucumber leaves subjected to light chilling stress exhibit a preferential inactivation of photosystem(PS) I relative to PSII, resulting in the photoinhibition of photosynthesis. In light chilled cucumber leaves, Cu/Zn-Superoxide dismutase(SOD) is regarded as a primary target of the light chilling stress and its inactivation is closely related to the increased production of reactive oxygen species. In the present study, we further explored that inactivation of PSI in cucumber leaves is not a light chilling specific, but general to various oxidative stresses. Oxidative stress in cucumber leaves was induced by treatment of methylviologen(MV), a producer of reactive oxygen species in chloroplasts. MV treatment decreased the maximal photosynthetic O$_2$ evolution, resulting in the photoinhibition of photosynthesis. The photoinhibition of photosynthesis was attributable to the decline in PSI functionality determined in vivo by monitoring absorption changes around 820 nm. In addition, MV treatment inactivated both antioxidant enzymes Cu-Zn-superoxide dismutase and ascorbate peroxidase known sensitive to reactive oxygen species. From these results, we suggest that chloroplast antioxidant enzymes are the primary targets of photooxidative stress, followed by subsequent inactivation of PSI.

Effects of Shading, Chilling and Gibberellin Treatments on the Flowering of Azalea (Rhododendron spp.) (차광, 저온 및 지베렐린 처리가 Azalea의 개화에 미치는 영향)

  • This research was conducted to control flowering of azalea (Rhododendron spp.) by shading, chilling and gibberellin treatments. Higher shading among treatments resulted in extended duration of flowering (DF) and increased number of flowers (NF) in all varieties tested. Number of days to flower (NDF) was increased in 'Ripple' by shading treatment, but this was not affected in other was vice verse in 'Reinhold Ambrosia' resulting in decreased NDF. Chilling treatment followed by high shading increased NDF and shortened DF in 'Inga' and 'Ripple', but that extended NDF and DF and increased NF in 'Reinhold Ambrosia'. In flowering response to $GA_3$ resulted in decreased NDF and increased NF. These results indicated that treatments of shading, chilling and gibberellin to azalea extended DF and increased NF, but NDF appeared different according to varieties.

  • Comparative tolerance of the new rice varieties from Indicax Japonica cross to elastic and plastic chilling injuries was examined at the 3rd leaf-stage, the most sensitive stage to chilling during nursery, using a chilling chamber set 11\circ C, 13hrs. 20K lux day/5$^{\circ}C$ night. Seedlings were reared until 35 days after seeding in a 26\circ C, 13hrs. day / 18\circ C night chamber except the period of chilling treatment for 0, 3, 4, 6, 8 or 10 days. The new varieties were approximately twice susceptible to elastic injuries, and thrice susceptible to plastic injuries than the Japonica varieties in the term of chilling duration. Among the variables indicating apparant chilling injuries the reduction in dry matter responded most sensitively and differentiated well the varietal difference in chilling resistance even with shorter chilling period. Sequential aspects of the cause and effect relations involved in the development of the apparant chilling injuries are discussed and a schematic diagram is presented as Fig. 5 in the text.

Effect of Several Treatments on Chilling Injury of Paprika Fruits during Low Temperature Storage (몇 가지 처리가 파프리카 과실의 저온장해에 미치는 영향)

  • Paprika fruits should be stored and distributed at above $7^{\circ}C$ to prevent chilling injury but the small amount of paprika that transports with other horticultural products in refrigerated container by ship usually stored less than $5^{\circ}C$ for other products. In this case, paprika fruits cannot help exposing chilling temperature, so that the paprika must be lost marketable value during a long period of transfer. This study was conducted to compare the alleviated effects of high $CO_2$ treatment (passive MAP), heat (hot water dipping), and UVc treatment on chilling injury of paprika fruits due to low temperature storage, and also to decide if these treatments can be used for transporting under $5^{\circ}C$. After each treatment the paprika were put in the low temperature storage ($4^{\circ}C$) for 20 days and afterwards change the in room temperature ($20^{\circ}C$) for 5 days. The fresh weight loss of all the treatments except the high $CO_2$ treatment showed around 7~12% after 25 days of storage and the ethylene concentration showed periodical increases and decreases as around 3 ${\mu}l/l$. The $CO_2$ concentration was rapidly increased 33% carbon dioxide in high $CO_2$ treatment during room temperature storage after cold storage for 20 days. The firmness which is key quality characteristics during storage and is decreasing caused by chilling injury was not significantly different among all treatments. However, the firmness of stored paprika was maintained highest in the treated with hot water dipping. Therefore, HWD and UVc treatment that showed 60% of electrolyte leakage in the $4^{\circ}C$ control (chilling injury control) and similar level with the $7^{\circ}C$ control (non-chilling injury control) would be effective to alleviate chilling injury in the stored paprika.

