Chemical Components of White and Red Lotus (목련과 홍련의 화학성분)

  • To get basic data for the utilization of white and red lotus as a raw material in functional food, chemical components of its leaf and root were investigated. Leaf had the highest level of nitrogen free extract and root had the highest level of moisture. The mineral analysis showed K(380.44~1,516.34 mg/100g), Ca(36.67~1,323.92 mg/100g), P(84.02~473.13 mg/100g) and Na(57.73~304.07 mg/100g). The rich free sugars in white and red lotus were glucose(161.12~765.15 mg/100g) and frutose (100.57~901.12 mg/100g). Total amino acid contents in leaf and root of white lotus and leaf and root of red lotus were 6,385.57, 1,162.93, 6,003.01 and 1,242.20 mg/100g, respectively. Although the free amino acid compositions of white and red lotus were different, their major free amino acids were glutamic acid, phenylalanine arginine and tyrosine. The ascorbic acid were 248.65 and 20.99 mg/100g in leaf and root of white lotus, 156.92 and 9.32 mg/100g in leaf and root of red lotus. The leaf of white lotus exhibited the highest total phenolic contents at 24.33 mg/g.

Assessment of Bio-corrosive Effect and Determination of Controlling Targets among Microflora for Application of Multi-functional CFB on Cement Structure (다기능 탄산칼슘 형성세균의 시멘트 건축물 적용위한 부식능 평가 및 건축물 정주미생물 중 방제 대상 결정)

  • The use of calcite-forming bacteria (CFB) in crack remediation and durability improvements in construction materials creates a permanent and environmentally-friendly material. Therefore, research into this type of application is stimulating interdisciplinary studies between microbiology and architectural engineering. However, the mechanisms giving rise to these materials are dependent on calcite precipitation by the metabolism of the CFB, which raises concerns about possible hazards to cement-based construction due to microbial metabolic acid production. The aim of this study was to determine target microorganisms that possibly can have bio-corrosive effects on cement mortar and to assess multi-functional CFBs for their safe application to cement structures. The chalky test was first used to evaluate the $CaCO_3$ solubilization feature of construction sites by fungi, yeast, bacterial strains. Not all bacterial strains are able to solubilize $CaCO_3$, but C. sphaerospermum KNUC253 or P. prolifica KNUC263 showed $CaCO_3$ solubilization activity. Therefore, these two strains were identified as target microorganisms that require control in cement structures. The registered patented strains Bacillus aryabhatti KNUC205, Arthrobacter nicotianae KNUC2100, B. thuringiensis KNUC2103 and Stenotrophomonas maltophilia KNUC2106, reported as multifunctional CFB (fungal growth inhibition, crack remediation, and water permeability reduction of cement surfaces) and isolated from Dokdo or construction site were unable to solubilize $CaCO_3$. Notably, B. aryabhatti KNUC205 and A. nicotianae KNUC2100 could not hydrolyze cellulose or protein, which can be the major constituent macromolecules of internal materials for buildings. These results show that several reported multi-functional CFB can be applied to cement structures or diverse building environments without corrosive or bio-deteriorative risks.

Physicochemical Properties of Sweet Potato (Ipomoea batatas) Starch by Heat-Moisture Treatment (수분-열처리에 따른 고구마 전분의 이화학적 성질)

  • Physicochemical properties on heat-moisture treatment of sweet potato starch were investigated. Starch granules of sweet potato observed by photomicroscope, polarized-light microscope and scanning electron microscope were round and polygonal. X-ray diffraction pattern was changed from Ca-pattern to A-pattern upon heat-moisture treatment. Water binding capacity was drastically increased as the moisture level was increased. The swelling power and solubility at the same temperature were decreased by heat-moisture treatment. Transmittance of 0.1% starch suspensions was increased rapidly from $65^{\circ}C$ in case of untreated starch and from $70^{\circ}C$ in case of treated starch. The starch on heat-moisture treatment was gelatinized over higher temperature range than the untreated starch.

