Effects of pressure and pressing time of soyprotein-CaSO4 coagulates on the yield, water holding capacity and textural characteristics of SPI (soy protein isolated) tofu were investigated. The tofu was prepared by addition of CaSO4 into SPI suspension at 9$0^{\circ}C$ and then compressed. As the compressing pressure increased from 8.66g /cm2 to 49.43g /cm2, the volume yield and moisture absorbed on filter paper were decreased and the textural properties of hardness, adhesiveness, cohesiveness and gumminess were increased. Increase in hardness and gumminess were more significant at high pressure than those at low pressure.
Two experiments were conducted to assess the effect of replacing a conventional concentrate with mixed cassava (Manihot esculenta) foliage and legume (Phaseolus calcaratus) foliage. In Exp. 1, three rumen fistulated crossbred cows were used for in sacco rumen degradability studies. In vitro gas production was also studied. In Exp. 2, 11 crossbred F2 heifers (Red Sindhi$\times$Holstein Friesian), with initial live weight of $129{\pm}6kg$ and aged six months, were allocated in a Completely Randomized Design (CRD) to evaluate a mixture (ratio 3:1) of cassava and legume foliage (CA-LE feed) as a protein source compared to a traditional concentrate feed (Control) in diets based on fresh elephant grass (Pennisetum purpureum) and urea treated rice straw ad libitum. The Control feed was replaced by the CA-LE feed at levels of 0% (Control), 40% (CA-LE40), and 60% (CA-LE60) based on dry matter (DM). The in sacco degradation of CA-LE feed was higher than Control feed (p<0.05). After 48 h incubation the degradation of CA-LE feed and Control feed was 73% vs. 58% of DM and 83% vs. 65% of CP, respectively. The gas production of CA-LE feed was also significantly higher than of Control feed during the first 12 h of incubation. The results of the performance study (Exp. 2) showed that the level of CA-LE feed in the concentrate had no effect on total dry matter intake (p>0.05), but live weight gains (LWG) in CA-LE40 and CA-LE60 were significantly higher (551 and 609 g/d, respectively) than in the Control group (281 g/d). The intake of CP was higher (p<0.05) for the treatments CA-LE40 and CA-LE60 (556 and 590 g/d, respectively) compared to that of Control (458 g/d), while there was no significant difference in ME intake. The feed conversion ratio was 16.8, 9.0 and 7.9 kg DM/kg LWG in Control, CA-LE40 and CA-LE60, respectively. The feed cost of CA-LE40 and CA-LE60 corresponded to 43% and 35%, respectively, of the feed cost of Control feed. The best results were found when CA-LE feed replaced 60% of DM in Control feed and considerably decreased feed cost. It is concluded that feeding cassava foliage in combination with Phaseolus calcaratus legume as a protein supplement could be a potentially valuable strategy which leads to reduced feed costs and a more sustainable system in smallholder dairy production in Vietnam.
The objective of this study was to investigate the effect of dietary supplementation with calcium salts of soybean oil fatty acids (CaSO) and linseed oil fatty acids (CaLO) on c9,t11-CLA production in ruminal fluid and milk fat from Holstein dairy cows. Rumen fermentation, lactational performances and fatty acid profiles in ruminal fluid and milk fat were also investigated. Twenty multiparous Holstein dairy cows were allotted randomly into two groups consisting of ten cows in each group according to calving date and average milk yield. The first group of cows was fed a control (without calcium salts) diet and a treatment as 1.0% of CaSO (on DM basis) for 30 days in each period. In the second group, cows were fed the same control diet and 1.0% of CaLO as a treatment in the same manner. The forage: concentrate ratio was 52:48, and diets were formulated to contain 17% crude protein (DM basis) for both groups. Ruminal pH, protozoal numbers and the concentration of total volatile fatty acids were unchanged, however, the ruminal ammonia-N decreased by feeding CaSO or CaLO treatment compared to the control diet. The vaccenic acid (trans-11 C18:1; VA) in rumen fluid increased (p<0.01) by 169% and 153%, and the c9,t11-CLA content of rumen fluid increased (p<0.01) by 214% and 210% in the CaSO and CaLO treatments, respectively, compared to the control diet. In milk fatty acids, the VA content increased by 130% and 132% in the evening and morning milking times, respectively, and the c9,t11-CLA content increased by 125% in both milking times for the CaSO supplementation than that of control diet. In the case of CaLO supplementation, the VA increased by 117% and 114%, and the c9,t11-CLA increased by 96% and 94% in the evening and morning milking times, respectively, compared to the control diet. The contents of VA and c9,t11-CLA of milk fatty acids were numerically higher in the evening milking time compared to the morning milking time for control and both treatments. Finally, these results indicated that the supplementation of CaSO or CaLO treatment increased the VA and the c9,t11-CLA in both ruminal fluid and milk fat of Holstein dairy cows.
