초고압 추출 처리에 의한 더덕 및 발효더덕의 항산화 증진 (Enhancement of Antioxidant Activities of Codonopsis lanceolata and Fermented Codonopsis lanceolata by Ultra High Pressure Extraction)

  • 본 연구에서는 초고압 추출공정을 이용하여 전통적인 기존 추출공정과 비교함으로써 초고압 추출공정에 의한 등급 외 더덕 및 발효더덕의 항산화 활성의 증진을 확인하고자 연구를 수행하였다. 총 페놀화합물 함량은 초고압 발효더덕 추출물이 35.11 mg/100 g으로 초고압 더덕 추출물 14.78 mg/100 g, 일반더덕 추출물 14.56 mg/100 g에 비해 총 페놀화합물 함량이 약 2배 이상 높은 수치를 나타내었다. 또한, 플라보노이드 함량은 페놀화합물과 유사한 경향으로 초고압 발효더덕 추출물이 280.86 mg/100 g으로 초고압 더덕 추출물의 198.35 mg/100 g과 일반더덕 추출물 193.05 mg/100 g에 비해 약 1.4배 정도 높은 함량을 나타내었다. 항산화도는 초고압 발효더덕 추출물의 경우 0.6 mg/mL에서 약 70%의 항산화도를 나타내었으며, 이 값은 초고압 더덕 추출물 60.35%와 일반더덕 추출물 30.92%보다 높은 항산화도를 나타내었다. 초고압 발효더덕 추출물의 환원력은 최종농도 1.0 mg/mL에서 일반더덕의 경우 0.733, 초고압 더덕 추출물의 경우 0.881, 초고압 발효더덕 추출물의 경우 0.926 으로서 초고압 발효더덕 추출물의 환원력이 다른 추출물보다 약 0.193까지 높은 결과를 나타내었다. 따라서 초고압 추출공정이 등급 외 더덕 및 발효더덕의 기능성을 증가시킬 수 있으며, 초고압 추출공정의 최적화를 통한 활성물질의 추출 극대화를 통해 높은 경제적 가치를 부여해 줄 수 있을 것으로 사료된다.

발효한 더덕 차의 열처리에 의한 항산화 활성 및 품질특성 (Antioxidant Activities and Quality Characteristics of Fermented Codonopsis lanceolata Tea according to Heating Processes)

  • 본 연구는 독특한 식감과 풍미로 인하여 가공성이 낮은 C. lanceolata의 응용성을 높이기 위해 발효차를 제조하고, 열처리가 미치는 영향에 대하여 이화학 특성 및 항산화력 분석을 통하여 확인하였다. 열처리 온도 및 시간이 증가함에 따라 pH는 감소하는 경향을 보였으나, 갈색도, 탁도, 환원당, 총 폴리페놀 및 총 플라보노이드 함량은 유의한 수준으로 증가하는 경향을 보였다. 특히 200$^{\circ}C$ 15분 열처리한 시료의 총 폴리페놀 함량은 713.71mg/100ml로 대조구에 비해 약 4.5배 증가하였다. 항산화력 측정 결과, DPPH radical 소거능, FRAP 및 환원력에서 열처리 온도 및 시간이 증가함에 따라 항산화력이 뚜렷하게 증가되는 경향을 나타냈다. DPPH radical 소거능에서 $200^{\circ}C$ 15분 열처리한 시료의 경우, 85.40%로 대조구 45.17%에 비해 약 2배 이상 증가되었으며, FRAP 및 환원력 또한 $200^{\circ}C$ 15분 열처리한 경우, 대조구에 비해 약 2~3배 이상 증가하였다. 열처리에 따른 관능검사 결과, 열처리 온도 및 시간이 증가함에 따라 선호도 역시 높게 나타났다.

