The [M(cod)(PPh$_3)_2] PF_6$ (M = Rh, Ir; cod = 1,5-cyclooctadiene) Mediated Activiation of Aldehyde C-H Bond

  • Acetone solution of quinoline-8-carbaldehyde reacts with $[Rh(cod)(PPh_3)_2] PF_6$and $[Ir(cod)(PPh_3)_2] PF_6$ to yield $[Rh(NC_9H_6CO)(H)(PPh_3)_2(CH_3COCH_3)] PF_6$ (1) and $[Ir(NC_9H_6CO)(H)(PPh_3)_2(CH_3COCH_3)] PF_6$ (2), respectively. The compound $[Ir(cod)(PPh_3)_2] PF_6$ also reacts with $Ph_2PC_6H_4-o-CHO$ in the acetone / $H_2O$ mixture to give $[Ir(Ph_2PC_6H_4-o-CO)(H)(PPh_3)_2(CH_3COCH_3)] PF_6$ (3). Compounds 1, 2, and 3 were characterized by infrared, $^1H$ NMR, $^{31}P$ NMR spectra and conductivity measurement. The $^1H$ NMR spectra of 1, 2, and 3 support the presence of a terminal hydride that is cis to the phosphine. The IR band of 3 at 2185 $cm^{-1}$, which is assigned to $\nu$(Ir-H), and the hydride cleavage reaction of 3 with $CCl_4$, provide evidence for the Ir-H bond.

A study on ITZ percolation threshold in mortar with ellipsoidal aggregate particles

  • The percolation of interfacial transition zone (ITZ) in cementitious materials is of great importance to the transport properties and durability issues. This paper presents numerical simulation research on the ITZ percolation threshold of mortar specimens at meso-scale. To simulate the meso-scale model of mortar as realistically as possible, the aggregates are simplified as ellipsoids with arbitrary orientations. Major and minor aspect ratios are defined to represent the global shape characteristics of aggregates. Some algorithms such as the burning algorithm, Dijkstra's algorithm and Connected-Component Labeling (CCL) algorithm are adopted for identification of connected ITZ clusters and percolation detection. The effects of gradation and aspect ratios of aggregates on ITZ percolation threshold are quantitatively studied. The results show that (1) the ITZ percolation threshold is mainly affected by the specific surface area (SSA) of aggregates and shows a global decreasing tendency with an increasing SSA; (2) elongated ellipsoidal particles can effectively bridge isolated ITZ clusters and thus lower the ITZ percolation threshold; (3) as ITZ volume fraction increases, the bridging effect of elongated particles will be less significant, and has only a minor effect on ITZ percolation threshold; (4) it is the ITZ connectivity that is essentially responsible for ITZ percolation threshold, while other factors such as SSA and ITZ volume fraction are only the superficial reasons.

Effect of ChungHuyl-Plus on inflammatory factors in Human Umbilical Vein Endothelial Cells (HUVECs) (청혈플러스가 혈관내피세포에서 염증 지표인자에 미치는 영향)

  • Objectives : Coronary and cerebrovascular disease with high mortality is a major factor in arteriosclerosis. Pro-inflammatory cytokines damage vascular endothelial cells, leading to vascular inflammation. These vascular inflammation can build up cholesterol and thrombus to cause atherosclerosis. Methods : In this study, we researched the effect of ChungHyul-Plus for vascular inflammation in human umbilical vein endothelial cells (HUVECs) stimulated with tumor necrosis $factor-{\alpha}$ ($TNF-{\alpha}$). Change in mRNA expression of inflammatory cytokines (CCL5, CXCL8, CX3CL1, and MCP-1), cell adhesion molecules (VCAM-1 and ICAM-1), and anti-inflammation modulators (KLF2 and eNOS) were quantified by qRT-PCR. Results : ChungHyul-Plus decreased expression of inflammatory cytokines and cell adhesion molecules and increased anti-inflammation modulators expression in $TNF-{\alpha}$ stimulated HUVECs. Conclusions : These results suggest that ChungHyul-Plus can be used in the treatment and prevention of vascular inflammation and arteriosclerosis.

