Evaluation of habitat range and physical habitat of black shinner (Pseudopungtungia nigra) from Yongdam to Daecheong Dam in Geum River (금강의 용담댐에서 대청댐까지 서식하는 감돌고기 (Pseudopungtungia nigra)의 서식범위 및 물리적 서식지 평가)

  • In this study, a fish survey was conducted to investigate the habitat status of black shinner (Pseudopungtungia nigra) designated as endangered species level from the lower part of Yongdam Dam to the upper part of Daecheong Dam. Biodiversity analysis was performed on the basis of the number of fish emerged, and biological health and physical habitat environment were quantitatively analyzed using collected P. nigra. According to the survey, the habitat range of P. nigra was found from Banguri-myeon, Buri-myeon, Geumsan-gun, downstream of Yongdam Dam. The biodiversity analysis results of all species appeared to be low in overall dominance index and relatively high in species diversity and uniformity index. In addition, P. nigra collected in this study was confirmed to have normal growth and nutritional status by the full-length-weighted relativities and condition factor. The physical habitat environment of P. nigra was 0.3 - 0.6 m in depth, 0.3 - 0.7 m/s in flow rate, and bed materials showed high frequency of occurrence in the range of cobbles (64.0 - 256.0 mm) to boulders (>256.0 mm). These results are expected to be used as data for habitat restoration and management in the future as basic data on the spatial range and preferred physical habitat environment of P. nigra in Geumgang.

  • The study on the nerve cells in the pars intercerebralis(IP) of 5-day-old cabbage butterfly Pieris rapae L. was performed to observe their ultrastructures and classify them on the basis. of the differences in size, shape and relative distribution cf cell organelles. The brain-subesophageal ganglion complex was fixed in 1% paraformaldehyde-1% gluaraldehyde mixture and embedded in araldite mixture. The transverse thin sections of IP were stained with uranyl acetate and lead citrate and examined by Hitachi 500 and ]EM 100B electron microscope. Five distinct types. of nerve cells are recognized and are arbitrarily designated as Type I, Type II Type III, Type IV and Type V. Type I neurone: These neurones are neurosecretory cells. Several neurosecretory cells are. recognized in the pars intercerebralis. They are roughly round or peach-shaped cells measuring $13{\sim}25{\mu}m$ in diameter. The rounded nucleus shows about $5{\sim}10{\mu}m$ in diameter. The chromatin is predominantly diffused with only occasional dense patches. The perikaryon contains numerous. mitochondria, free polyribosomes and neurosecretory granules. The neurosecretory granules are relatively uniform in electron density, and each one is about $100{\sim}400{\mu}m$ in diameter and surrounded by a single membrane. The granules are also observed mostly as in groups. In one group of neurones the cisternae of endoplasmic reticulum are distended or in other group of neurones are not distended. Golgi saccules are slightly dilated at their lateral extremities and contains. frequenty dense rounded materials. Type II neurone: Thes have the largest soma in the pars intercerebralis about $30{\sim}35{\mu}m$ in diameter. They also show roughly polygonal in shape. The nucleus is elongated or sickle-shaped. The chromatin is mainly in the euchromatin form. The perikarya in these cells are well populated with populated with free ribosomes and contain numerous mitochondria and Golgi bodies. The cisternae of granular endoplasmic reticulum are also well distributed. Type III neurone: They are oval or spindle-shaped and also medium-sized. neurones approximately $15{\sim}17{\mu}m$ in length. The nucleus is oval or slightly elongated in shape and $8{\sim}9{\mu}m$ in length. The chromatin occurs in diffused form. The cytoplasm contains many filamentous or oval mitochondria. The perikaryon has also numerous free polyribosomes and cisternae of granular endoplasmic reticulum. Type VI neurone: They are roughly polygonal in shape probably due to the close approximation of the adjacent cells. The soma is about $7{\sim}8{\mu}m$ in diameter. The nucleus is round or oval in shape and $5.0{\sim}5.8{\mu}m$ in diameter. The necleus also occupies a large proprion of the cell body. The perikaryon is well populated with free ribosomes and contains several mitochondria and cistenae of granular endoplasmic reticulum. Type V neurone: These neurones are similar to Type VI neurones in various respects such as cell size and cell inclusion, but they differ from Type IV neurones in shape. The soma is oval or slightly elongated. The cell body contains several filamentous and oval mitochondria.

