  • In order to achieve poly-beta-hydroxybutyric acid (PHB) production using recombinant DNA in various host bacterial cells, the isolation of genes for PHB biosynthesis was attempted. As a result, a 5.2kb DNA fragment containing phbCAB operon of Alcaligenes eutrophus was isolated by colony hybridization using synthetic oligodeoxyribonucleotides as probes. The constructed recmbinant plasmid pSK(+)-phbCAB operon was transferred to Escherichia coli, and the obtained transformant accumulated considerable amount of PHB.

  • High level of tractor noise may reduce operator's work performance and cause a hearing problem. This study was carried out to investigate the cab noise of a selected agricultural tractor. The noise was measured and analyzed in terms of sound and sound intensity levels. Form the results it was known that a structure-born noise was induced from the feet-rest and rear floor panel of the cab. The air-borne noise caused mainly by the engine was transmitted into the operator's position through the small openings of cab structures in the operator's lower front area.

  • Currently, the marketable truck has the cab tilting system which highly absorb the vibrations and impacts of cab. The cab tilting system is equipped with the hydraulic system for maintenance and inspection of truck. And it is very important to improve the user's feeling of driving and convenience. To have the better performance, it is important to confirm the dynamic characteristics of each parts of the system. In this study, a mathematical model of the system was derived and the parameters in the model were identified from experimental data. The results of the simulation were compared with those of experiments. These results of the computer simulation and experiments show that the proposed analysis can be applied effectively to design the system. Our results can be utilized for the future research to improve the capability of the truck cab tilting system.

  • Since the introduction of vonoprazan, a potassium-competitive acid blocker (P-CAB), it has been demonstrated to reversibly inhibit gastric acid secretion by engaging in potassium-competitive ionic binding to H+/K+-ATPase. In contrast, proton pump inhibitors (PPIs) achieve H+/K+-ATPase inhibition through covalent binding to cysteine residues of the proton pump. Reported cases have indicated an emerging trend of P-CAB-related gastropathies, similar to those associated with PPIs, as well as unique gastropathies specific to P-CAB use, such as the identification of web-like mucus. Pathologically, parietal cell profusions, which show a positively correlated with hypergastrinemia, have a higher incidence in P-CAB users compared to PPI users. Thus, this review aims to summarize the endoscopic and pathological findings reported to date concerning P-CAB-related gastric mucosal lesions. Additionally, it seeks to discuss the differences between the PPIs and P-CABs in terms of the formation and frequency of associated gastropathies. This review highlights the evident differences in the mechanism of action and potency of acid inhibition between P-CABs and PPIs, notably contributing to differences in the formation and frequency of associated gastropathies. It emphasizes the necessity to distinguish between P-CAB-related and PPI-related gastropathies in the clinical setting.

Toxicity Evaluation of Burkholderia pyrrocinia CAB08106-4 in Cyprinus carpio and Daphnia magna (Burkholderia pyrrocinia CAB08106-4 원제가 잉어 및 물벼룩에 미치는 영향 연구)

  • Bukholderia pyrrocinia CAB08106-4 has an anti-fungal effect on Garlic White Rot caused by Sclereotium cepivorum and Sclereotium sp. It is environmentally friendly microbial product that prevents and controls a variety of phytopathogens including Garlic White Rot caused by Sclereotium cepivorum and Sclereotium sp. The aim of this study was to assess the environmental toxicity using Cyprinus carpio and Daphnia magna. Bukholderia pyrrocinia CAB08106-4 ($1.0{\times}10^9cfu/mL$) was adminatrated to Cyprinus carpio and Daphnia magna according to the toxicity test guideline for peciticide. $LC_{50}$ of Bukholderia pyrrocinia CAB08106-4 is over $6.67{\times}10^4cfu/mL$ in Cyprinus carpio and Daphnia magna and no adverse effect was observed. Based on these results, we concluded that Bukholderia pyrrocinia CAB08106-4 has no toxiciy for Cyprinus carpio and Daphnia magna.

Photosynthetic Characterization of Transgenic Tobacco Plant, by Transformation of Chlorophyll a/b Binding Protein Gene of Korean Ginseng (인삼의 Chlorophyll a/b Binding Protein유전자를 도입한 연초의 광합성 특성)

  • A CAB cDNA vector(pKGCAB), encoding the light harvesting chlorophyll a/b binding protein in Korean ginseng (Panax ginseng C. A. Meyer), was constructed with the CaMV35S promoter of plant expression vector. The chimeric vector was transformed into tobacco(Nicotiana tabacum cv. NC 82) using Agrobacterium tumefaciens LBA 4404 strain, and the transgenic tobacco plant CAB-TP2 was selected. Photosynthetic rates of the CAB-TP2 plant at before-flowering stage were increased about 20% under low irradiance conditions of quantum 100 and 500 $\mu$mol.m$^{-2}$ s$^{-1}$ , however, the rates were similar to those of NC 82 under quantum 1000 and 2000 $\mu$mol.m$^{-2}$ s$^{-1}$ conditions. The plants were germinating under low- or normal irradiance condition and the quantum yield of photosystem III were measured. The differences of the Fv/Em values between conditions were 0.07 and 0.01 in NC 82 and CAB-TP2, respectively. The mature leaves in the position 8-10 of the CAB-TP2 at before-flowering stage revealed l0% higher Fv/Fm values in range of 0.759 to 0.781 and 40% more chlorophyll contents of 70-93mg/$m\ell$ than those of normal NC 82. These data suggest the possibility that the increase in photosynthetic activity of leaves under low light intensity in the canopy of CAB-TP2 transgenic tobacco might lead to increase the quality of lower tobacco leaves.

