Association of the Genetic Polymorphisms for CD247 Gene and Tuberculosis Case

  • Tuberculosis is airborne disease caused by Mycobacterium tuberculosis (MTB). Host genetic factors of these tuberculosis play an important role in determining individual difference in susceptibility or resistance to infectious diseases including tuberculosis. CD247 is named CD3zeta chain or CD3ζ. CD247 gene is a protein-coding gene involved in phagocytosis and signal transduction of the T cell receptor (TCR). Also, downregulation of the CD3ζ chain has been associated to chronic inflammation. The aim of this study was to research association of the genetic polymorphisms for CD247 gene and tuberculosis. We analyzed association of CD247 and Mycobacterium tuberculosis using 149 imputed single nucleotide polymorphisms (SNPs) with Korean population. And the results of this study show that seven SNPs of CD247 were identified to associate with tuberculosis. The most significant SNP was rs858545 (OR=1.22, CI: 1.05~1.42, P=0.009481). This study suggests that polymorphisms of CD247 may affect the T cell receptor signaling pathway, which may associate the infection of tuberculosis.

Complexation between Venlafaxine Hydrochloride and β -Cyclodextrin:Structural Study by Nuclear Magnetic Resonance Spectroscopy

  • A detailed spectroscopic study ($^1H$ NMR, COSY, ROESY) of complexation of venlafaxine hydrochloride (VEN) with $\beta$-cyclodextrin ($\beta$--CD) was carried out in solution. The stoichiometry of the complex was determined to be 1 : 1 and penetration of aromatic ring into $\beta$-Cyclodextrin cavity was confirmed from primary rim side, with the help of ROESY spectral data. The structure of the venlafaxine hydrochloride-$\beta$-CD complex has been proposed. The association constant was determined to be 234 $M^{-1}$.

Resveratrol의 CD4+ T 세포 활성과 분화 억제 효과 (Resveratrol Suppresses CD4+ T Cell Activation and Differentiation in vitro)

  • Resveratrol은 천연 stilbene으로 안전성 있는 항염증 활성을 가진 화합물로 알려져 있다. 최근의 연구들에서 resveratrol이 천식, 만성 대장염, 류마티스성 관절염과 같이 염증에 의해 발생하는 다양한 질병을 억제한다고 보고되었다. 이러한 연구들은 resveratrol이 $CD4^+$ helper T cells (Th cells)에 의한 면역반응을 조절할 것이라고 제시하였다. 그러나 resveratrol이 직접적으로 Th cells의 활성화와 분화를 조절하는지 완전히 밝혀지지 않았다. C57BL/6에서 Th cells을 분리하여 다양한 농도의 resveratrol을 세포에 처리하였다. 본 연구에서는 resveratrol이 직접적으로 Th cells의 활성화와 증식을 억제하는 것을 확인하였다. Th cells에 resveratrol을 처리하였을 때 IFN-${\gamma}$, IL-4, IL-17 사이토카인 생성이 농도에 따라 유의하게 감소하였고 또한 Th cells이 이러한 사이토카인들을 분비하는 Th1과 Th2과 Th17으로 분화되는 것이 억제되었다. 그리고 고농도의 resveratrol이 Th cells의 세포사멸을 유도하는 것으로 확인되었다. 본 연구에서는 resveratrol이 Th cells의 활성화와 분화를 직접적으로 억제하는 것을 확인하였으며, 이는 resveratrol이 $CD4^+$ Thcells에 의해 발생되는 자가면역질환의 효과적인 치료법이 될 수 있을 것이라고 제시한다.

