Analgesic Effects of Sokyungwhalhyul-tang on Constriction Nerve Injury-Induced Neuropathic Pain in Rats (말초 신경병증성 통증 모델에서 소경활혈탕의 진통 효과)

  • Nardostachys chinensis;Anti-proliferation;Cell cycle arrest;Differentiation;U937 cells; This study was conducted to determine the analgesic effect of Sokyungwhalhyul-tang(SKWHT) using the model of peripheral neuropathic pain model. A model of neuropathic pain was made by ligating left 5th lumbar spinal nerve of rats. After 1 days, the extract of SKWHT was orally administered daily. Rats were divided into four groups; (1) Control group(n=6), (2) Experimental group I(SKWHT-OA1, 100 mg/kg, n=6), (3) Experimental group II(SKWHT-OA2, 300 mg/kg, n=6), (4) Experimental group III(SKWHT-OA3, 500 mg/kg, n=6). After that, we examined the withdrawl response of neuropathic rats legs by von Frey filament and Hot plate at pre, $1^{th}$, $4^{th}$, $7^{th}$, $14^{th}$, $21^{th}$ days after the induction of neuropathic pain. And also we examined c-fos, GOT, GPT and histological study of Liver at 21th days. von Frey filament and Hot plate were increase in experimental group I, II, III than Con. especially group III was most significantly analgesic effect than the other groups at $14^{th}$, $21^{th}$ days. In c-fos protein expression on spinal cord, group III was most significantly reduction immunoreactivity at $21^{th}$ days and in blood serum GOT & GPT levels and histologic finding of Liver in all experimental groups were no significant difference with Con at $21^{th}$ days. According to the above results, SKWHT(500 mg/kg) may have a significant analgesic effect on the neuropathic pain.

A Retrospective Analysis of Use in Hospitalized Children with Upper Respiratory Tract Infection (상기도 감염으로 입원한 소아환자에서 항생제 사용에 대한 후향적 분석)

  • Purpose: The inappropriate prescription of antibiotics in children with upper respiratory tract infection (URTI) is common. This study evaluated the factors that influence antibiotics use in hospitalized children with viral URTI confirmed by reverse transcriptase-polymerase chain reaction (RTPCR) assay. Methods: The medical records of admitted patients who performed RT-PCR assay for respiratory virus pathogens from January 2013 to November 2014 were examined. The demographic and clinical features were compared between patients who were administered antibiotics at admission and those who were not. We also investigated differences between children who continued antibiotics and those who stopped antibiotics after a viral pathogen was identified. Results: In the total 393 inpatients, the median age was 23 months (interquartile range, 13 to 41.3 months). Antimicrobial agents were prescribed in 79 patients (20.1%) at admission. Patients with acute otitis media (AOM) had higher rates of antibiotics prescription than those without AOM (48.1% vs. 2.2%, P<0.001), with an adjusted odds ratio of 91.1 (95% confidence interval, 30.5 to 271.7). Level of high-sensitivity C-reactive protein and the proportion of acute rhinosinusitis were also significantly associated with antibiotics use (P<0.001). Among the 44 patients with viruses identified using the RT-PCR method during hospitalization, antibiotic use was continued in 28 patients (63.6%). AOM was statistically associated with continued antibiotic use in the patients (P=0.002). Conclusions: Although the respiratory virus responsible for URTI etiology is identified, clinicians might not discontinue antibiotics if AOM is accompanying. Therefore, careful diagnosis and management of AOM could be a strategy to reduce unjustified antibiotic prescriptions for children with URTI.

Molecular Cloning and Expression of a Cu/Zn-Containing Superoxide Dismutase from Thellungiella halophila

  • Superoxide dismutases (SODs) constitute the first line of cellular defense against oxidative stress in plants. SODs generally occur in three different forms with Cu/Zn, Fe, or Mn as prosthetic metals. We cloned the full-length cDNA of the Thellungiella halophila Cu/Zn-SOD gene ThCSD using degenerate RT-PCR and rapid amplification of cDNA ends (RACE). Sequence analysis indicated that the ThCSD gene (GenBank accession number EF405867) had an open reading frame of 456 bp. The deduced 152-amino acid polypeptide had a predicted molecular weight of 15.1 kDa, an estimated pI of 5.4, and a putative Cu/Zn-binding site. Recombinant ThCSD protein was expressed in Escherichia coli and assayed for SOD enzymatic activity in a native polyacrylamide gel. The SOD activity of ThCSD was inactivated by potassium cyanide and hydrogen peroxide but not by sodium azide, confirming that ThCSD is a Cu/Zn-SOD. Northern blotting demonstrated that ThCSD is expressed in roots, stems, and leaves. ThCSD mRNA levels increased by about 30-fold when plants were treated with sodium chloride (NaCl), abscisic acid (ABA), and indole-acetic acid (IAA) and by about 50-fold when treated with UVB light. These results indicate that ThCSD is involved in physiological pathways activated by a variety of environmental conditions.

