2008년 남해동부해역의 Cochlodinium polykrikoides 적조발생 특성 (Characteristics of Cochlodinium polykrikoides Bloom in Southeast Coastal Waters of Korea, 2008)

  • 2008년 한반도 남해동부해역에서 Cochlodinium polykrikoides 적조의 발생, 진행 및 소멸현상을 식물플랑크톤 종조성, 물리 화학적 환경요소와 기상자료를 이용하여 분석하였다. 2008년 C. polykrikoides 적조는 7월말에 조기 발생하여 저밀도로 장기간 연안해역에 집중되어 발생하였다. 7월 초부터 동해남부해역에서 발생한 냉수대의 영향으로 외해로부터 C. polykrikoides 가입이 어려웠으며, 연안 저층의 포자(cyst) 발아에 의한 적조발생 이후 약한 바람 및 가뭄으로 인한 낮은 영양염 농도로 적조가 확산 성장하지 못하고 연안해역에 제한되어 발생한 것으로 판단된다.

2000년 여름 남해안에 나타난 Cochlodinium polykrikoides 우점 적조의 발생 특성 (The Outbreak, Maintenance, and Decline of the Red Tide Dominated by Cochlodinium polykrikoides in the Coastal Waters off Southern Korea from August to October, 2000)

  • 2000년 한반도 남해안에서 발생한 Cochlodinium polykrikoides 적조 초기발생, 진행과 소멸현상을 현장과 NOAA 위성 표층수온 자료를 이용하여 검토하였다. 일반적으로 C. polykrikoides 적조는 1995년에서 1999년에는 적조 띠가 고흥 나로도해역에서 최초 발생되었으나, 2000년에는 여수해만 해역에서 최초로 발생되었다. 이는 한반도 남해해역에서 여러 해수가 영향을 미쳤기 때문으로 생각된다 : 1) 대마난류와 남해연안류, 2) 진도냉수대와 남서해 연안수 및 3) 남동연안의 저온수와 외양난류수, 이 기간동안 실시한 현장조사에서는 C. polykrikoides의 유영세포가 이들의 경계역에 집적되는 경향을 나타내었는데, 특히 8월말에 대마난류와 남해연안류 사이의 남해 해양전선이 여수해만에서 형성되어 2000년 C. polykrikoides 적조가 바로 이 지점에서 시작하는 것을 관찰할 수 있었다.(8월 22일). 이 C. polykrikoides 우점 적조는 Alexandrum tamarense, Gymnodinium mikimotoi, Skletonema coastatum, 그리고 Chaetoceros spp.와 혼합하여 발생하였다. 적조발생 기간 중에 한반도에는 'Prapiroon'과 'Saomai' 2개의 태풍이 C. polykrikoides 적조세력에 영향을 미쳤다. 태풍 'Prapiroon'이 통과한 후 C. polykrikoides 유영세포의 수는 최대를 보였고, 태풍 'Saimai' 통과 후 적조는 소멸하였다(9월 20일). 10월 5일에는 진해만과 한산-거제만에서 C. polykrikoides 적조가 발생하였는데, 이때의 위성사진은 22$^{\circ}C$의 수온전선이 한산-거제만을 거쳐 진해만까지 깊숙이 관입되어 있었으며, 적조발생해역 및 현장수온도 일치하고 있었다. 또한 이 지역에서 가을과 겨울철에 우점 출현하는 Akashiwo sanguinea(=Gym, sanguineum)와 C. polykrikoides이 함께 적조를 형성하였다.

An advanced tool, droplet digital PCR (ddPCR), for absolute quantification of the red-tide dinoflagellate, Cochlodinium polykrikoides Margalef (Dinophyceae)

  • To quantify the abundance of the harmful dinoflagellate Cochlodinium polykrikoides in natural seawaters, we developed the innovative procedure using a droplet digital PCR (ddPCR) with C. polykrikoides-specific primers targeting the internal transcription sequence (ITS). The abundance of C. polykrikoides was estimated by the specific copy number of target ITS DNA segments per cell in cultures and natural water samples. The copy number per C. polykrikoides cell as acquired by ddPCR was $157{\pm}16$, which was evaluated against known cell numbers through a simplified protocol preparing DNAs. The abundances of C. polykrikoides in the waters of different locations estimated by ddPCR agreed with the number of cells visually counted under a microscope. This protocol was used to measure the abundance of C. polykrikoides close to and further off the southern coast of Korea in August of 2016 and 2017. The practical application showed that this method can reduce time for analysis and increase accuracy.

