Increase of L-type Calcium Current by cGMP-dependent Protein Kinase Regulates in Rabbit Ventricular Myocytes

  • Background: We have previously reported that not only cGMP but also 8-Br-cGMP or 8-pCPT-cGMP, specific and potent stimulators of cGMP-dependent protein kinase (cGMP-PK), increased basal L-type calcium current $(I_{Ca})$ in rabbit ventricular myocytes. Our findings in rabbit ventricular myocytes were entirely different from the earlier findings in different species, suggesting that the activation of cGMP-PK is involved in the facilitation of $I_{Ca}}$ by cGMP. However, there is no direct evidence that cGMP-PK can stimulate $I_{Ca}}$ in rabbit ventricular myocytes. In this report, we focused on the direct effect of cGMP-PK on $I_{Ca}}$ in rabbit ventricular myocytes. Methods and Results: We isolated single ventricular myocytes of rabbit hearts by using enzymatic dissociation. Regulation of $I_{Ca}}$ by cGMP-PK was investigated in rabbit ventricular myocytes using whole-cell voltage clamp method. $I_{Ca}}$ was elicited by a depolarizing pulse to +10 mV from a holding potential of -40 mV. Extracellular 8-(4-Chlorophenylthio)-guanosine-3',5'-cyclic monophosphate (8-pCPT-cGMP), potent stimulator of cGMP-dependent protein kinase (cGMP-PK), increased basal $I_{Ca}}$. cGMP-PK also increased basal $I_{Ca}}$. The stimulation of basal $I_{Ca}}$ by cGMP-PK required both 8-Br-cGMP in low concentration and intracellular ATP to be present. The stimulation of basal $I_{Ca}}$ by cGMP-PK was blocked by heat inactivation of the cGMP-PK and by bath application of 8-(4-chlorophenylthio)-guanosine-3',5'-cyclic monophosphate, Rp-isomer (Rp-pCPT-cGMP), a phosphodiesterase-resistant cGMP-PK inhibitor. When $I_{Ca}}$ was increased by internal application of cGMP-PK, IBMX resulted in an additional stimulation of $I_{Ca}}$. In the presence of cGMP-PK, already increased $I_{Ca}}$ was potentiated by bath application of isoprenaline or forskolin or intracellular application of cAMP. Conclusions: We present evidence that cGMP-PK stimulated basal $I_{Ca}}$ by a direct phosphorylation of L-type calcium channel or associated regulatory protein in rabbit ventricular myocytes.

비정질 p-SiC/i-SiC/i-Si/n-Si 박막 태양전지에서 i-SiC 완충층의 역할 (Roles of i-SiC Buffer Layer in Amorphous p-SiC/i-SiC/i-Si/n-Si Thin Film Solar Cells)

  • 플라즈마 화학증착 (PECVD) 장비를 이용하여 $SnO_2$ 투명전도막이 피막된 유리기판 위에 p-SiC/i-Si/n-Si 이종접합 태양전지를 제작하였다. p-SiC 층의 증착중에 기체조성 x=$CH_4/\;(SiH_4+CH_4)$의 변화에 대한 태양전지의 광기전 특성을 관찰하였다. 기체조성(x)이 0~0.4의 범위에서 p-SiC 창층의 광학적 밴드갭의 증가로 인하여 태양전지의 효율은 증가하였으나, 그 이상의 기체조성에서는 p-SiC/i-Si 계면에서의 조성불일치가 증가하여 태양전지의 효율이 감소하였다. 이러한 계면문제는 p-SiC 층과 i-Si 계면에서의 조성불일치가 증가하여 태양전지의 효율이 감소하였다. 이러한 계면문제는 p-SiC 층과 I-Si 층 사이에 I-SiC 완충층을 삽입함으로써 크게 감소하였다. 그 결과 유효면적이 $1cm^2$인 glass/$SnO_2$/p-SiC/i-SiC/i-Si/n-Si/Ag 구조의 박막 태양전지는 100mW/$cm^2$ 조도 하에서 8.6%의 효율을 나타내었다. ($V_{oc}$=0.85V, $J_{sc}$=16.42mA/$cm^2$, FF=0.615)

합성 윤활유 제조를 위한 퍼옥사이드계 개시제를 이용한 이소파라핀의 단독 알킬레이션 (Single-molecule Alkylation of Isoparaffin Using Peroxide Initiator for Making Synthetic Lubricant)

