MiR-141-3p regulates myogenic differentiation in C2C12 myoblasts via CFL2-YAP-mediated mechanotransduction

  • Skeletal myogenesis is essential to keep muscle mass and integrity, and impaired myogenesis is closely related to the etiology of muscle wasting. Recently, miR-141-3p has been shown to be induced under various conditions associated with muscle wasting, such as aging, oxidative stress, and mitochondrial dysfunction. However, the functional significance and mechanism of miR-141-3p in myogenic differentiation have not been explored to date. In this study, we investigated the roles of miR-141-3p on CFL2 expression, proliferation, and myogenic differentiation in C2C12 myoblasts. MiR-141-3p appeared to target the 3'UTR of CFL2 directly and suppressed the expression of CFL2, an essential factor for actin filament (F-actin) dynamics. Transfection of miR-141-3p mimic in myoblasts increased F-actin formation and augmented nuclear Yes-associated protein (YAP), a key component of mechanotransduction. Furthermore, miR-141-3p mimic increased myoblast proliferation and promoted cell cycle progression throughout the S and G2/M phases. Consequently, miR-141-3p mimic led to significant suppressions of myogenic factors expression, such as MyoD, MyoG, and MyHC, and hindered the myogenic differentiation of myoblasts. Thus, this study reveals the crucial role of miR-141-3p in myogenic differentiation via CFL2-YAP-mediated mechanotransduction and provides implications of miRNA-mediated myogenic regulation in skeletal muscle homeostasis.

탄소섬유판을 이용한 철근콘크리트 보의 휨 보강 성능 (Flexural Rehabilitation Performance of Reinforced Concrete Beams Strengthened with Carbon Fiber Laminate)

  • In this study, the behavior of R/C beams strengthened with carbon fiber laminate (CFL) is analyzed from the test results. CFL is attractive for this application due to its good tensile strength and low weight. Test parameters are the width and the thickness of CFL and repair of damaged specimen. The failure mode and ultimate load are analyzed from these measured data. Test results show that the peak load of specimens strengthened with CFL is increased to 1.27~2.04 times that of non-rehabilitation specimen. The wider lap width, larger amount of CFL, the larger strength is obtained. But the ductile behavior of the rehabilitated specimens is inversely proportional to the CFL thickness.

장일처리 광원의 종류 및 광조사 시간에 따른 페튜니아와 팬지의 개화 및 형태학적 반응 (Flowering and Morphological Responses of Petunia and Pansy as Influenced by Lamp Type and Lighting Period to Provide Long Days)

  • 자연단일기에 장일식물의 개화를 촉진하기 일반적으로 사용되어 온 백열등(INC)이 낮은 에너지 효율 때문에 생산이 금지되었는데, 이를 대체할 수 있는 광원 중 하나는 컴팩트 형광등(CFL)이다. 본 연구에서는 몇 가지 장일식물에 대해 9시간 단일(SD) 조건의 $20^{\circ}C$ 온실에서 INC, CFL, 그리고 이 둘의 혼합광원(INC + CFL)을 이용하여 2시간(2-h) 또는 4시간(4-h) 야파(NI)나 6시간 명기연장(DE) 처리를 한 후 개화 및 형태학적 반응을 비교하였다. 전조 광원의 적색광(R):원적색광(FR) 비율은 각각 0.60, 8.46, 0.91이었으며, PPFD는 모두 $2.3{\pm}0.3{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$로 맞추었고, 대조구로 9시간 단일처리(9-h SD)를 두었다. 처리 10주 후, 페튜니아 'Wave Purple'는 단일 하에서 전혀 개화하지 않았으며, 모든 장일처리에서는 100% 개화하였다. 개화는 INC 및 INC + CFL 하에서 FL보다 앞당겨졌으며, DE와 4-h NI가 2-h NI보다 개화를 더 촉진하였다. 페튜니아 'Single Dreams Red'는 처리 후 65일까지 모두 개화하였는데, DE와 4h-NI가 2-h NI보다 개화를 더 앞당겼고 INC와 INC + CFL에서 빨리 개화하였으며 INC 6-h DE에서 개화소요일수가 가장 짧았다. 초장은 페튜니아 두 품종 모두 개화가 빨랐던 INC 하에서 가장 길었고, INC + CFL는 중간 정도였다. 팬지 'Coiossus Yellow'와 'Delta Blue Blotch' 두 품종 모두 INC 처리가 FL보다 개화를 촉진시켰는데, 전조시간별 차이는 작게 나타났다. 줄기 신장도 INC가 촉진하였으나 광원 및 전조시간에 따른 차이는 페튜니아보다 작게 나타났다. 결국 R:FR이 낮은 INC나 INC + CFL 하에서 개화 및 줄기신장이 촉진되었으며 장일처리시간이 길수록 그 효과가 더 컸다. 결론적으로, INC의 대체 전조광원으로써 CFL도 페튜니아 및 팬지의 개화를 촉진하였고 줄기신장을 억제하였으나, 개화 촉진 효과는 INC에 비하여 작았다.

