Chl a Fluorescence Characterization and Biomarker Selection from Ricciocarpos natans under Cadmium Stress (카드뮴 독성 평가를 위한 은행이끼의 엽록소형광 분석 및 환경지표 선발)

  • The effects of cadmium ions ($Cd^{2+}$) on the Chl a fluorescence of Ricciocarpos natans were investigated in order to determine whether Chl fluorescence can be used as a biomarker to estimate the physiological responses of plants to cadmium stress. In all plants treated with $Cd^{2+}$, the image of Fv/Fm, which represents the maximum photochemical efficiency of PSII, changed as the $Cd^{2+}$ concentration increased, when treated for 48 h or more. Changes of ${\Phi}_{PSII}$ and $Q_P$ images were recognized even at 10 ${\mu}M$ $Cd^{2+}$. The Chl a O-J-I-P fluorescence transient was also affected even at 10 ${\mu}M$ $Cd^{2+}$. The fluorescence yield decreased considerably in steps J, I and P in plants treated with $Cd^{2+}$, although a typical polyphasic rise was observed in non-treated plants. The Chl fluorescence parameters, Fm, Fv/Fo, Sm, SFIabs, PIabs and ETo/CS, decreased as the $Cd^{2+}$ concentration increased, while the Mo and Kn parameters increased. Peroxidase activity decreased significantly and catalase activity increased as the $Cd^{2+}$ concentration increased. Because of its sensitivity to $Cd^{2+}$ Ricciocarpos natans is useful in experiments investigating the responses of plants to cadmium exposure. Several parameters (Fm, Fv/Fo, Sm, SFIabs, PIabs, ETo/CS, Mo and Kn) can be applied to determine quantitatively the physiological states of plants under cadmium stress.

Stabilization Behavior of Heavy Metal ions by Treatment Conditions (처리조건에 따른 중금속 이온의 안정화 거동)

  • Cation exchange capacity of clay, white clay and zeolite was measured by the adsorption test for 3 different heavy metal (Cd, Cr, Zn) standard solutions whose concentrations were varied by 10, 20, 30 ppm and pH were varied by 3, 5, 7, 9, respectively. The adsorption rate of Cd and Zn increased with increasing pH and slowly increased with increasing pH above pH 5. However, adsorption rate of Cr did not increase with increasing pH. Especially, Cr adsorption rate of the mixture of clay and white clay at pH 5 showed an half decrease compared to that at pH 3. The adsorption rate of mixed heavy metal solutions was in the order of Cd, Zn > Cr; however, the order was changed by Fe>Pb, Cu>Cr>Zn>Cd in case of Cu, Fe and Pb addition.

GM-CSF Grown Bone Marrow Derived Cells Are Composed of Phenotypically Different Dendritic Cells and Macrophages

  • Granulocyte-macrophage colony stimulating factor (GM-CSF) has a role in inducing emergency hematopoiesis upon exposure to inflammatory stimuli. Although GM-CSF generated murine bone marrow derived cells have been widely used as macrophages or dendritic cells in research, the exact characteristics of each cell population have not yet been defined. Here we discriminated GM-CSF grown bone marrow derived macrophages (GM-BMMs) from dendritic cells (GM-BMDCs) in several criteria. After C57BL/6J mice bone marrow cell culture for 7 days with GM-CSF supplementation, two main populations were observed in the attached cells based on MHCII and F4/80 marker expressions. GM-BMMs had $MHCII^{low}F4/80^{high}$ as well as $CD11c^+CD11b^{high}CD80^-CD64^+MerTK^+$ phenotypes. In contrast, GM-BMDCs had $MHCII^{high}F4/80^{low}$ and $CD11c^{high}CD8{\alpha}^-CD11b^+CD80^+CD64^-MerTK^{low}$ phenotypes. Interestingly, the GM-BMM population increased but GM-BMDCs decreased in a GM-CSF dose-dependent manner. Functionally, GM-BMMs showed extremely high phagocytic abilities and produced higher IL-10 upon LPS stimulation. GM-BMDCs, however, could not phagocytose as well, but were efficient at producing $TNF{\alpha}$, $IL-1{\beta}$, IL-12p70 and IL-6 as well as inducing T cell proliferation. Finally, whole transcriptome analysis revealed that GM-BMMs and GM-BMDCs are overlap with in vivo resident macrophages and dendritic cells, respectively. Taken together, our study shows the heterogeneicity of GM-CSF derived cell populations, and specifically characterizes GM-CSF derived macrophages compared to dendritic cells.

  • Accumulation of cadmium (Cd) was investigated in organs (gill, liver, kidney, intestine) of olive flounder (Paralichthys olivaceus) exposed to sub-lethal concentration (5, 10, 50, 100 $\mu$g/L) of Cd for 30 days. Cd exposure resulted in an increased Cd accumulation in liver of flounder for exposure periods and Cd concentration, and Cd accumulation increased linearly with the exposure time. At 20 days of Cd exposure. the order of Cd accumulation in organs was Bill>intestine>liver>kidney and after 30 days of exposure, Cd accumulation in organs was intestine>liver>gill>idney. An inverse relationship was observed between the accumulation factor (AS) and the exposure level, but AE shows an increase with exposure time.