Isolation and Characterization of Thioredoxin cDNA from Codonopsis lanceolata (S. et Z.) Trautv

  • A thoredoxin (CTRX) gene was cloned and characterized from a full length cDNA library prepared from taproot of three-year old Codonopsis lanceolata. A CTRX was 666 nucleotides long and has an open reading frame of 372 bp with 124 amino acid residues (pI = 4.92). The deduced amino acid sequence of the CTRX matched to the previously reported plant thioredoxin h genes. The deduced amino acid sequence of CTRX exhibited the similarity of 33-67% among previously registered thioredoxin genes. The expression of CTRX in leaves of Codonopsis lanceolata was increased by wounding and 1 mM $H_2O_2$, but decreased by 0.1 mM cadmium.

The Improvement of skin barrier function and anti-obesity effect of Codonopsis lanceolata by supercritical carbon dioxide extraction

  • The root of Codonopsis lanceolata has been used in traditional medicine. This study was conducted to confirm the comparative effect of ethanol solvent extraction (CLE) and supercritical carbon dioxide extraction (CLS) of C. lanceolata roots. CLS had higher antioxidant than CLE. For supercritical co-solvent modified carbon dioxide extraction (CLS), it were extracted at 250 bar 50℃ 150 min at a flow rate of ethyl alcohol 3 mL/min for 90min. In addition, CLS inhibited the adipocyte differentiation of 3T3-L1 cells. When treated with the extract at a concentration of 100 ㎍/mL, the Wnt/β-catenin pathway reporter luciferase activity of HEK 293-TOP cells increased approximately by 3-folds compared to that of the untreated control group. Also, the treatment by CLS (50 ㎍/mL) showed a significant increase of involucrin expression. These results indicate that supercritical carbon dioxide extract of C. lanceolatamay serve as a cosmeceutical agent for improving skin barrier function and the treatment of obesity.

  • Proximate compositions of the roots of cultured and wild Codonopsis lanceolata were examined as tile basic reasearch for the study of their source of processed foods.No significant difference in the contents was found between the wild root and thecultivated at 3$0^{\circ}C$ in biotronroom. In view of the results to have measured mineralswhich is included in 14 sorts of Codonopsis lanceolata and surveyed their distribution.12 kinds of minerals including T1, Co, Ge, Sm, Mo, Sc, Be, Cd, As, Ga, Bi, ph are ne-vel or little included in almost source. Other twenty-one sorts of minerals (Ni, Se,Ba, Sb, Si, Ti, B, Li, Ifs, Ca, Sr, Mn, Fe, Cu, Zn, p, Al, Na, V, Cr, K) are more or less in-cluded in all source and Ca, Mg, p, K, and Fe are metals that are included in large qu-entities in comparison with others. No minerals difference in the contents was foundbetween the cultivated temperature. The content of elements of inorganic metal differsaccording to the part of C. lanceolata.

Simple sequence repeat marker development from Codonopsis lanceolata and genetic relation analysis

  • In this study, we developed 15 novel polymorphic simple sequence repeat (SSR) markers by SSR-enriched genomic library construction from Codonopsis lanceolata. We obtained a total of 226 non-redundant contig sequences from the assembly process and designed primer sets. These markers were applied to 53 accessions representing the cultivated C. lanceolata in South Korea. Fifteen markers were sufficiently polymorphic, and were used to analyze the genetic relationships between the cultivated C. lanceolata. One hundred three alleles of the 15 SSR markers ranged from 3 to 19 alleles at each locus, with an average of 6.87. By cluster analysis, we detected clear genetic differences in most of the accessions, with genetic distance varying from 0.73 to 0.93. Phylogenic analysis indicated that the accessions that were collected from the same area were distributed evenly in the phylogenetic tree. These results indicate that there is no correlative genetic relationship between geographic areas. These markers will be useful in differentiating C. lanceolata genetic resources and in selecting suitable lines for a systemic breeding program.