A novel F-box protein with leucine-rich repeats affects defecation frequency and daumone response in Caenorhabditis elegans

  • Targeted degradation of proteins through ubiquitin-mediated proteolysis is an important control mechanism in various cellular processes. The process of ubiquitin conjugation is achieved by three enzyme complexes, among which the ubiquitin ligase complex (E3) is in charge of substrate specificity. The SCF (SKP1-CUL1-F-box) family portrays the largest and the most characterized member of the E3 ligases. For each SCF complex, the ubiquitination target is recognized by the F-box protein subunit, which interacts with the substrate through a unique C-terminal domain. We have characterized a novel F-box protein CFL-1 that represents a single LRR-type F-box (FBXL) in the Caenorhabditis elegans genome. CFL-1 is highly homologous to FBXL20 and FBXL2 of mammals, which are known to regulate synaptic vesicle release and cell cycle, respectively. A green fluorescence protein (GFP)-reporter gene fused to the cfl-1 promoter showed restricted expression around the amphid and the anus. Modulation of CFL-1 activity by RNAi affected the time interval between defecations. RNAi-treated worms also exhibited reduced tendency to form dauer when exposed to daumone. The potential involvement of CFL-1 in the control of defecation and pheromone response adds to the ever expanding list of cellular processes controlled by ubiquitin-mediated proteolysis in C. elegans. We suggest that CFL-1, as a single LRR-type F-box protein in C. elegans, may portray a prototype gene exerting diverse functions that are allocated among multiple FBXLs in higher organisms.

탄소섬유판으로 횡보강된 콘크리트 압축부재의 보강성능 (Confined Effect of Concrete Compressive Members Strengthened with Carbon Fiber Laminate)

  • 본 연구에서는 에폭시 모르터를 접착제로 사용한 탄소섬유판 접착공법을 사용하여 철그콘크리트 구조물의 주요부재인 압축부재를 간격, 두께, 폭, 겹수 및 이음길이의 변수로 하여 횡보강한 콘크리트 압축부재의 구조적 거동에 관한 연구를 실험적으로 진행하였다. 파괴양상은 보강된 시험체 모두가 보강재인 탄소섬유판의 인장파단과 동시에 급작스러운 취성파괴로 파괴를 보여주었다. 연구결과 압축부재 시험체를 동일한 조건으로 횡보강할 경우, 시험체의 지름에 대한 길이의 비가 다르더라고 횡보강도니 시험체의 구조적 거동은 같게 나타났다. 압축부재 시험체의 보강효과는 보강재의 간격이 좁을수록 동일하중에서의 변위는 작아지고, 연성이 커지는 것으로 나타났으며, 보강재의 폭이 다르더라도 보강량과 보강겹수가 같으면 횡보강효과는 거의 같은 것으로 나타났다. 또한 횡보강 시, 보강재 두께가 증가하면 그와 비례하여 횡보강효과도 증가하는 것으로 나타났다. 보강재의 겹수가 증가할 경우 보강재 겹수 증가에 의한 보강효과 증가는 비례적이지 않을 것으로 나타났다.

Impact of Chronic Lateral Ankle Instability with Lateral Collateral Ligament Injuries on Biochemical Alterations in the Cartilage of the Subtalar and Midtarsal Joints Based on MRI T2 Mapping

  • Objective: To quantitatively assess biochemical alterations in the cartilage of the subtalar and midtarsal joints in chronic lateral ankle instability (CLAI) patients with isolated anterior talofibular ligament (ATFL) injuries and combined calcaneofibular ligament (CFL) injuries using MRI T2 mapping. Materials and Methods: This study was performed according to regulations of the Committee for Human Research at our institution, and written informed consent was obtained from all participants. Forty CLAI patients (26 with isolated ATFL injuries and 14 with combined ATFL and CFL injuries) and 25 healthy subjects were recruited for this study. All participants underwent MRI scans with T2 mapping. Patients were assessed with the American Orthopedic Foot and Ankle Society (AOFAS) rating system. The subtalar and midtarsal joints were segmented into 14 cartilage subregions. The T2 value of each subregion was measured from T2 mapping images. Data were analyzed with ANOVA, the Student's t test, and Pearson's correlation coefficient. Results: T2 values of most subregions of the subtalar joint and the calcaneal facet of the calcaneocuboid joint in CLAI patients with combined CFL injuries were higher than those in healthy controls (all p < 0.05). However, there were no significant differences in T2 values in subtalar and midtarsal joints between patients with isolated ATFL injuries and healthy controls (all p > 0.05). Moreover, T2 values of the medial talar subregions of the posterior subtalar joint in patients with combined CFL injuries showed negative correlations with the AOFAS scores (r = -0.687, p = 0.007; r = -0.609, p = 0.021, respectively). Conclusion: CLAI with combined CFL injuries can lead to cartilage degeneration in subtalar and calcaneocuboid joints, while an isolated ATFL injury might not have a significant impact on the cartilage in these joints.

