Influence of Nitrate on Growth, Chlorophyll Content, Content and Activity of Rubisco and Rubisco Activase of Tobacco Plant Treated with Cadmium in vitro (Cadmium을 처리하여 기내 배양한 담배 식물의 생장, 엽록소 함량 및 rubisco와 rubisco activase의 함량과 활성에 미치는 질산염의 영향)

  • Influence of nitrate on growth, chlorophyll content, content and activity of rubisco and rubisco activase of tobacco plant cultured on MS medium treated with cadmium in vitro was studied. In vitro growth and chlorophyll content reduced at 0.2 mM Cd was recovered by nitrate and this recovery was most significant at 80 mM nitrate. Rubisco content at 80 mM nitrate was more increased compared to that at other concentrations. A similar change was also shown in rubisco activity. These resultsindicate that the activation and induction of rubisco reduced by Cd were recovered by nitrate. The degree of intensity of 55 and 15 kD polypeptides identified as the large and small subunits of rubisco by SDS-PAGE analysis at 80 mM nitrate was significantly higher than that at other concentrations. The content and activity of rubisco activase at 80 mM nitrate was significantly increased than that at other concentrations. These data suggest that the recovery effects of rubisco by nitrate may be associated with rubisco activase.

Effects of Dietary ${\beta}$-Cyclodextrin on Plasma Lipid and Tissue Cholesterol Content in Swine

  • This study examined the effects of dietary ${\beta}$-cyclodextrin (${\beta}CD$) on the cholesterol of blood and tissues of swine. Thirty six male castrated swine ($Landrace{\times}Yolkshire{\times}Duroc$) weighing 50 kg were randomly assigned to one of four dietary groups until their weight reached 110 kg. The groups were: basal diet without ${\beta}CD$ (control) and basal diets containing 1.5%, 3.0%, or 5.0% ${\beta}CD$. Diets and water were offered ad libitum. No significant difference was found between treatments in terms of feeding performance measured by daily intake, daily weight gain, and feed efficiency. Addition of ${\beta}CD$ to the diets significantly reduced total lipid, triglyceride and total cholesterol levels in swine blood, particularly in the group receiving 5.0% ${\beta}CD$, which showed decreases (p<0.05) of 21.9%, 55.6% and 27.7%, respectively. Cholesterol levels in back fat, loin, belly and ham portions of swine fed ${\beta}CD$ significantly differed (p<0.05) from controls, especially in the 5.0% ${\beta}CD$-fed group, with reductions of 26.0%, 27.5%, 17.9% and 18.3%, respectively. These results suggested that the addition of ${\beta}CD$ to the diet of swine could reduce their body cholesterol by decreasing the migration of cholesterol through the blood.

Dynamic lipopolysaccharide transfer cascade to TLR4/MD2 complex via LBP and CD14

  • Toll-like receptor 4 (TLR4) together with MD2, one of the key pattern recognition receptors for a pathogen-associated molecular pattern, activates innate immunity by recognizing lipopolysaccharide (LPS) of Gram-negative bacteria. Although LBP and CD14 catalyze LPS transfer to the TLR4/MD2 complex, the detail mechanisms underlying this dynamic LPS transfer remain elusive. Using negative-stain electron microscopy, we visualized the dynamic intermediate complexes during LPS transfer-LBP/LPS micelles and ternary CD14/LBP/LPS micelle complexes. We also reconstituted the entire cascade of LPS transfer to TLR4/MD2 in a total internal reflection fluorescence (TIRF) microscope for a single molecule fluorescence analysis. These analyses reveal longitudinal LBP binding to the surface of LPS micelles and multi-round binding/unbinding of CD14 to single LBP/LPS micelles via key charged residues on LBP and CD14. Finally, we reveal that a single LPS molecule bound to CD14 is transferred to TLR4/MD2 in a TLR4-dependent manner. These discoveries, which clarify the molecular mechanism of dynamic LPS transfer to TLR4/MD2 via LBP and CD14, provide novel insights into the initiation of innate immune responses.

Heat-treatment Effects of CdS Thin Films Fabricated by Thermal Evaporation Method (진공증착법으로 제작한 CdS 박막의 열처리효과)

  • 열증착법에 의해서 온도 85$^{\circ}C$인 glass기판 위에 CdS 박막을 제작하였다. 두께가 200nm정도로 측정된 CdS박막은 공기 중에서 온도 25$0^{\circ}C$-55$0^{\circ}C$범위에서 각각 30분간 열처리 되었으며 이들 시료에 대하여 4-point probe, XRD, SEM, UV-Spectrophotometer 및 광발광 측정으로 전기적 광학적 특성을 조사하였다. 이들의 일련의 실험값은 열처리 온도 37$0^{\circ}C$ 근처에서 구조의 변화를 보여주었는데, 열처리 온도에 대한 비저항아니 XRD, SEM 의 측정은 cubic로부터 hexagonal구조로의 변환을 나타내었다. 특히 상온에서 측정한 광발광에서 green edge emission(GEE)피이크가 2.42eV를 나타내었는데 이 때의 발광 중심은 열처리할 때 생긴 S-vacancy에 보상된 산소로 이루어진 'CdO'의 악셉터준위에 기인하는 것으로 해석되며 그 이온화 에너지는 약 0.16eV이었다.