  • Immune responses associated with bacterial infection involve various inflammatory cells. Clinical symptoms and pathologic features are particularly influenced by the predominant cells Among inflammatory cells, T cells have the heterogenity. T cells may develop into the mature cells expressing the cell surface markers with different functions and T helper cells are categorized into Th1 and Th2 cells based on their different patterns of cytokine production. The objective of this study was to investigate the change of expression of surface markers on T cells and the Th1/Th2 immune response in pulpal inflammation associated with specific bacteria. We experimentally induced pulpal inflammation in rat incisors by drilling without coolant and innoculated with Streptococcus mutans (S.M. group), Porphyromonas endodontalis (P.E. group), or only sterile cotton (control group). After 1, 2, and 5 days, mandibular incisors were extracted and the pulp tissues were extirpated The expressions of IL-2 recepters (CD25) and ICAM-1 (CD54) on CD4+ and CD8+ cells in the pulps were determined using a flow cytometer, and the concentration of IL-2, IFN-$\gamma$ and IL-4 was measured by enzyme-linked immunosorbent assay. The results were as follows: 1 In the S.M. group, CD4+ cells were more increased at 2nd day than 1st day and in the P.E. group, CD8+ cells were more increased at 2nd day than 1st day. 2. The percentages of CD4+, CD4+25+ and CD4+54+ cells were decreased in the pulp tissues at 5th day after irritation in all groups. 3. The ratios of CD4+/CD8+, CD4+/CD4+25+ and CD4+/CD4+54+ in the pulps at 2nd day after irritation by P. endodontalis were significantly lower than the other groups. 4. The higher concentrations of IFN-$\gamma$ than IL-4 in the pulps at 2nd day after irritation by P. endodontalis showed that T helper 1 reaction were predominant in the early stage of the pulpal inflammation induced by P. endodontalis. 5. The higher concentrations of IL-4 than IFN-$\gamma$ in the pulps at 1st day and 5th day after irritation by S. mutans were measured but the differences were not significant.

아스파르트산 킬레이트 칼슘의 칼슘 결핍쥐에서의 생물학적 유용성 (Bioavailability of Aspartic Acid Chelated Calcium in Calcium Deficient Rats)

  • Calcium (Ca) is an essential element to maintain body homeostasis. However, many factors disturb calcium absorption. Aspartic acid chelated calcium (AAC) was synthesized by new methods using calcium carbonate and aspartic acid. This study was carried out to investigate the bioavailability of AAC in Ca-deficient rats. The experimental groups were as follows: NC; normal diet control group, CD-C; untreated control group of Ca-deficient (CD) rats, CD-$CaCO_3$; $CaCO_3$ treated group of CD rats, CD-AAC; AAC treated group of CD rats, and CD-SWC; and seaweed-derived Ca treated group of CD rats. The Ca content of various types of Ca was held constant at 32 mg/day, and the four CD groups were fed for 7 days after randomized grouping. Ca content in serum, urine, and feces within feeding periods were analyzed to confirm Ca absorption. Serum Ca content was significantly higher in the CD-AAC (11.24 mg/dL) and CD-SWC (10.12 mg/dL) groups than that in the CD-C (8.6 mg/dL) group 2 hours following the first administration. The Ca content in feces was significantly lower in the CD-AAC (35.4 mg/3 days) and CD-SWC (71.1 mg/3 day) groups than that in the CD-$CaCO_3$ (98.7 mg/3 days) group (p > 0.05). AAC had a 2.3-fold higher absorption rate of Ca than that of SWC. No differences in fibula length were observed in the NC and CD groups. The fibula weights of the CD-AAC (0.33 g) and CD-SWC (0.33 g) groups increased compared to those in the CD-C (0.27 g) group; however, no significant difference was observed between the CD groups. We conclude that bioavailability of AAC is higher than that of seaweed-derived Ca or inorganic Ca. Thus, these findings suggest the AAC has potential as a functional food material related to Ca metabolism.

  • The purpose of this study is to suggest the schemes to enhance information service functions by the utilization of CD-ROM in medical school libraries. The results of the study are summarized as follows : 1. The selection and evaluation of CD-ROM database are necessary steps in the planning of a CD-ROM. Before the CD-ROM is selected, therefore, medical libraries must make a practical evaluation criteria in important order of information services environment, characteristics of hardware/software, service requirements, price and cost, etc. 2. If possible, CD-ROM MEDLINE must be suited for the information services environment. 3. In case of the popular core journals, full-text CD-ROM should be gradually purchased. 4. In order to reduce the time required from search of bibliographic informations to receipt of original articles, CD-NET system and library holding administration program must be built up and developed. And channel of information search and order/receipt of original article should be varied. 5. Search education program for medical librarians and users should be enforced, and librarians must play an important role in CD-ROM retrieval consultant and intermediator.

