Shelf-Life Extension of Sing Sing Hoe (Fresh Sliced Raw Fish) by Frozen Storage (냉동저장을 통한 싱싱회 저장기간 연장에 관한 연구)

  • Changes in pH, viable microbial count, chemical freshness, texture, and sensory qualities of Sing Sing Hoe (SSH, fresh-sliced raw fish) were measured over 15 days at $4^{\circ}C$, $-20^{\circ}C$, and $-80^{\circ}C$. The initial pH of SSH was 6.25 at all three storage temperatures, and pH increased slightly after 12 days to pH 6.48 and pH 6.55 at $-20^{\circ}C$ and $-80^{\circ}C$, respectively. The range in viable cell count was 104-106 CFU/g, regardless of storage temperature. The initial content of volatile basic nitrogen (VBN) was 5.8 mg/100 g and became 8.2 mg/100 g or less, and 7.9 mg/100 g or less after 15 days at $-20^{\circ}C$ and $-80^{\circ}C$, respectively. However, pH and VBN values increased significantly after 3 days of storage at $4^{\circ}C$. At this temperature, the K-value was 22.3% after 6 days and 40% or more after 15 days. At $-20^{\circ}C$, the K-value was 9.6% or less after 6 days and 21% or less after 15 days of storage. At $-80^{\circ}C$, the K-value was 8.5% or less after 9 days and 20% or less after 15 days of storage. Compared with the K-value of live fish muscle (10%), freshness similar to that of live fish was maintained for 6 days under both $-20^{\circ}C$ and $-80^{\circ}C$ storage conditions. There was no significant change in texture during storage of SSH at $-20^{\circ}C$ or $-80^{\circ}C$, but SSH stored at $4^{\circ}C$ showed a decrease in texture quality during storage. Sensory scores were high for material stored for up to 3 days at $4^{\circ}C$ and 6 days at $-20^{\circ}C$ or $-80^{\circ}C$. The overall freshness of SSH was maintained for up to 6 days, in comparison with fresh-sliced raw fish, under both frozen storage conditions.

Continuous Production of Authentic Human Growth Hormone from Methionyl Human Growth Hormone Using the Column Reactor of Immobilized Aminopeptidase M (고정화 Aminopeptidase M 컬럼 반응기를 이용한 메치오닐 인간성장호르몬으로부터 천연형 인간성장호르몬의 연속생산)

  • The characteristics of aminopeptidase M(ApM) immobilized covalently on Cellufine Formyl and the continuous production of authentic human growth hormone(hGH) from methionyl human growth hormono(met-hGH) using the column reactor packed with immobilized ApM were investigated. Immobilized ApM with the proportion of 2.3mg ApM per 1g Cellufine Formyl gel had the highest met-hGH conversion activity. The optimum pH(7.0) and temperature($55^{\circ}C$) showed no appreciable difference between free and immobilized enzymes and the optimum temperature in continuous operation of the column reactor was also found to be $55^{\circ}C$. Under the conditions at which met-hGH was converted completely to hGH, the yield and productivity were about 77% and 0.8mg hGH/ml$.$h, respectively. In two column reactors of different sizes, met-hGH was converted to hGH with the same conversion rates and hGH yields at the same space velocities. The half-life of the reactor systems at $45^{\circ}C$ and $55^{\circ}C$ were projected from the continuous operations for 90 days to be 225 days and 81 days, respectively.

Influence of elevated temperatures on the physiological response of hemolymph from two species of abalone, Haliotis gigantea and Haliotis discus discus (Reeve, 1846) (수온 증가에 따른 말전복, Haliotis gigantea과 둥근전복, Haliotis discus discus (Reeve, 1846) hemolymph의 생리학적 변화)

  • This study was conducted to examine the effects of alterations in water temperature (WT) on biochemical and immunological factors in the hemolymph of the abalones, Haliotis gigantea and H. discus discus. The abalone were exposed to various WT; 18, 20, 22, 24, 26 and $28^{\circ}C$ for 96 hours. In biochemical factors, total-protein (TP), glucose, magnesium (Mg), aspartate aminotransferase (AST) and alanine aminotransferase (ALT) were not significant changes in hemolymph of H. gigantea and H. discus discus. But calcium was significantly increased by high WT (${\geq}24^{\circ}C$). In immunological factor, The phenoloxidase (PO) activity was decreased in hemolymph of H. gigantea and H. discus discus exposed to high temperature (${\geq}22^{\circ}C$) compared to the control (P < 0.05). Whereas alkaline phosphatase (ALP) was not significantly changed. These results suggested that high temperature adversely affects the immunity of H. gigantea and H. discus discus.

