백서(白鼠)의 B16-F10 Melanoma에 대한 한련초약침(旱蓮草藥鍼)의 항암(抗癌) 및 면역증강효과(免疫增强效果) (The Effect of Anti-Cancer and Immune Response Improvement of ELP(Ecliptae Herba) Herbal-Acupuncture into Chung-wan(CV12))

  • Objective & Methods : To study the effects of anti-cancer, anti-metastasis and immune response improvement of herbal-acupuncture with Eclipta prostrata diffusae herba infusion solution(ELP-HAS), we injected ELP-HAS into Chung-wan(CV12) of C57BL/6 mice implanted intravenously with B16-F10 melanoma. We have reached the following conclusion through the effect on the percentage of $CD25^+/CD4^+$, $CD8^+/CD3e^+$, $CD69^+/B220^+$, $NK1.1^+/CD3e^+$cells in mouse PBMCs, the effect on the pulmonary colony formation number, and the effect on MST(Median Survival Time) and ILS(Increase of Life Span) of C57BL/6 mice implanted intravenously with B16-F10 melanoma. Results : The results were obtained as follows : 1. In the experiment groups treated with ELP(Ecliptae Herba) Herbal acupuncture, the spleen cell proliferation in BALB/c mouse was significantly increased compared with control group. 2. In the experiment groups treated with ELP(Ecliptae Herba) Herbal acupuncture, the percentage of $CD25^+/CD4^+$, $CD8^+/CD3e^+$, $CD69^+/B220^+$, $NK1.1^+/CD3e^+$cells in C57BL/6 mouse PBMCs was increased compared with control group. 3. In the experiment groups treated with ELP(Ecliptae Herba) Herbal acupuncture, the pulmonary colony formation number of C57BL/6 mice implanted intravenously with B16-F10 melanoma was decreased significantly compared with control group. 4. In the experiment groups treated with ELP(Ecliptae Herba) Herbal acupuncture, MST(Median Survival Time) and ILS(Increase of Life Span) of C57BL/6 mice implanted intravenously with B16-F10 melanoma were increased significantly compared with control group. Conclusions : ELP Herbal acupuncture injection into Chung-wan(CV12) of C57BL/6 mice is considered to be effective in anti-cancer, anti-metastasis and immune response improvement.

  • 본 연구에서는 광전자 소자에 폭넓게 사용되는 ZnCdSe 화합물 반도체의 end-point인 CdSe의 유전함수 spectrum을 Vacuum Ultra Violet spectroscopic ellipsometry(타원편광분석법) 측정하여 분석하였다. 측정 결과는 변수화 모델을 이용하여 분석하였으며 그 결과 6 eV 이상에 존재하는 전자전이점들을 확인할 수 있었고 CdSe의 Critical Point(CP) 구조를 수치화 함으로써 온도나 화합물 함량에 따른 광특성 의존성 연구 등에 활용될 수 있는 database를 확보하였다.

가미삼령백출산(加味蔘苓白朮散)의 B16 흑색종 암모델에 대한 항종양효과(抗腫瘍效果)와 면역증강효과(免疫增强效果)에 관한 연구(硏究) (The Effect of Gamisamryungbaekchul-san(加味蔘苓白朮散) on the Tumor and Immune Response in Mouse B16 Melanoma Tumor Model)

  • Background : Gamisamryungbaekchul-san(加味蔘?白朮散) is a herbal medicine which has been used for the traditional therapeutic agent of augmentation of the spleen and reinforcement of the Qi. Objective : This Study was performed to investigate the effect of Gamisamryungbaekchul-san on the tumor and immune response in the moose B16 melanoma tumor model. Materials and Methods : The tumor was induced by subcutaneous inoculation of B16BL6 melanoma cells in the shaved dorsal region of mice. Mice were orally administered with Gamisamryungbaekchul-san extract(26.3mg/mouse) for 14days after inoculation. For making examination of antitumor effect, the Increase of life span, Tumor growth inhibition rate, change of body weight were measured and evaluated. For the immune response increasing effect, the percentage of T lymphocyte and B Lymphocyte in the peripheral blood, the percentage of CD4+ T-cell, CD8+ T-cell and CD4+/CD8+ ratio in the peripheral blood and spleen, interleukin-2 productivity were measured and evaluated. Results : Gamisamryungbaekchul-san showed 16.59% increase of life span, 31.64% tumor growth inhibition rate and increase of body weight. Gamisamryungbaekchul-san increased the percentage of T lymphocyte in the peripheral blood, CD4+ T cell percentage of peripheral blood and spleen, and Interleukin-2 productivity as compared with the Control group. Whereas Gamisamryungbaekchul-san had no effect on the percentage of B lymphocyte in the peripheral blood, the percentage of CD8+ T cell, CD4+/CD8+ T-cell ratio in both of peripheral blood and spleen as compared with the Control group. Conclusion : This study shows that Gamisamryungbaekchul-san has anti-tumor effects and immunoregulatory effects on the B16 melanoma tumor model. It is suggested that Gamisarmyungbaekchul-san could be a useful immunomodulator and anti-tumor agent.

