Methyl Jasmonate 및 Salicylic Acid 처리에 의한 더덕(Codonopsis lanceolata) 부정근의 항산화 활성 증가 (Increasement of antioxidative activity in Codonopsis lanceolata adventitious root treated by Methyl jasmonate and salicylic acid)

  • 본 연구의 목적은 더덕부정근 배양에서 MeJA 및 SA의 처리가 페놀화합물, 플라보노이드의 생산에 미치는 영향과 이러한 이차대사산물의 증가에 따른 더덕배양근의 항산화활성의 효과를 조사하기 위해 수행되었다. 다양한 농도의 MeJA 및 SA를 더덕배양근에 처리한 결과, 20 uM MeJA에서 총페놀 화합물의 생산은 74.53 mg/g으로 무처리구보다 2배 높은 함량을 보였다. 하지만, MeJA 및 SA의 처리는 부정근의 생장을 감소시키는 것으로 나타났다. 총플라보노이드의 함량 역시 20 uM MeJA의 처리구에서 38.45 mg/g으로 가장 높은 함량을 보였다. 20 uM의 MeJA 처리에 의한 총페놀화합물과 총플라보노이드 함량의 증가는 결과적으로 더덕부정근의 항산화활성을 $IC_{50}$ 값 수준에서 MeJA 무처리구에 비해 2배, 3년생 재배더덕에 비해서는 11배 이상 증가시키는 것으로 나타났다.

  • 더덕(Codonopsis lanceolata)은 주로 한국, 중국 등 동아시아 지역에 재배되고 있으며, 더덕의 뿌리는 기침, 기관지염, 천식, 결핵, 소화 불량의 증상을 치료하기 위한 기능성 식품 및 전통 의학으로 사용되어져 왔다. 보고된 바에 의하면 phenylpropanoids, polyacetylenes, saponins, flavonoids와 같은 다양한 식물 천연물 성분들이 항비만, 항염, 항암, 항산화, 항미생물 활성과 같은 약리학적 작용에 관여한다고 보고되어 있다. MS기반 대사체학 분석을 이용한 주산지의 마커 성분을 선정하는 것은 다른 지역에서 재배된 약용 식물의 안전성뿐만 아니라 화학적 조성과 생물학적 효능의 변화와도 관련이 있기 때문에 부작용 없이 더덕의 유익한 효과만을 보장하는데 중요하다. 본 연구에서는 국내산 더덕의 주산지 특성을 구별하기 위해 UPLC-QTOF-MS를 기반으로 하는 대사체 프로파일링과 다변량 통계분석 기법인 PCA 분석을 수행하여 판별모델을 확립하였다. 그 결과 인제(강원도), 횡성(강원도), 무주(전라북도)의 3개 그룹이 PCA와 loading plot 분석결과 tangshenoside I, lancemaside A, lancemaside G는 더덕 주산지를 구별하기 위한 잠재적 대사체 마커들로 제안하였다.

Decreased Triglyceride and Cholesterol Levels in Serum, Liver and Breast Muscle in Broiler by the Supplementation of Dietary Codonopsis lanceolata Root

  • Effects of the dietary supplementation of Codonopsis lanceolata root on triglyceride and cholesterol levels in the serum, liver, breast muscle and bile in male Cobb$\times$Cobb chicks were investigated. The chicks (15-42 days old) were fed diets supplemented with 0, 0.25 and 0.5% Codonopsis lanceolata root. No differences were observed in body weight, feed conversion ratio, gall bladder weight or abdominal fat deposition among the control group and the two treatment groups. Liver weights were higher in chicks fed a 0.5% Codonopsis lanceolata diet than in those fed the control diet (p<0.05). However, serum levels of both glutamic oxaloacetic transaminase (GOT) and glutamic pyruvic transaminase (GPT) were not different among the three groups. Broiler chicks fed either 0.25% or 0.5% dietary Codonopsis lanceolata root showed decreased serum levels of triglyceride, total cholesterol and low density lipoprotein cholesterol compared to the control group (p<0.05). Supplementation with either 0.25% or 0.5% dietary Codonopsis lanceolata root decreased the triglyceride and total cholesterol levels in liver and breast muscle compared to the control group (p<0.05). Biliary cholesterol increased by 15% in chicks fed 0.5% dietary Codonopsis lanceolata root, suggesting that the biliary excretion of cholesterol had been elevated by dietary Codonopsis lanceolata root (p<0.05). In conclusion, these results indicate that dietary Codonopsis lanceolata root can decrease triglyceride and cholesterol levels in the serum, liver and breast muscle of broilers.

