Failure Modes and Effects Analysis for Electric Power Installations of D University (D대학 수변전설비의 고장모드 영향 분석)

  • The purpose of this paper is to carry out Failure Modes and Effects Analysis (FMEA) and use criticality in order to determine risk priority number of the components of electric power installations in Engineering college building of D university. In risk priority number, GROUP A had 7 failure modes; more specifically, Transfomer had 4 modes, Filter(C)(1 mode), LA(1 mode), and CB(MCCB)(1 mode), and thus 4 components had failure modes. In terms of criticality, high-grade group a total of 16 failure modes, and 7 components-LA(1 mode), CB(MCCB)(1 mode), MOF(2 modes), PT(1 mode), Transformer(7 modes), Cable(3 modes), and Filter(C)(1 mode)-had failure modes. Comparison of risk priority number and criticality was made. The components which had high risk priority number and high criticality were Transformer, Filter(C), LA, and CB(MCCB). The components which had high criticality were MOF and cable. In particular, Transformer(RPN: 4 modes, Criticality: 7 modes) was chosen as an intensive management component.

Effect of Cooking Methods with Various Heating Apparatus on the Quality Characteristics of Chicken (가열기구에 따른 조리방법이 닭고기의 품질특성에 미치는 영향)

  • This study showed the quality characteristics of chicken breast meat(CB) and leg meat(CL) with various kinds of existing cooking methods and double layer pan filled with Phase Change Materials(PCM) heating. Steaming resulted the highest moisture contents of 63.9% and 62.1% each in CB and CL. Also, steaming showed the lowest with 1.3% and 8.6% of crude fat in CB and CL(p<0.05) respectively. Crude protein content of CB in charcoal heating and grilling of CL had the highest values of 37.9% and 30.5% each. In the test of crude ash, grilling showed the highest with 2.4% in CB(p<0.05) and oven heating and charcoal heating was the highest with 1.3% in CL(p<0.05). In the test of cooking loss, charcoal heating showed much higher with 33.52% and 41.16% in CB and CL each than the other cooking treatments. And in case of shear force test, $5.93kg/cm^2$ in CB and $6.80kg/cm^2$ in CL were the highest scores in grilling. In the test of color, L value of CB prepared by steaming showed the highest scores of 78.31(p<0.05) while CL by oven heating was the highest of 10.00 in a value. In the overall acceptability test of 9 point-scale sensory evaluation, CB prepared by charcoal heating showed the highest score of 7.25 points in boiling, but the lowest score of 6.00 points in steaming(p<0.05). CL by charcoal heating resulted the highest score of 7.71 points but had no significant difference.

Characterization of Leuconostoc mesenteroides B-742CB Dextransucrase Expressed in Escherichia coli

  • Recombinant E. coli DH5$\alpha$ harboring a dextransucrase gene (dsrB742) produced an extracellular dextransucrase in a 2% sucrose medium. The enzyme was purified by DEAE-Sepharose and Phenyl-Sepharose column chromatographies upto a 142.97-fold purification with a 11.11% recovery to near homogeneity. The enzyme had a calculated molecular mass of 168.6 kDa, which was in good agreement with the activity band of 170 kDa on a nondenaturing SDS-PAGE. An expression plasmid was constructed by inserting the dsrB742 into a pRSET expression vector. The activity after expression in E. coli BL21(DE3)pLysS increased about 6.7-fold compared to the extracellular dextransucrase from L. mesenteroides B-742CB. The expressed and purified enzyme from the clone showed similar biochemical properties (acceptor reaction, size of active dextransucrase, optimum pH, and temperature) to B-742CB dextransucrase, however, the ability to synthesize ${\alpha}$-(1$\rightarrow$3) branching decreased in comparison to that of L. mesenteroides B-742CB dextransucrase.

Purification and Characterization of Very Low Density Lipoprotein in Commercial Broiler and Crossbred Village Chickens by Fast Protein Liquid Chromatography

  • Very low density lipoprotein (VLDL) of commercial broiler (CB) and crossbred village chicken (AK) was purified using Fast Protein Liquid Chromatography (FPLC). The fraction collected was then confirmed as VLDL using 4% polyacrylamide gel electrophoresis and transmission electron microscopy (TEM). The particle size of VLDL is 46.8${\pm}$8.6 nm. The VLDL fraction was then subfractionated and the apolipoprotein (apo) profile was studied by sodium dodecyl sulphate polyacrylamide gel electrophoresis (SDSPAGE). The CB and AK have almost similar types of apo in both subfractions 1 and 2. The AK showed the presence of apoAI, AIV, D and E whereas the CB had apo AIV, D, E and H. The apo AIV and apo E were present in both subfractions of AK and CB.

