Risk based pollution level of Pb and Cd in metal contaminated soils depending on physicochemical properties of soil in a target site was assessed using biotic ligand model. Heavy metal activity in soil solution defined as exposure activity (EA) was assumed to be toxic to Vibrio fischeri and soil organisms. Predicted effective activity (PEA) determined by biotic ligand model was compared to EA value to calculate risk quotient. Field contaminated soils (n = 10) were collected from a formes area and their risk based pollution levels were assessed in the present study using the calculated risk quotient. Concentrations of Pb determined by aqua regia were 295, 258, and 268 mg/kg in B, H and J points and concentrations of Cd were 4.73 and 6.36 mg/kg in G and I points, respectively. These points exceeded the current soil conservation standards. However, risk based pollution levels of the ten points were not able to be calculated because concentrations of Pb and Cd in soil solution were smaller than detection limits or one (i.e., non toxic). It was because heavy metal activity in soil solution was dominant toxicological form to organisms, not a total heavy metal concentration in soil. In addition, heavy metal toxicity was decreased by competition effect of major cations and formation of complex with dissolved organic carbon in soil solution. Therefore, it is essential to consider site-specific factors affecting bioavailability and toxicity for estimating reliable risk of Pb and Cd.
Amyloid-${\beta}$-peptide ($A{\beta}$) is important in the pathogenesis of Alzheimer's disease (AD). Calpain ($Ca^{2+}$-dependent protease) and caspase-8 (the initiating caspase for the extrinsic, receptor-mediated apoptosis pathway) have been implicated in $AD/A{\beta}$ toxicity. We found that $A{\beta}$ promoted degradation of calpastatin (the specific endogenous calpain inhibitor); calpastatin degradation was prevented by inhibitors of either calpain or caspase-8. The results implied a cross-talk between the two proteases and suggested that one protease was responsible for the activity of the other one. In neuron-like differentiated PC12 cells, calpain promotes active caspase-8 formation from procaspase-8 via the $A{\beta}$ and CD95 pathways, along with degradation of the procaspase-8 processing inhibitor caspase-8 (FLICE)-like inhibitory protein, short isoform (FLIPS). Inhibition of calpain (by pharmacological inhibitors and by overexpression of calpastatin) prevents the cleavage of procaspase-8 to mature, active caspase-8, and inhibits FLIPS degradation in the $A{\beta}$-treated and CD95-triggered cells. Increased cellular Ca2+ per se results in calpain activation but does not lead to caspase-8 activation or FLIPS degradation. The results suggest that procaspase-8 and FLIPS association with cell membrane receptor complexes is required for calpain-induced caspase-8 activation. The results presented here add to the understanding of the roles of calpain, caspase- 8, and CD95 pathway in $AD/A{\beta}$ toxicity. Calpain-promoted activation of caspase-8 may have implications for other types of CD95-induced cell damage, and for nonapoptotic functions of caspase-8. Inhibition of calpain may be useful for modulating certain caspase-8-dependent processes.
Cadmium (Cd) is non-essential heavy metal that negatively affects plant metabolism. Nitric oxide (NO) is an increasingly important molecule for plant metabolism that makes signaling. In this study, it was aimed to investigate the alleviating effect of sodium nitroprusside (SNP) application as NO donor in white poplar (Populus alba) under Cd stress conditions. SNP and without SNP treatments increased the Cd accumulation in root tissue. While photosynthetic pigments (Chl a, Chl b, Chl a+b, and carotenoid) content decreased by only Cd application, SNP+Cd application decreased the rate of photosynthetic pigments reduction. When the results of Cd and Cd+SNP applications were evaluated for mineral (Fe, Zn, Mn and Cu) uptake, it was found that the positive effect of SNP was heterogeneously affected. Depending on SNP application, it was found that malondialdehyde (MDA) amount decreased in leaf in 100 µM Cd applications while hydrogen peroxide (H2O2) amount decreased in 100 and 500 µM Cd applications. When antioxidant enzyme activities were examined, it was found that catalase (CAT) and ascorbate peroxidase (APX) enzyme activities increased with 100 µM SNP applications under all Cd applications. As a result, it was found that SNP application under Cd stress generally supports physiological processes positively in white poplar, suggesting that NO molecule plays important alleviating roles in plant metabolism.
