The Inheritance of Jumping Activity in Reciprocal Cross of Two Subspecies of Mice

  • The objective of this study was to describe the nature of the inheritance of jumping as a behavioral trait and to analyze quantitatively the jumping height as a measure of vigor in two subspecies of mice. Two subspecies of mice, Yonakuni wild mouse (Y) and $CF_{{\sharp}1}$ laboratory mouse (C), were used as the parental types. Reciprocal mating between these two subspecies was made to produce subsequently the first and second generations. The first generation was $F_1$ (YC) resulting from $Y\;male{\times}C\;female$, and $F_1{^\prime}$ (CY) from $C\;male{\times}Y\;female$. The second generation $F_2$ (YCYC) was from mating $F_1{\times}F_1$ and $F_2{^\prime}$ (CYCY) from $F_1{^\prime}{\times}F_1{^\prime}$. Individuals were treated with a set of direct current shock apparatus at six weeks of age to evoke jumping. The results showed that the ratio between jumping and non jumping mice (J: NJ) for C was 0%:100% (0:1), which means that all C did not jump throughout the experiment, whereas Y was 68%:32% (2:1); and the $F_1$ and $F_2$ showed 65%:35% (2:1) and 51%:49% (1:1), respectively. All $F_1{^\prime}$ and $F_2{^\prime}$ individuals jumped as indicated by the ratio 100%:0% (1:0) for both these two genetic groups. Of the jumped mice, average height of the first three jumping observed for pooled sexes in Y, $F_1$, $F_2$, $F_1{^\prime}$ and $F_2{^\prime}$ were 19.3 cm, 19.3 cm, 18.0 cm, 19.9 cm and 16.4 cm, respectively. The distribution of jumping height showed a tendency to be a normal distribution. The jumping activity and jumping height may be affected by some major genes and polygenes, respectively.

The Effect of Formaldehyde Treatment of Solvent and Mechanical Extracted Cottonseed Meal on the Performance, Digestibility and Nitrogen Balance in lambs

  • The effect of formaldehyde treatment of solvent and mechanical extracted cottonseed meal on the performance, digestibility and nitrogen balance was assessed in lambs. Four total mixed rations viz., A, B, C and D containing 40% untreated and treated solvent and mechanical extracted meal were prepared. Sixteen male lambs with average BW of 20-22 kg were randomly allocated to experimental rations and were fed individually during ninety days growth trial. The treatment of solvent extracted cottonseed meal resulted in a linear decrease in ruminal protein degradation. Maximum decrease (64%) in protein degradation was observed at 4 h incubation time with 0.3% formaldehyde treatment. Highest daily BW gain was observed in lambs fed on rations Band D compared to lambs fed on rations A and C. Daily BW gain was higher on rations having 0.3% for fromaldehyde treated cottonseed meals. Higher DM digestibility was observed on ration D compared to other rations. Higher (p<0.05) CP and CF digestibility was observed on rations Band D compared to rations A and C. Nitrogen retention as % age of nitrogen intake was (p<0.05) higher for lambs fed rations B and D compared to rations A and C. Similar pattern was observed for nitrogen retention as percent of nitrogen absorbed. The present study suggested that oil extraction methods of cottonseed did not alter their meal utilization in lambs, however, formaldehyde (0.3%) treatment of meals enhanced its efficiency for growth, digestibility and nitrogen balance in lambs.

Analysis of In-Cylinder Flow Characteristics of a High Speed D.I. Diesel Engines (고속 직접분사식 디젤 엔진의 실린더내 유동 해석)

  • Recently, HSDI (High Speed Direct Injection) diesel engine has been spotlighted as a next generation engine because it has a good potential for high thermal efficiency and fuel economy. This study was carried out to investigate the in-cylinder flow characteristics generated in a 4-valve small diesel cylinder head with a tangential and helical intake port. The flow characteristics such as coefficient of flow rate(Cf), swirl ratio (Rs), and mass flow rate (ms) were measured in the steady flow test rig using the impulse swirl meter and the analysis of in-cylinder flow field was conducted by experiment using the PIV and calculation using the commercial CFD code. As the results from steady flow test indicate, the mass flow rate of the cylinder head with a short distance between the two intake ports is increased over 13% than that of the other head. However, the non-dimensional swirl ratio is decreased approximately 15%. From in-cylinder flow characteristics obtained by PIV and CFD calculation, we found that the swirl center was eccentric from the cylinder center and the velocity distribution became uniform near the TDC. In addition, the results of the calculation are good agreement with the experimental results.

