CCNU, Vinblastine and Prednisone Treatment for Grade II Dermal Mast Cell Tumor in a Yorkshire terrier dog (CCNU, Vinblastine과 Prednisone으로 병용 치료한 요크셔 테리어 개의 Grade II 피부 비만세포종 증례)

  • An 11-year-old, castrated male Yorkshire terrier dog was presented with multiple plaques on right inguinal region. Grade II mast cell tumor was diagnosed. The dog was treated with Vinblastine and prednisone(PDS) initially. Because of poor response of the dog, CCNU was added for more aggressive treatment. After 5 weeks treatment of with CCNU, vinblastine and PDS, the lesion was improved. Moderate leukopenia was shown after 4 cycles of chemotherapy. The chemotherapy was re-administered since the patient recovered from the leukopenia. Though the same protocol was applied, no improvement of the lesion was observed. Moreover, the general body condition of the dog became worse and was euthanized by the owner's request. Necropsy was not permitted. The survival time was 330 days after start of the chemotherapy.

Ploidy Characteristics of NS-1 Cells Resistant to BCNU or CCNU and Immune mediated Cytotoxicity in vitro (항암제와 세포독성에 내성을 나타내는 생쥐 임파종 세포(NS-1)의 염색체 분포특성)

  • In order to study the relationship between resistance of tumor cells to anticancer drugs and immunologic cytotoxicity and their chromosome number, a line of cancer cells (NS-1) was exposed to BCNU and CCNU in vitro. Characteristics of the distribution of chromosome number of the survived cells were then comparatively analyzed. Effect of immune mediated cytotoxicity, i.e. complement and cell-mediated cytotoxicity, on the ploidy characteristics was observed in the same way. NS-1 cells were found to be a population of neoplastic cells of heterogeneity having 5 to 115 chromosomes per cell in metaphase. The majority of the cells were belong to the class of chromosome number 56 to 60 which were considered as the stem cell line. Dramatic changes in the distribution of chromosome number following drug treatment were not observed. However the range of chromosome distribution was slightly changed. Characteristics of chromosomal distribution of drug treated cells were not significantly varied by different doses of drug treated. Changed chromosomal distribution patterns of drug treated cells were reversible, especially the cells having 56 to 60 chromosomes recovered rapidly. Cells having 41-60 and 61-80 chromosomes among cells treated with BCNU and cells with 41-60 chromosomes after CCNU treatment were the major population which regenerated continuously. Following BCNU treatment cells having 61-80 chromosomes were not varied much whereas CCNU treatment affects the population in the same class. Chromosomal aberrations were significantly enhanced by BCNU and CCNU treatment. The frequency of chromosomal aberrations was greater in cells having more than 40 chromosomes compared with that in cells having less than 40 chromosomes. Changes in ploidy characteristics of the cells following complement mediated and cell mediated cytotoxicity were not significant. Therefore it was tentatively concluded that association of numerical distribution pattern of NS-1 cells with the response to the treatment used in this experiment was not recognized.

  • A 13-year-old, neutered male, chihuahua was referred to the Veterinary Medical Teaching Hospital of Chungnam National University with a history of a bilateral mandibular mass, intermittent vomiting, cough, and left sided staggering starting ten days prior. Clinical examination along with radiography, neurologic examination and magnetic resonance imaging (MRI) demonstrated a well-circumscribed and solitary mass in the brain stem. The dura mater was thickened with marked linear enhancement after contrast administration. Based on diagnostic image analysis and histopathologic examination, this case was diagnosed as intracranial meningioma. The patient's symptoms were controlled for 18 months by a combination therapy of prednisolone and lomustine (CCNU), and then he died of at home.

  • Cholesteryl p-[3-(2-chloroethyl_-3-nitrosoureido] phenylthioaccetate (2) was synthesized : an intermediate, p-[3(2-chloroethyl_-3-nitrosoureido] phenylthioacetic acid (1) is a congener of an antitumor chlorambucil which both the -CH$_{2}$CH$_{2}$-linkage and -N(CH$_{2}$CH$_{2}$Cl)$_{2}$ group of chlorambucil molecule is doubly modified into the respective -S-linkage and -NH-CO-NNO-CH$_{2}$CH$_{2}$CL group. The attackment of cholesterol moiety as a carrier group to p-[3-(2-chloroethyl-3nitrosoureido] phenylthioacetic acid was accomplished through the esterification of cholesterol with p-[3-(2-chloroethyl-3-nitrosoureido] phenylthioacetyl chloride which was obtained from the treatment of p-[3-(2-chloroethyl)-3-nitrosourei-do] phenylthioaceticacid with SOCL$_{2}$, p-[3-(2-chloroethyl)ureido]]-phenythioacetic acid was nitrosated with NaNO$_{2}$ IN 98-100% HCOOH to give exclusively p-[3-(2-chloroethyl_-3-nitrosoureido] phenylthioacetic acid. Surgery and Metronomic Chemotherapy in a Pet Rabbit (Oryctolagus cuniculus) with Mammary Gland Adenocarcinoma

  • An 8-year-old female pet rabbit presented at the veterinary clinic for mammary gland palpation due to the presence of a mass. Upon physical examination, a mass was identified in the left fourth mammary gland. Abdominal ultrasonography revealed a 3 × 2 cm mass in the right uterus and general thickening of the endometrium, suggesting uterine sinusitis. Multiple pulmonary nodules suspected to be metastatic lesions were identified on chest radiography. Surgery was performed to mastectomy and ovariohysterectomy (OHE). The histopathological examination of the tumor revealed mammary gland adenocarcinoma (simple-type) with multiple nodules consisting of the proliferation of tumor cells forming tubules containing secretory materials, cellular debris, and solid nests with a central area of necrosis. Metronomic chemotherapy was performed with cyclophosphamide and lomustine (CCNU) based on the histopathological findings. The quality of life has been well maintained, with no specific clinical symptoms observed for 8 months after metronomic chemotherapy. To the best of authors' knowledge, this study is the first to examine the effects of metronomic chemotherapy with cyclophosphamide and lomustine in a pet rabbit.

  • Steroidal nitrosoureas have been synthesized and their antitumor activity on L 1210 cells was evaluated, N-(2-Chloroethyl)-N-nitrosocarbamoyl-3-aza-A-homo-5$\alpha$-cholestane (5a) showed significantly low $ED_{50}$ value of 1.6 $\nu$g/ml whose activity is equivalent to that of methyl-CCNU ($ED_{50}$ = 1.7 mg/ml).

Cytotoxic and Cytotoxicity-Potentiating Effects of the Curcuma Root on L1210 Cell

  • A cytotoxic sesquiterpene against L1210 cell has been isolated from the root of Curcuma domestica. Its structure was identified as ${\beta}-sesquiphellandrene$. The cytotoxicity-potentiating substance was (+)-ar-turmerone. (+)-ar-Turmerone potentiated the cytotoxicity of ${\beta}-sesquiphellandrene$(5 fold in $ED_{50}$ value) and an unknown sesquiterpene which was isolated from the root as well, and that of aurapten(6.3 fold) isolated from the unripe fruit of Poncirus trifoliata. Moreover, it potentiated the cytotoxic activities of MeCCNU 10 fold and cyclophosphamide 10 fold. Except the fact that all the effective cytotoxic substances possess relatively good lipophilicity, no relationship between structures of the cytotoxic substances and the cytotoxicity-enhancing effect of (+)-ar-turmerone could be observed.