Plant Growth-Promoting Activity of Endophytic Fungi Isolated from the Roots of Native Plants in Dokdo Islands (독도 자생식물의 뿌리로부터 분리된 내생균의 식물생장촉진 활성)

  • Endophytic fungal strains were isolated from the roots of six species plants in the Dokdo islands. Native plant samples, such as Artemisia japonica, Chenopodium album and Solanum nigrum were isolated from Dongdo, and those such as Cyrtomium falcatum, Dianthus longicalyx and Tetragonia tetragonoides were isolated from Seodo. In total, thirty two fungal strains were isolated from these native plants. To identify the fungal strains, polymerase chain reaction (PCR) amplification of internal transcribed spacer (ITS: containing ITS1, 5.8s and ITS2 region) regions was done with universal primers ITS1 and ITS4. Endophytic fungi of four species were isolated from A. japonica, eight species from C. album, three species from S. nigrum, three species from C. falcatum, three species from D. longicalyx and eleven species from T. tetragonoides. Culture filtrates (CF) of isolated endophytic fungi were used to treatwaito-c rice seedlings to test plant growth-promoting activity. As a result of bioassay, Ca-5-2-2 strain isolated from C. album expressed highest plant growth-promotion activity. Of all the endophytic fungi isolated, Penicillium sp., Fusarium sp. and Aspergillus sp. were the most abundantly distributed fungal strains in the six plants used in this study.

TREATMENT OF TOOTH DISCOLORATION ASSOCIATED WITH TRIPLE ANTIBIOTIC THERAPY: CASE REPORTS (미성숙영구치의 복합항생제 적용에 의한 변색의 처치: 증례보고)

  • The concept of revascularization of necrotic pulps regained interest and became an alternative conservative treatment option for young permanent teeth with immature roots. Revascularization of immature teeth with apical periodontitis depends mainly on disinfection of the canal. Since the infection of the root canal system is considered to be polymicrobial, a combination of drugs would be needed to treat the diverse flora. A triple antibiotic mixture of metronidazole, ciprofloxacin, and minocycline was used as an intracanal medicament. However, discoloration was developed after applying the triple antibiotic mixture. It is believed that the marked discoloration is related to the use of minocycline. The aim of this article was to present cases of coronal discoloration after triple antibiotic therapy in immature tooth and was treated with bleaching technique to control coronal discoloration. In conclusion, revascularization by using triple antibiotics promotes a paradigm shift in treating endodontically involved permanent teeth. However, we should understand that triple antibiotics containing minocycline induces tooth discoloration. Further research to prevent coronal discoloration should be investigated and suggested for the safe use of triple antibiotics.

  • Annual reproductive cycle of small yellow croaker, Larimichthys polyactis were histologically investigated based on samples captured nearby the coast around Chujado, Youngkwang and Kanghwa, South Korea from February $2004{\sim}February$,2005. By histological survey, the annual reproductive cycle was classified into the following successive stages: The primary growth stage from September to October, the second growth stage from October to December, the mature stage from January to April, the final mature, ovulation and spawning stage from May to June, the degeneration stage from June to July and the rest stage from August to September. Throughout anatomical observation of the ovary, it is found that the small yellow croaker, L. polyactis belongs to an asynchronous and multiple spawner from late spring to early summer.

