Gene Cloning, Expression, and Characterization of Glucose-1-Phosphatase from Enterobacter cloacae B11

  • A bacterial strain with phytase and glucose-1-phosphatase activity was isolated from seawater. The colony was identified as an Enterobacter cloacae strain and named E. cloacae B11. A gene, agpEnB11, coding for an intracellular acid glucose phosphatase was cloned from the strain and sequenced. It comprised 1,242 nucleotides and encoded a polypeptide of 413 amino acids. Recombinant glucose-1-phosphatase (AgpEn) was overexpressed in Escherichia coli and purified using Ni-NTA column under native conditions. Purified protein displayed a single band of 47 kDa on SDS-PAGE. AgpEn hydrolyzed a wide variety of phosphorylated compounds, with high activity for glucose-1-phosphate and glucose-6-phosphate. Optimum pH and temperature for enzyme activity were pH 5.0 and $50^{\circ}C$, respectively. Enzyme activity was stimulated by $Ca^{2+}$ and $Co^{2+}$, and inhibited by $Cu^{2+}$.

A Case of Abdominal Tuberclosis Mimicking Ovarian Cancer (난소 종양의 임상적 양상을 보인 소아 복막 결핵)

  • We report the case of a girl with peritoneal tuberculosis (Tb) who presented with mimicking features of ovarian cancer including massive ascites, elevation of CA-125 and ovarian masses. These ovarian masses cannot be ruled out malingnacy with image study. Gram stain, acid-fast stain, bacteriological and mycological culture of centrifuged ascites cells were negative. Tumor markers revealed an elevated CA-125 level of 112 U/mL and adenosine deaminase level were significantly high with 118 U/L. Diagnostic laparoscopy was done. The histology of the peritoneum and the ovary showed multiple granulomatous nodules with epithelioid cells and multi-nucleated giant cells. And there were no evidence of malignancy. Furthermore acid-fast bacilli (AFB) was identified in this specimen. Peritoneal Tb was difficult to diagnosis in our patient because it mimicked ovarian cancer. And this is the first case of abdominal Tb mimicking ovarian cancer in children in Korea.

Chemical properties of Watercress(Oenanthe javanica D.C.) Depend upon Cultivating Methods (재배방법이 다른 미나리의 성분 특성)

  • Chemical properties of the different parts of watercress(Oenanthe javanica D.C.) grown and harvested from the culture fields under different cultivating methods were studied. In proximate analyses of watercress in parts, moisture contained more in stem than in root or in leaf, but crude protein and crude lipid contents were lower in stem than in root or in leaf. Crude ash in root contained up to about double amount in leaf or stem. Major mineral elements detected in watercress were Fe, Mg, Ca, and K, and their contents in root was higher than those in stem or leaf. Especially, Fe in root was significantly higher than that in stem or in leaf. Total free sugar composed mainly with fructose and glucose, was the highest in watercress from Hwasoon and followed the watercress from Jeonju and Donggok in order. Free sugar content was highest in leaf and the lowest in root. Major water-soluble vitamins were vitamin C, thiamin and biotin and the content of vitamin C was higher than others. These vitamins contained more in leaf than in stem or root. Niacin contained 6.09 mg/100 g in leaf of watercress from Hwasoon, which was much higher than others, but it was not detected in stem of watercress from Jeonju. Organic acids detected were oxalic acid, citric acid and malic acid and other 12 organic acids were not detected. In fatty acid composition, there were significant differences among watercresses from different parts and different culture fields. Linoleic acid, linolenic acid and palmitic acid were major fatty acids contained in watercress and it took about 80% of the total content. Amino acid content in leaf was higher than that in root and in stem. Glutamic acid and proline were major amino acids in stem of watercress from Jeonju and in stem of watercresses from Hwasoon and Donggok, respectively. In leaves of all three watercresses glutamic acid content was the highest.

  • A fusion protein of human interleukin-2(hiL-2) and glucagon which was expressed in Escherichia coli. was digested with enterokinase for recovery of hIL-2 from the fused protein. To obtain hIL-2 of optimum recovery hydrolysis reaction were performed under various conditions of urea additives and reaction time. hIL-2 was finally purified by RP-HPLC(reversed phase-HPLC) to remove cleaved G3 fusion partner and residual uncleaved G3-IL-2 HIL-2 was eluted in a single peak at 100% acetonitrile at 28 min. Optimum urea concentration was found to be 0.5 M and 24 h reaction time was sufficient without any additive such as CaCl2 and Tween-20.