Influence of Detailed Structure and Curvature of Woven Fabric on the Luminescence Effect of Wearable Optical Fiber Fabric (직물의 세부 구조 및 굴곡이 웨어러블 광섬유의 발광 효과에 미치는 영향)

  • The two main requirements of wearable optical fiber fabrics are that they must presuppose a high degree of flexibility and they must maintain the luminance effect in both flat and bent conformations. Therefore, woven optical fiber fabrics that satisfy the above conditions were developed by both weaving and by using computer embroidery. First, we measured the brightness of the wearable optical fiber fabric in the flat state at a total of 10 measurement points at intervals of 1 cm. Second, the wearable optical fiber fabric was placed horizontally on the forearm, where three-dimensional bending occurs, and the luminance values were recorded at the same 10 measurement points. For the woven fabric in the flat state, the maximum, minimum, average, and standard deviation luminance values were $5.23cd/m^2$, $2.74cd/m^2$, $3.56cd/m^2$, and $1.11cd/m^2$, respectively. The corresponding luminance values from the bent forearm were $7.92cd/m^2$ (maximum), $2.37cd/m^2$ (minimum), $4.42cd/m^2$ (average), and $2.16cd/m^2$ (standard deviation). In the case of the computer-embroidered fabric, the maximum, minimum, average, and standard deviation luminance values in the flat state were $7.56cd/m^2$, $3.84cd/m^2$, $5.13cd/m^2$, and $1.04cd/m^2$, respectively, and in the bent forearm state were $9.6cd/m^2$, $3.63cd/m^2$, $6.13cd/m^2$, and $2.26cd/m^2$, respectively. Therefore, the computer-embroidered fabric exhibited a higher luminous effect than the woven fabric because the detailed structure reduced light-loss due to the backside fabric. In both types of wearable optical fiber fabric the luminance at the forearm was 124% and 119%, respectively, and the light emitting effect of the optical fiber fabric was maintained even when bent by the human body. This is consistent with the principle of Huygens, which defines the wave theory of light, and also the Huygens-Fresnel-Kirchhoff principle, which states that the intensity of light increases according to the magnitude of the angle of propagation of the light wavefront (${\theta}$).

Inhibitory Effect of a Decoction of Eucommiae ulmides OLIVER and Dipsacus asperoides C. Y. Cheng et T.M.Ai on Collagen II-induced Arthritis Mice (Collagen II-induced Arthritis 생쥐에 대한 두충(杜仲).속단(續斷) 배합약물의 관절염 억제 효과)

  • Objectives : The object of this study was to verify the inhibitory effect of a decoction of Eucommiae ulmides OLIVER (EU) and Dipsacus asperoides C. Y. Cheng et T.M.Ai (DA) on Collagen II-induced Arthritis Mice (CIA mice). Methods : DBA/1OlaHsd mice were immunized with bovine type II collagen. Boostnig same collagen 21 days later, arthritis was induced and then administrated orally the extract of EU+DA (200 or 50 mg/kg) once a day for 4 weeks and compared with that of methotrexate (MTX, 0.3 mg/kg) as a positive control. Results : Administration of EU+DA suppressed the inflammatory progression of CIA mice and the results were 1. Arthritis index of CIA mice was decreased. 2. EU+DA decreased the production of TNF-$\alpha$, IL-6, IL-$1{\beta}$ in the serum of CIA mice. 3. EU+DA decreased the level of IgM. 4. EU+DAincreasaed $CD3^+$, $CD4^+$, $CD4^+$/CD25 but decreased $CD19^+$, $CD3^+/CD49b^+$(NKT), $CD3^-/CD49b^+$(NK), $B220^+/CD23^+$ in PBMC of CIA mice. 5. EU+DA decreased $CD3^+$, $CD4^+$, $CD3^+/CD69^+$ of paw joint in CIA mice. 6. EU+DA decreased subsynovial inflammation. Conclusions : This results demonstrated that extract of EU+DA suppressed the inflammatory progression of CIA mice and supported further studies are required to clarify a mechanism of therapeutic role.