Body content bone mined density and nutrient intake status of 129 college women in Daegu area are analyze4 The mean age was 23.1 years. Bone mineral density was measured in their right heel by SONOST-2000 ultrasound bone densitometer, and nutrient intake was assessed by a Questionnaire. Diet intake data were obtained by using a 24 hours recall method to evaluate the subject's usual diet. The average energy, Ca, Ee, Zn and folate intakes were 64.2, 47.5, 54.7, 62.9 and $57.4\%$ of RDA, respectively. Ca/P and Ca/Protein ratio were 0.46 and 6.7, respectively. The mean adequacy ratio(MAR), an index of overall dietary quality wag 0.6. The index of nutritional quality was under 1.0 for protein(0.7), vitamin A(0.9), E(0.7), $B_1(0.8)$, niacin(0.8), $B_6(0.6)$ and phosphorus(0.6). The less the protein and energy intake, the less the bone mineral density(p<0.05). The intakes of protein(p<0.05) and that of phosphorus(p<0.001) were positively correlated with bone quality index(BQI). The calcium intake was positively correlated with Z-score(p<0.05). The phosphorus intake was positively correlated with T-score(p<0.001) and Z-score(p<0.0001). In conclusion, this study indicates that nutrient intake of the college women is considerably lower than the RDA. Also the BQI is lower than standard.
To determine relationships of biochemical parameters involved in Ca metabolism with eggshell quality, serum Ca level, duodenal and uterine calcium binding protein (CaBP) and uterine carbonic anhydrase (CA) activities were measured using 102-week old hens. Three groups of chickens were selected, those showing high quality (HQ) and low quality (LQ) eggshells and non-laying activity (NE). NE hens exhibited significantly (p < 0.05) lower serum Ca levels than laying hens. HQ and LQ hens were not different in the Ca level, indicating that serum Ca level was not good indicator of hen's ability to produce different quality eggshells. Duodenal CaBPs was highest in HQ and lowest in NE (p < 0.10). Uterine CaBPs of LQ and NE were not different and lower significantly (p<0.10) than that of HQ, suggesting that CaBP played an important role in determining eggshell quality. Uterine CA activities of the three groups were significantly different (p < 0.01) ; highest in HQ and lowest in NE, suggesting intimate relationship between CA and Eggshell quality.

Protein-bound polysaccahrides(PBP) were isolated, purified, and characterized from the sawdust media after harvesting the fruit body of Flammulina velutipes. The yield of the crude PBP(Fr.CA) extracted from the sawdust media, was 0.367% relative to the original sawdust media. The total sugar and protein contents of Fr.CA were 19.8% and 23.8% respectively. Using the membrane filtration, the fraction of which the molecular weight is over 300 kDa(Fr.A) was isolated from the Fr.CA and the yield was 44.6% relative to the Fr.CA. This result indicates that high molecular PBP is the dominant components of the Fr.CA. The Fr.A was separated into three fractions (Fr.A-1, Fr.A-2 and Fr.A-3) whose yields are 5.8%, 8.5% and 13.2% respectively. These fractions were further purified using gel filtration, obtaining a single peak in each fraction that considered as pure PBP Among them, the yield of Fr.A-1-${\alpha}$ was 20.9% relative to the Fr.A-1, and the molecular weight was 800 kDa. Monosaccharide components such as glucose, galactose, mannose, xylose and fucose could be detected all fractions by a HPLC analysis. Especially in the Fr.A-1-${\alpha}$ fraction, the content of glucose and galactose appeared to be high.
Three concentrations of P (0.33, 0.43 and 0.54%) and two concentrations of Ca (0.60 and 0.97%) in ration dry matter were evaluated for effects on dry matter intake and on milk yield and composition using 24 Holstein cows. Cows were arranged in a $3{\times}2$ factorial experiment as an incomplete randomized block design with three 28-day periods. Each cow consumed at least one ration with each concentration of Ca. Dry matter intake, yield of 3.5% Fat Corrected Milk, and milk composition were not affected by concentration of P, but milk yield was greater when lowest concentration of P was fed (22.8 vs. 22.1 kg/day; p<0.07). Cows fed rations containing 0.60% Ca had greater milk (22.7 vs. 21.9 kg/day; p<0.02) and 3.5% Fat Corrected Milk yields (p<0.03) and slightly greater protein content than when fed 0.97% Ca. Dietary Ca:P ratios between 1.1:1 and 2.9:1 had no effect on dry matter intake, milk yield, or composition. Concentrations of P in plasma were within the normal range for all rations. Because cows had high dry matter intake, mean daily intakes of both P and Ca were greater than required for their level of milk yield.