  • Corrosion inhibitors including calcium hydroxide have been used to prevent corrosion in the pipes for tap water supply. The corrosion index (i.e., Langelier Index) differs by area and water quality. The corrosion indices of the areas studied differed by more than 2.0. The 'homogenized' calcium hydroxide was added to the treated water at the K water treatment plant, in order to increase the value of the corrosion index and the concentration of calcium. As the result, the concentration of calcium was increased while the turbidity and pH changed little. The corrosion rate of the tap water with the 'homogenized' calcium hydroxide could be slowed down pretty much. The results suggested that the technology of 'homogenization' of calcium hydroxide can applied to tap water and desalinated water to prevent corrosion in water pipes even in corrosive pipes.

Effect of Yanggyuksanhwa-tang on Ischemic Damage in Organotypic Hippocampal Slice Culture (양격산화탕(凉膈散火湯)이 뇌해마 조직배양의 허혈손상에 따른 신경세포손상에 미치는 영향)

  • Objectives : We can find out the experimental reports of Yanggyuksanhwa-tang, which has the function of regulating blood pressure related with cerebral disease, and increasing local cerebral blood stream volume, also has the recoveries for the damage of vessel endothelium, and endothelium hypertrophy caused by angiospasm after subarachnoid hemorrhage, and reduces the contraction of smooth muscle, so simultaneously improves necrosis. The aim of this study is to investigate effect of Yanggyuksanhwa-tang protecting neuronal cells from being damaged by brain ischemia through using organotypic hippocampal slice cultures. Methods : We caused ischemic damage to organotypic hippocampal slice cultures by oxygen and glucose deprivation, and Yanggyuksanhwa-tang extract was added to cultures. Thereafter we measured area percentage of propidium iodide (PI)-stained neuronal cell, lactate dehydrogenase (LDH) levels in culture media and terminal deoxynucleotidyl transferase-mediated dUTP nick-end labeling (TUNEL)-positive cells. Results : Area percentage of PI-stained neuronal cells and count of TUNEL-positive cells in CA1 and DG area of organotypic hippocampal slice culture were significantly decreased in pertinent density level of Yanggyuksanhwa-tang extract. LDH levels in culture media of organotypic hippocampal slice culture were significantly decreased in pertinent density level of Yanggyuksanhwa-tang extract. Conclusions : Within pertinent density level, Yanggyuksanhwa-tang has cell protection effect that prevents brain ischemia damaging neuronal cells and apoptosis increasing.

  • Invertase (${\beta}$-D-fructofuranosidase; EC 3.2.1.26) catalyzes the conversion of sucrose into glucose and fructose and is involved in an array of important processes, including phloem unloading, carbon partitioning, the response to pathogens, and the control of cell differentiation and development. Its importance may have caused the invertases to evolve into a multigene family whose members are regulated by a variety of different mechanisms, such as pH, sucrose levels, and inhibitor proteins. Although putative invertase inhibitors in the Arabidopsis genome are easy to locate, few studies have been conducted to elucidate their individual functions in vivo in plant growth and development because of their high redundancy. In this study we assessed the functional role of the putative invertase inhibitors in Arabidopsis by generating transgenic plants harboring a putative invertase inhibitor gene under the control of the CaMV35S promoter. A transgenic plant that expressed high levels of the putative invertase inhibitor transcript when grown under normal conditions was chosen for the current study. To our surprise, the stability of the invertase inhibitor transcripts was shown to be down-regulated by the phytohormone ABA (abscisic acid). It is well established that ABA enhances invertase activity in vivo but the underlying mechanisms are still poorly understood. Our results thus suggest that one way ABA regulates invertase activity is by down-regulating its inhibitor.