The Correlation between Uneven Ripening and Respiration Rate of Tomato at Breaker Stages (Breaker 단계 토마토의 착색불량과 호흡률간 상관관계)

  • Breaker stage tomato fruits that were stored at low temperatures show typical chilling symptoms such as uneven ripening and a high respiration. Experiments were performed to assess and compare these chilling injury symptoms of breaker stage tomato fruits, and to gather basic data that can be used to decide whether horticultural crops receive chilling injuries. Tomato fruits that had been sorted in the breaker stage were stored at $2^{\circ}C$ for 0, 3, 6, and 9 days, and then their respiration rates were measured at 3, 6, 9, 12, 24, 36, and 48 h after moving them to room temperature. This treatment was repeated twice on the same procedures, except the storage periods, which were changed to 0, 1, 2, 3, and 10 days. The respiration rate was increased in a 1 day storage treatment, and the increasing rate rose higher with extended storage periods. The $a^*$ value, which represents the surface color of tomato fruits, was measured 10 days after moving to them into room temperature. Those with an increased $a^*$ value rate got dull and showed uneven coloring after 2 days' storage treatment. These two factors, the respiration rate and $a^*$ value of the surface, showed a high correlation (r = 0.9716, p < 0.001). Therefore, the chilling injury of breaker stage tomato fruits can be diagnosed by measuring the respiration rate after moving them into room temperature, and the degree of chilling injury can also be assessed in terms of the respiration increase rate.

The effect of chemicals and chilling treatments on the frequency of metaphase in root tips of some economic crops (화학약품과 저온처리가 작물근단세포의 Metaphase 출현빈도에 미치는 영향)

  • Root tips of wheat, soybean, cotton and barley were treated with cold temperature (12 or 24 hrs in $0^{\circ}C$ or 2$^{\circ}C$), 8-Hyd.exyquinolin (2 or 10 hrs in 0.03 or 0.1%) and colcllicine (2 or 10 hrs in 0.2 or 1, 0%), and the frequency of metaphase were observed. The results were summarized as follow; 1. Chilling the seminal roots before or after sooting from the seed significantly increased tile number of mitotic cells and the rate of metaphase cells to mitotic cells. The optimal duration of chilling seemed to be differ depending on the kinds of plant and 24 hours to be too long except wheat so far examined here. 9. 8-Hydroxyquinolin treatment, about 2 hours in 0.03%, increased the rate of metaphase cells. The higher concentration and the longer treatment of this chemical caused the lower frequency of mitotic cells generally. 3. Colchicine treatment, 2 to 10 hrs in 0.2 to 1.0%, increased the frequency of mitotic cells and the rate of metaphase cells. Colchicine treatment was same or superior than any other treatments on the increase of metaphase cells. 4. All the treatments examined here caused chromosome contraction with most serious in colchicine following by 8-Hydroxyquinolin and chilling. 5. The feasibility of general application of chilling under the check of proper temperature and proper duration depending on the kinds of crops were discussed.

Effects of Hot Water and Chilling Treatments of Bulblets Propagated by Tissue Culture on Sprouting and Bulb Development in Korean Native Lilies (조직배양 유래 자생나리 소인경의 온탕 및 저온처리가 맹아 및 비대에 미치는 효과)

  • This study was carried out to investigate the effects of hot solution soaking and chilling treatment on sprouting and enlargement of bulblets obtained through in vitro culture in Korean native lilies with ornamental values. In vitro bulblets of Lilium cernuum, L. hansonii, L. hansonii for. mutatum, L. leichtlinii, and L. tsingtauense were soaked in distilled water or 100 $mg{\cdot}L^{-1}$ $GA_3$ and $GA_{4+7}$ solution maintained at $35^{\circ}C$ for two hours (hot water treatment) and/or exposed at $4^{\circ}C$ for 0, 4, or 8 weeks (chilling treatment) and then planted in plastic trays filled with media and grown in a greenhouse at $20^{\circ}C$ and under 16 h photoperiod. In all species, no bulblet propagated by tissue culture sprouted without chilling or hot water treatment due to dormancy. For dormancy breaking, $GA_{4+7}$ hot solution treatment increasing sprouting by 55-96%, whereas distilled water or GA was not effective in sprouting. Chilling treatment for 4 weeks induced sprouting by 50-70% in L. cernuum and L. leichtlinii, whereas 8 weeks was needed for sprouting of L. hansonii and L. hansonii for. mutatum. Combined treatment of hot water and chilling treatments synergistically promoted sprouting. Especially, in L. cernuum and L. hansonii, $GA_{4+7}$ hot solution soaking prior to chilling for 4 weeks promoted sprouting by 35-45% compared with the reverse order. Enlargement of bulblets resulted from increase in fresh weight and diameter was promoted by the treatments that increased the sprouting percentage of bulblets. Only in L. cernuum, shoots emerged from bulblets soaked in hot $GA_{4+7}$ solution or chilled at $4^{\circ}C$ and shoot emergence rate was highest in bulblets soaked in hot $GA_{4+7}$ solution and then chilled for 8 weeks. From these results, the most effective method for bulblet sprouting and enlargement was to soak in hot $GA_{4+7}$ solution and then chill for 4 weeks in L. hansonii, hansonii for. mutatum, and leichtlinii, and to soak in hot $GA_{4+7}$ solution and then chill for 4 weeks in L. cernuum and tsingtauense.

