Optimum Chain Extension and Change of the Average Particle Size of Aqueous Polyurethane Dispersion (수분산 폴리우레탄 제조시 최적 사슬연장 조건과 입도 변화에 관한 연구)

  • In the synthesis of water-based polyurethane using self-emulsification process, after being neutralized, polyurethane pre-polymers containing ionic pendant groups are dispersed by simple convective mixing. Preparation of dispersion is followed by chain extension reaction, which is considered as important step for growth of the molecular weight of polyurethane. In this work, pH variations in the aqueous phase were monitored during the chain extension process in the presence of water-soluble diamines. The optimum degree of chain extension and the average particle size in the polyurethane dispersions were examined with varying ionic pendent group contents, type of chain extenders, and feed rate of chain extenders, The initial pH value in the aqueous phase linearly increased and the optimum chain extension point could be obtained from the intersection of two linear lines having different slopes, All average particle sizes before chain extension reaction were almost same, however, the final average particle size increased as feed rates of chain extenders increased, In addition, as the ionic pendant group contents increased, the particle size decreased since the hydrophilicity and hydrodynamic volume increased. As carbon numbers of the chain extenders increased, the final particle size increased significantly. From the results, it was concluded that the chain extension reaction took place among the particles not only in a particle.

Treatment of Genomic DNA with Restriction Enzyme(s) Improves Amplification Efficiency by Polymerase Chain Reaction (제한효소 처리된 Genomic DNA에 의한 Polymerase Chain Reaction 증폭 효율에 관한 연구)

  • Polymerase chain reaction (PCR) is a powerful tool for precisely amplifying selected DNA sequences that have had a broad impact on genomic studies. When examining human $\alpha$- and $\beta$- tryptase genes which have 95% DNA homology, inconsistent PCR amplification of genomic sequences hampered our progress. This study suggests that long PCR technique on the original DNA digested with restriction enzymes improves both efficiency and sensitivity of PCR. These improved results seem to derived from the effective denaturation of the original genomic DNA template or reduction of formation of secondary structures that block either primer annealing or extension in PCR. Elimination of homo- or hetero-duplex products derived from highly homologous genes provides an additional advantage in this study. This communication describes how the use of restriction enzymes improved these efficiencies, and also facilitated studies of highly homologous genes including tryptase genes.

Rapid Detection of Enterotoxigenic Staphylococcus aureus by Polymerase Chain Reaction (중합효소 연쇄반응에 의한 식중독성 황색포도상구균의 신속한 검출)

  • Staphylococcal food poisoning is the major cause of bacterial food poisoning occurring in this country. Therefore government regulates commercial foods through Official Dictionary of Food that there should be free of enterotoxigenic Staphylococcus aureus in Korean rice cakes, bread, and a box lunch. Since at least 5 days are required to identify the S. aureus by the official method in the Dictionary it is difficult to prevent the food poisoning and the investigation of the outbreaks. In this report an improved determination method of the S. aureus has been developed using polymerase chain reaction (PCR) technique. Sense and antisense primers for specific amplification of genes encoding staphylococcal enterotoxins were designed and synthesized for the PCR. Rapid chromosomal DNA isolation method was also developed from S. aureus using lysostaphin. The PCR condition was developed as follows. Reaction solution $(50\;{\mu}l)$ consisted of target DNA $2\;{\mu}l$ (about 20ng), 10X buffer $5\;{\mu}l$, primer 100pmole, dNTP (10 mM) $4\;{\mu}l$ and Taq DNA polymerase 2.5 unit in a thin-wall tube. Operation condition of the PCR was 5 min pre-denaturation at $94^{\circ}C$, 15 sec denaturation at $94^{\circ}C$, 15 sec annealing at $50^{\circ}C$, 20 sec extension at $72^{\circ}C$, and 5 min post-extension at $72^{\circ}C$, and 30 cycles of denaturation-annealing- extension. Using the PCR with Perkin Elmer GeneAmp PCR system 2400, types of enterotoxigenic S. aureus could be identified from Ddok or bread in a day.

  • The cure mechanicsm and cure kinetics of diglycidyl ether of bisphenol A(DGEBA)/4,4'-methylene dianiline(MDA)/nitrile(MN, SN, GN) systems were studied by FT-IR and DSC to develop new applications in the biomedical polymer fields. The network structure of the DGEBA/MDA system was changed to the chain-extended network structure by the addition of nitriles. The reactions contributed to the chain extension were the primary amine-nitrile and hydroxyl-nitrile reactions. The chain-extended network structure could be indirectly proved by the decrement of T\ulcorner and the increment of impact strength with the increasing nitrile content. The cure rate of DGEBA/MDA/nitrile system was lower than that of DGEBA/MDA system due to the disturbance of nitrile group in the reaction of primary amine and epoxide groups.

Chain extension effects of para-phenylene diisocyanate on crystallization behavior and biodegradability of poly(lactic acid)/poly(butylene terephthalate) blends (파라-페닐렌 다이이소시아네이트의 사슬 연장이 PLA/PBT 블렌드의 결정화 거동과 생분해성에 미치는 영향)

  • Blends of poly(lactic acid) (PLA) and poly(butylene terephthalate) (PBT) were prepared by reaction extrusion with para-phenylene diisocyanate (PPDI). The crystallization behavior and biodegradability were investigated by using a differential scanning calorimeter (DSC), a wide angle X-ray diffractometer (WAXD), a contact angle goniometer, and a buffer solution containing esterase. The addition of PBT into PLA polymer matrix induced the cold crystallization of PLA phase, and the crystallization rate of PLA phase was significantly accelerated when both PBT and PPDI participated in the reaction with PLA simultaneously. But the chain extension caused by PPDI decreased the crystallinity and hydrophilicity of PLA and PBT phases. The crystallinity and hydrophilicity did not affect the biodegradability of PLA/PBT blends. However, phase separation between PLA and PBT in PLA/PBT blends increased the interfacial area exposed to the hydrolysis of enzyme, resulting in the improved degradability rate of PLA phase. In contrast, the improved interfacial adhesion between PLA and PBT matrices by the reaction with PPDI reduced the area exposed to the enzyme to decrease the degradation rate of PLA phase.