Comparison of Nutrient Intakes between Korean and United States Adults (한국성인과 미국성인의 영양섭취 실태 비교 연구)

  • Dietary Variety Score (DVS) or Nutrient Adequacy Ratio (NAR) offers a way of comparing eating habits across populations and across countries. This study compared nutrients intakes, food intakes, DVS, and NAR between Korean and US adults using the large national survey data. 4662 Korean adults (men: 2201, women: 2461) aged 20-49 years were selected from the 'Korean National Health and Nutrition Survey,2001' and 4199 US adults (men: 2127, women: 2072) aged 20-49 years were selected from the 'US National Health and Nutrition Examination Survey (NHANES IV,1999-2002)'. The total plant food intakes were high in Korean adults, but the total animal food intakes were high in US adults. Intakes of legumes and nuts, vegetables and mushrooms were higher in Korean than in US adults. On the other hand, intakes of meats, milk and dairy products of US adults were higher than Korean. Beverage intakes were 8 - 10 times higher in US adults than in Korean. The mean DVS of Korean men and women were higher than US men and women. Intakes of fat, calcium and vitamin $B\_{2}$ of US adults were 1.5 - 2 times higher than those of Korean, which could probably related to larger consumption by US adults on milk and dairy product. While the intakes of carbohydrate and vitamin C of the Korean were higher than the US adults, which could presumably related to higher grain and vegetable consumption. Percent energy intakes from carbohydrate, protein, and fat were $64.1\%$, $15.8\%$, and $20.1\%$ for Korean men, $52.4\%$, $15.0\%$, and $32.6\%$ for US men, $65.9\%$, $14.9\%$, $19.2\%$ for Korean women, and $52.1\%$, $14.7\%$, $33.2\%$ for US women respectively, which showed higher $\%$energy from carbohydrate in Korean and higher $\%$energy from fat in US adults. Most nutrient intakes except energy and vitamin C expressed as $\%$RDA were higher in US than in Korean adults. Mean Adequacy Ratio (MAR) of US men and Korean men were both 0.85, while the MAR was 0.81 for Korean women,0.79 for US women. The Korean women's diet showed more diverse and adequate than the US women's diet.

Effects of luteolin on chemical induced colon carcinogenesis in high fat diet-fed obese mouse (고지방식이를 급여한 비만 마우스에서 luteolin이 화학적으로 유도한 대장암 발생에 미치는 영향)

  • Purpose: Colorectal cancer, which is one of the most commonly diagnosed cancers in developing and developed countries, is highly associated with obesity. The association is largely attributed to changes to western style diets in those countries containing high-fat and high-energy. Luteolin (LUT) is a known potent inhibitor of inflammation, obesity, and cancer. In this study, we investigated the effects of LUT on chemical-induced colon carcinogenesis in high fat diet (HFD)-fed obese mice. Methods: Five-week-old male C57BL/6 mice received a single intraperitoneal injection of azoxymethane (AOM) at a dose of 12.5 mg/kg body weight. Mice were then divided into four groups (n = 10) that received one of the following diets for 11 weeks after the AOM injection: normal diet (ND); HFD; HFD with 0.0025% LUT (HFD LL); HFD with 0.005% LUT (HFD HL). One week after AOM injection, animals received 1~2% dextran sodium sulfate in their drinking water over three cycles consisting of five consecutive days each that were separated by 16 days. Results: Body weight, ratio of colon weight/length, and tumor multiplicity increased significantly in the HFD group compared to the ND group. Luteolin supplementation of the HFD significantly reduced the ratio of colon weight/length and colon tumors, but not body weight. The levels of plasma $TNF-{\alpha}$ and colonic expression of inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 protein increased in response to HFD, but were suppressed by LUT supplementation. Immunohistochemistry analysis also showed that iNOS expression was decreased by LUT. Conclusion: Consumption of LUT may reduce the risk of obesity-associated colorectal cancer by suppression of colonic inflammation.

