Molecular and Crystal Structure of' Metalaxyl, $C_{15}H_{21}NO_4$ (Metalaxyl, $C_{15}H_{21}NO_4$의 분자 및 결정구조)

  • The molecular and crystal structure of metalaxyl C/sub15/H/sub21/NO₄, was determined by single crystal x-ray diffraction study. Crystallographic data for, title compound P2₁/c, a=7.849(4) Å, b=13.081(5) Å, c=15.100(3) Å, β=101.8(2)°, V= 1517.6(3) ų, Z=4. The molecular. Structure model was solved by direct method and refined by full-matrix least- squares. The final reliable factor, R, is 0.067 for 1694 independent reflections (F/sub o//sup 2/>4σ(F/sub o//sup 2/)). The molecular structure of title compound shows an intramolecular hydrogen bond: Cl2-Hl2A…O1.

Isolation and Characterization of Cyclodextrin Glycosyl Transferase Producing Alkalophilic Bacillus sp. (Cyclodextrin glycosyltransferase를 생산하는 호알칼리성 Bacillus속 미생물)

  • A strain of alkalophilic Bacillus sp. YC-335 has been isolated from soil. The strain was capable of producing large amount of cyclodextrin glycosyl transferase (CGTase) in the culture broth. The preferable medium composition has been determined to be as follows : 1.5% soluble starch, 5% corn steep liquor, 0.1% $K_2$HPO$_4$, 0.02%mgSO$_4$.7$H_2O$, 1% CaCO$_3$and 1% Na$_2$CO$_3$(pH 10.3). The highest enzyme production was observed after 48 hours of cultivation at 31$^{\circ}C$. The optimum pH and temperature for the activity of crude enzyme were 6.0 and 5$0^{\circ}C$, respectively. The enzyme was stable between pH 5 and 9, and upto 5$0^{\circ}C$. The enzyme converted starch into $\alpha$-, $\beta$- and ${\gamma}$-CD in the relative amounts of 1:10:1.5, respectively.

Influence of pH on Leaching Behavior of Phosphorous from Steelmaking Slag (제강슬래그에서 인의 침출 거동에 대한 pH의 영향)

  • In this study, leaching process to extract phosphorus from the steelmaking slag was investigated for using the fertilizer resources of agriculture. In general, the phosphorus of steelmaking slag is formed as $C_2S-C_3P$ solid solution, and also, this solid solution is soluble in water more than the other phase in slag, and less than free CaO phase. In the present experiment, the influence of pH on the leaching behavior of various elements from the steelmaking slag was investigated by using multi-component steelmaking slag. When the pH was decreased, the concentration of Ca, Si, P and Fe in solution from the steelmaking slag was increased. Furthermore, at a pH of 3, the concentration of P ion in solution was decreased as leaching time increased. It is considered that the decrement of P was caused from the precipitation reaction between P ion and Fe ion in solution.

A Mechanistic Study on Reactions of Aryl Benzoates with Ethoxide, Aryloxides and Acetophenone oximates in Absolute Ethanol

  • Second-order rate constants have been measured spectrophotometrically for the reactions of aryl benzoates (X-C6H4CO2C6H4-Y) with EtO-, Z-C6H4O- and Z-C6H4C(Me)=NO- in absolute ethanol at 25.0 ℃. All the reactions have been performed in the presence of excess 18-crown-6 ether in order to eliminate the catalytic effect shown by alkali metal ion. A good Hammett correlation has been obtained with a large ρ- value (-1.96) when σ- (Z) constant was used for the reaction of p-nitrophenyl benzoate (PNPB) with Z-C6H4O-. Surprisingly, the one for the reaction of PNPB with Z-C6H4C(Me)=NO- gives a small but definitely positive ρ- value (+0.09). However, for reactions of C6H5CO2C6H4-Y with EtO-, correlation of log k with σ- (Y) constant gives very poor Hammett correlation. A significantly improved linearity has been obtained when σ0 (Y) constant was used, indicating that the leaving group departure is little advanced at the TS of the RDS. For reactions of X-C6H4CO2C6H4-4-NO2 with EtO-, C6H5O- and C6H5C(Me)=NO-, correlations of log k with σ (X) constants for all the three nucleophile systems give good linearity with large positive ρ values, e.g. 2.95, 2.81 and 3.06 for EtO-, C6H5O- and C6H5C(Me)=NO-, respectively. The large ρ values clearly suggest that the present reaction proceeds via a stepwise mechanism in which the formation of the addition intermediate is the RDS.