Molecular Characterization of Cold-Inducible ${\beta}$-Galactosidase from Arthrobacter sp. ON14 Isolated from Antarctica

  • A psychrotrophic bacterium, Arthrobacter sp. ON14, isolated from Antarctica, was shown to exhibit a high ${\beta}$-galactosidase activity at a low temperature. A genomic library of ON14 was constructed and screened for ${\beta}$-galactosidase genes on functional plates containing 5-bromo-4-chloro-3-indolyl-${\beta}$-D-galactopyranoside (X-gal) as the substrate. Two different ${\beta}$-galactosidase genes, named as galA, galB, were found in ON14. Computational analyses of the genes revealed that the encoded protein GalA belongs to family 2 of glycosyl hydrolysases and is a cold-active protein, whereas GalB belongs to family 42 of glycosyl hydrolysases and is a mesophilic protein. Reverse transcription analyses revealed that the expression of galA is highly induced at a low temperature ($4^{\circ}C$ ) and repressed at a high temperature ($28^{\circ}C$ ) when lactose is used as the sole carbon source. Conversely, the expression of galB is inhibited at a low temperature and induced at a high temperature. The purified GalA showed its peak activity at $15^{\circ}C$ and pH 8. The mineral ions $Na^+$, $K^+$, $Mg^{2+}$, and $Mn^{2+}$ were identified as enzyme activators, whereas $Ca^{2+}$ had no influence on the enzyme activity. An enzyme stability assay revealed that the activity of GalA is significantly decreased when it is incubated at $45^{\circ}C$ for 2 h, and all its activity is lost when it is incubated at $50^{\circ}C$.

Effects of Herbal-Acupuncture with Evodiae Fructus at KI10 on Osteoporosis in Ovariectomized Mice (음곡(陰谷) 오수유(吳茱萸) 약침(藥鍼)이 난소적출(卵巢摘出)생쥐의 골다공증(骨多孔症)에 미치는 영향(影響))

  • Objective : This study is aimed to evaluate the effects of Evodiae Fructus herbal-acupuncture (EF-HA) at KI10 on osteoporosis induced by ovariectomy in mice. Method : Mice underwent bilateral ovariectomy. After recovering, the ovariectomized (OVX) mice were treated by needle prick, saline injection, herbal acupuncture with Evodiae Fructus (EF-HA) at KI10 for 8 weeks. Result : 1. EF-HA at KI10 significantly inhibited the overgrowth of tibia in ovariectomized mice. 2. NP at KI10 significantly restored the tibial BMD (bone mineral density) in ovariectomized mice. 3. EF-HA at KI10 significantly restored the phosphorus and creatinine levels in ovariectomized mice serum. 4. EF-HA at KI10 significantly restored the tibial Ca and P levels in ovariectomized mice. 5. EF-HA at KI10 significantly reduced the tibial osteoclast-like cells in ovariectomized mice. 6. EF-HA at KI10 significantly inhibited the overgrowth of tibial GPL (growth plate length) in ovariectomized mice. Conclusion : EF-HA at KI10 has protective and therapeutic effect for osteoporosis in ovariectomized mice. Thus, it is suggested that EF-HA can be an useful therapeutics in clinical field after further researches.

Effects of Surface Water Chemistry and Physicochemical Characteristics of Humic Acid on Fouling of Membrane (원수의 수질화학과 HA의 물리화학적 특성이 막 오염에 미치는 영향)

  • In this study, we investigated the removal efficiencies of pollutants and permeate fluxes depending on chemistry of feed water, various molecular weight cut-offs (MWCOs) and materials of membrane, operating pressure. We used seven MWCO membranes of YC0.5, YM1, YM3, YM10, YM30, YM100 and PM30, humic acid solution and surface water as feed water, and examined variation in permeate flux. Results of TOC removal experiment demonstrate that MWCO lower 1,000daltons could remove humic acid effectively. As increasing solution pH and decreasing divalent cations ($Ca^{2+}$) concentration, TOC removal increased. But $UV_{254}$ removal efficiency increased with higher divalent cation concentration and solution pH. Membrane fouling increased with increasing electrolyte (NaCl), divalent cation concentration and decreasing solution pH. In spite of initial permeate flux of the hydrophobic membrane (PM30) was higher than that of the hydrophilic membrane (YM30), flux decline of PM30 was significant during operation. At higher operating pressure, compactness of the cake layer on the membrane surface increased, resulting in gradual increase in hydraulic resistance.
