Analysis for Cyclodextrins to Entrap with Hexanal using Electronic Nose (전자코를 이용한 헥사날과 싸이클로덱스트린의 결합 분석)

  • The effectiveness of cyclodextrin(CD) on binding and/or entrapping hexanal in model solution was investigated. The types and concentration of CDs for entrapping hexanal were studied using electronic nose, which composed of metal oxide sensor or was based on GC with surface acoustic wave sensor. ${\alpha}-CD$ was the most effective for lowering headspace concentration of hexanal in model solution. As concentration of CD increased, hexanal concentration in the headspace decreased significantly. Addition of 5% ${\beta}-CD$ to hexanal in model system resulted in 86% reduction of hexanal in the headspace. There was no difference between control and treatment at the initial stage of binding CD with hexanal while reduction of hexanal in the headspace was found during storage time. This could be estimated by electronic nose.

Anticancer Effect of Activated Natural Killer Cells on Human Colorectal Tumor (결장암에 대한 활성 자연살해세포의 항암효능)

  • Colorectal cancer is one of the most common alimentary malignancies. In this study, the antitumor activity of activated human natural killer (NK) cells against human colorectal cancer was evaluated in vivo. Human NK cells are the key contributors of innate immune response and the effective functions of these cells are enhanced by cytokines. Human peripheral blood mononuclear cells (PBMC) were cultured with interleukin-2 (IL-2)-containing medium for 14 days and resulted in enriched NK cell population. The resulting populations of the cells comprised 7% $CD3^+CD4^+$ cells, 25% $CD3^+CD8^+$ cells, 13% $CD3^-CD8^+$ cells, 4% $CD3^+$CD16/$CD56^+$ cells, 39% $CD3^+$CD16/$CD56^-$ cells, and 52% $CD3^-$CD16/$CD56^+$ cells. Tumor necrosis factor alpha (TNF-$\alpha$), interferon gamma (IFN-$\gamma$), IL-2, IL-4, and IL-5 transcripts of the activated NK cells were confirmed by RT-PCR. In addition, activated NK cells at doses of 2.5, 5 and 10 million cells per mouse inhibited 10%, 34% and 47% of SW620-induced tumor growth in nude mouse xenograft assays, respectively. This study suggests that NK cell-based immunotherapy may be used as an adoptive immunotherapy for colorectal cancer patients.

Electrical and Optical Properties of CdS Films prepared by Electrodeposition (전착법으로 제조한 CdS 막의 전기 및 광학적 특성)

  • Polycrystalline CdS films were grown on glass substrate with conducting indium tin oxide(ITO) by electrodeposition. The average size of the plate-shape grains o the CdS films was from 0.3um to 0.05um, and the adhesion to ITO was excellent. The optical band gap of the electrodeposited CdS films was in the range from 2.51eV to 2.68 eV. The optical transmittance was 80% and the electrical resistivity varied from 10$^3$ to 10$\^$5/$\Omega$-cm depending on the deposition condition.

  • Long-term immunity to many viral and bacterial pathogens requires$ CD8^+$ memory T cell development, and the induction of long-lasting$ CD8^+$ memory T cells from a $na{\ddot{i}}ve$, undifferentiated state is a major goal of vaccine design. Formation of the memory$ CD8^+$ T cell compartment is highly dependent on the early activation cues received by $na{\ddot{i}ve}$ $CD8^+$ T cells during primary infection. This review aims to highlight the cellularity of various niches within the lymph node and emphasize recent evidence suggesting that distinct types of T cell activation and differentiation occur within different immune contexts in lymphoid organs.