  • 폴리알파올레핀은 낮은 유동점, 높은 점도지수와 같은 물리화학적 특성이 기존의 광유보다 우수한 합성윤활유이다. 본 논문은 퍼옥사이드계 개시제를 이용하여 올레핀을 사용하지 않고 이소파라핀 분자간의 알킬레이션 반응을 통한 합성 윤활유 제조에 관한 새로운 경로를 제시한다. 평균 탄소수가 i-$C_{16}$, i-$C_{18}$, 그리고 i-$C_{23}$인 세가지 장쇄 이소파라핀의 알킬레이션과 i-$C_{18}$과 2-pentene의 알킬레이션을 di-tert butyl peroxide 라디칼 개시제를 사용하여 여러가지 반응조건에서 수행하여 단독 알킬레이션 반응 경로를 확인하였다. 세가지 유분으로 제조된 알킬레이트를 합성 윤활유로의 적용 가능성을 알아보기 위하여 동점도, 점도지수, 및 유동점을 조사하였다. i-$C_{16}$ 및 i-$C_{18}$ 유분으로 제조된 단독 알킬레이트는 극초고점도지수(>140)와 낮은 유동점(< $-30^{\circ}C$)을 보였다.

외상 환자에서의 cardiac troponin I 상승군에서의 기초 조사 (Primary Survey of Cardiac Troponin I Elevated Groups in Trauma Patients)

  • Background: Cardiac troponin I (cTnI) is a sensitive cardiac marker of myocardial injury. In normal coronary angiogram, positive cTnI values may be detected in various events such as sepsis, stroke, trauma and so on. To investigate characteristics of cTnI positive group in trauma patients, we designed this study between cTnI positive group and cTnI negative group. Method: Trauma patients who visited emergency room within 24 hours after accidents were included. Patients who had renal failure, acute coronary syndrome, sepsis, spontaneous SAH were excluded. Retrospective study of 97 trauma patients was done. We investgated ISS (injury severity score), positive cTnI, EKG abnormality, shock class, ICU admission rate and mortality. Result: In comparing with non chest trauma group, chest trauma group, whose chest AIS (Abbreviated Injury Score) is more than 3 point, had significant values in ISS, positive cTnI, EKG abnormality, shock class and ICU admission rate. Also, in non chest trauma group, we found several patients whose cTnI level was positive. When non chest trauma group was divided into two subgroups, the mortality and shock class of positive cTnI group were higher than that of negative cTnI group. When all trauma patients were divided into two groups, a positive cTnI group had higher values in ISS, shock class, ICU admission rate and mortality than that in a negative cTnI group. Conclusion: We found that cTnI were positive in patients of cardiac contusion but also in various trauma cases. In non chest trauma patients, we assumed that hypotension caused cTnI elevating. The cTnI could play a role in predicting prognosis in trauma patients.

변압기 지역 코로나 전자파 간섭에 의한 계측제어설비 영향에 관한 연구 (A Study on the Effects of I&C Systems by EMI Generating from Corona Discharge at Transformer Area)

  • The Electromagnetic Interference(EMI) generating from corona discharge of transformer area can interference the digital Instrument and Control(I&C) systems located nearby transformers. When the potential gradient of the electric field around the conductor is high enough to form a conductive region but not high enough to cause electrical breakdown to nearby objects, the EMI of corona discharge emits with the conducted and radiated noise and it interferences the signals of the I&C systems. Since digital I&C systems have an efficiency and competitive price, the analog I&C systems have been upgraded and displaced with the digital I&C systems but which have less EMI Immunity. There was no assessment to I&C systems by EMI generating corona discharge nearby transformers. When the safety-related I&C systems are installed in plants, the verification of equipment EMI should be done not in site-specific test but in test facilities. There are the need to do the site-specific EMI evaluation of corona discharge nearby transformers. This paper assesses the margin between plant emission limits and the highest composite plant emission of corona. When the non safety-related I&C systems are placed in transformer area, it suggests the appropriate radiated susceptibility level to EMI of corona discharge.