Current Increase Effect and Prevention for Electron Trapping at Positive Bias Stress System by Dropping the Nematic Liquid Crystal on the Channel Layer of the a-InGaZnO TFT's

  • The effect of nematic liquid crystal(5CB-4-Cyano-4'-pentylbiphenyl) on the amorphous indium gallium zinc oxide thin film transistors(a-IGZO TFTs) was investigated. Through dropping the 5CB on the a-IGZO TFT's channel layer which is deposited by RF-magnetron sputtering, properties of a-IGZO TFTs was dramatically improved. When drain bias was induced, 5CB molecules were oriented by Freedericksz transition generating positive charges to one side of dipoles. From increment of the capacitance by orientation of liquid crystals, the drain current was increased, and we analyzed these phenomena mathematically by using MOSFET model. Transfer characteristic showed improvement such as decreasing of subthreshold slope(SS) value 0.4 to 0.2 and 0.45 to 0.25 at linear region and saturation region, respectively. Furthermore, in positive bias system(PBS), prevention effect for electron trapping by 5CB liquid crystal dipoles was observed, which showing decrease of threshold voltage shift [(${\delta}V$]_TH) when induced +20V for 1~1000sec at the gate electrode.

Cluster-based Minimum Interference Channel Assignment for Multi-Radio Multi-Channel Wireless Mesh Networks (멀티 라디오 멀티 채널 무선 메쉬 네트워크를 위한 클러스터 기반 최소 간섭 채널 할당)

  • Total performance is improved by minimizing the channel interference between links in wireless mesh networks (WMNs). The paper refines on the CB-CA [1] to be suitable for multi-radio multi-channel (MRMC) WMNs. The CB-CA is the cluster-based channel assignment algorithm for one radio three channel WMN based on IEEE 802.11b/g. The CB-CA does not perform the channel scanning and the channel switching between the cluster heads (CHs) and the edge gateway nodes (EGs). However, the use of co-channel for links between CHs and EGs brings the problem of channel interference among many nodes. We propose and evaluate an improved CB-CA algorithm to solve this problem in MRMC WMNs. The proposed algorithm discriminates between transmission channel and receive channel and assigns channels to each interface randomly and advertises this information to neighbor clusters in order to be assigned no-interference channel between clusters. Therefore, the proposed algorithm can minimize the interference between clusters and also improve QoS, since it can use multiple interfaces and multiple channels.

Karyotype analysis of cryopreserved mononuclear cells from cord blood (제대혈 단핵세포의 냉동 전.후의 염색체 핵형분석의 실험적 연구)

  • Purpose : The ability to perform chromosome analysis of cryopreserved cord blood mononuclear cells is important for future retrospective studies. We compared the karyotypes of cryopreserved cells with cells before cryopreservation. Methods : One cord blood (CB) sample was obtained from normal healthy volunteer. Karyotype analysis was performed before cryopreservation. After mononuclear cell separation with Ficoll-Hypaque, the mononuclear cells were cryopreserved by programmed controlled-rate freezer and then transferred into the liquid nitrogen ($-196^{\circ}C$) for 3 days. After rapid thawing, cytogenetic analysis was performed as the same method for each sample by different conditions. The samples were divided by three groups. The first group was no culture before cryopreservation, the second group was 72 hours culture before cryopreservation, but no 24 hours culture after thawing and the third group was 72 hours culture before cryopreservation and 24 hours culture after thawing. Results : The chromosome analysis was successful in the second and third groups of CB sample. Conclusion : The successful result from CB samples may suggest the usefulness of long-term cryopreservation for retrospective study in various clinical settings including hematologic malignancies.