Chemical composition and in vitro digestibility of corn stover during field exposure and the fermentation characteristics of silage prepared with microbial additives

  • Objective: To effectively use corn stover resources as animal feed, we explored the chemical composition and in vitro digestibility of corn stover during field exposure and the fermentation characteristics of silage prepared with lactic acid bacteria (LAB) and cellulase. Methods: Corn ears including the cobs and shucks were harvested at the ripe stage. The corn stover was exposed in the field under natural weather conditions. Silages were prepared after 0, 2, 4, 7, 15, 30, and 60 d of exposure. Corn stover was chopped into approximately 1 to 2 cm lengths and then packed into 5 liter plastic silos. The ensiling density was $550.1{\pm}20.0g/L$ of fresh matter, and the silos were kept at room temperature ($10^{\circ}C$ to $25^{\circ}C$). Silage treatments were designed as follows: without additives (control), with LAB, with cellulase, and with LAB+ cellulase. After 45 d of fermentation, the silos were opened for chemical composition, fermentation quality and in vitro digestion analyses. Results: After harvest, corn stover contained 78.19% moisture, 9.01% crude protein (CP) and 64.54% neutral detergent fiber (NDF) on a dry matter (DM) basis. During field exposure, the DM, NDF, and acid detergent fiber (ADF) contents of corn stover increased, whereas the CP and water-soluble carbohydrate contents and in vitro digestibility of the DM and CP decreased (p<0.05). Compared to the control silage, cellulase-treated silage had lower (p<0.05) NDF and ADF contents. The pH values were lower in silage treated with LAB, cellulase, or LAB+cellulase, and lactic acid contents were higher (p<0.05) than those of the control. Silage treated with cellulase or LAB+cellulase improved (p<0.05) the in vitro DM digestibility (IVDMD) compared to that of the control or LAB-treated silage. Conclusion: Corn stover silage should be prepared using fresh materials since stover nutrients are lost during field exposure, and LAB and cellulase can improve silage fermentation and IVDMD.

Comparisons of Physicochemical Composition of Korean and Chinese Crataegi Fructrus (한국산 산사와 중국산 산사의 이화학적 성분 비교)

  • This study was conducted to compare the major chemical components of Korea Crataegi fructrus (KCF) and Chinese Crataegi fructrus (CCF). Among the proximate compositions, the curde fat content of KCF was lower than that of CCF, whereas their crude protein, crude ash and carbohydrate contents were similar. CCF had a higher total free sugar content than KCF. The major free sugars of KCF and CCF were identified as fructose and glucose. The value of glutamic acid was greater in the amino acids of KCF and CCF, and KCF had higher total amino acids and essential amino acids contents than CCF. KCF also had a higher level unsaturated fatty acids than CCF. CCF had a higher organic acid content, but both KCF and CCF had high citric acid levels. and Chinese The vitamin C contents of KCF and CCF were 272.69 mg per 100 g and 262.38 mg per100 g, respectively. The mineral content of KCF was higher than that of CCF, in the following order : K > Ca > Mg > Fe. The results showed that KCF had higher total amino acid, essential amino acid, unsaturated fatty acid and mineral contents and CCF had higher free sugar and organic acid contents.

Effect of keratinase on ileal amino acid digestibility in five feedstuffs fed to growing pigs

