Assay of Human Chorionic Gonadotropin in Urine of Athletes and Evaluation of Assay Kit Performance (운동선수들의 뇨 중 Human Chorionic Gonadotropin 분석 및 분석킷트 평가)

  • Special attention has been paid to human chorionic gonadotropin (hCG) for athlete doping control because it stimulates the endogenous production of testosterone and epitestosterone without increasing the T/E ratio which is a doping indicator for the exogenous administration of testosterone. Even though the IOC banned the use of hCG, a detection method has not been decided upon since there are a variety of immunoassay kits available on the market. We evaluated three kits in terms of their performance characteristics. The assay value of the control sample varied depending on the kit, resulting in 198 mIU/ml for the MAIA kit, 172mIU/ml for the IRMA kit, and 143 mIU/ml for the MEIA kit. Considering the IOC inter-lab distribution of results(55-312 mIU/ml) using 27 different kits and the mean value (178$\pm$56 mIU/ml), all three kits are within the range of -15.8% - +5.6% of the mean value, which proves them useful for the hCG assay. The MEIA kit resulted in lower hCG values because it detects only intact hCG molecules, in contrast to the other two kits which detect intact hCG and -hCG together. However, it is suitable for screening purposes because of its advantage of being an automated system. When 123 urine samples of athletes were analyzed in 22 batches using this system, the variation of control values fell within $\pm$ 10% of the mean values, and an specimens tested negative with hCG values less than the detection limit of 2 mIU/ml.

Structural and Biochemical Analysis of 3-Dehydroquinate Dehydratase from Corynebacterium glutamicum

  • Dehydroquinate dehydratase (DHQD) catalyzes the conversion of 3-dehydroquinic acid (DHQ) into 3-dehydroshikimic acid in the mid stage of the shikimate pathway, which is essential for the biosynthesis of aromatic amino acids and folates. Here, we report two the crystal structures of type II DHQD (CgDHQD) derived from Corynebacterium glutamicum, which is a widely used industrial platform organism. We determined the structures for CgDHQDWT with the citrate at a resolution of 1.80Å and CgDHQDR19A with DHQ complexed forms at a resolution of 2.00 Å, respectively. The enzyme forms a homododecamer consisting of four trimers with three interfacial active sites. We identified the DHQ-binding site of CgDHQD and observed an unusual binding mode of citrate inhibitor in the site with a half-opened lid loop. A structural comparison of CgDHQD with a homolog derived from Streptomyces coelicolor revealed differences in the terminal regions, lid loop, and active site. Particularly, CgDHQD, including some Corynebacterium species, possesses a distinctive residue P105, which is not conserved in other DHQDs at the position near the 5-hydroxyl group of DHQ. Replacements of P105 with isoleucine and valine, conserved in other DHQDs, caused an approximately 70% decrease in the activity, but replacement of S103 with threonine (CgDHQDS103T) caused a 10% increase in the activity. Our biochemical studies revealed the importance of key residues and enzyme kinetics for wild type and CgDHQDS103T, explaining the effect of the variation. This structural and biochemical study provides valuable information for understanding the reaction efficiency that varies due to structural differences caused by the unique sequences of CgDHQD.

Effects of Follicle Stimulating Hormone and Human Chorionic Gonadotrophin on the In Vitro Maturation of Canine Oocytes

