A Study on Quality Characteristics of pimpinella brachycarpa Kimchi during Storage at Different Temperatures (숙성 온도를 달리한 참나물 김치의 품질특성 조사)

  • This study was conducted to enhance the value of chamnamul (Pimpinella brachycarpa (Komarov) $N_{AKAI}$) as an useful food resource. Hunter L, a, b values (lightness, redness, yellowness) of chamnamul leaf were 33.28$\pm$1.94, -10.98$\pm$0.74, 14.05$\pm$1.29 and shearing force was 2745.2g. Contents of tannin and dietary fiber were 100.9 mg%, 24.0% (freeze drying base). The minerals identified in chamnamul were Ca 7.85 g/kg, K 76.31 g/kg, Mg 4.78g/kg, Fe 0.35g/kg, Na 2.35 g/kg. Chamnamul kimchi was packed in polyethylene film (200g) and fermented at 2$0^{\circ}C$ and 4$^{\circ}C$. In color changes kimchi fermented at 2$0^{\circ}C$ showed more increase in Hunter L, a, b values than kimchi fermented at 4$^{\circ}C$. The pH of kimchi decreased and acidity increased with storage time at both temperature. Ascorbic acid contents decreased sharply with storage time. Loss of ascorbic acid contents was about 81.9% in kimchi fermented at 2$0^{\circ}C$ after 5 days, and kimchi fermented 4$^{\circ}C$ lost 77.3% of ascorbic acid after 30 days. Also reducing sugar contents decreased with storage time at 2$0^{\circ}C$ and 4$^{\circ}C$. The results of sensory evaluation showed that optimum ripening time of chamnamul kimchi was 1~3 days at 2$0^{\circ}C$ and more than 20 days at 4$^{\circ}C$.>.

Nucleotide Insertion Fidelity of Human Hepatitis B Viral Polymerase

  • The hepadnaviruses replicate their nucleic acid through a reverse transcription step. The MBP-fused HBV polymerase was expressed in E. coli and purified by using amylase affinity column chromatography. The purified protein represented DNA-dependent DNA polymerase activity. In this report, the MBP-HBV polymerase was shown to lack 3'$\rightarrow$5' exonuclease activity, like other retroviral RTs. The ratio of the insertion efficiency for the wrong versus right vase pairs indicates the misinsertion frequency (f). The nucleotide insertion fidelity (1/f), observed with the MBP-HBV polymerase and HIV-1 RT, was between 60 and 54,000, and between 50 and 73,000, respectively, showing that they are in close range. A relatively efficient nucleotide incorporation by the MBP-HBV polymerase was observed with a specificity of three groups: (1) A : T, T : A>C : G, G : C (matched pairs), (2) A : C, C : A>G: T, T : G (purine-pyrimidine and pyrimidine-purine mispairs), and (3) C : C, A : A, G : G, T : T>T : C, C : T>A : G, G : A (purine-purine or pyrimidine-pyrimidine mispairs), and their order is (1)>(2)>(3). The data from the nucleotide insertion fidelity by the MBP-HBV polymerase suggest that the HBV polymerase may be as error-prone as HIV-1 RT.

Food Composition of Crab(Charybdis japonica) Preserved in Brine (민꽃게(Charybdis japonica)장의 식품 성분)

  • Pickled crabs made using Charybdis japonica are becoming part of our dietary habits. Yet research on this food is disappointingly scarce. It is generally accepted that the original food composition of pickled crabs defies scrutiny because various seasonings are added to soy for the production of pickled crabs in most regions. Accordingly, this study attempted to measure the food composition of the Charybdis japonica preserved in brine according to different storage days (10, 15, 20, 25, and 30 days) and different temperatures ($5{^{\circ}C}$, $15{^{\circ}C}$, and ambient temperature). It was found that Charybdis japonica is comprised of moisture (77.5%), ash (2.3%), crude protein (19.7%), and crude lipid (0.5%). The content of crude protein and crude lipid during the storage period tended to decrease over time.

Determination of C3G Content in Blackish Purple Rice using HPLC and UV-Vis Spectrophotometer

  • Cyanidin 3-glucoside (C3G) content contained in the grains of blackish purple rice varieties, Heugjinjubyeo, Kilimheugmi, Heugnambyeo, Sanghaehy-anghyeolla, and the progenies derived from their crosses was evaluated by HPLC and UV-Vis spectroscopy. C3G content was higher in the range of 10-30% by using UV-Vis method compared to HPLC method. A significant linear relationship was, however, observed between two analytical methods. The correlation coefficient was 0.98. Thus, this results suggested that it would be able to use UV-Vis spectroscopy to determine C3G content which does not demanded precise value like selection.