Separation and Elution Behavior of Some Iron(Ⅲ)porphyrin Complexes by Reversed-Phase Liquid Chromatography (역상 액체 크로마토그래피에 의한 Iron(Ⅲ)porphyrin 착화합물들의 분리 및 용리거동에 관한 연구)

  • Some iron(III)porphyrin complexes were prepared, and identified by the spectroscopic methods. Elution behavior of iron(III)porphyrin complexes was investigated by reversed-phase HPLC. The optimum conditions for the separation of iron(III)porphyrin complexes were examined with respect to flow rate and mobile phase strength. These complexes were successfully separated on NOVA-PAK $C_{18}$ column using methanol / water(95/5) for $[T_pCF_3PP)Fe(R)]$ and methanol / water (98/2) for $[(P)Fe(C_6F_5)]$ as a mobile phase. It was found that these complexes were largely eluted in an acceptable range of capacity factor value ($0{\leq}logk'{\leq}1$). The dependence of the capacity factor (k') on the volume fraction of water in the binary mobile phase as well as the dependence of k' on the liquid-liquid extraction distribution ratio$(D_c)$ in methanol-water / n-pentadecane extraction system showed a good linearity. It means that the retention of iron(III)porphyrin complexes on NOVA-PAK $C_{18}$ column is largely due to the solvophobic effect. Also, there was a good linear dependence of the capacity factor(k') on the column temperature and enthalpy calculated by van't Hoff plot. From these results, it was confirmed that the retention mechanism of iron(III)porphyrin complexes in reversed-phase liquid chromatography was invariant under the condition of various temperature, and the solvophobic binding process exhibited isoequilibrium behavior.

Evaluation of Enteral Foods Prepared with Soybean Dietary Fiber for Patients with Diarrhea (대두 식이섬유를 이용한 특수 영양식품의 유효성 평가)

  • In the development of enteral foods for the patients with diarrhea, soybean hull, by-products of soybean processing, was used to prepare crude dietary fiber extracts (soybean hull fiber, SHF). Total dietary fiber content of SHF is 85% and their composition are 86.1% cellulose, 8.1% hemi-cellulose, and 4.7% lignin. The effects of SHF on the prevention of diarrhea were studied in animal. Spraque-Dawley (SD) rats were fed AIN93G diets containing 5% dietary fiber for 3days simultaneously inducing diarrhea with the phenolphthalein Mg citrate solution. On day 4, feces were collected at different time point. Dietary fibers used for the animal study were SHF, soybean cotyledon fiber (SCF), psyllium husk fiber (PHF), and chicory fiber (CF). ${\alpha}$-cellulose was used as a control. Body weight gain, calorie consumed and food efficiency ratio among the experimental groups were not different. However, water content in the feces of SHF group was significantly lower by 10%, compared with other groups at 24hrs. time point. This effect was even greater in the feces collected later than 24 hrs. time point. SHF seems to have a greater effects on slow the symptom of diarrhea. Based on the previous results, enteral food enriched with SHF were prepared and its effect was compared with other commercially available products from domestic or imported ones. Weight changes among experimental groups were not different, but the moisture content of feces consumed SHF enriched products were lower than that of other products. Approximately 10% decrease in water content was observed from feces collected at 24 hr time point. According to the sensory evaluation, overall acceptability of the enteral food enriched with SHF was 3.24 out of 5 indicating that taste of this product is acceptable.

  • Heterogeneity in antigenic composition of Aspergillus fumigatus isolates from clinical specimens and in antibody response of patients infected with this fungus was investigated by immunoblotting. A considerable quantitative and qualitative difference was found in composition of the culture filtrate antigens derived from a reference strain (ATCC 13073) and 8 clinical isolates of A. fumigatus on SDS-PAGE and immunoblots. The crude CF antigen of a strain AFG7 was selected to identify the serologically reactive and specific components by immunoblotting. Out of more than 36 components separated by electrophoresis, transblotted to nitrocellulose sheet, and reacted with sera that showed a positive reaction to A. fumigatus or other fungal antigens on immunodiffusion tests, merely four or so were found useful to serodiagnosis of aspergillosis. An antigen of 82KD was found most reactive and specific component so as to be contained in the standard preparation. Several other components, for example 11KD, 26KD, 30KD and 31KD, also possessed relatively high reactivity and specificity and seemed to be worth while purifying and characterizing. Antibody binding activity (reactivity) of the antigenic components was clearly shown on immunoblots because some were faintly stained with Coomassie blue but darkly stained on immunoblots, while some others behaved contrary to them. A number of components seemed to carry not only species specific but cross reactive antigenic determinants. Immunoblotting proved very useful to identify serologically reactive and specific components that should be present in the antigen to be employed to the serodiagnosis of aspergillosis.