  • Gaucher disease (GD, OMIM #230800 OMIM#230800) is a rare, autosomal recessive inherited metabolic disorder caused by mutation in GBA1 encoding the lysosomal enzyme, glucocerebrosidase. The deficiency of glucocerebrosidase leads to an accumulation of its substrate, glucosylceramide in macrophages of various tissues. Common clinical manifestations include cytopenia, splenomegaly, hepatomegaly, and bone lesions. The phenotype of GD is classified into three clinical categories: Type 1 (non-neuronopathic) is characterized by involvements on the viscera, whereas types 2 and 3 (neuronopathic) are associated with not only visceral symptoms but also neurological impairment, either severe in type 2 or variable in type 3. A diagnosis of GD can be confirmed by demonstrating the deficiency of acid glucocerebrosidase activity in leukocytes. Mutations in the GBA1 should be identified as they may be of prognostic value in some cases. Biomarkers including Chitotriosidase, CCL18, and glucosylsphingosine (lyso-GL1) are useful in diagnosis and treatment monitoring. Currently available disease-specific treatment in Korea consists of intravenous enzyme replacement therapy and substrate reduction therapy. For enhancing long-term prognosis, the onset of Parkinson's disease and Lewy body dementia, or the occurrence of a blood disease or cancer (hepatocellular carcinoma) should be monitored in older patients. The development of new strategies that can modify the neurological phenotype are expected, especially in Asia including Korea, where the prevalence of neuronopathic GD is relatively higher than that in western countries.

Heat-Killed Lactiplantibacillus plantarum LRCC5314 Mitigates the Effects of Stress-Related Type 2 Diabetes in Mice via Gut Microbiome Modulation

  • The incidence of stress-related type 2 diabetes (stress-T2D), which is aggravated by physiological stress, is increasing annually. The effects of Lactobacillus, a key component of probiotics, have been widely studied in diabetes; however, studies on the effects of postbiotics are still limited. Here, we aimed to examine the mechanism through which heat-killed Lactiplantibacillus plantarum LRCC5314 (HK-LRCC5314) alleviates stress-T2D in a cold-induced stress-T2D C57BL/6 mouse model. HK-LRCC5314 markedly decreased body weight gain, adipose tissue (neck, subcutaneous, and epididymal) weight, and fasting glucose levels. In the adipose tissue, mRNA expression levels of stress-T2D associated factors (NPY, Y2R, GLUT4, adiponectin, and leptin) and pro-inflammatory factors (TNF-α, IL-6, and CCL-2) were also altered. Furthermore, HK-LRCC5314 increased the abundance of Barnesiella, Alistipes, and butyrate-producing bacteria, including Akkermansia, in feces and decreased the abundance of Ruminococcus, Dorea, and Clostridium. Thus, these findings suggest that HK-LRCC5314 exerts protective effects against stress-T2D via gut microbiome modulation, suggesting its potential as a supplement for managing stress-T2D.

Characterization of composite prepared with different mixing ratios of TiO2 to activated carbon and their photocatalytic activity

  • In this work, pitch/activated carbon/$TiO_2$ composite were prepared by $CCl_4$ solvent method with different mixing ratios. The BET surface area of pitch/activated carbon/$TiO_2$ composite has a significantly increase with increasing activated carbon content in pitch/activated carbon/$TiO_2$ composite. The surface structure and elemental compositions of the composite were studied by SEM and EDX, respectively. The SEM results were presented to the characterization of porous texture on the pitch/activated carbon/$TiO_2$ composite. And EDX data was shown the presence of C, O, S, Ti and other elements. The structural properties of the composite were studied in XRD measurements. The $TiO_2$ crystal phases of the pitch/activated carbon/$TiO_2$ composite had lots of rutile-type structure which transforms from anatase-type with a little of anatase-type structure. The photocatalytic activities of the composite were evaluated using a photo-decomposition method under UV lamp. The pitch/activated carbon/$TiO_2$ composites were observed better photocatalytic activity than that of pristine $TiO_2$.