A Case Study of Metastatic Cholangiocarcinoma with Sepsis who Showed Symptomatic Improvement after Treated with Handayeolso-tang, Fel Tauri, and Antibiotics (전이된 담도암 환자의 패혈증을 한다열소탕 및 우담과 항생제를 병행하여 치료한 환자 1례)

  • Objectives Despite the treatment with antibiotics, patients with sepsis has a high mortality (80%) in the underlying disease group. The aim of this study was to report the improvement of septic condition of the cholangiocarcinoma patient after the treatment with Handayeolso-tang, Fel Tauri, and antibiotics. Methods We retrospectively reviewed the medical records. The patient's subjective symptoms such as chilling and abdominal pain were evaluated by NRS and the performance status was evaluated by ECOG. This case was literally compared with relevant published studies on prognosis of sepsis. Results Despite poor prognostic factor(MEDS score 18), the patient's symptoms such as fever, chilling, abdominal pain, and diarrhea and ECOG(Eastern Cooperative Oncology Group) improved. The patient was hemodynamically stabilized on 3rd day from the treatment, and her laboratory test results were normalized on 7th day. Conclusions A female patient of metastatic cholangiocarcinoma came to the hospital for cholangitis, later causing septic shock. Both her symptoms and laboratory tests showed significant improvement after the treatment of antibiotics, Handayeolso-tang and Fel Tauri. To our knowledge, this is the first case reporting the synergistic combination of Korean oriental medicine and Western medicine approaching to sepsis.

1-Methylcyclopropene and Carbon Dioxide Absorber Reduce Chilling Injury of Eggplant ($Solanum$ $melongena$ L.) during MAP Storage (1-Methylcyclopropene과 이산화탄소 제거제 처리에 의한 가지($Solanum$ $melongena$ L.) 과실의 MAP 저장 중 저온장해 경감)

  • This study was conducted to extend the postharvest life of eggplant ($Solanum$ $melongena$ L.) by modified atmosphere packaging using a OPP bag with 1-MCP to evaluate the alleviation effects of that treatment on external chilling injury during at low temperature. 1-MCP treatment at the level of $1.0{\mu}L{\cdot}L^{-1}$ were very effective in delaying the progress of surface bronzing of eggplant during 10 days of storage at $8^{\circ}C$, but not significant at $1^{\circ}C$. Also, the treatment of 1-MCP at the levels of $0.1{\sim}1.0{\mu}L{\cdot}L^{-1}$ are beneficial to complement low temperature storage and maintain quality indices such as higher firmness and lower weight loss than untreated control even at a critical low temperature 4 to $8^{\circ}C$. The treatment of carbon dioxide using dry ice (DI, 5 g/3 fruits) significantly promoted chilling injury of eggplant stored at $4^{\circ}C$, symptoms were appeared from 3 days after treatment and combination treatment of 1-MCP did not block the development of chilling injury symptom. Meanwhile, single treatment of $CO_2$ absorber (CA, 5 g/3 fruits) or combination treatment with 1-MCP at the level of $1.0{\mu}L{\cdot}L^{-1}$ prevented effectively the external and internal chilling injury at $4^{\circ}C$ during 6 days and 15 days of MAP storage, respectively. Results suggest that 1-MCP and carbon dioxide absorber treatments delay fruit deterioration and are beneficial to broaden storage temperature of eggplant fruits.

Influence of Chilling Duration on Oxygen Consumption and Hatchability in Eggs of the Silkworm, Bombyx mori

  • The rate of oxygen consumption of the silkworm eggs was measured to set up the barometer for measuring the effect of egg chilling on diapause termination. In diapause eggs, $O_2$ uptake showed a maximum of 79.2 ${mu}ell$/mg eggs/hr, at one day after oviposition and then gradually decreased to 2.2${mu}ell$/mg eggs/hr at 9days. The rates of oxygen uptake of eggs raised immediately after HCl-treatment and reached to a maximal level of 484.5 ${mu}ell$/mg eggs/hr in 9-day-old eggs, which corresponds to 220 fold that of diapause eggs, and the hatching ability was also over 50%. In order to break diapause, eggs incubated at $25^{\circ}C$ for 30 days after oviposition had to be kept at least for 45 days at 5$^{\circ}C$, but chilled eggs for 90 days or longer hatched at 10 days as normal hatching periods. We also investigate periods which the value of $O_2$ uptake of eggs chilled during 15 to 120 days at 5$^{\circ}C$ reached at 200 ${mu}ell$/mg eggs/hr. As a result of that, $O_2$ uptake of eggs chilled during 15 to 45days were not reached at 200 ${mu}ell$/mg eggs/hr and longer the chilling durations are, the higher the rate of $O_2$ uptake is and the longer the chilling durations are, the shorter the periods up to hatching is. And also, hatch-ability increased rapidly as $O_2$ uptake reaches over 20/eggs/hr.