Characterization of New Bacillus thuringiensis Isolated with Bioactivities to Tobacco Cutworm, Spodoptera litura (Lepidoptera: Noctuidae) (담배거세미나방에 살충효과를 나타내는 새로운 Bacillus thuringiensis 균주의 특성)

  • Bacillus thuringiensis with selected high toxicities against tobacco cutworm, Spodoptera litura were isolated from domestic soils. When being observed under a phase-contrast microscope, the insecticidal crystal proteins were showed a bipyramidal crystal types. New CAB 109 isolate was identified to B. thuringiensis subsp. aizawai in the H serotype. As a results of insecticidal activities between CAB 109 isolate and 3 existing ready-made products against 3rd larva of S. litura, CAB 109 isolate showed 100% mortality with spore concentration $(1.3{\times}10^7cfu/ml)$. It was a very high insecticidal activity compared with a existing ready-made B. t. products. $LD_{50}$ values of CAB 109 isolate was $9.78{\times}10^5,\;6.87{\times}10^6\;and\;1.83{\times}10^7cfu/ml$ spore concentration against 2nd, 3rd and 4th larva of S. litura, respectively. Unlike Plutella xylostella, S. litura was slowly died after application up to 7 days. The weight of S. litura larva applied with CAB 109 isolate were 6-7 times less than controlled group. Even though it didn't die, it did not grow into next larva. The result observed with scanning electron microscope was that CAB 109 isolate of B. t. aizawai formed a typical bipyramidal crystal protein type. Otherwise, when CAB 109 isolate was examined with SDS-PAGE and with trypsin, there was no difference between CAB 109 strain and ready-made products of B. thuringiensis.

  • Eight Bacillus thuringiensis strains activated against mosquito larva were compared their characterization. Spherical-shaped parasporal inclusion of B. thuringiensis subsp. tohokuensis CAB167 was observed by phase-contrast microscopy and scanning electron microscopy. $LC_{50}$ values of B. thuringiensis subsp. tohokuensis CAB167 against Culex pipiens molestus, Culex pipiens pallens, and Aedes aegyti were 173, 190 and 580 ng/ml, respectively. B. thuringiensis subsp. tohokuensis CAB167 had a parasporal inclusion containing 4 major protein components, for example, 135, 80, 49 and 28-kDa by SDS-PAGE. Otherwise, after trypsin digestion of parasporal inclusion, SDS-PAGE was showed new protease-resistant peptides at 72 and 63-kDa. Activated toxins of isolated CAB167 were different from other reference strains on a serological by immuno-diffusion test.

Biological Control of White Rot in Garlic Using Burkholderia pyrrocinia CAB08106-4 (Burkholderia pyrrocinia CAB08106-4 균주를 이용한 마늘 흑색썩음균핵병의 생물학적 방제)

  • White rot caused by Sclerotium cepivorum was reported to be severe soil-born disease on garlic. Disease progress of white rot of garlic (Allium sativum L.) was investigated during the growing season of 2009 to 2011 at Taean and Seosan areas. The white rot disease on bulb began to occur from late April and peaked in late May. The antifungal bacteria, Burkholderia pyrrocinia CAB08106-4 was tested in field bioassay for suppression of white rot disease. As a result of the nucleotide sequence of the gene 16S rRNA, CAB008106-4 strain used in this study has been identified as B. pyrrocinia. B. pyrrocinia CAB080106-4 isolate suppressed the white rot with 69.6% control efficacy in field test. These results suggested that B. pyrrocinia CAB08106-4 isolate could be an effective biological control agent against white rot of garlic.

  • The leading-cab (high energy absorption area) of rolling stock directly is impacted on the frontal crash unlike other cabs. Thus, leading-cab has a structurally complex shape to solve getting concentrated loads. However, in order to enhance structural performance and to achieve the weight reduction of cab, changing the sizes and adjusting the distance of members do not take an effective result. Therefore, in design phase, to find the material arrangement which helps structural capacity be better should be done. This research applies the topology optimization to concept design of protective shell frame on strategy of crush energy absorption with considering pressure and vertical loads acting on the principal part of leading-cab. In this research, topology optimization method focuses on structural design, and which yields optimal material arrangement under given loads and boundary conditions using density method which has the density of material as design variables. Finally, this research presents optimal material arrangement and structure of protective shell frame on given loads with applying topology optimization.