Quality Characteristics of Cod Bone Stock Containing Various Amounts of Tomatoes (토마토 첨가량을 달리한 대구뼈 육수의 품질특성에 관한 연구)

  • This study was performed to develop cod bone stock using various nutritional elements in cod bone with the addition of various amounts of tomatoes (2%, 4%, 6% and 8%). Moisture contents, pH, L value and Na contents decreased, while the salinity, sugar contents, a value, b value and Mg, K, Ca, Fe contents increased with increased rates of tomatoes. Fourteen types of free amino acid were detected, and stock containing 6% of tomato addition had the highest free amino acid content. In the test on different attributes, tomato content significantly affected the properties of the stock including color intensity, savory flavor, tomato flavor, tomato taste, savory taste and after taste. In the acceptance test, 6% of tomato addition was preferred for appearance, flavor, taste, texture and overall quality. Consequently, the optimal tomato content for maximizing the overall quality of cod bone stock was 6%.

  • These experiment were conducted to investigate the characteristics of the purified lipase of Rhizopus japonicus. 1. The optimum pH for the lipase action was 6.8, the stable pH ranged $5.0{\sim}8.0$ and the optimum temperature was $35^{\circ}C$. The Purified lipase was stable below $45^{\circ}C$ and inactivated abruptly above $45^{\circ}C$ 2. Coconutoil castor oil and palm oil were most favorably hydrolyzed by the purified lipase, wheres olive oil and soybean oil were slightly less than the former three, and Tween 80 was hydrolyzed only 30% compared with coconut oil. 3. The purified lipase was activated by $Ca^{++}$ and $Mg^{++}$, whereas it was inhibited by $Hg^{++}$ and $Fe^{++}$.

  • The crystal structure of K92-X (K92Al92Si100O384), a=25.128(1) Å, dehydrated at 360 ℃ and 2X 10-6 Torr, has been determined by single-crystal X-ray diffraction techniques in the cubic space group Fd&bar{3} at 21(1) ℃. The structure was refined to the final error indices R1=0.044 and Rw=0.039 with 242 reflections for which I<3σ(I). In this structure, ninety-two K+ ions are located at the five different crystallographic sites. Sixteen K+ ions are located at the centers of the double six rings (site I; K(1)-O(3)=2.65(2) Å and O(3)-K(1)-O(3)=92.0(6)°). About twelve K+ ions lie at site I' in the sodalite cavity opposite double six rings (D6R's) and these K+ ions are recessed ca. 1.62 Å into the sodalite cavity from their O(3) plane (K(2)-O(3)=2.74(2) Å, O(3)-K(2)-O(3)=88.5(8)°). About thirty-two K+ ions are located at the site II in the supercage and these K+ ions are recessed ca. 1.20 Å into the supercage from their O(2) plane (K(3)-O(2)=2.64(2) Å, and O(2)-K(3)-O(2)=101(1)°). About twenty-two K+ ions lie at the site III in the supercage opposite 4-ring ladder and the remaining ten K+ ions lie at the site III' near the 4-ring ladder in the supercage (K(4)-O(4)=2.88(3) Å, O(4)-K(4)-O(4)=79.8(9)°, K(5)-O(4)=2.8(2) Å, and O(4)-K(5)-O(4)=68(5)°).

Chemical Properties and Heavy Metal Content of Forest Soils around Abandoned Coal Mine Lands in the Mungyeong Area (문경지역 폐탄광지 주변 산림토양의 화학적 성질 및 중금속 함량)

  • Chemical properties and heavy metal concentrations of forest soils of four abandoned coal mine lands affected by coal mining activities in the Mungyeong area were investigated to provide basic information for revegetation of abandoned coal mine lands. Soil pH in abandoned coal mine lands ranged from 5.30 to 6.76 it in the control site was 5.23. Contents of organic matter and total N in abandoned coal mine lands were $4.46\~7.19\%\;and\;0.07\~0.15\%$, respectively. Available P contents were 6.54 for A (Samchang), 6.52 for B (Bongmyeong),3.94 fur C (Kabjung), 5.45 mg/kg for D (Danbong coal mine land) and 5.25 mg/kg for the control site, which had a positive correlation with soil pH. Contents of exchangeable Ca, Mg, K and Na in abandoned coal mile lands averaged 196.1, 88.7, 88.2 and $10.2cmol^+/kg$, with a range of $132.1\~242.1,\;24.2\~138.\; 64.9\~120.8\;and\;8\~12.2cmol^+/kg$, respectively. Those of the control site were 192.8, 95.8, 104 and $21.2 cmol^+/kg$, respectively. Heavy metals such as Al, As, Cd, Cr, Cu, Fe, Mn, Ni, Pb and Zn of forest soil in abandoned coal mine lands have a larger content than those of the control site. Al, Mn and fb content was especially high in abandoned coal mine lands. The Al content of forest soil in abandoned coal mine lands ranged from 397 to 917 ppm, which was considered to be high enough to inhibit tree growth. Therefore, it is suggested that soils of abandoned coal mine lands contaminated by mining activities need to be properly treated for remediation of environmental problems.