Feeding the Larvae of the Sea Urchin Strongylocentrotus intermedius on a Red-Tide Dinoflagellate Cochlodinium polykrikoides

  • This study is the first attempt to understand the feeding physiology of a sea-urchin larva on a red-tide dinoflagellate. Fifteen day old larvae of S. intermedius capture C. polykrikoides cells by localized reversal of ciliary beats. No failure to transporte the algal cells from theciliated band to mouth and no rejection at the mouth suggest that C. polykrikoides has no feeding deterrence to S. intermedius larvae. The trend obtained for the clearance rate of S. intermedius larvae is similar to that of other sea urchin larvae. Thus, the clearance rate decreased as the algal concentration increased. Maximum clearance rate of S. intermedius on C. polykrikoides was 17.7 $\mu l$/larva/hr. Ingestion rate rapidly increased at lower algal concentrations and saturated at higher concentrations. There was no inhibition in ingestion rate at the highest prey concentration of ca. 3000 cells/ml. Maximum ingestion rate of S. intermedius on C. polykrikoides was 131 ngC/larva/d, which is higher than that reported for the larvae of the mussel Mytilus gal-lotrovincialis, but lower than that of the ciliate Strombidinopsis sp. The grazing rate, calculated by combining the field data on algal abundances with experimental data on ingestion rate, suggests that due to its low abundance, sea urchin Iarva has no significant grazing impact on C. polykrikoides population.

적조생물 Cochlodinium polykrikoides의 세포표면 특이항원 단백질의 분리 (Isolation of a Specific Antigen Protein on Cell Membrane of Cochlodinium polykrikoides, Red Bloom)

  • 우리나라 연안에 주로 발생되는 적조인 Cochlodinium polykrikoides를 빠르고 정밀한 생화학적인 방법으로 측정하기 위한 일종의 maker로서 C. polykrikoides의 세포막에 존재하는 특이 항원성을 가진 막 단백질을 분리하였다. 먼저, C. polykrikoides와 gymnodium sangineum의 세포를 삼투압으로 터뜨린 후 원심분리하여 세포막을 모았다. 그 후 양 적조생물의 세포막은 1 mM DTT가 함유된 50 mM Na-carbonate로 용해하고 SDS-PAGE행하여 용해된 막 단백질을 분리하였다. SDS-PAGE로 분리된 막 단백질은 C. polykrikoides의 세포 막 단백질로 제조한 항혈청을 사용하여 immune-blot한 결과 C. polykrikoides의 120 kDa의 단백질이 G. sangineum의 동일한 크기의 막 단백질과는 서로 다른 항원성을 나타내었다.

동해안의 Cochlodinium polykrikoides 적조 발생에 미치는 해황의 특성 (The Influence of Oceanic Conditions on the Occurrence of Cochlodinium polykrikoides Blooms in the East Sea)

  • Harmful dinoflagellate Cochlodinium polykrikoides blooms have been frequently occurred in coastal areas of the East Sea since 1995. We compared the oceanic conditions in years 1995, 2001 and 2003 when the C. polykrikoides bloom was strong, and in years 1998 and 2004 when the C. polykrikoides bloom was not appeared. We studied temporal and spatial variation of upwelling and geostrophic currents on the western channel of Korean Strait, an entrance of the East Sea. The period and occurrence area of C. polykrikoides bloom was depended on variation of upwelling in summer. In the distributions of geostrophic current, southward current was dominant near the coast in August, 1998 and 2000. Whereas northward current was dominant near and off the coast in August, 1995 and 2003 which the C. polykrikoides bloom was strong. When compared dominant phytoplankton of the coastal areas in each year, Kuroshio indicator species Proboscia alata and Chaetoceros affine were dominant, respectively, in 2001 and 2003 at every stations. However, the dominant species was variable at each coastal area in 1998 and 2000. In 2003, the abundance of Sagitta elegans which is known as the cold water indicator was low, but the abundance of S. enflata, warm water indicator, was very high in Gangneung compared to Sokcho. It seemed that the distribution of S. elegans is restricted by strong warm water current. In conclusion, it was estimated that the distribution of C. polykrikoides bloom in the coastal area of the East Sea was closely related with the strength of East Korea Warm Current and upwelling.

적조생물 Cochlodinium Polykrikoides에 대한 유류 및 유처리제의 영향 (Effects of Oils and Dispersant on the Red Tide Organism Cochlodinium Polykrikoides)

  • Oil spill caused severe effects on the marine fauna and flora due to direct contact of organisms with the oil and even in regions not directly affected by the spill. This study was conducted to understand the effects of the oil spill accidents and the use of dispersant on the red tide of Cochlodinium polykrikoides. Crude oil produced in Kuwait, bunker-C, kerosene and diesel oil, and a chemical dispersant produced in Korea, were added with a series of 10 ppb to 100 ppm in the f/2-Si medium at $20^{\circ}C$ under a photon flux from cool white fluorescent tubes of $100\;mol\;m^{-2}\;s^{-1}$ in a 14: 10 h L:D cycle for the culture of C. polykrikoides. In low concentrations of ${\leq}$ 1 ppm of examined oils no impact on the growth of C. polykrikoides was recorded, while in high concentration of ${\geq}$ 10 ppm, cell density was significantly decreased with the range of 10 to 80% in comparison with the control. The growth of C. polykrikoides after the addition of the dispersant and the mixtures combined with oils and a dispersant of ${\geq}$ 10 ppm appeared to decrease, whereas the growth of C. polykrikoides exposed to ${\leq}$ 100 ppb showed little serious impact. However, almost all the C. polykrikoides cells were died regardless of a dispersant and combined mixtures within a few days after the addition of high concentrations.