  • Objective: This study was conducted to evaluate the effect of keratinase (KE) on the apparent ileal digestibility (AID) and standardized ileal digestibility (SID) of amino acids (AA) in rice bran, cottonseed meal (CSM), rapeseed meal (RSM), corn distillers dried grains with solubles (DDGS), and peanut meal (PNM). Methods: Twelve crossbred barrows (Duroc${\times}$Landrace${\times}$Yorkshire, $50.5{\pm}1.4kg$ body weight [BW]) fitted with T-cannulas at the terminal ileum were allotted to a $12{\times}6$ Youden Square design with 12 diets and 6 periods. The treatment diets included rice bran, CSM, RSM, corn DDGS, PNM, or corn-soybean meal (cSBM) supplemented with 0.05% KE or not. Diets were given to pigs at a level of 3% BW in two equal meals. The endogenous AA losses were the mean results of three previously experiments determined by a same nitrogen-free diet fed to pigs. Pigs had free access to water during the experiment. Results: The KE supplementation improved (p<0.05) the AID and SID of Met, Thr, Val, Asp, Cys, and Tyr in rice bran. Inclusion of KE increased (p<0.05) the AID and SID of Met and Val in CSM. The KE supplementation decreased (p<0.05) the AID and SID of His in RSM and all measured AA except for Arg, Met, Trp, Val, Gly, and Pro in corn DDGS. There was an increase (p<0.05) in AID and SID of Leu, Ile, Met, Ala, Cys, Ser, and Tyr in PNM supplemented with KE compared with that without KE. Inclusion of KE increased (p<0.05) the AID and SID of crude protein, Leu, Ile, Phe, Thr, Asp, and Ser in cSBM. Conclusion: This study indicated that KE had different effects on ileal AA digestibility of feedstuffs for growing pigs, which can give some usage directions of KE in swine feed containing those detected feedstuffs.

Relative Association of Overhydration and Muscle Wasting with Mortality in Hemodialysis Patients: Assessment by Bioelectrical Impedance Analysis (혈액투석 환자에서 Bioelectrical Impedance Analysis를 활용하여 측정한 과수분량과 근육량 감소와 사망률의 상관관계)

  • Background/Aims: Assessment of fluid status in hemodialysis patents is very important. Overhydration in hemodialysis is associated with generalized edema, cardiovascular complications, and hypertension. The aim of this study was to determine the factors correlated with mortality of hemodialysis patients, assessing body muscle mass and fluid status using bioelectrical impedance analysis (BIA). Methods: This study enrolled 93 patients who underwent hemodialysis between January 2010 and May 2015 at CHA Bundang Medical Center. Medical records of enrollees up to June 2017 were reviewed retrospectively. These included laboratory results (serum albumin, C-reactive protein [CRP], lipid profile, etc.) and BIA data (extracellular water, intracellular water, total body water, soft lean mass, fat free mass, skeletal muscle mass, etc.). Results: Eleven of 93 patients had expired by May 2017. Among the surviving subjects, mean age was younger, CRP levels were lower, albumin levels were higher, and extracellular water/total body water (ECW/TBW) ratios were lower than in the expired patient group. Kaplan-Meier survival analysis revealed that overhydration (ECW/TBW > 0.4) was associated with higher mortality. Conclusions: In hemodialysis patients, overhydration is an important factor in mortality, and BIA could be a reliable modality in its assessment. We suggest that, for hemodialysis patients, overhydration is more of a risk factor for mortality than is muscle wasting.

The comparative study of predictive factors for prolonged length of stays that adult patients with acute appendicitis in emergency department (응급의료센터를 방문한 성인 급성 충수염 환자에서 재실 기간의 연장을 예측하는 인자에 대한 비교연구)

  • Objective: This study examined the predictive factors for prolonged length of stays of adult patients with acute appendicitis (AA) in an emergency department (ED). Methods: This was a retrospectively clinical study including patients in an ED. All patients were diagnosed from the clinical symptoms and a typical physical examination, and had undergone a computed tomography (CT) evaluation on the ED visiting date. All data were collected from the electrical medical records. The clinical parameters analyzed were the laboratory data, including the white blood cell count with differential values, C-reactive protein (CRP) level, initial vital signs, duration of admission, coexisting perforation of the appendix in the CT findings. The relationship between the clinical parameters and length of stay was assessed. Results: A total of 547 patients with AA were enrolled in this study. Among them, there were 270 male patients with a mean age of $40.7{\pm}15.8years$. The baseline characteristics, initial clinical features, laboratory, and imaging studies results of 129 patients in the prolonged length of stay (pLOS) group, and 418 patients of the non-pLOS group in AA were compared. Multivariable logistic regression analysis revealed the predictive factors related to pLOS in AA to be as follows: age 40 years or older, body temperature over $37.3^{\circ}C$, CRP level greater than 5.0 mg/dL, and evidence of perforation in CT findings (P<0.001). Conclusion: If we check age, fever, CRP level and find evidence of perforation, it might be helpful for predicting the increasing period of length of hospital stay for patients with AA in ED.