Purification and Properties of Cyclodextrin Glucanotrnsferase Synthesizing $2-O-{\alpha}-D-Glucopyranosyl{\;}_{L}-Ascorbic$ Acid from Paenibacillus sp. JB-13

  • A Gram-positive bacterium (strain JB-13) that was isolated from soil as a producer of cyclodextrin glucanotransferase (CGTase) [EC 2.4.1.19] was identified as Panibacillus sp. JB-13. This CGTase could catalyze the transglucosylation reaction from soluble starch to L-ascorbic acid (AA). A main product formed by this enzyme with ${\alpha}-glucosidase$ was identified as $2-O-{\alpha}-D-glucopyranosyl{\;}_{L}-ascorbic$ acid (AA-2G) by the HPLC profile and the elemental analysis. CGTase was purified to homogeneity using ammonium sulfate fractionation, ion-exchange chromatography on DEAE-Seohadex A-50, and gel chromatography on Sephacryl S-200HR. The molecular weight was determined to be 66,000 by both gel chromatography and SDS-PAGE. The isoelectric point of the purified enzyme was 5.3. The optimum pH and temperature was PH 7.0 and $45^{\circ}C$ respectively. The enzyme was stable in the range of pH 6-9 and at temperatures of $75{\circ}C$ or less in the presence of 15 mM ${CaCl_2}.\;{Hg^2+},\;{Mn^+2},{Ag^+},\;and\;{Cu^2+}$ all strongly inhibited the enzyme's activity.

ABSORPTION LINE GRADIENTS OF AN ELLIPTICAL GALAXY

  • Archival long-slit spectra, covering the wavelength range 4050~5150$\AA$, have been used to investigate the radial behavior of absorption line features (G4300, Fe4383, Ca4455, Fe44531, Fe4668, and H$\beta$) of an eliptical galaxy NGC 5322. The heliocentric recession velocity of NGC 5322 has been derived as 1888$\pm$51kms-1. Metallic absorption lines of NGC 5322 show significant radial gradients through the major axis. The minor axis shows much smaller radial metal line gradients than the major axis. The minor axis shows much smaller radial metal line gradients than the major axis. The mean slopes of Fe line gradients to the major and minor axes of NGC 5322 were estimated as -0.433$\pm$0.064 and -0.242$\pm$0.096, respectively. Significant radial gradients of H$\beta$ absorption of NGC 5322 are also detected both on the major and minor axes. It is shown that the radial metallicity gradients in NGC 5322 are smaller than expected in a simple dissipative collapse model. Rather, dissipationless collapse, such as hierarchical merhing, could have contributed during the initial stage of the galaxy formation.

A Study on the Joining of Different Al Alloys by Centrifugal Casting (원심주조를 이용한 2종 알루미늄의 접합에 대한 연구)

  • To improve the quality of the product and the cost efficiency, the joining of A356 alloy to an Al-18wt%Si alloys has been performed by centrifugal casting. The influence of the mold preheating temperature, the pouring temperature and the rotational velocity of the mold on the microstructures of the shell in the centrifugal casting was investigated using the experimental and simulation methods. In the present study, the cellular automaton (CA) technique and the finite volume method (FVM) were adopted to simulate the evolution of the macro structures and to calculate the temperature profiles, respectively. The evolution of the microstructures was also simulated using a modified cellular automaton (MCA) model. The optimal rotational speed of the mold for obtaining the sound shape of the shell was estimated experimentally to be over 1200 rpm. For the uniform microstructure, the outer shell needs to be cast with higher preheated mold temperature and lower pouring temperature, and the melt was poured at lower temperature in the inner shell. In order to obtain the sound shape of the joining, the different materials were poured simultaneously.