CD40-CD40 Ligand Interactions in the Production of IL-12 and IFN-γ by Tuberculous Pleural Mononuclear Cells

  • Background: Our previous study showed that purified protein derivative (PPD)-stimulated pleural mononuclear cells (PMC) from tuberculous pleurisy (Tbp) produced significantly more $IFN-{\gamma}$ (10- to 70-fold) after in vitro PPD stimulation than freshly isolated pleural cells from malignant pleurisy. The present study was designed to determine whether blocking the CD40-CD40 ligand (CD40L) interaction decreases $IFN-{\gamma}$ production by altering IL-12 levels. Methods: IL-12 and $IFN-{\gamma}$ production after neutralizing anti-CD40L antibody treatment was compared to the efficacy of anti-CD80, anti-CD86, and a combination of anti-CD80 and CD86 (CD80+86) monoclonal antibodies (mAb). These activities were measured by enzyme-linked immunosorbent assays (ELISAs) and reverse transcription-polymerase chain reaction (RT-PCR), after in vitro stimulation with PPO antigen (Ag). Results: Neutralization of CD80, CD86 and CD80+86 did not decrease $IFN-{\gamma}$ and IL-12 production in Tbp-PMC, whereas neutralization of CD40L significantly depressed IL-12 p40 and $IFN-{\gamma}$. In addition, neutralization of CD40L completely inhibited IL-12 p40 and $IFN-{\gamma}$ mRNA expression. Conclusion: The CD40-CD40L interaction might play a major role in IL-12 and $IFN-{\gamma}$ production in Tbp-PMC, thus contributing to protective immunity in human tuberculosis.

Photo-conductive properties of CdS thin film deposited on glass substrate (글라스 기판위에 증착한 CdS 박막의 광전특성 평가)

  • Photo-conductive properties of CdS films deposited on glass substrates by a reactive sputtering in Ar atmosphere were characterized as a function of working pressure and the film thickness. The XRD measurements of CdS films revealed obvious (002) preferred orientation. In 300nm-thick of films, difference between dark and photo-resistance increases with increasing working pressure within the films. The films at 5 mTorr of working pressure show a dark resistance of approximately $1\;{\times}\;10^6\;{\Omega}/{\square}$ and a photo-resistance of $3\;{\times}\;10^4\;{\Omega}/{\square}$. The decrease dark- and photo-resistance of films as thickness decrease were $1.4\;{\times}\;10^6$ and $3\;{\times}\;10^4\;{\Omega}/{\square}$, respectively. CdS films deposited on glass substrates are considered tobe suitable for photo-conductivity materials in stealth radome applications.

Cd(II) and Zn(II) Complexes Containing N,N'-Bidentate N-(Pyridin-2-ylmethylene)cyclopentanamine: Synthesis, Characterisation and Methyl Methacrylate Polymerisation

  • The reaction between $[CdBr_2{\cdot}4H_2O]$ and anhydrous $[ZnCl_2]$ with N,N'-bidentate N-(pyridin-2-ylmethylene)-cyclopentanamine (impy) in ethanol yields dimeric $[(impy)Cd({\mu}-Br)Br]_2$ and monomeric $[(impy)ZnCl_2]$ complexes, respectively. The X-ray crystal structure of Cd(II) and Zn(II) complexes revealed that the cadmium atom in $[(impy)Cd({\mu}-Br)Br]_2$ and zinc in $[(impy)ZnCl_2]$ formed a distorted trigonal-bipyramidal and tetrahedral geometry, respectively. Both complexes showed moderate catalytic activity for the polymerisation of methyl methacrylate (MMA) in the presence of modified methylaluminoxane (MMAO), with polymethylmethacrylate (PMMA) syndiotacticity of about 0.70.

  • The electrodeposition in acidic aqueous electrolyte bath of cadmium telluride on gold electrodes has been studied by electrochemical analysis. Conventional cyclic voltammetry using potentiostat is considered as a reliable method to study electrochemical behavior of electrodeposition of CdTe. In this paper, the mechanism of CdTe deposition and its cyclic voltammetry were studied with the Te ion concentration, temperature, potential, and scan rate. We also investigated surface morphologies using FESEM and atomic composition of Cd and Te using EDS. Atomic composition of Cd and Te were varied with Te ion concentration in the electrolyte.

Optical Constants and Dispersion Parameters of CdS Thin Film Prepared by Chemical Bath Deposition

  • CdS thin film was prepared on glass substrate by chemical bath deposition in an alkaline solution. The optical properties of CdS thin film were investigated using spectroscopic ellipsometry. The real (${\varepsilon}_1$) and imaginary (${\varepsilon}_2$) parts of the complex dielectric function ${\varepsilon}(E)={\varepsilon}_1(E)+i{\varepsilon}_2(E)$, the refractive index n(E), and the extinction coefficient k(E) of CdS thin film were obtained from spectroscopic ellipsometry. The normal-incidence reflectivity R(E) and absorption coefficient ${\alpha}(E)$ of CdS thin film were obtained using the refractive index and extinction coefficient. The critical points $E_0$ and $E_1$ of CdS thin film were shown in spectra of the dielectric function and optical constants of refractive index, extinction coefficient, normal-incidence reflectivity, and absorption coefficient. The dispersion of refractive index was analyzed by the Wemple-DiDomenico single-oscillator model.

Development of a CD-ROM based Educational Program for the Prevention of Sexual Harassment and Violence in Middle School Students (중학생을 위한 성희롱.성폭력 예방교육 CD-ROM 개발)

  • Purpose: The purpose of this study was to develop a CD-ROM title for the prevention of sexual harassment and violence in middle school students. Method: The process for developing the CD-ROM title consisted of 10 steps proposed by Alessi and Trollip. Results: This CD-ROM title was composed of 4 domains, 11 subjects, and 31 detailed subjects. The 4 domains were: 1) understanding of sexual harassment and violence, 2) cases of sexual harassment and violence, 3) prevention and coping strategies, and 4) sexual knowledge. The subjects included texts, and the detailed subjects included graphics, animations, videos, and pictures. Conclusion: This CD-ROM may reduce the occurrence of sexual harassment and violence and accommodate enriched humanity in middle school students.