Kinetic Study of Cadmium Adsorption by Sewage Sludge (하수오니에 의한 Cd 흡착의 속도론적 연구)

  • To investigate the adsorption-desorption reaction of Cd by sewage sludge, the adsorption of Cd from $Cd(NO_3)_2$ solutions of concentrations ranging from 5 to $50{\mu}g\;Cd\;mL^{-1}$ by sewage sludge was examined for reaction periods up to 48 hours. The amount of Cd adsorbed as a function of time was measured. The adsorption between Cd in solution and the solid phase could be described by two stages. The initial adsorption of Cd was very rapid, that is, approximately 95% of the added Cd was removed from solution within the first 30 minutes. Further, the greater the concentration of Cd added, the greater was the amount of Cd adsorbed. After the rapid initial decrease of Cd, a slower decline in the Cd concentration resulted which followed first order reversible kinetics. The equilibrium concentrations for the reactions, as well as the time period for the equilibrium reactions were dependent on the initial Cd concentrations. If equilibrium is reached, the amount of Cd remaining in solution is greater when the amount adsorbed is higher, although the percentage of Cd in solution is constant relative to the initial concentration of Cd. Some of the adsorbed Cd was released back to solution since the concentration of Cd after 48 hours was higher than the equilibrium concentration of Cd. However, despite the increased amount of Cd measured, the overall net reaction was a significant adsorption of Cd from solution by sewage sludge.

Evaluation of BCL-6, CD10, CD138 and MUM-1 Expression in Diffuse Large B-Cell Lymphoma patients: CD138 is a Marker of Poor Prognosis

  • The diffuse large B-cell lymphoma (DLBCL) encompasses two major groups of tumors with uneven survival outcomes - germinal center B-cell (GCB) and non-germinal center B-cell (non-GCB). In the present study, we investigated the expression of GCB markers (BCL-6 and CD10) and non-GCB markers (CD138 and MUM-1) in an effort to evaluate their prognostic value. Paraffin-embedded tumor biopsies of 46 Jordanian DLBCL patients were analyzed, retrospectively, by immunohistochemistry to investigate the expression of BCL-6, CD10, CD138 and MUM-1. In addition, survival curves were calculated with reference to marker expression, age, sex and nodal involvement. Positive expression of BCL-6, CD10, CD138 and MUM-1 was shown in 78%, 61%, 39% and 91% of the cases, respectively, that of BCL-6 being associated with better overall survival (p = 0.02), whereas positive CD138 was linked with poor overall survival (p = 0.01). The expression of CD10 and MUM-1 had no impact on the overall survival. Among the clinical characteristics studied, diagnosis at an early age, nodal involvement and maleness were associated with a higher overall survival for DLBCL patients. Our results underline the importance of BCL-6 as a marker of better prognosis and CD138 as a marker of poor prognosis for DLBCL patients.

  • The properties of Fe ions in the $ CdFe_2$$O_4$ and $CdFeAlO_4$ were investigated using the Mossbauer spectroscopy, in the temperature range from 300K to 900K. The calculated Debye temperature, recoil free fraction, mean square displacement and velocity are, 476.5K, 0.835, 3.39$\times$10\ulcorner$\textrm{cm}^2$, and 1.11$\times$10\ulcorner$\textrm{cm}^2$sec$^2$for Fe ions in the $CdFe_2$$O_4$ and 369.0K, 0.741, 5.65$\times$10\ulcorner$\textrm{cm}^2$ and 1.33$\times$10\ulcorner$\textrm{cm}^2$/$sec^2$in the $CeFeAlO_4$. The difference of the properties in the two samples were explained by the variation of the Fe\ulcorner-O\ulcorner bonding length with the substituted Al ions.

  • Bulk type radiation detector of Au/Cd$_{1-x}$ Zn$_{x}$Te(x=20%)/Au structure using Cd$_{1-x}$ Zn$_{x}$Te(x=20%) wafer(3x4xl mm$^{3}$) grown by high pressure Bridgman method has been developed. We etched wafer surfaces with 2% Br-methanol solution and coated gold thin film on the surfaces by electroless deposition method for 5 min. in 49/o HAuCI$_{3}$ 4H20 solution. Initial etch rates of Cd, Zn and Te were 46%, 12% and 42% respectively. After etched, the surface of wafer was slightly revealed to Te rich condition. The leakage current was increased with etch time, but it didn't exceed 3nA at 50volt. The thickness of Au film was about 100nm by Rutherford Backscattering Spectroscopy(RBS). The resolution were 6.7% for 22.1 keV photon from 109 $^{109}$ Cd and 8.2% for 59.5 keV photon from $^{241}$ Am. The radiation detector such as Au/Cd$_{1-x}$ Zn$_{x}$Te(x=20%)/Au structure was more effective to monitor the low energy gamma radiation.iation.