Effect of Replacing Til Oil Cake by Poultry Excreta on Growth and Nutrient Utilization in Growing Bull Calves

  • An experiment was conducted for 90 days using 9 growing bull calves (initial LW 71.5 kg) to investigate the effect of replacing til oil cake by poultry excreta on growth performance and nutrient utilization. The animals were randomly divided into three groups. The control group A was fed with conventional concentrate mixture containing til oil cake, rice bran, wheat bran, bone meal and common salt and the groups B and C were offered diets in which 50 and 100 percent of til oil cake of diet A were replaced by dried poultry excreta. All the animals were fed urea soaked rice straw ad libitum and concentrate mixture was given at the rate of 10 g per kg LW. Towards the end of growth trial a conventional digestibility trial was conducted. Average daily live weight gain was 216, 211 and 188 g for animals fed diets A, B and C, respectively. Average daily dry matter intake in groups A, B and C was 3.42, 3.37 and 3.30 kg per 100 kg LW, respectively. The daily live weight gain and dry matter intake did not differ significantly (p > 0.05) among the dietary groups. The digestibility coefficient for DM or NFE was almost similar but that for OM, CP, CF and EE was significantly different (p < 0.01) among the dietary groups. TDN percent in diets A, B and C was 57.3 53.3 and 50.8, respectively and the difference was significant (p < 0.01). Animals in all the groups were in a state of positive nitrogen balance. The results indicated that til oil cake can be replaced by dried poultry excreta in bull calf ration.

Amine and Olefin Complexes of Pt(II) Having a PCP-Pincer Ligand

  • $Pt(2,6-(Cy_2PCH_2)_2C_6H_3)(OTf)\;(OTf=CF_3SO_3^-)$ readily reacts with various amines to afford cationic amine complexes $[Pt(2,6-(Cy_2PCH_2)_2C_6H_3)(amine)](OTf)\;(amine=NH_3,\;NHMe_2,\;NHC_4H_8,\;NH_2Ph,\;NH_2(Tol-p))$ in high yields. These complexes have been fully characterized by IR, $^1H-,\;^{19}F{^1H}-,\;and\;^{31}P{^1H}-NMR$ spectroscopy, and elemental analyses. Reaction of $Pt(2,6-(Cy_2PCH_2)_2C_6H_3)(OTf)$ with acrylonitrile quantitatively produced the ${\pi}$-olefinic complex $Pt(2,6-(Cy_2PCH_2)_2C_6H_3)(CH_2=CHCN)](OTf)$ which is only stable in solution in the presence of acrylonitrile. Attempt at isolating this complex in the pure solid state was failed due to partial decomposition into $Pt(2,6-(Cy_2PCH_2)_2C_6H_3)(OTf)$ The equilibrium constants $(K_{eq}=[Pt(PCP)-(NH_2R)^+][CH_2=CHCN]/[Pt(PCP)(CH_2=CHCN)^+][NH_2R]:\;[Pt(2,6-(Cy_2PCH_2)_2C_6H_3)(CH_2=CHCN)]^++NH_2R{\rightleftarrows}[Pt(2,6-(Cy_2PCH_2)_2C_6H_3)(NH_2R)]^++CH_2=CHCN=Ph,\;p-tolyl)$ were calculated to be 0.28 (for R = Ph) and 3.1 (R = p-tolyl) at $21^{\circ}C$. The relative stability of the ${\sigma}$-donor amine versus the ${\pi}$-olefinic acrylonitrile complex has been found largely dependent upon the amine-basicity $(pK_b)$, implicating that acrylonitrile practically competes with amine in the platinum coordination sphere. On the contrary to the formation of the acrylonitrile complex, no reaction of $Pt(2,6-(Cy_2PCH_2)_2C_6H_3)(OTf)$ with other olefins such as ethylene, styrene and methyl acrylate was observed.

