Synthetic Curcumin Derivatives Inhibit Jun-Fos-DNA Complex Formation

  • Jun/Fos, a crucial factor in transmitting the tumor-promoting signal from the extracellular environment to the nuclear transcription machinery, has a dimerization interface possessing several coiled structural properties. Jun and Fos can interact with the DNA regulatory region, AP-1 (Activator Protein-1), which is composed of 5'-TGAC/GTCA-3'.$^1$ Curcumin is a well-known anticancer and anti-inflammatory compound.$^{2,3}$ It also acts as an inhibitor of the Jun-Fos function. c-Fos and c-Jun with a bZIP region are overexpressed in BL21 E. coli and purified with an $Ni^{2+}$ affinity column. The inhibitors of Fos-Jun-AP-1 complex formation were searched through the EMSA (electrophoresis mobility shift assay) experiment, and new curcuminoids were synthesized and investigated as to their inhibitory effect on the same system. Two curcuminoids showed a stronger inhibitory effect than curcumin. This inhibitory activity was quantified with EMSA. 1,7-bis(4-methyl)-1,6-heptadiene-3,5-dione (BJC003) and 1,7-bis(4-hydroxy-5-methoxy-3-nitrophenyl)-1,6-heptadiene-3,5-dione (BJC005) showed remarkably high inhibitory activities. $IC_{50}$ of 1,7-bis(4-methyl)-1,6-heptadiene-3,5-dione (BJC003) and 1,7-bis(4-hydroxy-5-methoxy-3-nitrophenyl)-1,6-heptadiene-3,5-dione (BJC005) are 8.98 ${\mu}M$ and 5.40 ${\mu}M$, respectively. However, 1,7-bis(4-methyl-3-nitrophenyl)-1,6-heptadiene-3,5-dione (BJC004) did not show inhibitory activity.

Antimicrobial Activity and Components of Extracts from Agastache rugosa during Growth Period

  • Antimicrobial activities of volatile flavor, water and methanol extracts from Agastache rugosa were investigated. The volatile flavor extract was obtained from A. rugosa by simulataneous distillation-extraction (SDE) method. Antimicrobial activity was investigated by disc diffusion method against several microorganisms, four species of Gram positive, three species of Gram negative and tow species of yeast. The volatile flavor extracts had strong antimicrobial activity againstc. utilisand S. cerevisiae. During the growth period, a difference in antimicrobial activity among volatile flavor extracts from A. rugosa was not shown. The water extract of above 10 mg/disc showed antimicrobial activity. Methanol extracts from A. rugosa harvested in June showed antimicrobial activity against tested Gram positive and Gram negative bacteria, showed weak antimicrobial activity against the bacteria from those harvested in July and August. In particular, antimicrobial activity against V. parahaemolyticus was stronger than that against other bacteria. Water and methanol extracts did not inhibit yeast. C. utilis and S. cerevisiae. To further elucidate the effective components, volatile flavor extracts was analyzed by GC/MS. harvested in June, the components included 8 phenols (93.031%), 18 terpenes (5.230%), 12 alcohols (1.300%) 8 alkanes (0.181%), 1 ester (0.056%), 2 ketones (0.033%), 2 aldehydes (0.011%) and 1 pyrrole (0.007%). In July, the components included 6 phenols (94.366%), 19 terpenes (3.394%), 11 alcohols (2.045%), 1 ester (0.039%), 2 ketones (0.028%), 1 furan (0.005%) and 1 aldehyde (0.005%). And in August, the components included 7 phenols (95.270%), 19 terpenes (2.951%), 13 alcohols (1.399%), 1 ester (0.063%), 2 aldehydes (0.016%), 2 ketones (0.011%), 1 alkane (0.006%), 1 acid (0.005%) and 1 pyrrole (0.005%). The major component of volatile flavors was estragole, a phenolic compound.

  • In this paper, the $0.995(Li_{0.04}(Na_{0.56}K_{0.44})_{0.96}(Nb_{0.90}Ta_{0.10})_{0.998}Zn_{0.005}O_3+0.005KNbO_3+xwt%\;TeO_2$ lead-free piezoelectric ceramics for energy harvesting devices were fabricated by the conventional mixed oxide method. The microstructure, dielectric, and piezoelectric properties were investigated as a function of the $TeO_2$ addition. All the specimens showed an orthorhombic phase structure. At the composition ceramics doped with 0.1 wt%$TeO_2$, the optimum values of $d_{33}$= 212 pC/N, $d_{33}{\cdot}g_{33}=9.54pm^2/N$, and kp=0.448 were obtained, respectively. The results indicate that the composition ceramics is a promising candidate for energy harvesting devices applications.

리놀레산에 의한 인체암세포의 성장 억제효과 및 암세포막 인지질 지방산 변화 (Growth Inhibitory Effect and Changes in Membrane Phospholipid Fatty Acid Composition on MG-63 and AZ-521 Human Cancer Cells by Linoleic Acid)

  • MG-63 골육암세포 및 AZ-521 인체 위 암세포 성장에 미치는 리놀레산(LA)의 성장 억제효과와 이때 암세포막의 인지질 유리지방산 조성 변화에 대한 연구를 하였다. LA 첨가 농도 0.005%에서 MG-63 암세포와 AZ-521 암세포는 대조군에 비해 각각 54% 및 52%의 암세포 성장이 억제되었다. 또한 LA 첨가(0.01%)시 이들 암세포의 [$^3$H] thymindine 결합능은 90% 이상이 억제되어 암세포의 DNA합성이 크게 억제되었다. 한편 LA(0.005 %) 첨가시 각 암세포의 형태학적인 변형을 유발시켰으며 세포막의 인지질 지방산 조성의 변화가 관찰되었다. LA처리는 이런 세포의 형태적 변형과 함께 세포막의 인지질 중 16:0과 18:0의 지방산은 현저히 감소되었으나 탄소수가 20 이상의 탄소수를 가지거나 불포화 도가 높은 지방산(20 : 4, 22 6, 24 : 4)은 증가되었다.