Molecular Cloning and Characterization of Type 2 Metallothionein cDNA from Codonopsis lanceolata (S. et Z.) Trautv

  • A class I type 2 metallothionein (CMet2) cDNA from taproot of Codonopsis lanceolata was isolated and characterized. A CMet2 cDNA was 572 nucleotides long and had an open reading frame of 234 bp with a deduced amino acid sequence of 78 residues (pI = 4.99). The deduced amino acid sequence of CMet2 matched to the previously reported type 2 metallothionein-like protein genes and showed 74% identity with that of G. max (BAD18377) and C. arietinum (CAA65009). Expression of CMet2 by the RT-PCR was increased at 1 hr after cadmium and hydrogen peroxide treatment, respectively.

증숙가공공정을 통한 더덕의 항산화 활성 증진 (Enhancement of Antioxidative Activity of Codonopsis lanceolata by Stepwise Steaming Process)

  • In general, stepwise hot steaming process is known to be effective in improving its biological activities; however, not much employed in processing Codonopsis lanceolata due to its hardness. In this study, C. lanceolata was first pretreated with warm water at $50^{\circ}C$ and $60^{\circ}C$ for two hours, then steamed for 3 hours. Antioxidant activities of 70% ethanol extracts were compared with the extract from the water solvent: 41.58% vs 8.98% of DPPH radical scavenging activity in adding $1.25mg/m{\ell}$ of steamed extract and water extract, respectively. Reducing power of steamed and fresh C. lanceolata were also measured as 1.39 and 0.71. Total poly phenolic of the steamed extract was estimated as 12.11mg/g, compared to 3.98mg/g fresh C. lanceolata. Total flavonoid contents were also obtained as 11.48mg/g, compared to 7.11mg/g of fresh C. lanceolata. In comparing phenolic acids profiles in the extract, in general higher amounts of gallic acid, trans-ferulic acid, vanillic acid were obtained possibly by easy release of active components during thermal processing, which results in better antioxidant activities than that of water extract. This findings can also be supported by result that the ethanol extract showed better activities than the water extract.

Priming과 파종후(播種後) 광질처리(光質處理)에 따른 도라지, 더덕, 만삼의 발아율(發芽率) (Effect of Priming and Light Quality on Seed Germintion in Three Campanulan Plants)

  • 본 연구는 전보(前報)에 이어 초롱꽃과 약용작물 중에서 농가에서 재배되고 있는 도라지, 더덕, 만삼 종자의 발아 및 유근신장에 미치는 priming 방법(재료, 농도, 기간)과 Priming 후 발아과정에서의 광질(光質) (적색광(赤色光), 백색광(白色光), 암상태(暗狀態)) 효과(效果)를 조사하여 입묘율 향상에 관한 정보를 제공하고자 실시하였던 바 그 결과를 요약하면 다음과 같다. 1. 평균발아율(平均發芽率)은 priming 후 발아시 암조건(暗條件)에서는 도라지, 만삼, 더덕 순으로 감소하였던 반면, Priming 후 광질처리(光質處理)로 인하여 만삼에서는 발아가 억제되고 더덕에서는 발아가 향상되어 도라지, 더덕, 만삼 순으로 감소하였다. 2. 암상태(暗狀態)로 수행된 priming 처리에서 처리기간 간에는 차이가 없었다 할지라도 $KNO_3$ 보다는 $Ca(NO_3)_2$가, 무처리(無處理) 또는 타처리농도(他處理濃度)에 비하여 $50{\sim}150mM$에서 발아율과 유근신장이 향상되었다.3. Priming 후 광질처리(光質處理)로 인한 발아율은 도라지에서는 priming 유무(有無)에는 차이가 없었던 반면, 더덕에서는 발아초기에, 만삼에는 시간이 경과할수록 발아율이 향상되었다. 4. 발아중 백색광(白色光) 처리(處理)는 더덕과 만삼에서 발아를 억제하였는데 이러한 경향은 만삼에서 큰 반면, 도라지에서는 초기발아가 조장되는 것으로 나타나 초롱꽃과 약용작물의 발아는 priming 후 주어지는 광질(光質)에 따라 변화됨을 알 수 있었다.