Synthesis and Characterization of Group VI Metal Carbonyl Complexes Containing closo-1,2-$(PPh_2)_2$-1,2-$C_2B_1_0H_1_0$ and Their Conversion to Metal Carbene Complexes

  • The complexes M(CO)4-1,2-(PPh2)2-1,2-C2B10H10 (M=Cr 2a, Mo 2b, W 2c) have been prepared in good yields from readily available bis-diphenylphosphino-o-carboranyl ligand, closo-1,2-(PPh2)2-1,2-C2B10H10 (1), by direct reaction with Group Ⅵ metal carbonyls. The infrared spectra of the complexes indicate that there is an octahedral disposition of chelate bis-diphenylphosphino-o-carboranyl ligand around the metal atom. The crystal structure of 2a was determined by X-ray diffraction. Complex 2a crystallizes in the monoclinic space group P21/n with cell parameters a = 12.2360(7), b = 17.156(1), c = 16.2040(6) Å, V = 3354.1(3) Å3, and Z =4. Of the reflections measured a total of 2514 unique reflections with F2 > 3σ(F2) was used during subsequent structure refinement. Refinement converged to R1 = 0.066 and R2 = 0.071. Structural studies showed that the chromium atom had a slightly distorted pseudo-octahedral configuration about the metal center with two phosphine groups of o-carborane occupying the equatorial plane cis-orientation to each other. These metal carbonyl complexes are rapidly converted to the corresponding metal carbene complexes, [(CO)3M=C(OCH3)(CH3)]-1,2-(PPh2)2-1,2-C2B10H10 (M= Cr 3a, Mo 3b, W 3c), via alkylation with methyllithium followed by O-methylation with CF3SO3CH3.

Synthesis and Anti Bacterial and Anti-ulcer Evaluation of New S-mannich Bases of 4,6-diaryl-3,4-dihydropyrimidin-2(1H)-thiones

  • The synthesis of title compounds were accomplished by synthetic sequence shown in Scheme 1. Chalcones on cyclocondensation with thiourea in ethanol and potassium hydroxide under reflux yielded the respective dihydropyramidin-2(1H)-thiones. Each of the dihydropyrimidin thiones was, then subjected to the Mannich condensation in alkaline medium using three different secondary amines, viz., dimethylamine, diethylamine and morpholine to obtain a new series of S-Mannich bases. All the synthesised compounds ($C_1-C_{15}$) were evaluated for their antiulcer and antibacterial activities. Compounds $C_4$, $C_5$, $C_6$, $C_{14}$ and $C_{15}$ exhibited relatively more potent antiulcer activity but not comparable to the standard; Omeprazole, while $C_1$, $C_2$, $C_3$ and $C_{13}$ were moderate in activity at 100 mg/kg p.o. All the compounds ($C_1-C_{15}$) showed mild to moderate activity against both Gram-positive (S.aureus, L.delbrueckii) and Gram-negative (P.vulgaris, E.coli) bacteria. Amongst the compounds tested, only $C_6$, $C_9$, $C_{12}$ and $C_{15}$ were found to be potent.

Isolation and Identification of Sesquiterpene o-Naphthoquinones, Mansonones E, F and H, from the Root Bark of Ulmus davidiana Planch (당느릅나무로부터 Sesquiterpene o-Naphthoquinone류 화합물, Mansonone E, F 및 H의 분리와 구조결정)

  • Three sesquiterpene ortho-naphthoquinones were isolated from the methanolic extract of root bark of Ulmus davidiana Planch whose stem and root bark have been used as an oriental medicine for the treatment of edema, mastitis, gastric cancer and inflammation. The structures of these compounds were established on the basis of spectral data obtained from UV-vis, IR, HR-EIMS and NMR spectrometry, including the pulse field gradient (PFG)-HMQC and HMBC techniques. Their structures were determined as 2,3-dihydro-3,6,9-trimethylnaphtho(1,8-b,c)pyran-7,8-dione, 3,6,9-trimethylnaphtho(1,8-b,c)pyran-7,8-dione and 2,3-dihydro-4-hedroxy-3,6, 9-trimethylnaphtho(1,8-b,c)pyran-7,8-dione, which were identified as mansonones E. F and H, respectively. These compounds have originally been isolated from Mansonia altissima Chev, but have never been isolated from Ulmus davidiana Planch. Especially, mansonone H was isolated for the first time from Ulmaceae. The mismatched carbon chemical shifts of mansonones E and F in the reported literature were corrected by the aid of the PFG-HMBC spectral data.

Nickel Complexes Having (N-C-N) Tridentate Ligands ((N-C-N) 세자리 리간드를 가지는 니켈 착물)

  • Monomeric complexes of nickel(II) having terdentate bis(imino)aryl ligands (N,C,N-pincer) are reported. New complexes (2,6-(ArN=CH)2C6H3)NiBr (Ar=2,6-dimethylphenyl (1), 2,6-diisopropylphenyl (2)) have been synthesized through oxidative addition of 1,3-(ArN=CH)2C6H3Br (bis(N-Ar)-2-bromoisophthalaldimine: Ar=Ph-2,6-Me2, Ph-2,6-iPr2) to Ni(COD)2 (COD=1,5-cyclooctadiene), in high yields. The development of a synthetic route to ligands and nickel complexes is outlined. The complexes were characterized by IR, 1H-NMR and elemental analysis. Full characterization of complexes 1 and 2 is discussed. An investigation into the catalytic activity of the complexes in ethylene polymerization was performed, resulting in no formation of polyethylenes but producing a small amount of oily oligomers. Preliminary results indicate that the pincer complexes were found to be inactive as catalysts in ethylene polymerization.