퇴적물 내 황화물(acid volatile sulfide, AVS)이 저서 생물의 금속 생물 축적 및 독성 반응에 어떠한 영향을 미치는 지를 이해하기 위해서 해양 다모류인 Neanthes arenaceodentata를 이용한 퇴적물 노출실험을 수행하였다. 이를 위해서 세 개의 다른 AVS 농도군에 대조구를 포함한 다섯 개의 농도 구배로 Cd, Ni, Zn를 오염시킨 퇴적물에서 N. arenaceodentata를 20일간 배양한 후 실험생물의 체내 금속 축적량과 그에 따른 사망률 및 성장률을 조사하였다. N. arenaceodentata에 의한 금속의 생물축적은 Cd과 Zn의 경우 AVS 농도의 영향을 받아서 해수(overlying water, OW)내 용존 금속 농도에 비례해서 증가했다. Ni은 AVS농도에 영향을 받지 않고 퇴적물 내 금속(simultaneously extracted metals. SEM)농도에 비례해서 증가했다. N. arenaceodentata의 사망과 성장률 저해현상은 SEM과 AVS 간의 몰농도차가 영보다 큰 조건([SEM-AVS]>0)에서만 관찰되었는데 용존 Zn에 의한 결과로 추정되었다. OW-Zn의 20-d LC50값은 9.3(8.0$\pm$11.0) $\mu$M이었다. 사망률에 대한 체내 Zn 농도의 최소영향농도(LOEC)는 7.8 $\mu$mol/g이었고, 최대무영향농도(NOEC)는 6.2$\mu$mol/g이었다. 성장률 저해에 대한 체내 Zn 농도의 LOEC는 5.9$\mu$mol/g이었고, NOEC은5.1 $\mu$mol/g토이었다. 본 실험에서는 실험실 조건에서 인위적으로 오염시킨 퇴적물 내 Zn의 입자상 Zn 농도와 용존 Zn농도의 비 (K$_{d}$ )가 현장 퇴적물에 비해서 10배 정도 감소함으로써 결국 용존 Zn에 의한 독성이 과대평가된 것으로 보인다.
We recently showed that polylysine, the polymer of lysines, retains anti-prion activity. Although the effectiveness of prion inhibition by polylysine was demonstrated with the regimen tolerated in mice, a determination of quantitative polylysine toxicity is necessary to precisely address the in vivo toxicity level of polylysine. In this communication, we report the results of body weight monitoring and hematologic tests performed in CD-1 mice that received two different tolerable dosages of polylysine for an either 5-day or 4-week period. We found that there was no significant alteration in overall serum chemistry, blood cell count, and body weight gain compared with controls. The only notable quantitative change with statistical significance was the decrease of platelet numbers in mice subchronically administered with polylysine. Our results suggest that polylysine is harmless in mice if administered for a short period, but administrations of polylysine in mice may require considerate attention for safety in future investigations as mice chronically receive tolerable doses of polylysine.
This study was performed to investigate the applicability of 9 chromosome aberration and sister chromatid exchange analysis for the assessment of cytotoxicity and cytogenetic effects of cadmium. Induction of chromosome aberration and sister chromatid exchange in CHO-K1 cells and human peripheral lymphocytes by 2 hour-treatment of CdCl$_{2}$ with various concentrations was observed in relation to their frequencies and types of aberration. The frequency of chromosome aberration in CHO cells treated with CdCl$+{2}$ at G$_{1}$ was increased with dose-dependent manner. When human peripheral lymphocytes were treated with cadmium at G0 and harvested at 72 hours there after, the response was dose-dependent and all the aberrations were also chromatid types. There was no significant increase in frequencies of sister chromatid exchange in both CHO cells and human lymphocytes treated with different concentrations of cadmium. It was suggested that SCE analysis was not a good assessment method for cadmium toxicity.