  • CTAB(cetyltrimethylammonium bromide)-capped CdSe nanoparticles were prepared by using a 4 : 1(v/v) distilled water-isopropyl alcohol mixture. The cadmium chloride and sodium selenosulfate were used as the cadmium and selenium source. By the analysis of XRD and XPS, the resultant particle was confirmed to be cubic CdSe phase. TEM image showed CdSe nanoparticles with empty core. The CTAB-capped sample showed an maximum absorption at 418nm, blue-shifting compared with bulk CdSe, which indicated stronger quantum confinement effect compared with uncapped sample. From FT-IR analysis, it was found that the presence of the new peaks in the $850{\sim}1250cm^{-1}$ range indicated the existence of chemical bonding between CTAB and surface of CdSe nanoparticles. Also TG analysis indicated that there were two weight-loss steps for the CTAB-capped CdSe nanoparticles. It was suggested that CTAB played a significant role in protecting CdSe nanoparticles.

카드뮴화합물별(化合物別) 수도(水稻) 흡수(吸收) 및 생육(生育)에 미치는 영향(影響) (Effect of Various Cadmium Compounds on the Growth and Cadmium Uptake of Paddy Rice)

  • 수도(水稻)에 대(對)한 cadmium화합물(化合物)의 형태(形態)에 따른 피해농도(被害濃度) 및 cadmium흡수정도(吸收程度)를 구명(究明)하기 위(爲)하여 $Cd(NO_3)_2$, $CdCl_2$, $CdSo_4$, $CdCo_3$ 및 CdS 등(等)의 화합물(化合物)을 토양(土壤)에 Cd로서 0, 5, 10, 25, 50 ppm처리(處理)하여 수도(水稻)의 수양(收量)에 미치는 농도(濃度)와 수도체중(水稻體中)의 cadmium함량(含量)을 조사(調査)한 바 그 결과(結果)는 다음과 같다. 1) 토양중(土壤中) cadmium처리농도(處理濃度)가 높을수록 식물체(植物體) 경엽(莖葉) 및 현미중(玄米中) cadmium함량(含量)은 증가(增加)하였으나 수량(收量)은 높은 농도(濃度)에서 감소(減小)되었다. 2) 수양감수(收量減收)를 가져오는 토양중(土壤中) Cd의 농도(濃度)는 $Cd(NO_3)_2$ : 12.9ppm, $CdSO_4$ : 21.5ppm, $CdCl_2$ : 25.8ppm $CdCO_3$ : 33.2ppm, CdS : 97.6ppm 이었다. 3) 현미중(玄米中) cadmium함량(含量)이 1.0ppm이 되는 토양중(土壤中) Cd의 농도(濃度)는 $cd(NO_3)_2$ : 13.8ppm, $CdCl_2$ : 14.4ppm, $CdSO_4$ : 16.9ppm, $CdCO_3$ : 19.2ppm이었다. 4) cadmium의 현미흡수양/지상부흡수량비(玄米吸收量/地上部吸收量比)는 최고(最高) 분얼기(分蘖期) 28%, 유수형성기(幼穗形成期) 20%, 수확기(收穫期) 15%이었다. 5) 유수형성기(幼穗形成期) 식물체(植物體) 경엽중(莖葉中) cadmium함량(含量)과 현미중(玄米中) cadmium함량(含量)과의 관계(關係)는 $Y=0.067-0.0028x+0.007x^2$으로 고도(高度)의 유의성(有意性)이 있었다. 6) 용해도(溶解度)가 낮은 cadmium화합물(化合物)이 시험후(試驗後) 토양중(土壤中) 가용성(可溶性) cadmium합량(合量)이 낮았다.

