Effect of cAMP on the Replication of Human Cytomegalovirus (Human cytomegalovirus 증식에 미치는 cAMP의 영향)

  • Since the 'promoter/enhancer region of the major immediate early (IE) ~ene of human cytomegalovirus (HCMV) contains the cyclic AMP (cAMP) response element (CRE) consensus sequence, it was reasonable to hypothesize that cAMP might affect HCMV replication. Cyclic AMP modulating drugs such as 8-bromoadenosine 3',5'-cyclic monophosphate (BrA), and papaverine were used to affect the intracellular levels of cAMP, and the effects of the drugs on HCMV replication were studied. While papaverine effectively inhibited HCMV multiplication and DNA synthesis, BrA exerted little effect on the production of infectious HCMV yields. The synthesis of DNA in HCMV-infected cells appeared to be stimulated by BrA In order to understand the effect of cAMP on the expression of HCMV major IE gene, plasmid (pCMVIE/CAT) containing a reporter gene driven by HCMV IE promoter was transfected into either permissive human embryo lung (HEL) cells or nonpermissive cells. PL,Javerine, which has been reported to block the HCMV-induced increase in cAMP, reduced the expression of pCMVIE/CA T in permissive HEL cells. Treatment of transfected cells with BrA increased the expression of HCMV major IE promoter not only in HEL cells, but also in nonpermissive HeLa and Vero cells. Therefore, it seems that the expression of HCMV major IE gene is regulated by cAMP.

  • Cellular automata (CA), which is known as a pseudo random number generator due to its excellent randomness, has various applications. Cho et al. designed a CA-based shrinking generator to generate a long period of nonlinear sequence. In addition, chaotic cat maps have been studied by many researchers as the complex nonlinear dynamics systems with sensitivity in initial conditions and unpredictable characteristics. In this paper, we propose a new image encryption method using nonlinear sequence generated by CA-based shrinking generator with maximum period and 3D chaotic cat map for high security.

Overexpression and Purification of Monooxygenases Cloned from Arthrobacter chlorophenolicus A6 for Enzymatic Decomposition of 4-Chlorophenol (4-Chlorophenol 분해박테리아 Arthrobacter chlorophenolicus A6로부터의 monooxygenase의 복제 및 대량발현과 정제 그리고 기질분해활성도 분석)

  • Arthrobacter chlorophenolicus A6 possesses several monooxygenases (CphC-I, CphC-II, and CphB) that can catalyze the transformation of 4-chlorophenol (4-CP) to hydroxylated intermediates in the initial steps of substrate metabolism. The corresponding genes of the monooxygenases were cloned, and the competent cells were transformed with these recombinant plasmids. Although CphC-II and CphB were expressed as insoluble forms, CphC-I was successfully expressed as a soluble form and isolated by purification. The specific activity of the purified CphC-I was analyzed by using 4-CP, 4-chlorocatechol (4-CC), and catechol (CAT) as substrates. The specific activities for 4-CP, 4-CC, and CAT were determined to be 0.312 U/mg, 0.462 U/mg, 0.246 U/mg, respectively. The results of this study indicated that CphC-I is able to catalyze the degradation of 4-CC and CAT in addition to 4-CP, which is a primary substrate. This research is expected to provide the fundamental information for the development of an eco-friendly biochemical degradation of aromatic hydrocarbons.

  • 강원도 설악산 국립공원에는 국내 최장의 토왕성 폭포가 있다. 유역면적이 $0.54km^2$에 불과하여 평소에는 폭포의 물줄기를 볼 수 없지만, 비 온 뒤에는 며칠간 하얀 물줄기의 장관을 연출한다. 본 연구에서는 물리적 기반의 수문모형인 CAT3.0을 이용하여 강우시 토왕성 폭포의 유출량을 분석하고 강우량에 따른 유출지속기간을 산정하여 강우의 누적량 및 지속기간에 따른 폭포의 물줄기를 볼 수 있는 기간을 추정하고자 하였다. CAT 3.0은 다양한 침투방정식중에 Rainfall Excess, Horton Eq., Green-Ampt. Eq.을 이용하여 강우시 토양특성(토양깊이, 투수계수)과 잠재증발산량과 토양수분량에 의한 실제증발산량을 산정하고 유출량을 계산하는 수문모형이다. 토왕성 폭포의 유출 특성을 분석하기 위하여 속초지점의 19년간(2010~2018) 관측자료를 이용하였으며, 토양의 깊이는 10 cm, 30 cm를 가정하여 비교하였다. 유출해석을 위하여 일단위 자료를 사용하였으며, 강우지속일수 및 누가강우량에 따른 유출을 산정하고 폭포의 물줄기가 확연히 보일 수 있는 한계유출량을 추정하였다. 폭포로부터의 유출량을 측정하는 관측지점이 없으므로, 폭포의 물줄기를 볼 수 있는 기간은 방송사 뉴스, 신문기사, SNS 등의 다양한 정보원으로부터 유추해석하였다. 이러한 분석결과로부터 강우에 의한 토왕성 폭포의 물줄기를 볼 수 있는 기간을 제시하였다.