  • The present study investigated the effects of follicle stimulating hormone (FSH) and human chorionic gona-dotrophin (hCG) on the nuclear maturation of canine oocytes. Oocytes were recovered from mongrel female ovaries in various reproductive states; follicular, luteal or anestrous stage. Oocytes were cultured in sserum-free tissue culture medium (TCM)-199 supplemented with various concentrations of FSH (Exp. 1: 0, 0.5, 1.0 or 10 IU) or hCG (Exp.2:0, 0.5, 1.0 or 10 IU) or both (Exp. 3:1 IU FSH +1 IU hCG) for 72 hr to determine the effective concentration of these hormones, and to examine their combined effect. After maturation culture, oocytes were denuded in PBS containing 0.1% (w/v) hyaluronidase by gentle pipetting. The denuded oocytes were stained with $1.9\;{\mu}M$. Hoechst 33342 in glycerol and the nuclear state of oocytes was evaluated under UV light. More (p<0.05) oocytes matured to MII stage when follicular stage oocytes were supplemented with 1 IU FSH (6.2%) compared with the control, 0.1 or 10.0 IU FSH (0 to 1.2%). Significantly higher (p<0.05) maturation rate to MII stage was observed in follicular stage oocytes supplemented with 1.0 IU hCG (7.2%) compared with the control or other hCG supplemented groups (0 to 1.5%). However, the combination of FSH and hCG did not improve the nuclear maturation rate of canine oocyte (2.4 %) compared with FSH (6.2%) and hCG alone (7.2%). In conclusion, FSH or hCG alone significantly increased the maturation of canine oocytes to MII stage.

Study on the Freezing of Bovine Embryos I. Ovarian response to the administration of gonadotrophins (우 수정란의 동결보존에 관한 연구 I. 성선자극홀몬과 $PGF_{2\alpha}$의 투여에 따른 난소반응)

  • This study was carried out with 46 cows to investigate the ovarian response to the administration of PMSG, PGF2$\alpha$ and hCG, and the developmental stage of embryos recovered. Superovulation was induced by the injection of 2,000IU to 3,000IU PMSG on the days of 7-13 of the estrus cycle followed 48 hours later by the injection of 22.5mg PGF2$\alpha$. Of 29 cows treated with 3,000IU PMSG and 22.5mg PGF2$\alpha$ 18 cows were given 2,000IU hCG at the onset and 7 after artificial insemination. The results obtained were summarized as follows; 1. The number of developed follicles per cow after an injection of 2,000, 2,500, 3,000IU of PMSG and 3,000 PMSG-2,000IU hCG in combination with 22.5mg PGF2$\alpha$ were 12.6, 19.6, 21.5 and 29.3, respectively. This result indicated that the no. of developed follicles per cow was increased according to the increase of PMSG dosage and the combination with hCG injection. 2. The highest number of ovulation was 17.1 in cows treated with PMSG-hCG and the number of matured corpus luteum was increase as the dosage of PMSG was higher. 3. Ovulation rate from cows treated with 2,500IU PMSG was 71.0% and this reulst was higher than the average of ovulation rate (59.3%). 4. Average recovery rate was 36.8%(232/631), and the number of ova per cow was 5.0. 5. Of 232 recovered embryos, the number of morulae and blastocysts were 76 (32.8%) and 83 (35.8%), respectively. 6. 28.4% of total recovered embryos was abonormal morphologically.

Influences of Hormone Treatment on the Ovulation Rates, Maturation and In Vitro Fertilization of Mouse (Hormone의 처리방법이 Mouse의 배란, 핵 성숙도 및 체외수정에 미치는 영향)

  • This research was conducted to investigate the interrelationship among methods of injection of PMSG-hCG to the number of ovulated eggs, percentage of matured oocytes and in vitro fertilization using out-bred ICR mice. The results obtained are as follows, 1) The optimurn dose was 5 IU for both PMSG and hCG, while the number of ovulated eggs was 42$\pm$8, percentage of M II was 73% and in vitro fertilization rate was 81 %. 2) The optimum injection interval of PMSG-hCG was 48 hours, while the number of ovulated eggs was 48 $\pm$ 8, percentage of M II was 80% and in vitro fertilization rate was 81%. 3) The optimum time for collecting eggs was between 16 and 18 hours after hCG injection, while the numbers of ovulated eggs were 44$\pm$8, 42$\pm$7 and 43$\pm$7 in 14,16 and 18 hours after hCG injection respectively, and percentages of M II were 79 and 81 %, and in vitro fertilization rates were 81 and 80% in 16 and 18 hours after hCG injection, respectively. 4) The repeat of superovulation decreased with the number of ovulated eggs, percentage of M II and in vitro fertilization rate, than in control. But it was recovered by increasing the repeat interval.