Research on Effects of Cordyceps Sinensis in Spleen Cells of Mouse. (동충하초 추출물이 생쥐 비장세포에 미치는 영향)

  • Objectives : This study was carried out to know the effect of Cordyceps sinensis(CS) on the immune inflammatory responses of spleen cells and function or immunocytes of the normal mouse. Methodes: We investigated effects of Cordyceps sinensis(CS) on normal immunocytes, gene expression of IL-12, IFN-$\gamma$ and surface-receptor expression of $CD3_{\epsilon}+$, CD4+, CD8+ and CD19+ cells were measured by PCR and FACS. Results : CS activated adhisive splenic cells morphologically as compared with the control group in the normal spleen cells of BALB/C mice. CS enhanced gene expression of interleukin-12 and interferon-gamma in a dose-dependent manner in the normal spleen cells of BALB/C mice. CS reduced the number of activating cells and surface-receptor expression of CD4+, CD8+ and CD19+. Conclusion : Cordyceps sinensis will be used as a stable remedium in the auto-immune diseases.

Temperature Effect on Effective Diffusion Coefficients of Zn and Cd through Column Diffusion Tests (칼럼 확산 실험을 통한 아연 및 카드뮴의 유효확산계수에 미치는 온도영향)

  • In this study, column diffusion tests for Cd and Zn were conducted at $15^{\circ}C$ and $55^{\circ}C$ to investigate a temperature effect on effective diffusion coefficient. An increase in temperature from $15^{\circ}C$ to $55^{\circ}C$ caused up to ten times larger diffusion coefficient for each heavy metal. Besides, it caused the increased retardation of heavy metals, and hence the effective diffusion coefficient should be overestimated as we use an overestimated retardation factor to calibrate the coefficient. The results of sequential extraction analyses showed that Zn was occluded in carbonate phase and this trend was getting prominent with the increase in temperature. As for Cd, it was partitioned mainly in the exchangeable phase(over 60%) at any temperature.

Purification and Some Properties of Cyclodextrin Hydrolase (Cyclodextrin분해효소의 정제 및 그 특성)

  • Cyclodextrin hydrolase from Bacillus stearothermophilus KFCC 21203 was purified and the properties of the purified enzyme were investigated. The enzyme was purified 15 folds with 77 % recovery by ammonium sulfate fractionation, DEAE-cellulose chromatography, and Ultro AcA 34 gel filtration. The specific activity and the molecular weight of the enzyme were 1.30 units/mg protein and about 29,500, respectively, The maximum activity of the enzyme was shown at $55^{\circ}C$ and pH 5.5. However, stable temperature and pH were $40^{\circ}C$ and $5.0{\sim}8.0$, respectively. The Km value for ${\gamma}-cyclodextrin$ was $3.78{\times}10^{-3}$ M. The degradation activity of the enzyme was selectively high for ${\gamma}-cyclodextrin$, and very low for ${\beta}-cyclodextrin$, but not for ${\alpha}-cyclodextrin$. The decomposed products of ${\gamma}-cyclodextrin$ were mainly glucose and maltose, and a little mlatotriose. The activity of the enzyme was very high for amylose, potato starch, corn starch, amylopectin and maltooligomer, and relatively high for glycogen and dextrin. The decomposed products of them were mainly glucose and maltose.

Reference Values for Peripheral Blood Lymphocyte Subsets in a Healthy Korean Population

  • Flow cytometric immunophenotyping of peripheral blood lymphocyte subsets is a powerful tool for evaluating cellular immunity and monitoring immune-mediated diseases. The numbers and proportions of blood lymphocyte subsets are influenced by factors such as gender, age, ethnicity, and lifestyle. This study aimed to establish reference ranges for peripheral blood lymphocyte subsets in a healthy Korean population. Blood samples from 294 healthy adults were collected. Lymphocyte subsets were analyzed using a single-platform method with a flow cytometer; white blood cells and lymphocytes were analyzed using an automated hematology analyzer. The mean value of the white blood cell count was $5,665cells/{\mu}l$, and the mean values of the subtype counts (percentages) were as follows: lymphocytes, $1,928cells/{\mu}l$ (35.08%); $CD3^+$ cells, $1,305cells/{\mu}l$ (67.53%); $CD3^+CD4^+$ cells, $787cells/{\mu}l$ (40.55%); $CD3^+CD8^+$ cells, $479cells/{\mu}l$ (25.23%); $CD3^-CD19^+$ cells, $203cells/{\mu}l$ (10.43%); and $CD3^-CD56^+$ cells, $300cells/{\mu}l$ (15.63%). Additionally, the $CD4^+/CD8^+$ ratio was 1.81. In this study, gender and age significantly influenced blood lymphocyte subsets. Our results demonstrate that, as with other populations, a healthy Korean population has its own, region-specific, lymphocyte subset reference ranges.