돼지고기의 콜레스테롤 함량 감소에 관한 β-Cyclodextrin의 급여효과 (Influence of Feeding β-Cyclodextrin on Reducing the Content of Cholesterol in Pork)

  • 본 연구의 주목적은 돼지고기의 콜레스테롤 함량 감소에 관한 사료 내 ${\beta}CD$의 급여 효과를 조사하는 것이었다. 생체중 50 kg의 3원교잡종 거세 수퇘지 12마리를 이용하여서 4처리구 3반복으로 완전임의배치한 후, 시판체중인 110 kg 도달시까지 약 9주 동안 실험사료를 급여하였다. 일일 사료섭취량, 일일 증체량 및 사료효율은 ${\beta}CD$를 첨가하지 않은 대조구와 비교할 때, ${\beta}CD$ 1.5%, 3.0% 및 5.0% 첨가구의 각 처리구간 통계적인 유의차가 나타나지 않았다. 혈액 총지질, 중성지방 및 총콜레스테롤 함량은 대조구와 비교할 때 ${\beta}CD$ 첨가구가 유의적으로 낮아졌으며, 특히 ${\beta}CD$ 5.0% 첨가구에서 각각의 감소율은 21.80%, 55.58% 및 27.69%로서 가장 높게 나타났다(p<0.05). 돼지고기 삼겹살의 콜레스테롤 함량은 대조구와 비교할 때 ${\beta}CD$ 1.5%, 3.0% 및 5.0% 첨가구에서 각각 5.33mg, 12.70mg및 15.23 mg의 유의적인 감소를 나타났으며, 특히 이 값을 콜레스테롤의 감소율로서 나타냈을 때 각각 6.44%, 15.36%, 18.42%로서 유의적인 차이를 나타냈다(p<0.05). 이 결과는 돼지에서 ${\beta}CD$첨가급여가 콜레스테롤 대사를 조절할 수 있는 식이섬유로서 사용될 수 있음을 시사해준다.

차가버섯, 상황버섯 및 영지버섯 복합추출물 복용이 인체의 혈중 조혈모세포와 면역세포에 미치는 영향 (The Effects of Extracts Mixture Drink from Inonotus Obliquus, Phellinus Linteus and Ganoderma Lucidum on Hematopoietic Stem Cells and Lymphocyte Subset of Blood in Human)

  • 본 연구는 차가버섯, 상황버섯, 영지버섯 혼합추출물(IPGE)음료 복용이 인체의 혈중 조혈모세포와 임파구 아형(Lymphocyte, $CD4^+T$ cell, $CD8^+T$ cell, Natural Killer cell) 증식에 미치는 영향을 관찰하기 위하여 수행하였다. 피험자는 건강한 지원자들로서 $40{\sim}70$대의 남여 일반인들로 하였다. 전체 39명을 모집하여 무작위 배정을 통해 27명, 12명 씩 2그룹으로 나누고 각각 버섯 복합추출물과 위약 음료를 따로 지급하여 4주 동안 매일 복용하게 하였다. 혈액은 복용 첫날부터 시작하여 2주 간격으로 피험자들로부터 채취되었으며 면역세포 수 측정에 사용되었다. 조혈모세포(hematopoietic stem cell)는 IPGE 음료 복용군에서 복용 전에 비하여 현저하게 증가하였으며 통계적으로 유의한 차이를 나타냈다(p<0.001). 임파구(lymphocyte)는 IPGE음료 복용 전과 후 간에 유의한 차이가 관찰되지 않았다. $CD4^+T$ 세포, $CD4^+T$ 세포 및 $CD4^+/CD8^+$ 비율은 시험 음료 복용 전과 후 간에 유의한 차이가 나타나지 않았다. 그리고 NK 세포도 IPGE 음료 복용 전과 후 간에 유의한 차이가 관찰되지 않았다. 본 연구결과를 종합해 볼 때 차가버섯, 상황버섯 및 영지버섯 혼합추출물(IPGE)음료는 조혈모세포의 증식효과를 현저하게 높게 나타내었기 때문에 총체적인 혈액세포 정상화를 통해 인체 건강증진에 긍정적인 효과를 나타낼 것으로 사료되었다.