Protoplast Fusion of Alkaline Protease Producing Bacillus subtilis (Alkaline Protease를 생산하는 Bacillus subtilis의 원형질체 융합)

  • To improve alkaline protease producing strain by protoplast fusion, a strain of Bacillus sp. was treated with $100{\mu}g/ml$ NTG (N-methyl-N'-itro-N-nitrosoguanidine) for 45 minutes and mutants of Bacillus subtilts $Arg^-,\;Try^-,\;His^-$ and $Ade^-$ were isolated. The frequency of protoplast formation was about 99%, when teas of exponential phase were treated with $200{\mu}g/ml$ lysozyme at $42^{\circ}C$ for $10{\sim}30$ minutes. In a regeneration medium containing 0.3M sodium succinate, 2.0% polyvinylpyrrolidone, 0.5% casamino acid, 10mM $MgCI_2$ and 20mM $CaCI_2$, regeneration frequency cf the isolated Bacillus subtilis strains was 25.2%. The fusion frequency between mutant was from $2.1{\times}10^{-5}$ to $8.1{\times}10^{-5}$ under optimum condition.

Synthesis of Diazacrown Ethers Containing Phenolic Side Arms and Their Complex with Divalent Metal Ions

  • The aminomethylation of phenols with para-substituents by the Mannich reaction has successfully been accomplished to produce the Mannich bases 2-6. The compounds 7-8 have also been synthesized in order to identify the effect of the side arms and t he macrocycle in the complex formation. Protonation constants and stability constants of the double armed diaza-18-crown-6 ethers 2-7 with metal ions have been determined by potentiometric method at 25 $^{\circ}C$ in 95 % methanol solution. Under a basic condition (pH > 8.0), the double-armed crown ethers 2-6 revealed stronger interaction with divalent metal ions than the simple diazacrown ether 1. The stability constants with these metal ions were Co 2+ < Ni2+ < Cu2+ > Zn 2+ in increasing order, which are in accordance with the order of the Williams-Irving series. The stability constants with alkali earth metal ions were Ca 2+ < Sr 2+ < Ba 2+ in increasing order, which may be explained by the concept of size effect. It is noteworthy that the hosts 2-6, which have phenolic side arms and a macrocycle, bind stronger with metal ions than the hosts 1 and 7. On the other hand, the host 8, which has phenolic side arms with a pyperazine ring,provided comparable stability constants to those with the host 3. These facts demonstrate that phenolic side arms play a more important role than the azacrown ether ring in the process of making a complex with metal ions especially in a basic condition. In particular, the log KML values for complexation of divalent metal ions with the hosts 2-6 had the sequence, i.e., 2 (R=OCH3) < 3 (R=CH3) < 4 (R=H) < 5 (R=Cl) < 6 (R=CF3). The stability constants of the hosts 5 and 6 containing an electron-withdrawing group are larger than those of the hosts 2 and 3 containing an electron-donating group. This substituent effect is attributed to the solvent effect in which the aryl oxide with an electron-donating group has a tendency to be tied strongly with protic solvents.

A Laboratory Test for Determining the Qulity of Imported pasture Plant and Forage Crop Seeds (수입목초 및 사료작물 종자의 품질에 관한 조사연구)

  • A series of tests was carried out to determine the purity and germination of pasture plant and forage crop seeds imported in 1986 and 1987 at the grassland and forage laboratory, College of Agriculture, Seoul National University, Suweon. At the same time, the results obtained were compared with the National Livestook Cooperatives Federation(NLCF1's seed standards as a guide for determining seed quality. 1. The germination percentages of Potomac orchardgrass, Fawn tall fescue, Aquila Ky bluegrass and California ladino clover among grasses and legumes tested were lower than those of the N1,CF's standards, but the germination percentages of forage crops were higher than those of the NLCF's standards. 2. The purity of Potomac orchardgrass and Aquila Ky bluegrass were relatively lower than those of the NLCF's standards. 3. The percentages of other crop seeds in Potomac orchardgrass, Fawn tall fescue, Aquila Ky bluegrass and Cayuse oats were lower than those of the NLCF's standards. 4. Percentage of inert matter in the seeds was lower compared with the NLCF's standards except Akela ' forage rape. 5. The seeds of Potomac orchardgrass, NK 367 sorghum hybrid and sorghum x sudangrass hybrids such as NC+855, G83F and TE Haygrazer were contaminated with one or more weed seeds. Rumex spp. was by far the most common weed in the seeds imported. The other weeds occurred at a low level were Rumex acestosella, Senecio jacobaea, Cirsium arvense and Cuscicta spp. 6. There were many variations in 1,000 seed weight among different species. A thousand seed weight of Aquila Ky bluegrass was 0.263g, but that of Tetrone Italian ryegrass, Akela forage rape and Pioneer 3160 sorghum x sudangrass hybrid was 3.875, 3.476 and 242.4578, respectively. 7. According to the fluorescence test of ryegrass seed, Tetrone Italian ryegrass was known as a true annual type, but an unknown cultivar produced in Cheju was contaminated with 6% of perennial ryegrass seed.