In this study, we examined the influences of pH and total concentration on the speciation of heavy metals (Cd, Cu, Zn) in acid mine drainage. Their labile concentrations were analyzed by Anodic Stripping Voltammetry (ASV) at both natural pH and adjusted pHs (from 2 to 8). We obtained regression equations for predicting labile concentrations as a function of the water pH and contamination level (total dissolved metal concentration). Our data show that labile Cu depends on both the total concentration and pH, while labile Cd and Zn concentrations are controlled mainly by their total concentration rather than pH. Therefore, the pH variation of AMD may significantly change the toxicity and bioavailability especially of Cu, owing to its speciation change.
This study was conducted to investigate the effect of dietary zinc (Zn) and/or cadmium (Cd) on hepatic microsomal and cytosol enzyme activities. Male Spraque-Dawley rats (110$\pm$10g ) received zinc (0, 30 and 300 ppm/) and Cd-treated groups were administered oral intubation with Cd chloride (5.0mg/kg of body weight 0 at the same time once a week. The effect of Cd on the activity of hepatic cytochromep-450 , xanthine oxidase(X. O) and superoxide dismutase (SOd) was studied in rats. Cd oral intubation resulted in a decrease in cytochrome P-450 content and SOD activity whereas a significant increase in the X.O. activity was observed was observed . Intake of excessive Zn led to an increased activity of microsomal alcohol dehydrogenase (ADH) , whereas Zn deficiency group led to a decreased group. The mechanism by which Zn induces the decreasing of Cd toxicity in rats, seems to rely on the protection of the enzyme systems P-450, ADH, aldehyde dehydrogenase (ALDH) and X.O. in the liver, possibly by forming non-toxic Cd metallothionein. These results indicate that Zn and Cd regulation might occur via inhibitory protein component of the $H_2O$$_2$ -generator system.
The standardized activated sludge for the biodegradation test of anion surfactants has been produced from the collected microorganisms in the soil and the wastewaters treatment plant. The activated sludge was kept under control of the pH, dissolved oxygen, microorganisms and inoculated the basal medium flasks with LAS and LAS mixed with heavy metals [Cd(II), Cu(II), Zn(II)]. Based of results, the inhibition effect(%) of heavy metals in LAS biodegradation were 1. All 1% when LAS 30mg/l-Cd(II), Cu(II) and Zn(II) 0.1mg/l, respectively 2. All 1${\sim}$10% when LAS 30mg/l-Cd(II), Cu(II) and Zn(II) 1mg/l, respectively 3. All 10${\sim}$40% when LAS 30mg/l-Cd(II), Cu(II) and Zn(II) 10mg/l, respectively 4. All 30${\sim}$65% when LAS 30mg/l-Cd(II), Cu(II) and Zn(II) 100mg/l, respectively And toxicity order of heavy metals to the microorganisms in LAS biodegradation were Cd>Cu>Zn in low concentration(0.1${\sim}$1mg/l)and Cd>Zn>Cu in high concentration(10${\sim}$100mg/l).
As cultured plant cells can grow in high oxidative stress conditions, they form an excellent system to study antioxidant mechanisms and the mass production of antioxidants. Oxidative stress is a major cause of damage in plants exposed to various types of environmental stress, including heavy metals, such as cadmium (Cd). Heavy metal accumulation can interfere with many cell functions and plant growth. To evaluate the contribution of oxidative stress to Cd-induced toxicity, cultured sweetpotato (Ipomoea batatas) cells were treated with increasing concentrations of Cd (0, 10, 25, and 50 μM) and cultured further. Cell growth was significantly inhibited by 25 and 50 μM of Cd, and the total protein content increased with 50 μM of Cd. Additionally, the activity of peroxidase (POD) and ascorbate peroxidase (APX), antioxidant enzymes that remove hydrogen peroxide (a reactive oxygen species), increased in the cells after treatment with 50 μM of Cd. The expression analysis of POD, APX, and peroxiredoxin (PRX) isolated from sweetpotato cultured cells in a previous study revealed the differential expression of POD in response to Cd. In this study, the expression levels of several acidic POD (swpa2, swpa3, and swpa4) and basal POD (swpb1, swpb2, and swpb3) genes were increased in Cd-treated cultured cells. These results indicate that Cd-mediated oxidative stress is closely linked to improved POD-mediated antioxidant defense capacity in sweetpotato suspension-cultured cells.
