Systemic lupus erythematosus (SLE) is the prototypic systemic autoimmune disease characterized by production of autoantibodies to various nuclear antigens and overexpression of genes regulated by IFN-I called IFN signature. Genetic studies on SLE patients and mutational analyses of mouse models demonstrate crucial roles of nucleic acid (NA) sensors in development of SLE. Although NA sensors are involved in induction of antimicrobial immune responses by recognizing microbial NAs, recognition of self NAs by NA sensors induces production of autoantibodies to NAs in B cells and production of IFN-I in plasmacytoid dendritic cells. Among various NA sensors, the endosomal RNA sensor TLR7 plays an essential role in development of SLE at least in mouse models. CD72 is an inhibitory B cell co-receptor containing an immunoreceptor tyrosine-based inhibition motif (ITIM) in the cytoplasmic region and a C-type lectin like-domain (CTLD) in the extracellular region. CD72 is known to regulate development of SLE because CD72 polymorphisms associate with SLE in both human and mice and CD72-/- mice develop relatively severe lupus-like disease. CD72 specifically recognizes the RNA-containing endogenous TLR7 ligand Sm/RNP by its extracellular CTLD, and inhibits B cell responses to Sm/RNP by ITIM-mediated signal inhibition. These findings indicate that CD72 inhibits development of SLE by suppressing TLR7-dependent B cell response to self NAs. CD72 is thus involved in discrimination of self-NAs from microbial NAs by specifically suppressing autoimmune responses to self-NAs.
Ssu72, a dual-specificity protein phosphatase, not only participates in transcription biogenesis, but also affects pathophysiological functions in a tissue-specific manner. Recently, it has been shown that Ssu72 is required for T cell differentiation and function by controlling multiple immune receptor-mediated signals, including TCR and several cytokine receptor signaling pathways. Ssu72 deficiency in T cells is associated with impaired fine-tuning of receptor-mediated signaling and a defect in CD4+ T cell homeostasis, resulting in immune-mediated diseases. However, the mechanism by which Ssu72 in T cells integrates the pathophysiology of multiple immune-mediated diseases is still poorly elucidated. In this review, we will focus on the immunoregulatory mechanism of Ssu72 phosphatase in CD4+ T cell differentiation, activation, and phenotypic function. We will also discuss the current understanding of the correlation between Ssu72 in T cells and pathological functions which suggests that Ssu72 might be a therapeutic target in autoimmune disorders and other diseases.
본 연구의 목적은 $CdCl_2$이 간 미세조직에 일으키는 독성에 대한 50의 억제효과를 연구하는데 있다. 본 실험에서는 총 40마리의 건강한 ICR계 mouse를 사용하였다. 실험군은 먼저 2개군(group A, and B)으로 구분하였다. A군에는 $CdCl_2$(40mg/kg)을 복강투여 하였다. B군에는 SQ (180 mg/kg, 2회/일)과 $CdCl_2$ (4.0 mg/kg)을 동시 복강 투여하였다. 각 군은 24, 48, 72, 96시간째에 관찰하여 다음과 같은 결과를 얻었다. 1. A군 : 핵막은 24시간대에서 매우 불규칙하게 관찰되었고, 48시간대부터는 대체로 원형을 유지하였다. 사립체는 72시간까지 내강이 파괴된 채로 관찰되었고, 96시간대에는 일부에서만 내강 파괴가 관찰되었다. 과립형질 내세망은 내강이 전 시간대에 걸쳐 팽대된 소견을 보였고, 72시간대부터 층판구조를 형성하였다. 용해소체가 24 시간대부터 세포질에서 관찰되었고, 72시간대부터는 글리코겐이 관찰되었다. 2. B군 : 핵막은 전 시간대에 걸쳐 비교적 규칙적인 형태를 보였다. 사립체는 전반적으로 원형 ,세장형 등 정상적인 형태로 관찰되었다. 과립형질내세망은 48시간군에서 내강이 팽대되었고 층판구조가 일부 분절된 소견을 보였지만, 72시간부터는 전형적인 층판구조가 관찰되었다. 용해소체와 glycogen은 24시간대부터 관찰되었다. 이상의 결과를 종합하면 50이 mouse간세포에 미치는 카드뮴의 독성을 감소시키는 것으로 사료된다.

Objectives: Chronic diseases (CDs) continue to increase due to advances in medicine and increase in lifespan, affecting quality of life and resulting in economic loss through treatment costs. This study addressed the perception characteristics of the effectiveness of Korean medicine (KM) based on the existence of a CD to determine the national preference for KM treatment. Methods: Using data from the 2017 National Survey of KM Usage, we evaluated the perception on treatment effect of 16 diseases by dividing them into the CD group and the non-CD group. Response reliability was verified by applying the chi-square test (χ2-test) analysis method of the Statistical Package for the Social Sciences (SPSS) statistical program. Results: The analysis of the perception on effectiveness of KM for patients with CD (n = 1,050, 21.0%) and for patients with non-CD (n = 3,950, 79.0%) showed an overall similar trend for all the 16 diseases. The response rates of having some treatment effect were high for nine diseases namely, disc-related disease (CD: 70.7%, non-CD: 73.1%), osteoarthritis (CD: 72.3%, non-CD: 72.4%), frozen shoulder and shoulder pain (CD: 79.6%, non-CD: 81.4%), back pain (CD: 84.6%, non-CD: 85.0%), sprain (CD: 84.8%, non-CD: 84.1%), facial nerve paralysis (CD: 73.5%, non-CD: 71.7%), stroke (CD: 66.2%, non-CD: 62.8%), digestive disease (CD: 53.3%, non-CD: 50.0%), and common cold and rhinitis (CD: 44.7%, non-CD: 44.8%). Conclusion: The present results found that there was little difference in the perception on effectiveness of KM for each of the 16 diseases in patients with CD and non-CD, however, 70% or more of the respondents recognized some treatment effect on musculoskeletal disorders, regardless of the existence of CD. Preferential political support for KM treatment of chronic musculoskeletal disorders is recommended.