  • 1-[Ethyl (E)-5,6-dideoxy-2,3-O-isopropylidene-$\beta$-D-ribo-hept-5-enofuranosyluronate] uracil(12) was synthesized. Other various heptofuranose uncleosides were also synthesized from uridine and adenosine by two-carbon chain extension using Witting reaction.

  • Glass transition temperatures($T_g$) of poly(phenylene sulfide sulfone)[PPSS], which were heat treated at various temperature and time, were studied by DSC. Up to $230^{\circ}C$, $T_g$ of heat treated PPSS were increased with increasing the heat treated time. When the sample was heated at $260^{\circ}C$ for 30 minute and $280^{\circ}C$ for 10 minute, however, $T_g$ of those samples were decreased. These phenomena can be explained by the factors of chain extension reaction, oxidative crosslinking, and thermal crosslinking.

Incidence of Virus Diseases in Major Cultivated Areas of Watermelon and Melon in Chungbuk Province (충북지역 주산지 수박, 멜론에서의 바이러스 발생현황)

  • To investigate the incidence status of viruses in major cultivated areas of watermelon and melon in Chungbuk Province, samples were collected from 2020 to 2021 in vinyl greenhouse of Jincheon and Eumseong and examined for virus infection using reverse transcription polymerase chain reaction. Of the six viruses on watermelon that was analyzed in this study, watermelons were infected with cucumber mosaic virus (CMV), watermelon mosaic virus (WMV), cucumber green mottle mosaic virus (CGMMV), and cucurbit aphid-borne yellows virus (CABYV). The incidence rate of CMV was 20.9-35.0%, WMV 0.4-15.8%, CGMMV 1.6-38.5%, and CABYV was 3.5-3.7% from 2020 to 2021. But strangely, there were no incidence of zucchini yellow mosaic virus and cucurbit chlorotic yellows virus (CCYV) during investigation. From this result, we knew the major virus was CGMMV on watermelon in Chungbuk Province. Molecular diagnosis assays of the two melon viruses, showed that melons were infected with CABYV and CCYV from 2020 to 2021. The incidence rate of CABYV was 53.9-92.2% and CCYV was 2.7-20.8%. The incidence of CABYV was high in melon cultivation of Jincheon and Eumseong, Chungbuk. Afterwards, it is necessary to establish a control management strategy for reduce the incidence of CABYV. Furthermore, we must pay attention that of CCYV even if the incidence was low.

Polymerase chain reaction for the detection of Newcastle disease virus (닭 뉴캐슬병 바이러스의 특이 검출을 위한 polymerase chain reaction 법)

  • To study the specific tools for the diagnosis of Newcastle disease virus (NDV) in chicken, polymerase chain reaction (PCR) and its presumable conditions were evaluated for the detection of hemagglutinin-neuraminidase (HN) gene of NDV RNA. For these purposes, Kyojeongwon strain of the NDV was propagated in allantoic cavity of SPF embryonating chicken eggs, and viral RNA was extracted from fractionated virus after the allantoic fluids were ultracentrifuged with sucrose gradient. The first-strand cDNA was then made for the HN gene of NDV RNA by reverse transcription at $42^{\circ}C$ for 1 hour using specific primer complementary to the HN gene. The single-stranded cDNA was used as template in the PCR of the HN-DNA, and various conditions of the PCR were evaluated to set up method for the specific detection of the HN-DNA. The PCR conditions promising for the detection of HN gene consist of preheating at $94^{\circ}C$, 5 min, 30 cycles of denaturation at $94^{\circ}C$, 1 min, annealing at $55^{\circ}C$, 1 min and polymerization at $72^{\circ}C$, 2 min, and a cycle of extension at $72^{\circ}C$, 5 min. when NDVs of allantoic fluids without fractionation were applied to the above PCR condition, the HN genes were detected effectively not only from Kyojeongwon but from other velogenic strains such as Herts and a field isolate.

Degradation Characteristics of Perfluoropolyether Lubricant for Computer Hard Disk (컴퓨터 하드디스크 윤활제로 사용되는 Perfluoropolyether의 분해거동)

  • The degradation characteristics of perfluoropolyether (PFPE) for computer hard disk drive have been investigated. Thermal degradation in PFPE started at $170\;^{\circ}C$ and it was completed at $450\;^{\circ}C$. If PFPE was contacted with wear fragment from slider made by $Al_2O_3{\cdot}TiC$, the thermal degradation was accelerated by the catalytic Lewis acid degradation. The Lewis acid degradation mainly took placed in methylene oride(fluoride) chain scission as well as methylene(fluoride) and hydroxy end chain. As a result, the degradation reaction accomplished as early as at $300\;^{\circ}C$. The photo oxidation due to UV exposure on PFPE caused the chain scission in methylene(fluoride), and end chain in PFPE without chain scission in methylene oxide(fluoride) and then the molecular weight of PFPE increased by expected secondary reactions between formed radicals in the photo oxidation.