Each of the three ring system for the stars with B,A and C ring in order whilst A and C ring of econazole lie close to the same plane which is nearly 60˚with B ring. The hydrogen binding nitrogen of C ring and oxygen of nitrate contributes to stailization of econazole nitrate. Intr and intermolecular distances and angles are within the values recorded for simiar compounds.

Purification and Characterization of Cellulolytic Enzymes from Aspergillus niger (Aspergillus niger가 생산(生産)하는 섬유소(纖維素) 분해효소(分解酵素)의 정제(精製) 및 특성(特性))

  • Three fractions of carboxymethyl-cellulase (F-I, F-II, and F-III) and ${\beta}-glucosidase$ form Aspergillus niger were partially purified by ammonium sulfate fractionation. Sephadex G-150 and DEAE-Sephadex column chromatography. The optimum conditions such as pH and temperature and thermal inactivation properties of the enzymes were investigated. Arrhenius plots of F-II and F-III appeared as straight lines, whereas that of F-I was biphasic. The Z-values of F-II and F-III were $8^{\circ}C$ and $10^{\circ}C$ respectively, while that of F-I was $4^{\circ}C$ over $60{\sim}70^{\circ}C$ and $383^{\circ}C$ over $70{\sim}98^{\circ}C$. Three fractions and the crude extract of carboxymethyl-cellulase exhibited a similar optimum pH 4.3 and temperature of $60^{\circ}C$, while Z-value of crude extract $(21.5^{\circ}C)$ was much higher than that of the purified enzyme. Maximum activity of both purified and crude extract of ${\beta}-glucosidase$ was shown at pH 4.7 and $60^{\circ}C$, and z-value of the enzyme was $7^{\circ}C$.

The effect of sigma phases formation depending on Cr/Ni equivalent ratio in AISI 316L austenitic stainless steel weldments. (AISI 316L 용접부의 시그마상 형성에 영향을 미치는 크롬/니켈 당량비)

  • AISI 316L 용접금속의 크롬/니켈 당량비에 따른 시그마상의 영향을 알아보기 위하여 응고모드가 다른 3종류의 플럭스 코어드 와이어를 제작하였다. AISI 316L 시편에 FCAW 프로세스를 적용한 용접재를 $650^{\circ}C$, $750^{\circ}C$, $850^{\circ}C$, $950^{\circ}C$에서 각 각 1H, 5H, 24H, 72H동안 열처리하였다. 크롬/니켈 당량비가 높을수록 즉, 크롬의 함량이 높아질수록 $\delta$-페라이트 함량은 증가하였으며, $\delta$-페라이트는 고온에서 시그마상으로 변태되었다. $\delta$-페라이트는 $650^{\circ}C$에서 가장 느리게 분해되었으며 $850^{\circ}C$에서 가장 활발히 분해되었다. 용접부의 특성상 크롬과 니켈 등의 합금원소에 의하여 응고온도범위가 넓어져 $950^{\circ}C$에서도 시그마상이 석출되었으며, 5시간 이상 유지 시 구형으로 존재하였다. 충격시험 시 시그마상에 의해 취약해진 inter-dendrite 를 따라 파면이 형성되었으며, $-100^{\circ}C$이하의 극저온에서는 시그마상의 양과 무관하게 충격흡수에너지는 0에 가까워졌다. 하지만 3%미만의 $\delta$-페라이트를 함유하는 AF모드에서 발생한 DDC와 미량의 시그마상은 충격흡수에너지에 결정적인 영향을 미치지 않았다.