동방결절에서 과분극에 의해 활성화되는 내향전류에 대한 Cyclic-GMP의 영향 (Effects of Cyclic-GMP on Hyperpolarization-activated inward Current $(I_f)$ in Sino-atrial Node Cells of Rabbit)

  • The aim of present study is to investigate the effects of cGMP on hyperpolarization activated inward current ($I_f$), pacemaker current of the heart, in rabbit sino-atrial node cells using the whole-cell patch clamp technique. When sodium nitroprusside (SNP, $80{\mu}M$), which is known to activate guanylyl cyclase, was added, $I_f$ amplitude was increased and its activation was accelerated. However, when $I_f$ was prestimulated by isopreterenol (ISO, $1{\mu}M$), SNP reversed the effect of ISO. In the absence of ISO, SNP shifted activation curve rightward. On the contrary in the presence of ISO, SNP shifted activation curve in opposite direction. $8Br-cGMP(100\;{\mu}M)$, more potent PKG activator and worse PDE activator than cGMP, also increased basal $I_f$ but did not reverse stimulatory effect of ISO. It was probable that PKG activation seemed to be involved in SNP-induced basal $I_f$ increase. The fact that SNP inhibited ISO-stimulated $I_f$ suggested cGMP antagonize cAMP action via the activation of PDE. This possibility was supported by experiment using 3-isobutyl-1-methylxanthine (IBMX), non-specific PDE inhibitor. SNP did not affect $I_f$ when $I_f$ was stimulated by $20{\mu}M$ IBMX. Therefore, cGMP reversed the stimulatory effect of cAMP via cAMP breakdown by activating cGMP-stimulated PDE. These results suggest that PKG and PDE are involved in the modulation of $I_f$ by cGMP: PKG may facilitate $I_f$ and cGMP-stimulated PDE can counteract the stimulatory action of cAMP.

  • We investigate the relationships between the space X and the hyperspaces concerning admissibility and connectedness im kleinen. The following results are obtained: Let X be a Hausdorff continuum, and let A, $B{\in}C(X)$ with $A{\subset}B$. (1) If X is c.i.k. at A, then X is c.i.k. at B if and only if B is admissible.   • By analyzing the voice communication performance of the existing tactical FM radios, the performance target of the newly developing TICN combat network radio system VHF band tactical voice communication waveform was derived. In addition, a vocoder and modulation method that can satisfy the performance target and additional requirements are presented, and the expected voice communication quality is analyzed.

  • 본 논문에서는 실내 음영지역에서 사용되는 모바일 와이맥스의 펨토셀 기지국(WFAP; WiMAX Femtocell AP)에 대한 성능을 분석하였다. 시뮬레이션을 통해 와이맥스 펨토셀 기지국의 패킷에러율(PER), Outage, throughput을 확인하였다.

참당귀에서 분리한 다당의 면역활성에 대한 당쇄의 역할 (Roles of sugar chains in immunostimulatory activity of the polysaccharide isolated from Angelica gigas)

  • 참당귀 유래 면역활성 다당의 활성과 구조의 상관관계를 규명하고자 조다당 AGE-0로부터 두 차례의 연속적인 컬럼 크로마토그래피를 실행하여 단일 정제 다당 AGE-2c-I을 얻었다. AGE-2c-I은 농도의존적으로 우수한 항보체 활성을 보여주었으며, 일반화학적 특성을 분석한 결과, 분자량 약 140 kDa의 고분자 다당으로 4종의 구성당과 13종의 당쇄 결합양식으로 구성되어 있음을 확인할 수 있었다. 이 다당은 ${\beta}$-glucosyl Yariv reagent와의 높은 반응성을 보임으로써 arabino-3,6-galactan의 구조를 가진 rhamnogalacturonan-I 구조임을 추정할 수 있었다. 한편, AGE-2c-I의 미세구조의 해명과 항보체 활성에 관여하는 다당 중의 활성부위 검토를 위해 ${\alpha}$-L-arabinofuranosidase와 endo-1,4-${\beta}$-galactanase를 이용한 연속적 가수분해를 행하고 얻은 단편획분들을 이용, 구성당 및 당쇄결합 양식 분석, ${\beta}$-glucosyl Yariv reagent와의 반응성 검토 및 항보체 활성 결과를 분석한 결과, 참당귀 유래 항보체활성 다당 AGE-2c-I은 rhamnogalacturonan-I과 유사한 구조를 소유하고 있음이 확인되었으며, AGE-2c-I의 측쇄 구조인 arabino-${\beta}$-3,6-galactan 사슬이 항보체 활성 발현에 주요 역할을 수행하며, 5-linked Araf와 3,5-branched Araf로 구성된 ${\alpha}$-arabinan 측쇄가 활성에 부분적으로 관여하고 있음을 최종 확인할 수 있었다.