  • The survival rates of Littorina brevicula exposed to experimental concentration regimes of TBTCl, HB and Cd on the large and the small size individuals during 80 days were $80\%$ at 0,9ppb TBTCl, 40 and $25\%$, respectively at 200ppb Hg, and 75 and $45\%$, respectively at 100ppb Cd. The growth rates of the experimental animals exposed to each concentration for 80 days was 0.023mm/day at control, 0.019mm/day at 0.1ppb and 0.014mm/day at 0.9ppb TBTCl, 0.022 mm/day at 5ppb, 0.008 mm/day at 200ppb Hg, and 0.017 mm/day at 5ppb, 0.008mm/day at 100ppb Cd. The respiration rates and excretion rates of the experimental animals exposed to chronic concentration of TBTCl, Hg and Cd were decreased until approximatively 40 days and increased after, Toxic effect of pollutants on L. brevicula was highest at TBTCl. The histological injury of L. brevicula exposed to TBTCl, Hg and Cd was shown at gill, digestive organ and muscle, respectively.

Suppressive Effects of JTT on Collagen-Induced Arthritis in Mice (정통탕(定痛湯)이 콜라젠으로 유도한 관절염 동물 병태 모델에 미치는 효과)

  • Rheumatoid arthritis is an autoimmune disease involving multiple joint. In order to access the suppressive effects of JTT on rheumatoid arthritis and it's effects on immune system we investigated whether JTT could suppress the disease progression of collagen-induced arthritis. DBA/1 mice were immunized with bovine type II collagen. After a second collagen immunization, mice were treated with DW, JTT (200 or 400 mg/kg) or methotrexate (MTX, 30 mg/kg) as a positive control. Oral administration of JTT significantly suppressed the progression of CIA, which extend is comparable to that of MTX. Histological examination reveled that JTT inhibited infiltration of inflammatory cells into affected paw joint and bone erosion and cartilage destruction were greatly reduced compared with control. Total cell number of spleen, lymph node and peripheral blood were significantly reduced. The absolute number of CD19$^+$, CD3$^+$/CD69$^+$, CD4$^+$/CD25$^+$ cell in spleen from JTT treated mice were significantly decreased. The absolute number of CD19$^+$, CD3$^+$, CD3$^+$/CD69$^+$, CD4$^+$, CD4$^+$/CD25$^+$ CD8$^+$, CD49b, CD3/CD49b cells in draining lymph node were significantly increased compared with control. In peripheral blood mononuclear cells of JTT treated mice, the absolute number of CD4$^+$, CD4$^+$/CD25$^+$, CD3$^+$/CD69$^+$ cells were significantly decreased compared with control, while that of CD49b$^+$ was slightly increased. Infiltration of CD3$^+$ cells and CD11b$^+$/Gr-1$^+$ cells into paw joint was significantly reduced in JTT treated mice. The levels of pathologic cytokines including TNF-a and IL-6 in serum were significantly decreased by oral treatment with JTT The levels of IFN-g in the culture supernatant of splenocyte stimulated with CD3$^+$/CD28$^+$ or collagen were dramatically decreased, while the levels of IL-4 was increased under CD3$^+$/CD28$^+$ or collagen stimulation. Rheumatoid factors including IgG, IgM and collagen specific antibody were present much lower in the serum of JTT treated mice than control. Taken together, JTT has suppressive effects on rheumatoid arthritis by modulating immune system, and has potential to use anti-rheumatic arthritic agent in human.