  • In this paper, proposes Kerberos certification mechanism that improve certification service of PKINIT base that announce in IETF CAT Working Croup. Also proposed Authentication Mechanism for reusability of Ticket that after Ticket's Lifetime is ended, message exchange that Local Client receives Remote Server's service. Since my suggestion to regional services are not described in Kerberos, authentication between regions can be performed via PKINIT(Public Key Cryptography for Initial Authentication) presented by IETF(Internet Engineering Task Force) CAT working group. The new protocol is better than the authentication mechanism proposed by IETF CAT Working group in terms of communication complexity and mechanism according to simplified Ticket issue processing.

Characterization of $\beta$-Ketoadipate Pathway from Multi-Drug Resistance Bacterium, Acinetobacter baumannii DU202 by Proteomic Approach

  • In this study, the biodegradative activities of monocyclic aromatic compounds were determined from the multi-drug resistant (MDR) Acinetobacter baumannii, which were studied in the form of clinical isolates from a hospital in Korea. These bacteria were capable of biodegrading monocyclic aromatic compounds, such as benzoate and p-hydroxybenzoate. In order to determine which pathways are available for biodegradation in these stains, we conducted proteome analyses of benzoate, and p-hydroxybenzoate-cultured A. baumannii DU202, using 2-DE/MS analysis. As genome DB of A. baumannii was not yet available, MS/MS analysis or de novo sequencing methods were employed in the identification of induced proteins. Benzoate branch enzymes [catechol 1,2-dioxygenase (CatA) and benzoate dioxygenase $\alpha$ subunit (BenA)] of the $\beta$-ketoadipate pathway were identified under benzoate culture condition and p-hydroxybenzoate branch enzymes [protocatechuate 3,4-dioxygenas $\alpha$ subunit (PcaG) and 3-carboxy-cis,cis-muconate cycloisomerase (PcaR)] of the $\beta$-ketoadipate pathway were identified under p-hydroxybenzoate culture condition, respectively, thereby suggesting that strain DU202 utilized the $\beta$-ketoadipate pathway for the biodegradation of monocyclic aromatic compounds. The sequence analysis of two purified dioxygenases (CatA and PcaGH) indicated that CatA is closely associated with the CatA of Acinetobacter radiresistance, but PcaGH is only moderately associated with the PcaGH of Acinetobacter sp. ADPI. Interestingly, the fused form of PcaD and PcaC, carboxymuconolactone decarboxylase (PcaCD), was detected on benzoate-cultured A. baumannii DU202. These results indicate that A. baumannii DU202 exploits a different $\beta$-ketoadipate pathway from other Acinetobacter species.

Validity and Reliability of the Person-centered Care Assessment Tool in Long-term Care Facilities in Korea (노인장기요양시설 종사자를 대상으로 한 인간중심돌봄 사정도구의 타당도 및 신뢰도 검증)

  • Purpose: The aim of this study was to evaluate the validity and reliability of the Korean version of the Person-centered Care Assessment Tool (P-CAT). Methods: The English P-CAT was translated into Korean with forward and backward translation. Survey data were collected from 458 staff in 17 long-term care facilities in Korea. Construct validity and criterion related validity were evaluated. Cronbach's alpha was used to assess reliability. Results: The Korean version of P-CAT was shown to be valid homogeneously by factor, item and content analysis. Internal consistency reliability was satisfactory in which the values of factor 1, factor 2 and the total scale were .84, .77 and .86 respectively. Exploratory factor analysis supported the construct validity with a two-factor solution. Factor loadings of the 13 items ranged in .34~.80. Criterion validity to the Person-centered Climate Questionnaire-staff (PCQ-S) was .74 (p<.001). Conclusion: The Korean version of the P-CAT was found to be an applicable instrument with satisfactory reliability and validity for further use in measuring successful person-centered care in long-term care facilities for older persons.