Development of Indole-3-Acetic Acid-Producing Escherichia coli by Functional Expression of IpdC, AspC, and Iad1

  • Biosynthesis of indole-3-acetic acid (IAA) via the indole-3-pyruvic acid pathway involves three kinds of enzymes; aminotransferase encoded by aspC, indole-3-pyruvic acid decarboxylase encoded by ipdC, and indole-3-acetic acid dehydrogenase encoded by iad1. The ipdC from Enterobacter cloacae ATCC 13047, aspC from Escherichia coli, and iad1 from Ustilago maydis were cloned and expressed under the control of the tac and sod promoters in E. coli. According to SDS-PAGE and enzyme activity, IpdC and Iad1 showed good expression under the control of $P_{tac}$, whereas AspC was efficiently expressed by $P_{sod}$ originating from Corynebacterium glutamicum. The activities of IpdC, AspC, and Iad1 from the crude extracts of recombinant E. coli Top 10 were 215.6, 5.7, and 272.1 nmol/min/mg-protein, respectively. The recombinant E. coli $DH5{\alpha}$ expressing IpdC, AspC, and Iad1 produced about 1.1 g/l of IAA and 0.13 g/l of tryptophol (TOL) after 48 h of cultivation in LB medium with 2 g/l tryptophan. To improve IAA production, a tnaA gene mediating indole formation from tryptophan was deleted. As a result, E. coli IAA68 with expression of the three genes produced 1.8 g/l of IAA, which is a 1.6-fold increase compared with wild-type $DH5{\alpha}$ harboring the same plasmids. Moreover, the complete conversion of tryptophan to IAA was achieved by E. coli IAA68. Finally, E. coli IAA68 produced 3.0 g/l of IAA after 24 h cultivation in LB medium supplemented with 4 g/l of tryptophan.

  • The followings are the results obtained from the investigation of isolation and cultural conditions of a fungus producing antibiotic from humus, spring soil, paddy, field and etc. 1. Among fungi which were capable of antimicrobial sub stance producing at the $45^{\circ}C$, Strain H-3 was selected as test st rain because of its efficiency in antimicrobial substance producing. 2. Strain H-3 was identified as a Aspergullus fumigatus by the mophological properties. 3. The compositions of optimun media for the production of antimicrobial substance were followed;glucose 20g, $NaNO_3$ 3g, $K_2HPO_4$ 1g, $CaCl_2$ 0.5g, $MgSo_4$ 0.5g, $FeSO_4$ 0.005g. 4. Optimum culture conditions were founded to be pH 5.0, $40^{\circ}C$ respectively. 5. The maximum antimicrobial substance producing obtained in this study was showed 14mm of clear zone after 72 hrs incubation under the optimum conditions. 6. AF-c substance was active against Gram-positive, Gram-negative bacteria, and yeast but not mold in its biological properties.

Yookmijihwang-Tang on the Plasma Corticosterone level in Mice exposed to Heat, Cold and Immobilization Stress (한(寒), 열(熱), 구속(拘束) 스트레스에 대한 사군자탕(四君子湯), 사물탕(四物湯), 육미지황탕(六味地黃湯)의 효능(效能))