재래누룩에서 분리한 곰팡이를 이용한 탁주의 성분분석

  • 쌀과 같은 곡류는 전분질을 당분으로 전환시켜 술을 제조하여야 하므로 미생물이 생성하는 효소가 필요한데 그 효소원이 누룩이며 누룩은 주류의 품질이나 생산량에 영향을 미치는 가장 중요한 요소라고 할 수 있다. 본 연구는 전국44개 지역의 누룩에서 순수 분리된 89종의 곰팡이 중 효소역가와 당화력이 비교적 뛰어난 10종(Aspergillus sp. SH-412 Heunghae, Aspergillus sp. SH-422 Ulsan, Rhizopis sp. SH-606 Imdeok, Aspergillus sp. SH-607 Yhesan, Aspergilus sp. SH-613 Wolseong, Rhizopus sp. SH-654 Uncheon, Aspergillus sp. SH-660 Jeonkok, Aspergillus sp. SH-667 Dongseong, Aspergillus sp. SH-669 Uncheon, Aspergillus sp. SH-696 Daecheon)과 대조균주로 Aspergillus kawachii CF1002 그리고 효모 Saccharomyces cerevisiae를 사용하셨으며 탁주제조 중 균주별 술덧의 산도, 환원당, 아미노산도와 제조된 완성주의 유기산, 유리아미노산, 유리당, 휴젤유 및 색도를 HPLC, GC Mass, 색차계로 측정한 결과 다음과 같은 결론을 얻었다. 균주별 술덧의 주정도는 모든 실험구에서 Aspergillus kawachii보다 높았으며 특히 Aspergillus sp. SH-422는 14.9%로 가장 수율이 좋았다. 환원당은 Aspergillus sp. SH-613이 0.49%로 가장 높았고 Aspergillus sp. SH-422는 0.37%로 가장 낮았으며 산도는 시간의 경과에 따라 감소했으며 술덧 발효중 아미노산도는 시간의 경과에 따라 약간 상승 후 다소 낮아지는 경향을 보였다. 각 균주별 술덧의 유기산은 tataric acid, malic acid, succinic acid, lactic acid, acetic acid가 검출되었으며 lactic acid 함량이 2.0∼3.2g/100ml로 가장 많이 검출되었다. 유기산은 Aspergillus sp. SH-669가 가장 높게 확인되었으며 Aspergillus sp. SH-607이 가장 낮게 분석되었다. 술덧의 주 아미노산은 histidine, alanine, glutamic acid, leucine, tryptophan순으로 검출되었으며 특히 alanine은 주류에 단맛을 주는 성분으로 모든 실험구에서 많은 양이 검출되었다. 각 균주별 술덧의 유리당은 glucose가 가장 많이 검출되었으며 Fusel oil은 iso-butyl alcohol과 iso-pentyl alcohol 이 가장 많이 검출되었다. 한편 균주별 술덧의 색차를 측정한 결과 L값은 63.33∼41.98, a값은 0.09∼-3.47, b값은 17.41∼4.90으로 나타났다.

SOUND PRODUCTION AND BEHAVIOUR OF CRABS, PORTUNUS TRITUBERCULATUS(MIERS) (II) (꽃게의 소리와 행동 (II))

  • For the development of acoustic fishing method, we had been researched a fundamental study which concerned on the sound production and behaviour of crabs was conducted. For specimen crabs such as Portunus trituberculatus and Charybdis japonica were selected. Croaking noise were recorded by the Cassette-recorder (Sony model CF-1600) through the under water monitor microphone, and analyzed in frequencies by Octave band analyzer, Rion SA-55, and sound pressure level of source by sound level meter, SM-5844. The following are the results obtained from the present investigations : When sound production of crabs (Portunus trituberculatus( female ) : carapace width $12.6\~15$cm) were attracted to another crabs in the water of anechoic aquarium, efficacy of phonotaxis was $84\~100\%$ and velocity by phonotaxis was $6.5\~7.2cm/sec$. The time required for copulation ranged from 90 minute to 95 minutes by Charybdis japonica, at that time there was no sound production with their copulation.