A case study of red tide detection around Korean waters using satellite remote sensing

  • Korea has experienced 10 a Cochlodinium polykrikoides red tide outbreaks during the last 10 years (1993-2002). The monitoring activities at National Fisheries Research and Development Institute (NFRDI) in Korea have been extended to all the coastal waters after the worst of fish killing by C. polykrikoides blooms in 1995. NFRDI is looking forward to finding out the feasibility of red tide detection around Korean waters using satellite remote sensing of NOAA/AVHRR, Orbview-2/SeaWiFS, IRS-P4/OCM and Terra/MODIS on real time base. In this study, we used several alternative methods including climatological analysis, spectral and optical methods which may offer a potential detection of the major species of red tide in Korean waters. The relationship between the distribution of SST and C. polykrikoides bloom areas was studied. In climatological analysis, NOAA, SeaWiFS, OCM satellite data in 20th and 26th August 2001 were chosen using the known C. polykrikoides red tide bloom area mapped by helicopter reconnaissance and ground observation. The 26th August, 2001 SeaWiFS chlorophyll a anomaly imageries against the imageries of non-occurring red tide for August 20, 2001 showed the areas C. polykrikoides occurred. The anomalies of chlorophyll a concentration from satellite data between before and after red tide outbreaks showed the similar distribution of C. polykrikoides red tide in 26th August, 2001. The distribution of the difference in SST between daytime and nighttime also showed the possibility of red tide detection. We used corrected vegetation index (CVI) to detect floating vegetation and submerged vegetation containing algal blooms. The simple result of optical absorption from C. polykrikoides showed that if we use the optical characteristics of each red tide we will be able to get the feasibility of the red tide detection.

2013년 하계 자란만 서부 연안의 Cochlodinium 적조 미발생 원인 : 영양염 자료를 중심으로 (Non-Outbreak Cause of Cochlodinium Bloom in the Western Coast of Jaran Bay in Summer, 2013 : On the Basis of Nutrient Data)

  • 2013년 하계 자란만 서부 연안에서 Cochlodinium polykrikoides 적조의 미발생 원인을 화학적 현장관측결과와 기존에 발표된 C. polykrikoides의 생리학적 자료를 이용하여 해석하였다. 조사기간 동안 식물플랑크톤 군집은 규조류가 우점하고 있었으며, Cerataulina pelagica, Chaetoceros spp., Navicula spp. 그리고 Nitzschia spp.가 주요 우점종으로 출현하였다. 자란만 서부 연안의 영양염 농도는 이전의 C. polykrikoides 적조 발생 시기에 비해서 DIP 농도는 유사하였지만, DIN 농도는 낮았다. 특히, C. polykrikoides는 자란만 서부 연안에서 우점종으로 출현하는 규조류들에 비해서 무기 영양염에 대한 반포화상수(Ks)가 낮아 이들과의 종 경쟁에서 불리한 위치에 있는 것으로 보였다. 또한 상대적으로 낮은 DIN 농도를 보인 자란만 서부 연안은 질소에 대한 의존성이 높은 C. polykrikoides가 증식하기 위해서 불리한 환경이었다. 따라서 자란만 서부 연안의 낮은 영양염 환경하에서 무기 영양염 경쟁에 대해 불리한 위치에 있는 C. polykrikoides는 규조류의 번무에 따라 출현이 억제된 것으로 생각된다.

Cochlodinium 적조생물을 포식하는 Protoperidinium sp. (Report on Protoperidinium sp. fed on Cochlodinium polykrikoides (Gymnodiniales, Dinophyceae))

  • In 2005, harmful dinoflagellate Cochlodinium polykrikoides was first occurred on July 18 and disappeared on September 12 in the Yeosu waters of the South Sea of Korea. During C. polykrikoides blooms, the species of Protoperidinium was isolated from Jabong Island in Yeosu in the middle of August, 2005. The surface water temperature at the sampling site was $25^{\circ}C$, and salinity was 33.1 psu. The specimen was somewhat large in size and was longer than its width. The shape was close to being ovoid, with an apical horn. This study discovered cells with colorless and transparent ingested chain-forming C. polykrikoides. However, this species was not abundant, implying low grazing impact on C. polykkoides.