Effect of CXCR4 and CD133 Co-expression on the Prognosis of Patients with Stage II~III Colon Cancer

  • Background: To explore the relationship between CXCR4, CD133 co-expression and clinicopathological features as well as prognosis of patients with phase II~III colon cancer. Materials and Methods: Forty-nine paraffin-embedded samples of tumor tissue and epithelial tissue adjacent to cancer were collected from patients with colon cancer undergoing radical surgery in Baotou Cancer Hospital from January, 2010 to June, 2011. CXCR4 and CD133 expression was detected using immunohistochemistry and its relationship with clinicopathological features and the 3-year survival rate was analyzed. Results: In the tumor tissue and colonic epithelial tissue adjacent to cancer, the positive expression rates of CXCR4 were respectively 61.2% (30/49) and 8.16% (4/49), while those of CD133 being 36.7% (18/49) and 6.12% (3/49). CXCR4 and CD133 expression in tumor tissue was not related to patient age, gender, primary focal sites, tumor size, TNM staging, histological type, tumor infiltration depth and presence or absence of lymphatic metastasis, but CXCR4 and CD133 co-expression was associated with TNM staging and lymphatic metastasis. The 3-year survival rate of patients with CXCR4 and CD133 co-expression was 27.3% (3/11), and that of the remainderwas 76.3% (29/38), the difference being significant ($X^2=7.0206$, p=0.0081). Conclusions: CXCR4 and CD133 co-expression may be a risk factor for poor prognosis of patients with stage II~III colon cancer.

Catharanthus roseus 세포 배양액에 deuterium이 치환된 tabersonine을 사용한 vindoline 생합성 경로 연구 (Studies of vindoline metabolism in Catharanthus roseus cell cultures using deuterium-labeled tabersonine)

  • Catharanthus roseus로부터 생산되는 빈카알칼로이드는 암을 치료하는 데에 사용되는 가장 중요한 천연물 중의 하나이다. 이러한 항암제는 두 단량체 인돌 알칼로이드인 catharanthine과 vindoline의 결합으로 합성될 수 있다. 이 중 vindoline의 생합성에 관계하는 경로를 조사하기 위해서 tabersonine의 메틸기에 중수소를 치환한 전구체인 tabersonine-$CD_3$ 1a를 합성하였다. 이는 중수소의 질량 증가로 인해 자연에서 발생하는 tabersonine과 뚜렷이 구별될 수 있도록 해 준다. 우리는 이 중소소가 치환된 tabersonine 1a가 성공적으로 vindoline 생합성경로에 편입되어 중수소가 치환된 3개의 새로운 vindoline 중간체(2a, 3a와 4a)를 생성함을 보였다. 세포현탁배양액에 tabersonine-$CD_3$ 1a를 주입한 5일과 13일째에 생성된 대사물인 16-Hydroxytabersonine-$CD_3$ (m/z 356) 2a, 16-Methoxytabersonine-$CD_3$ (m/z 370) 3a와 16-Methoxy-2,3-dihydro-3- hydroxytabersonine-$CD_3$ (m/z 388) 4a의 생성량을 UPLC/MS를 사용하여 측정하였다. 출발물 1a에서 생성물 2a, 그리고 2a에서 3a로의 전환은 빨랐던 반면 3a에서 4a로의 전환은 상대적으로 훨씬 느렸다. 따라서 각 대사물들의 상대적 전환속도를 서로 비교해 보았을 때 vindoline 생합성 과정의 첫 세 단계 중에서 가장 느린 마지막 단계가 속도결정단계임을 암시한다. 즉 대사물 3a에서 4a로의 느린 전환속도의 결과, 배양 후 13일까지도 3a의 축적률이 현저히 증가됨을 보였다. 배양 5일째의 대사물 2a, 3a와 4a의 축적비는 각각 1, 2와 0.1이었다. 그러나 desacetoxyvindoline-$CD_3$ 5a, deacetylvindoline-$CD_3$ 6a와 vindoline-$CD_3$ 7a의 피크는 배양 13일 후에도 발견되지 않아 세포현탁 배양액에 각 생합성단계와 관련된 효소들이 존재하지 않음을 알 수 있었다.

초음파 화학법에 의한 CdSe 나노 입자의 합성 (Sonochemical Synthesis of CdSe Nanoparticles from Mixed Aqueous Solution)

  • Cadmium selenide (CdSe) nanoparticles with the diameter of about 3.4nm have been synthesized from the mixed aqueous solution of distilled water and diethanolamine at room temperature. The cadmium chloride ($CdCl_2$), sodium selenosulfate ($Na_2SeSO_3$) were used as the cadmium and selenium source, respectively. The properties of CdSe nanoparticles were characterized by using transmission electron microscopy (TEM), X-ray photoelectron spectroscopy (XPS), X-ray diffraction (XRD) and UV-Vis measurements. CdSe nanoparticles were analyzed to be cubic phase with the absorption excition peaks between 540 and 600 nm. CdSe nanoparticles also showed red-shifted excition peaks with increasing the sonication time. This paper mainly presents the sonication effects on the formation of CdSe nanoparticles prepared from the mixed aqueous solution of distilled water and diethanolamine.