Successful Management of Hepatic Lipidosis Accompanied by a Feline Skin Fragility Syndrome-like Lesion in a Cat (고양이 피부유약증 유사병변을 동반한 지방간에 이환된 한 마리 고양이의 성공적인 치료증례)

  • A 7-year-old spayed female Somali-cross cat was admitted with a 4-week history of anorexia, vomiting, weight loss, and lethargy. The cat was diagnosed with hepatic lipidosis. With intensive care and nutritional support via a nasogastric feeding tube for 3 weeks, the clinical signs of hepatic lipidosis were improved. However, skin lesions were found in the left and right scapular regions during the treatment that were suspected to be due to feline skin fragility syndrome (FSFS). Intensive wound healing therapy with granulated sugar, laser therapy, and a surgical flap was conducted. Skin lesions improved uneventfully without other clinical signs or recurrence of any skin lesion for a year. To our knowledge, this is the first report of a good prognosis in a hepatic disorder and concurrent FSFS.

Effects of Aconiti tuber butanol fraction and several cardiotonics on the mechanical properties of cat papillary muscle preparation (수종강심약물(數種强心藥物)과 부자(附子)"부타놀" 분획(分劃)이 심장근(心臟筋)의 기계적(機械的) 성질(性質)에 미치는 영향(影響))

  • The effect of positive inotropic agents on the contractile properties of myocardial muscle were studied in the cat papillary muscle preparation. For the purpose, the effects of ouabain $(1{\times}10^{-6}g/ml)$, norepinephrine (0.05r/m1) and Aconiti tuber butanol fraction (AF(5), $1{\times}10^{-4}$, $5{\times}10^{-4}$, $1{\times}10^{-3}$, $2{\times}10^{-3}g/ml$) on the contractile dynamics of the papillary muscle preparation isolated from right ventricle of cat were observed in terms of the characteristics of isometric twitch and the lengh-tension relation, the force-velocity relation and the load-extension relation of the series elastic component of contractile model of A.V. Hill. All the studied inotropic drugs similary increased the rate and the intensity of the developed isometric tension, while shortened the time from onset of contraction to peak tension and the total duration of contraction. In the afterloaded simultaneous isotonic and isometric contraction, they also similary increased the maximal velocity of shortening accompanied with the increasing the maximum developed force. In the load-extension relation all the drugs, however, had no appreciable influence on the properties of the series elastic component. Increasing the concentration, Aconiti tuber butanol fraction produced more pronounced effect on all the studied parameters of isometric and isotonic contraction of cat papillary muscle preparation. From the aspect of contractile dynamics, it seemed that the positive inotropic effect of ouabain, norepinephrine and Aconiti tuber butanol fraction are similary achieved through an influence on the behavior of the contractile component only.

Establishment of In Vitro Test System for the Evaluation of the Estrogenic Activities of Natural Products

  • In order to evaluate estrogenic compounds in natural products, an in vitro detection system was established. For this system, the human breast cancer cell line MCF7 was stably trans-fected using an estrogen responsive chloramphenicol acetyltransferase (CAT) reporter plas-mid yielding MCF7/pDsCAT-ERE119-Ad2MLP cells. To test the estrogenic responsiveness of this in vitro assay system, MCF7/pDsCAT-ERE119-Ad2MLP cells were treated with various concentrations of 17f3-estradiol. Treatments of 10$^{-8}$ to 10$^{-12}$ M 17$\beta$-estradiol revealed significant concentration dependent estrogenic activities compared with ethanol. We used in vitro assay system to detect estrogenic effects in Puerariae radix and Ginseng radix Rubra extracts. Treat-ment of 500 and 50 $\mu\textrm{g}$/ml of Puerariae radix extracts increased the transcriptional activity approximately 4- and 1.5-fold, respectively, compared with the ethanol treatment. Treatment of 500, 50, and 5 $\mu\textrm{g}$/ml of Ginseng radix Rubra extracts increased the transcriptional activity approximately 3.2-,2.7, and 1.4-fold, respectively, compared with the ethanol treatment. These observations suggest that Puerariae radix and Ginseng radix Rubra extracts have effective estrogenic actions and that they could be developed as estrogenic supplements.