This study was performed to identify the characteristics of the OFC1 gene (locus: chromosome 6p24.3) in Korean patients, which is assumed to be the major gene behind the nonsyndromic cleft lip and palate. The sample consisted of 80 subjects: 40 nonsyndromic cleft lip and palate patients (proband, 20 males and females, mean age 14.2 years); and 40 normal adults (20 males and 20 females, mean age 25.6 years). Using PCR-based assay, the OFC1 gene was amplified, sequenced, and then searched for similar protein structures. Results were as follows: 1. The OFC1 gene contains the microsatellite marker 'CA' repeats. The number of the reference 'CA' repeats was 21 times, and formed as TA(CA)11TA(CA)10. But, in Koreans, the number of tandem 'CA' repeats was varied from 17 to 26 except 18, and 'CA' repeats consisted of TA(CA)n. 2. Nine allelic variants were found. Distribution of the OFC1 allele was similar between the patients and control group. 3. There was a replacement of the base 'T' to 'C' after 11 tandem 'CA' repeats in Koreans compared with Weissenbach's report. However, the difference did not seem to be the ORF prediction results between Koreans and Weissenbach's report. 4. The BLAST search results showed the Telomerase reverse transcriptase (TERT) and the Nucleotide binding protein 2 (NBP2) as similar proteins. The TERT was a protein product by the hTERT gene in the locus 5p15.33 (NCBI Genome Annotation; NT023089) The NBP2 was a protein product by the ABCC3 (ATP-binding cassette, sub-family C) gene in the locus 17q22 (NCBI Genome Annotation; NT010783). 5. In the Pedant-Pro database analysis, the predictable protein structure of the OFC1 gene had at least one transmembrane region and one non-globular region.
This study was conducted to provide basic information on the nutritional status and dietary behaviors of the deaf teenagers. The subjects of this study were 87 deaf-mute high school students in Pusan and Kyung-nam area and, in comparison,90 general high school students in Ham-an area. The survey was investigated by using a self-administered questionnaire. The results were as follows: Deaf-mute group was consisted of 63.2% male and 36.8% female, and mean of height and weight of male were 171.2 cm and 61.9kg, female were 158.0cm and 51.7kg. Obesity index and body mass index (BMI) were normal range in both deaf-mute and normal groups. The average nutrition knowledge score of the deaf-mute group was 6.8 $\pm$ 1.5 out of possible 10 points, that was significantly lower than 7.3 $\pm$ 0.8 of the normal group (p<0.01). The mean of self-control and conscious control scores in the eating behaviors of the deaf-mute group were 2.6 $\pm$ 1.2 and 2.4 $\pm$ 1.3 out of possible 5 points from each item, which was significantly higher than 2.1 $\pm$ 1.3 and 1.8 $\pm$ 1.4 of the normal group (p< 0,01). Nutrients consumed below 90% of Korean RDA were energy (79.9%), Ca (71.5%) for deaf-mute male students and Ca (88.5%) for deaf-mute female students. Energy (71.4%), protein (87.8%), Ca (74.8%), vitamin B$_1$ (83.4%) intake of normal male students and energy (72.8%), Ca (71.2%), Fe (78.7%) intake of normal female students were below 90% of Korean RDA. Energy, protein, fat, vitamin B$_1$, niacin intake of deaf-mute male students were significantly higher than normal male students and all nutrients intake of deaf-mute female students were significantly higher than normal female students. By the correlation of nutrients intakes with nutrition knowledge, there was positive correlation with the intakes of Ca, Fe, vitamin A, vitamin B$_2$, and vitamin C in the deaf-mute group, while there was negative correlation with the nutrients intakes (except for protein and fat) in the normal group. The amount of meal, breakfast, regularity of meal time, frequency of snacks showed a positive relation to nutrient intakes in deaf-mute group and amount of meal, breakfast, regularity of meal time, frequency of overeating showed a positive relation to nutrient intakes in normal group. The nutrition knowledge had no correlation with food habits or eating behaviors in both groups.(Korean J Nutrition 35(9) : 982~995, 2002)

Changes in surface hydrophobicity of soy protein isolate (SPI), which plays an important role in the functional characteristics of protein, were measured according to various SPI concentrations, pH levels, electrolytes concentrations, and alginate molecular weights by using 1-anilino-8-naphthalene sulfonic acid as a fluorescent probe. SPI surface hydrophobicity decreased as SPI concentrations increased. SPI surface hydrophobicity reached a maximum at pH 7.0. SPI surface hydrophobicity rapidly increased as the NaCl concentration of SPI solution increased up to 100 mM, and showed no large increases above 100 mM. However, SPI surface hydrophobicity radically decreased until the $CaCl_2$ concentration reached 50 mM and revealed no large variations above 50 mM. A similar trend was exhibited in the case of $MgCl_2$. As both the concentration and molecular weight of sodium alginate increased, SPI surface hydrophobicity decreased. The increasing rate of SPI surface hydrophobicity decreased as the molecular weight of sodium alginate increased.