Single Cell Transcriptomic Re-analysis of Immune Cells in Bronchoalveolar Lavage Fluids Reveals the Correlation of B Cell Characteristics and Disease Severity of Patients with SARS-CoV-2 Infection

  • The coronavirus disease 2019 (COVID-19) pandemic (severe acute respiratory syndrome coronavirus 2) is a global infectious disease with rapid spread. Some patients have severe symptoms and clinical signs caused by an excessive inflammatory response, which increases the risk of mortality. In this study, we reanalyzed scRNA-seq data of cells from bronchoalveolar lavage fluids of patients with COVID-19 with mild and severe symptoms, focusing on Ab-producing cells. In patients with severe disease, B cells seemed to be more activated and expressed more immunoglobulin genes compared with cells from patients with mild disease, and macrophages expressed higher levels of the TNF superfamily member B-cell activating factor but not of APRIL (a proliferation-inducing ligand). In addition, macrophages from patients with severe disease had increased pro-inflammatory features and pathways associated with Fc receptor-mediated signaling, compared with patients with mild disease. CCR2-positive plasma cells accumulated in patients with severe disease, probably because of increased CCL2 expression on macrophages from patients with severe disease. Together, these results support the hypothesis that different characteristics of B cells might be associated with the severity of COVID-19 infection.

Hepatoprotective Effects of Ginseng Intestinal Metabolite IH-901 on Chemical-Induced Hepatic Damage

  • Hepatoprotective effects of white ginseng extract (WGE), and IH-901 (20-O-${\beta}$-D-glucopyranosyl-20(S)-protopanaxadiol) derived from intestinal metabolite of ginsenoside $Rb_1$ were studied using two experimental animal models with chemical-induced hepatic damage. Administration of WGE (200 and 500 mg/kg) and IH-901 (0.01, 0.05, and 0.1 mM/kg) significantly decreased aspartate aminotransferase (AST) and alanine aminotransferase (ALT) levels in acute hepatitic mice induced by $CCl_4$. Administration of WGE (l00 mg/kg) and IH-901 (0.02, 0.04, and 0.08 mM/kg) significantly decreased AST and ALT levels in acute hepatitic rats induced by D-galactosamine. AST and ALT levels of IH-901 groups decreased. These results suggested WGE and IH-901 may have protective effects against chemical-induced hepatic damage.

The Study of Free Radical Scavenging Effect of Lycii Fructus by Liver Injury of Rats (백서 간손상에 의한 구기자의 유리자유기 소거능에 관한 연구)

  • In the present study, we investigated the protective effect of the Lycii Fructus water extracts (LFE) against CCl4-induced hepatotoxicity and the mechanism underlying these protective effects in the rats. The pretreatment of LFE has shown to possess a significant protective effect by lowering the serum alanine and aspartate aminoteansferase (AST and ALT) and alkaline phosphatase (ALP). This hepatoprotective action was confirmed by histological observation, In addition, the pretreatment of LFE prevented the elevation of hepatic malondialdehyde (MDA) formation and the depletion of reduced glutathione (GSH) content and catalase activity in the liver of CC1₄-injected rats. The LFE also displayed hydroxide radical scavenging activity in a dose-dependent manner (IC50 = 83.6 μg/ml), as assayed by electron spin resonance (ESR) spin-trapping technique. Moreover, the expression of cytochrome P450 2E1 (CYP2E1) mRNA, as measured by reverse transcriptase-polymerase chain reaction (RT-PCR), was significantly decreased in the liver of LFE-pretreated rats when compared with that in the liver of control group. Based on these results, it was suggested that the hepatoprotective effects of the LFE may be related to antioxidant effects and regulation of CYP2E1 gene expression.