FUNWAVE-TVD 수치모형을 이용한 수중천퇴를 통과하는 불규칙파의 수치모의에서 TVD 기법들에 의한 수치해 비교 (Comparison of Numerical Solutions by TVD Schemes in Simulations of Irregular Waves Propagating over a Submerged Shoal Using FUNWAVE-TVD Numerical Model)

  • 최근 개발된 FUNWAVE-TVD 파랑모형을 이용하여 적용되어 온 TVD 기법들의 수렴도와 수치적인 안정성을 비교하였다. Yamamoto and Daiguji(1993)의 minmod limiter를 사용하는 4차 정확도의 MUSCL-TVD 기법과 Erduran et al.(2005)의 van-Leer limiter를 사용하는 4차 정확도의 MUSCL-TVD 기법, Zhou et al.(2001)의 van-Leer limiter를 사용하는 2차 정확도의 MUSCL-TVD 기법을 비교하였으며, 수리실험 관측치가 제시되어 있는 Vincent and Briggs(1989)의 불규칙 파랑실험에 적용하였다. 불규칙 파랑의 비쇄파 실험 결과에서 minmod limiter를 사용하는 4차 정확도의 기법은 van-Leer limiter를 사용하는 기법이 요구하는 격자의 크기만큼 세밀한 격자를 요구하지는 않지만, 더 낮은 CFL을 사용해야 안정적인 모의가 가능하였다. 반면에 van-Leer limiter를 사용하는 기법에서는 numerical dissipation을 줄이기 위하여 보다 세밀한 격자를 필요로 하지만 비교적 높은 CFL을 사용할 수 있는 것으로 나타났다. 각 기법의 numerical dissipation의 영향을 최대한 줄이기 위하여 공간격자를 충분히 줄인 쇄파 모의 실험에서는 비쇄파 실험에 비하여 각 기법의 특성이 명확히 나타났다. Numerical dissipation이 상대적으로 작은 minmod limiter를 사용하는 기법으로 모의할 때는 격자를 충분히 줄이면 수치적인 불안정성이 나타나며 수치해가 발산하는 결과를 보였지만, van-Leer limiter를 사용하는 기법에서는 비교적 낮은 CFL을 사용하여 쇄파 모의가 완료되었으며, 관측치를 잘 재현하는 결과를 보였다.

한국 성인의 푸드 리터러시 실태 및 영양지수와의 관계 (Status of Food Literacy and Association with the Nutrition Quotient among Korean Adults)

  • Food literacy (FL) refers to the ability to understand and utilize food information. This concept consists of functional (FFL), interactive (IFL), and critical food literacy (CFL). In today's massive information-driven society, FL needs must be paid attention to as a determinant of health along with health literacy. This study sought to identify the FL of Korean adults, and to analyze the association between FL and nutritional status. Data was collected through an online self-reported survey among 1,355 Korean adults aged 19 to 64 years, stratified by gender and age. An χ2 -test, student's t-test, ANOVA, analysis of covariance (ANCOVA), and multinomial logistic regression were performed using IBM SPSS 25.0. There were significant differences in the subjects' FFL, IFL, and CFL according to their gender, age, residence area, work type, and monthly income. Furthermore, it was confirmed that subjects with a higher nutrition quotient (NQ) grade also had higher FLs. Additionally, FFL and CFL contributed to improving the NQ grade. This study is meaningful in that it investigated the FL of Korean adults and provided data to identify populations that are particularly vulnerable to a lack of FL. In addition, it was confirmed that FL can have a positive effect on the nutritional status of adults. In conclusion, this study suggests the possibility of using FL as an intervention strategy to improve nutrition.

Atomistic analysis of nano/micro biosensors

  • Dynamic analysis of nano/micro bio-sensors based on a multiscale atomistic/continuum theory is introduced. We use a generalized atomistic finite element method (GAFEM) to analyze a bio-sensor which has $3{\times}N_a{\times}N_p$ degrees of freedom, where $N_p$ is the number of representative unit cells and $N_a$ is the number of atoms per unit cell. The stiffness matrix is derived from interatomic potential between pairs of atoms. This work contains two studies: (1) the resonance analysis of nano bio-sensors with different amount of target analyte and (2) the dependence of resonance frequency on finite element mesh. We also examine the Courant-Friedrichs-Lewy (CFL) condition based on the highest resonance frequency. The CFL condition is the criterion for the time step used in the dynamic analysis by GAFEM. Our studies can be utilized to predict the performance of micro/nano bio-sensors from atomistic perspective.

Diagnostic and Active Filtering of Harmonics Generated by Compact Fluorescent Lamps

  • Use of nonlinear loads, such as power converters, fluorescent lamps and adjustable speed motor drives, is expected to grow rapidly. All of these loads inject harmonic currents. This paper presents the active filtering of the harmonic distortion generated by the compact fluorescent lamps (CFL). The Instantaneous active and reactive power theory (the p-q Theory) is used to design the control of parallel active filter. The control scheme has been verified using Matlab/Simulink with SimPower Systems through a set of simulation tests under different load conditions. Also, the tuning of the active power filter is performed to improve the quality of the electrical power supply.