Determination of Dietary Fiber Contents in Mushrooms (식용버섯 중 식이 섬유소의 함량 측정)

  • Recent epidemiological observations suggested beneficial effects of dietary fiber on man's health. The obsective of this study was to obtain the dietary fiber reference data of mushrooms. The dietary fiber contents of six different mushrooms (Cornellus edodes, Auricularia auriculajudae, Gyrophora esculanta, Agaricus bisporus, Pleurotus ostreatus, Collybia velutipes) were analyzed by Southgate method, modified neutral detergent fiber (NDF) method and Food Research Institute (FRI) method. Duplicate sample were used for each determination. The mean values of total dietary fiber by Southgate method, modified NDF method and FRI method were respectively $20.08\pm1.45g$/100g dry weight, $20.24\pm1.85g$/100g dry weight and $21.5\pm2.70g$/100g dry weight. The mean values of all mushroom samples by FRI method were significantly different from the mean values of the samples by modified NDF method and Southgate method. However, there was no difference in the mean values of the samples between modified NDF method and Southgate method. By Southgate method, total dietary fiber of mushrooms composed of 1.7-3.1% soluble fiber, 47.0-66.6% hemicellulose, 28.4-57.7% cellulose and 0.9-3.3% lignin. By modified NDF method, total dietary fiber of mushrooms composed of 61.8-79.1% hemicellulose, 5.4-32.9% cellulose and 4.5- l5.5% lignin. Therefore, dietary fiber contents of mushrooms were mainly hemicellulose. Our values for total dietary fiber for six mushrooms were 2~4 times higher than crude fiber in textbook.

Effect of Immersion in Water and Thermal Cycling on the Mechanical Properties of Light-cured Composite Resins (광중합형 수복용 복합레진의 기계적 성질에 미치는 수중침적과 Thermal Cycling의 영향)

  • This study was performed to investigate the effec% of immersion in water and thermal cycling on the mechanical peoperties of light cured restorative composite resins. Five commerically available light-cured composite resins(Photo Clearfil A : CA, Lite-Fil A . LF, Clearril Photo Posterior CP, Prisms AP.H.. PA, 2100 : ZH) were unto The specimens of 12 m in diameter and 0.7 m in thickness were made, and an immersion in $37^{\circ}C$ water for 7 days and a thermal cycling of 1000 cycles at 15 second dwell time each in $5^{\circ}C$ and $55^{\circ}C$ baths were performed. Biaxial flexure test was conducted using the ball-on-three-ball method at the crosshead speed of 0.5mm/min. In order to investigate the deterioration of composite resins during the thermal cycling test, Weibull analysis for the biaxial flexure strengths was done. Fracture surfaces and the surfaces before and after the thermal cycling test were examined by SEM. The highest Weibull modulus value of 10.09 after thermal cycling tests which means the lowest strength variation, was observed in the CP group, and the lowest value of 4.47 was obsered in the LF Group. Biaxial flexure strengths and Knoop hardness numbers significantly decreased due to the thermal cycling ($\textit{p}$< 0.01), however, they recovered when specimens were drie4 The highest biaxial flexure strength of 125.65MPa was observed in the ZH group after the thermal cycling test, and the lowest value of 64.86MPa was observed in the CA group. Biaxial flexure strengths of ZH and CP groups were higher than those of PA, CF, and CA groups after thermal cycling test($\textit{p}$< 0.05). Knoop hardness numbers of CP group after the thermal cycling test was the highest(95.47 $\pm$ 7.35kg/$mm^2$) among the samples, while that of CA group was the lowest(30.73 $\pm$ 2.58kg/$mm^2$). Knoop hardness numbers showed the significant differences between the CP group and others after the thermal cycling test(($\textit{p}$< 0.05). Fracture surfaces showed that the composite resin failure developed along the matrix resin and the filler/resin interface region, and the cracks propagated in the conical shape from the maximum tensile stress zone.