  • 연직 공기 기둥에 대한 복사 및 대류 과정이 포함된 일차원 복사-대류 평형 모형을 구축하였고, 이 모형을 이용하여 남극 세종기지에서 복사-대류 평형 온도를 계산 하고 분석하였다. 이 모형의 반응도에서 지표면 알베도와 태양 천정각의 코사인 및 이산화탄소 증가는 지표면에서의 복사-대류 평형 온도를 감소시켰다. 그리고 구름 광학두께가 비교적 큰 하층운은 지표면 온도를 감소시키나, 구름 광학두께가 작은 상층운은 온실효과 때문에 지표면 온도를 증가시켰다. 남극 세종기지의 44년 (1958∼2001)의 기간에 대하여 계산된 지표면에서의 복사-대류 평형 온도의 연변화는 0.012oC/년이었다. 마찬가지로 13년 동안(1989∼2001)의 자료에 대한 복사-평형 온도 변화는 0.01oC/월이었으며, 동일한 기간의 관측 자료 분석 결과로는 0.005oC/월의 변화를 나타내었다.

  • 옥살산과 아세트산 용액의 농도변화에 따른 우라늄과 바나듐의 음이온교환수지로부터의 용리거동 및, UV및 스펙트럼 변화로부터 이들 이온의 화학종과 평형관계를 연구하였다. 0.005~0.5M 옥산살산 용액속에서 우라늄과 바나듐은 각각 $UO_2(C_2O_4)_2^{2-}$, $UO_2(C_2O-4)_3^{4-}$$VO_2(C_2O_4)_2^{3-}$의 화학종으로 존재하고, 0.01~0.1M의 아세트산 용액에서 우라늄은 $VO_2(Ac)_2^0$, $UO_2(Ac)_3^{1-}$의 화학종으로 존재하며, 바나듐은 $VO_2^++2Ac^-=VO_2(Ac)_2^-$의 평형이 이루어짐을 확인하였다.

  • ZnO nano film for transparent thin film transistors is prepared by injection type source delivery system of atomic layer deposition. By using this delivery system the source delivery pulse time can dramatically be reduced to 0.005s in ALD system. ZnO nanofilms obtained at $150^{\circ}C$ are characterized.

도형의 변환학습의 순차성 고찰 (A Study of the Sequence of Figure Transformation Learning)

  • This study aimed at studying the sequence of the Figure Transformation Learning, inquiring relationship among these transformations and then researching whether there is the difference of the learning ability or not between by teaching them as it is independent and by teaching them as it is contains. (Hypothesis 1) It may be more effective to teach The Sequence of Transformation Learning by beginning with peculiar field, ending with general field than vice versa At the result of verification-C $R_{M}$=2.59, 0.005$R_{M}$=5.19, p<0.005-significant difference appeared. It is proved more effective to teach the Figure Transformation Learning the way it contains than the way it is independent. Synthesizing two hypothesises of the above, the conclusion is following The Figure Transformation Learning should be taught by beginning with peculiar field. ending with general field (congruent transformationlongrightarrowsimilar transformationlongrightarrowprojective transformationlongrightarrowtopological transformation). To teach it the way it contains is more effective.ive.

졸겔 $WO_3$박막의 특성 (Properties of Sol-gel $WO_3$ thin films)

  • $WO_3$ 박막은 졸겔방법으로 텅스텐 알코산화물 용액을 사용하여 마이크로 슬라이드유리와 ITO 유리기판위에 여러번 침적도포시켜 다층박막형으로 제작하였다. 침적도포와 시료제조 공정인자가 박막의 구조, 광학적 및 전기화학적 특성에 미치는 영향이 연구되었다. 텅스텐 알코산화물 용액을 사용하여 증착속도를 0.005m $s^{-1}$로 하여 제작된 박막은 매우 균일하였으나 증착속도를 0.007m $s^{-1}$ 보다 크게 하여 제작된 시료의 두께는 상당한 변화를 보였다. 전기화학적 착색 실험결과 박막은 $200^}\circ}C$보다 낮은 온도에서 열처리된 시료는 쉽게 착색이 되었다. 라더포드 후방산란법에 의해 착색된 시료에서의 $K^+$ 이온들은 막 속에 균일하게 분포되어 있었다.

  • Naetakcheonkeumsan(NCS) was a drug that treated carbuncle and cellulitis. So, the purpose of this Study was to investigate effect of NCS on the anti-cancer and proliferation of lymphocytes in normal mouse group, L1210 cells-transplanted mouse group and anti-cancer drug (vincristine) 0.005mg/kg were injected mouse(Ll210 cells-transplanted) group. We used NCS extract with freeze-dried, 8wks-old male mice, and Ll210 cell lines for this Study, The proliferation of cells was tested using a colorimetric tetrazoliun assay(MTT assay). The results of this Study were obtained as follow ; Group C(NCS plus Rehmanniae Radix Preparat administered group) inhibited proliferaion of lymphocytes in normal mouse group and Ll210 cells transplanted mouse group. Group A(NCS administered group) and Group B(NCS plus Cervi pantotrichum Cornu administered group) inhibited proliferation of Ll210 cells in Ll210 cells-transplanted mouse group and anti-cancer drug were injected mouse(Ll210 cells-transplanted) group. Group C incresed proliferation of L1210 cells in L1210 cells-transplanted mouse group, but inhibited in anti-cancer drug(vincristine) 0.005mg/kg were injected mouse(L1210 cells-transplanted) group.