  • In order to investigate the effect of Sagoonja-Tang, Samool-Tang and Yookmijihwang-Tang against the cold, heat and immobilization stress, each formula was injected intragastically to mice exposed to forced cold water swimming($4^{\circ}C$), forced hot water swimming($42^{\circ}C$) and immobilization stress before measuring the change of plasma corticosterone level of mice. The results were as follows: 1. One hour after each Sagoonja-Tang, Samool-Tang, Yookmijihwang-Tang with the dose of 1g/kg, 3g/kg each were administered to mice, we put cold stress($4^{\circ}C$) on mice for 3min. Then those prescriptions didn't show any significant effect on plasma corticosterone level. however, Yookmijihwang-Tang(1g/kg) showed a significant increase(p〈0.05), and Samool-Tang(3g/kg) showed a small increase. 2. One hour after Samool-Tang, Sagoonja-Tang and Yookmijihwang-Tang with the dose of 1g/kg, 3g/kg each were administerd, mice exposed to heat stress($42^{\circ}C$) for 3 min. Samool-Tang(1g/kg) and Yookmijihwang-Tang(lg/kg) showed significant decrease of corticosterone level(p〈0.01, p〈0.05) but Sagoonja-Tang didn't change it. In the case of the dose of 3g/kg, only Samool-Tang decreased corticosterone level(P〈0.01), but Sagoonja-Tang, Yookmijihwang-Tang didn't make significant effect on plasma corticosterone level of the mice exposed to heat stress($42^{\circ}C$ ). 3. Those mice which exposed to immobilization stress one hour after Sagoonja-Tang, Samool-Tang, Yookmijihwang-Tang with the dose of 1g/kg and 3g/kg didn't show effective decrease of corticosterone level. However, Sagoonja-Tang(3g/kg), Samool-Tang(lg/kg, 3g/kg) showed the effect of mild increase of corticosterone level rather than decrease. These data revealed that the effect-variation of prescriptions depended on the kinds of stress and the dose of prescriptions had a different effect on therapy.

Quality characteristics of semi-dried restructured jerky: combined effects of duck skin gelatin and carrageenan

  • The present study investigated the effects of duck skin gelatin and carrageenan on the quality of semi-dried restructured jerky. Restructured jerky was prepared as follows: G0 (control, without duck skin gelatin and carrageenan), G0C (0.3% carrageenan), G0.5 (0.5% duck skin gelatin), G0.5C (0.5% duck skin gelatin and 0.3% carrageenan), G1 (1.0% duck skin gelatin), and G1C (1.0% duck skin gelatin and 0.3% carrageenan). The moisture content was the highest for the semi-dried restructured jerky from G0.5C and G1C groups, which showed the lowest for shear force value (p < 0.05). The processing yield of semi-dried restructured jerky with carrageenan was higher compared to that of the control group (p < 0.05). The rehydration capacities of G0.5, G0.5C, and G1C groups were significantly higher than the rehydration capacity of the control group (p < 0.05). Water activity, lightness, yellowness, flavor score, texture score, and overall acceptability were the highest (p < 0.05) for the semi-dried restructured jerky from the G1C group. No significant (p > 0.05) difference was observed in appearance score among restructured jerky prepared from duck skin gelatin and carrageenan. Thus, the addition of 1.0% duck skin gelatin and 0.3% carrageenan to semi-dried restructured jerky formulations results in the optimization of quality characteristics.

  • After oral administration of $^{14}$ C-labelled $N^{G}$-mono[$^{14}$ C-methyl]-L-arginine into rats, 66.3% of the administered radioactivity has been recovered in the urine, and 86.2% of the total of the recovered radioactivity is recovered in the first 24-hr urine. Distributions of the radioactivity of the acidic, basic, and neutral portions and unmetabolized $N^{G}$-monomethyl-L-arginine are 33.3%, 40.2%, 12.5% , and 0.3%, respectively. The radioactivity corresponding N-monomethylurea is about 50% of the neutral portion and 6% of the administered radioactivity.ity.

Primary Production in the Osyter Farming Bay (굴 양식장 수역의 기초생산 연구)

  • The primary production of phytoplankton in oyster farming bays were seasonally measured by $^{14}C$ method in Hansan-Koje Bay and Kamagyang Bay for one year period. In Hansan-Koje Bay, mean daily prima production was $0.52gC/m^2/day\;(0.22{\~}1.02gC/m^2/day)$, and it was relatively high in summer and low in winter. Annual primary production was estimated to be $189gC/m^2/yr.$. Mean chlorophyll a content was $1.70mg/m^3$, and it was relatively high in summer. In Kamagyang Bay, mean daily prima production was $0.91gC/m^2/day(0.11{\~}3.61gC/m^2/day)$. and it was maximum in summer and minimum in fall. Annual primary production was estimated to be $334gC/m^2/yr$. Mean chlorophyll a concentration was $2.34mg/m^3$, and it was maximum in summer and relatively high in winter. Considering the water area of the two osyter farming bays, annual carbon production by phytoplankton were 9,450 ton in Hansan-Koje Bay and 37,000 ton in Kamagyang Bay. In general pattern of primary production in these bays, high production occurs in summer with increasing trend since spring.