Nuclear Maturation and In Vitro Fertilization of Mouse Eggs Recovered at Various Times after Superovulation (과배란처리후의 경과시간이 생쥐 난자의 핵성숙과 체외수정에 미치는 영향)

  • Mouse eggs recovered from oviducts at one hourly intervals between 10 and 20 hours after administration of hCG were fixed, stained and then investigated the rate of in vitro fertilization and nuclear maturation. In case of out- bred ICR mice, ovulations were occured between 11 and 13 hours after hCG injection. The stages of in vitro maturation of eggs recovered from female mice at various times after hCG injection were metaphase I, anaphase I, telophase I and metaphase II. However the majority was metaphase I(17.6 to 44.4%) and metaphase II(42.9 to 80.0%) stage. When the eggs were inseminated with epididymal spermatozoa, the fertilization rate was declined as the egg recovery time after hCG administration was delayed. That is, the proportion of eggs undergoing fertilization became higher(68.1 to 77.4%) in the eggs at 12 to 15hr after injection of hCG than those(17.5 to 56.4) at 16 to 20 hr after injection of hCG. Also, when nuclear maturation of the unfertilized eggs were observed at 8 hours after insemination, the majority was in metaphase I and metaphase II and no anaphase I and telophase I were observed.

  • In this study, we shall be concerned with computing in parallel a few of the smallest eigenvalues and their corresponding eigenvectors of the eigenvalue problem, $Ax={\lambda}Bx$, where A is symmetric, and B is symmetric positive definite. Both A and B are large and sparse. Recently iterative algorithms based on the optimization of the Rayleigh quotient have been developed, and CG scheme for the optimization of the Rayleigh quotient has been proven a very attractive and promising technique for large sparse eigenproblems for small extreme eigenvalues. As in the case of a system of linear equations, successful application of the CG scheme to eigenproblems depends also upon the preconditioning techniques. A proper choice of the preconditioner significantly improves the convergence of the CG scheme. The idea underlying the present work is a parallel computation of the Multi-Color Block SSOR preconditioning for the CG optimization of the Rayleigh quotient together with deflation techniques. Multi-Coloring is a simple technique to obatin the parallelism of order n, where n is the dimension of the matrix. Block SSOR is a symmetric preconditioner which is expected to minimize the interprocessor communication due to the blocking. We implemented the results on the CRAY-T3E with 128 nodes. The MPI(Message Passing Interface) library was adopted for the interprocessor communications. The test problems were drawn from the discretizations of partial differential equations by finite difference methods.

Adherence to Health-Related Lifestyle Behavior Recommendations and Association with Quality of Life among Cancer Survivors and Age-Matched Controls in Koreaaa

  • Background: This study was conducted to determine the relationship between lifestyle behavior and quality of life (QoL) among cancer survivors in Korea. Materials and Methods: Data for a total of 471 (173 men, 298 women) cancer survivors (CS) over 40 years old were obtained from the database of the $4^{th}$ Korean National Health and Nutrition Examination Survey (KNHANES IV). An identical number of subjects of the same age, sex, and education who had no restrictions in physical activity were randomly selected from the database and represented the control group (CG). Drinking, smoking, and exercise behavior were assessed. Results: The number of heavy drinkers was lower in CS (9.4%) than in CG (15.8%) (p<0.01); similarly, there were fewer smokers in CS (9.1%) than in CG (14.0%) (p<0.05). The percentage of individuals engaging in vigorous, moderate, and low-intensity exercise did not differ between CS (13.6%, 14.7%, and 50.0%) and CG (14.3%, 13.4%, and 49.7%, respectively). No differences in Euro QoL Questionnaire 5-Dimensional Classification (EQ-5D) scores on both drinking and smoking behaviors were noted. Compared to the non-exercisers, the low-intensity exercisers in CG ($0.91{\pm}0.10$ vs. $0.94{\pm}0.09$), vigorous-intensity exercisers in CS ($0.84{\pm}0.62$ vs. $0.91{\pm}0.11$), and low-intensity exercisers in CS ($0.82{\pm}0.22$ vs. $0.88{\pm}0.13$) scored higher on the EQ-5D. Conclusions: Although cancer survivors practiced more conscious health behavior in drinking and smoking, their engagement in exercise did not differ from that of non-cancer survivors. Since exercise engagement increases QoL in general, implementation of an educational program that promotes exercise engagement in cancer survivors may be required.