이 연구는 고령자의 근육수축양식 및 손상정도에 따른 염증물질, 면역적격세포 및 골수유래 전구세포의 변화를 검증하는데 그 목적이 있다. 연구대상은 최근 6개월간 저항성운동 프로그램을 실시하지 않은 65세 이상의 고령자 20명을 신장성운동 집단(ECC, n=10)과 단축성운동 집단(CON, n=10)으로 분류하였다. 모든 피험자는 팔꿈치 굽힘근의 1차 신장성운동(ECC 1) 및 단축성운동(CON)을 실시하였고, ECC는 4주후에 재차 2차 신장성운동(ECC 2)을 6회 10세트, 세트 사이에 2분의 휴식시간을 적용하여 실시하였다. 골격근손상 지표(ROM, VAS, CK), 염증물질(TNF-α, IL-1, IL-6), 면역적격세포(CD3+, CD4+, CD8+, CD19+), 골수유래 전구세포(CD34+) 및 백혈구수는 운동전, 운동 직후, 운동 시기의 2, 24, 48, 72 및 96시간 후에 각각 측정하였다. 주요결과는 다음과 같다. 고령자의 골격근손상 지표(ROM, VAS)는 CON(근육수축양식) 및 ECC 2(손상정도)와 각각 비교해서 ECC 1에서 손상이 크게 증가하였다(p<.05). IL-6 및 TNF-α는 CON 및 ECC 2와 각각 비교해서 ECC 1의 24, 48 및 72시간 후에 유의하게 높게 나타났다(p<.05). 호중구수는 CON 및 ECC 2와 각각 비교해서 ECC 1의 2시간 후에 유의하게 높게 나타났다(p<.05). 이 연구에서는 고령자에서도 근육수축양식은 단축성운동보다 신장성운동에서, 또한 손상정도는 2차 신장성운동보다 1차 신장성운동에서 보다 크게 손상이 발생하였다는 사실을 확인하였다. 특히 IL-6와 TNF-α 및 호중구수는 근손상 정도와 밀접한 관련성이 있다는 것이 시사되었다.

Ischemic and traumatic brain injuries are the major acute central nervous system disorders that need to be adequately diagnosed and treated. To find biomarkers for these acute brain injuries, plasma levels of some specialized pro-resolving mediators (SPMs, i.e., lipoxin A4 [LXA4], resolvin [Rv] E1, RvE2, RvD1 and RvD2), CD59 and interleukin (IL)-6 were measured at 0, 6, 24, 72, and 168 h after global cerebral ischemic (GCI) and traumatic brain injuries (TBI) in rats. Plasma LXA4 levels tended to increase at 24 and 72 h after GCI. Plasma RvE1, RvE2, RvD1, and RvD2 levels showed a biphasic response to GCI; a significant decrease at 6 h with a return to the levels of the sham group at 24 h, and again a decrease at 72 h. Plasma CD59 levels increased at 6 and 24 h post-GCI, and returned to basal levels at 72 h post-GCI. For TBI, plasma LXA4 levels tended to decrease, while RvE1, RvE2, RvD1, and RvD2 showed barely significant changes. Plasma IL-6 levels were significantly increased after GCI and TBI, but with different time courses. These results show that plasma LXA4, RvE1, RvE2, RvD1, RvD2, and CD59 levels display differential responses to GCI and TBI, and need to be evaluated for their usefulness as biomarkers.
본 연구는 효율적인 CD-ROM 네트워크 시스템 설계 및 운영을 위해 고려해야 할 요인들을 도출시켜 시스템 설계자 및 운영자에게 도움이 되는 지침을 제공하는데 목적을 두고, 국내 72개 대학도서관의 CD-ROM 네트워크 시스템 현황과 문제점을 파악하였으며, 3개 대학 64명의 실제 이용자들을 대상으로 시스템 이용행태와 시스템에 대한 만족도를 분석하였다. 또한 이용자 만족도와 시스템 운영정책, 시스템 자원, 시스템 성능 요인간의 영향관계를 밝히기 위해 $\chi^2(Chi-Square)$검증과 F검증, 회귀분석을 실시하여 이용자 만족도에 영향을 미치는 요인들을 분석하였다.
Man-made cadmium (Cd) emissions can be transported between environmental matrices and the food chain. Food is the primary source of Cd exposure among general population as a consequence of the bioconcentration of Cd from soil. Chronic Cd exposure has been reported to be associated with chronic kidney disease, osteoporosis, diabetes, cardiovascular disease and cancer. The Joint FAO/WHO Expert Committee on Food Additives (JECFA) established the safe level of Cd intake as provisional tolerable monthly intake (PTMI) of $25{\mu}g/kg\;bw$ in 2010. The major food groups that contribute to the most Cd exposure are rice and grains, shellfish and sea food, meat including edible offal, and vegetables. A number of studies reported the high Cd contaminated levels in foods from polluted areas in Thailand. The results are of high concern since the contaminations occur in foods that are major Cd contributors. Thus, in this review, the current situations of Cd contaminated foods in polluted areas of Thailand are summarized. In addition, the Cd intakes from selected scenarios are estimated to assess the potential health risk to consumers and the suggestions are also included.