Kinetics of Goblet Cells and Mast Cells in the Intestine of C3H/HeN and BALB/c Mice Infected with Echinostoma hortense

  • Mast cells and goblet cells have the ability to protect against parasites by increasing mucus production that traps and excludes worms and prevents their intimate contact with the gut mucosa in the host. In this study, we investigated the function of mast cells and goblet cells for the rejection of Echinostoma hortense (E. hortense). In addition, we used both C3H/HeN and BALB/c mice in order to examine whether mast cells and goblet cells function differentially according to the strains of mice. After an oral infection with 30 E. hortense metacercariae, the number of mucosal mast cells and goblet cells, as well as worm recovery rate, were observed in experimentally infected mice between 1 week and 8 weeks post-infection (PI). Worm recovery rates in C3H/HeN and BALB/c mice were 65.7% and 23%, respectively, in week 1 P.I., indicating that worm expulsion in C3H/HeN mice was higher than in BALB/c mice. Our results demonstrate that the period (week 3 P.I.) in which worm recovery falls rapidly is the same period that the number of goblet cells and mast cells reaches a peak. These results indicate that worm recovery significantly correlates with the growth rate of goblet cells and mast cells (P=0.0482). However, worm expulsion is not associated with goblet cells or mast cells in BALB/c mice.

Characterization of Protease Produced by Bacillus amyloliquefaciens HTP-8 Isolated from Korean Fermented Anchovy Sauce. (멸치 어간장으로부터 분리한 Bacillus amyloliquefaciens HTP-8 이 생산하는 단백질 분해효소의 특성)

  • For commercial production of Korean fermented anchovy sauce through rapid fermentation, a bacterial strain which showed the high protease activity was isolated from a commercially fermented anchovy sauce. The isolate was Bacillus amyloliquefaciens, and named as B. amyloliquefaciens HTP-8. The incubation temperature, initial pH, and cultivation time for optimal production of protease by B. amyloliquefaciens HTP-8 were $30^{\circ}C$, 7.0, and 3 days, respectively. In jar fermenter, B. amyloliquefaciens HTP-8 showed higher protease activity when grown at pH 7.0. The protease was partially purified by 80% ammonium sulfate precipitation and CM-Sephadex C-50 ion exchange chromatography. The partially purified enzyme had specific activity of 103.3 units/mg, yield of 0.4%, and purification fold of 43.0. The optimal pH and temperature for the protease activity were 10.0 and $50^{\circ}C$, respectively. The protease was relatively stable at the pH range of 7.0~12.0 and at the temperatures below 4$0^{\circ}C$. The activity of the enzyme was inhibited by $Ag^{+}$ /, $Ba^{2+}$ and selectively inhibited by PMSF, suggesting that it is a serine protease.

The Properties of Proteolytic Enzymes in Fruits (Pear, Kiwifruit, Fig, Pineapple and Papaya) (배, 키위, 무화과, 파인애플, 파파야에 존재하는 단백질 분해효소의 특성 비교)

  • This study was attempted to compare the properties of proteolytic enzymes in fruits(Pear, Kiwifruit, Fig, Pineapple and Papaya) for the application of scientific information to cooking. The results were as follows: 1. The optimum temperature of crude proteolytic enzymes in pear, fig and pineapple is 60$^{\circ}C$ and it was relatively active in 40-70$^{\circ}C$; papaya showed max. activity in 60$^{\circ}C$ and highly stable activity in 40-80$^{\circ}C$, but kiwifruit showed max. activity in 40$^{\circ}C$ and it maintained to 70$^{\circ}C$. 2.. The crude proteolytic enzymes of pear, fig, pineapple and papaya showed opt. pH at pH 7.0 and maintained at pH 5.0-8.0, but max. activity of kiwifruit observed in pH 3.0 and pH 5.0-8.0. 3. As a result of comparison of total activities of fruits per kg unit, the order of activities was pineapple > kiwifruit > papaya > fig > pear.