Determination and Isolation of Antioxidative Serotonin Derivatives, N-(p-Coumaroyl)serotonin and N-feruoylserotonin from Safflower Seeds (홍화종자에서 항산화성 Serotonin계 화합물, N-(p-Coumaroyl)serotonin과 N-feruoylserotonin의 분리 및 정량분석)

  • In the methanol extract from safflower seeds, two kinds of antioxidant were detected by preparative HPLC [$\mu$-Bondapak $C_{18}$ column ($7.8{\times}300\;mm$)]. Two unknown compounds were defined as CA and CB which had peaks at 22.1 min and 24.5 min, respectively. Antioxidant activity was measured by their scavenging ability on the stable tree radical of 1,1-diphenly-2-picrylhydrazyl (DPPH). For bulk extraction of antioxidants, the methanol extract was fractionated with hexan, chloroform, ethyl acetate and butanol. The ethyl acetate traction showing the highest DPPH radical scavenging activity was further purified by silica gel column chromatography to CA and CB. By NMR analysis, CA and CB were identified as N-(p-Coumaroyl)serotonin and N-feruoylserotonin, respectively. The content of N-(p-Coumaroyl)serotonin and N-feruoylserotonin were analyzed by reverse phase HPLC using a $\mu$-Bondapak $C_{18}$ column ($3.9{\times}300\;mm$) with linear gradient elution from 10% acetonitrile to 50% acetonitrile for 30min on UV detector at 300 nm. The contents of N-(p-Coumaroyl)serotonin and N-feruoylserotonin were 4.11 mg/g DW and 7.29 mg/g DW, respectively, and these two DPPH radical scavengers were detected only in the hull of seeds.

Eliminations from (E)-2,4-Dinitrobenzaldehyde O-Aryloximes Promoted by R3N in MeCN. Effects of β-Aryl Group and Base-Solvent on the Nitrile-Forming Transition-State

  • Nitrile-forming eliminations from (E)-2,4-$(NO_2)_2C_6H_2CH=NOC_6H_4-2-X-4-NO_2$ (1a-e) promoted by $R_3N$ in MeCN have been studied kinetically. The reactions are second-order and exhibit Br$\ddot{o}$nsted ${\beta}$ = 0.83-1.0 and ${\mid}{\beta}_{lg}{\mid}$ = 0.41-0.46. The results have been interpreted in terms of highly E1cb-like transition state with extensive $C_{\beta}$-H bond cleavage and limited $N_{\alpha}$-OAr bond cleavage. Comparison with existing data reveals that the structure of the transition state changes from E2-central to highly E1cb-like either by the change of the ${\beta}$-aryl group from Ph to 2,4-dinitrophenyl under the same condition or by the base-solvent system variation from $EtO^-$-EtOH to $Et_3N$-MeCN for a given substrate (1a-e).

Isolation and purification of protein-bound polysaccharides from the sawdust mycelia of Agrocybe cylindracea (톱밥배양한 버들송이의 균사체로부터 단백다당류의 분리 및 정제)

  • The characteristics of protein-bound polysaccharides (PBP) which were isolated and purified from the sawdust mycelia of Agrocybe cylindracea were investigated. The yield of crude protein-bound polysaccharides (Fr.CB) extracted with boiling water and precipitated with 95% ethanol, was 0.74% based on the original sawdust mycelia. The Fr.CB was purified by the membrane filtration, ion exchange chromatography and gel filtration. The Fr.B fraction of which the molecular weight is over 300 KDa, was isolated from the Fr.CB using membrane filtration, and the yield was 38.6% based on the Fr.CB. This result indicates that high molecular protein-bound polysaccharides are the dominent components of the Fr.CB. Two fractions (Fr.B-1, Fr.B-2) were also isolated from the Fr.B using ion exchange chromatography, and the yields were 17.3% (Fr.B-1) and 10.3% (Fr. B-2), respectively. The Fr.B-1 was concentrated and gel-filtrated, and the single peak, thought to be nearly pure protein-bound polysaccharides, was obtained. The yield of final fraction $(Fr.B-1-{\beta})$ was 42.5% based on the Fr.B-1. The molecular weight of $Fr.B-1-{\beta}$ was nearly 710 KDa. The monosaccharides' composition of $Fr.B-1-{\beta}$ was analized by HPLC, and glucose was the dominent component, and fucose and galactose were also detected. The result of amino acid analysis was that glutamic acid and analysis were detected to a significant level, and cysteine was not detected.