Induction and Characterization of Precocious Trimolting Silkworm, Bombyx mori (조숙 3면잠의 유발과 실용형질)

  • For fine-denier cocoon filament production, the precocious trimolting silkworms were induced by the treatments with both the imidazole compound "KK-42" hating anti-juvenile hormone activity and high temperature, and their growth, dietary efficiency, cocoon and cocoon filament qualities were compared with those of normal tetramolters as control. 1. The percentage of precocious trimolters was higher in the application of KK-42 than the treatment of high temperature. The effective concentration of KK-42 was 10$\mu\textrm{g}$/1arva. The high temperature treatment for 48 hour was more effective than 36 hour treatment, and silkworm larvae fed on mulberry leaves were more sensitive than artificial diet to induce trimolters. 2. The larval duration of the trimolters induced by KK-42 was 5.17 day shorter than that of normal tetramolters in mulberry leaves rearing, and the increasing pattern of body weight in the 4th inster larvae of trimotlers was similar to the 5th instar larvae of normal tetramolters. 3. The qualities such as cocoon weight, cocoon layer weight and cocoon layer ratio of precocious trimolter induced by KK-42 were much lower those of normal tetramolters: the cocoon weight and cocoon layer ratio were 0.78g, 14.2cg and 18.4% in mulberry leaves rearing, and 0.86 cg, 10.3cg and 12.3% in artificial diet rearing, respectively. 4. The size of cocoon and cocoon filament was smaller in the precocious trimolter, both KK-42 and high temperature, as compared with that of the normal tetramolters. 5. The efficiency of cocoon layer production of the precocious trimolters by the KK-42 was lower than that of the normal tetramolters: amount of the cocoon layer production per 1g of dry mulberry leaves ingested was 7.97cg in the precocious trimotors, while 9.20cg in the normal tetramolters.

Clomid/hMG/hCG Regimen or FSH/hMG/hCG Regimen : Differences in The Luteal Phase Serum Estradiol and Progesterone Levels Determined after In Vitro Fertilization (시험관아기 프로그램에서 과배란유도 약제의 종류에 따른 황체기 혈청 난포호르몬 및 황체호르몬의 변동에 관한 비교연구)

  • Steroid hormone profiles during luteal phase of clomiphene citrate(CC)/human menopausal gonadotropin(hMG)/human chorionic gonadotropin(hCG)-stimulated in vitro fertilization (IVF) cycles and of follicle-stimulating hormone(FSH)/hMG/hCG-stimulated IVF cycles were compared. In seventy three cycles stimulated with CC/hMG/hCG regimen, follicles were aspirated during exploratory laparotomy and yielded 7 pregnancies, and in 83 cycles stimulated with FSH/hMG/hCG regimen, follicles were aspirated by laparoscope and made 13 pregnancies. Serum estradiol($E_2$) and progesterone($P_4$) levels were determined on days 2, 5, 7, and 9 after follicle aspiration. The FSH/hMG/hCG regimen was more effective than the CC/hMG/hCG regimen in folliculogenesis, ie, ovarian stimulation, follicular phase $E_2$ peak levels, oocyte maturation, and the number of retrieved oocytes. There was no significant difference between luteal serum $P_4/E_2$ ratio of the two regimens, suggesting that secretory endometrial build-up ability for implantation may not differ each other. Several significant correlations were observed between follicular phase seum $E_2$ peak levels and luteal phase serum $E_2$ and $P_4$ levels in the FSH/hMG/hCG-stimulated cycles but any correlation was not significant in the CC/hMG/hCG-stimulated cycles, suggesting that somewhat more follicles may eventually fall in atresia even after attaining dominant stage in the CC/hMG/hCG-stimulated cycles than the FSH/hMG/hCG-stimulated cycles.