더덕 잎, 줄기, 뿌리 부위의 Triterpenoid 사포닌 함량 (Triterpenoid Saponin Contents of the Leaf, Stem and Root of Codonopsis lanceolata)

  • Codonopsis lanceolata (Campanulaceae) has been used in traditional medicines, as its roots contain several kinds of 3,28-bidesmosidic triterpenoid saponin with high medicinal values. In this study, we induced hairy root-derived transgenic plants of C. lanceolata and analyzed triterpenoid saponins from the leaf, stem and root. Transgenic plants were regenerated from the hairy roots via somatic embryogenesis. The saponins are lancemaside A, B and E, foetidissimoside A, and aster saponin Hb. Transgenic plants contained richer triterpenoids saponin than wild-type plants. Major saponin lancemaside A was the most abundant saponin in the stem from transgenic-plant, $4.76mg{\cdot}1^{-1}dry$ stem. These results suggest that transgenic plants of C. lanceolata could be used as medicinal materials for the production of triterpene saponins.

Isolation and Quantitative Analysis of Chemical Constituents in Codonopsis lanceolata

  • Codonopsis lanceolata has numerous chemical constituents that includes polyphenols, saponins, tannins, triterpene, alkaloids, and steroids. The extract of C. lanceolata was partitioned with Haxane, CH2Cl2, EtOAc, n-BuOH and MeOH. The determination of structure for lancemaside G, lancemaside B, lancemaside A were based on physicochemical and HPLC chromatogram data, including NMR and HR-MS. In addition, tangshenoside I and lobetyolin were identified in the material separation process for the extract of C. lanceolata, and content analysis was performed using HPLC. The compounds were confirmed as lancemaside G, lancemaside B, lancemaside A, tangshenoside I, and lobetyolin.

증숙 및 초고압 증숙 공정을 통한 더덕의 향장활성 증진 (Enhancement of Cosmeceutical Activity from Codonopsis lanceolata Extracts by Stepwise Steaming Process)

  • In general, stepwise hot steaming process is known to be effective in improving its biological activities; however, not much employed in processing Codonopsis lanceolata due to its hardness. The complex processed C. lanceolata showed highest free radical scavenging acitivity as 45.21%. Total phenol and flavonoid content were of complex processed C. lanceolata higher than conventional extract and alone steaming process. It was showed the lower melanogenesis rate on melanin production test by B16F10 cells as 27.46%. High inhibitory of tyrosinase was also measured as 28.61% by adding steamed Codonopsis lanceolata extracts by high pressure extraction of 1.0 $mg/m{\ell}$. And anti-wrinkle activity were 39.08%. In comparing phenolic acids profiles in the extract, in general higher amounts of polyphenol were obtained possibly by easy release of active components during thermal processing, which results in better antioxidant activities than that of general extract. This findings can also be supported by result that the extract by steaming process showed better activities than the general extraction extract.

산지별 더덕 추출물의 폐암 및 난소암에 대한 항암 효능 (Anti-tumor Effects of Codonopsis Lanceolata Extracts on Human Lung and Ovarian Cancer)

  • Codonopsis lanceolata L. (Campanulaceae) has long been used in traditional Korean medicine to treat bronchitis, cough, and inflammatory diseases, however, the efficacy of anti-tumor activities remains to be defined. In this study the effects of Codonopsis lanceolata (C. lanceolata) on proliferation, migration and adhesion in lung (A549, H1299) and ovarian cancer (SKOV-3) cells were investigated. To assess and compare the pharmacological effects and production places of C. lanceolata, the ethanolic extracts of C. lanceolata from different places in Korea (Hongseong, Yecheon, Yeongwol, Yanggu, Gangjin, and Hoengseong) were prepared. The extract from Hoengseong county did have only marginal anti-proliferative activity in all the cell lines tested, however, other extracts had little or no effect on cell proliferation. The extracts from Hongseong, Gangjin or Hoengseong county had partial anti-migratory activity in lung cancer cells, but not in ovarian cancer cells. In addition, the extract from Hoengseong county had partial anti-adhesive activity in ovarian cancer cells, however, other extracts did not affect cell adhesion in both lung and ovarian cancer cells. Taken together, these findings provide the first description of anti-tumor efficacy of C. lanceolata from different production places in Korea, and suggest that C. lanceolata from Hoengseong county may have therapeutic potential in lung and ovarian cancers.

Triterpenoid production and phenotypic changes in hairy roots of Codonopsis lanceolata and the plants regenerated from them

  • Codonopsis lanceolata (Campanulaceae) has been used in traditional medicines, as its roots contain several kinds of triterpenoid saponin with high medicinal values. In this work, we induced transgenic hairy roots of C. lanceolata and analyzed triterpenoid saponins from the hairy roots and hairy root-derived transgenic plants. Hairy roots were obtained from leaf explants by the transformation of Agrobacterium rhizogenes R1000. Transgenic hairy root lines were confirmed by the transcriptional activities of rolA, B, C, and D genes by RT-PCR. Transgenic root lines actively proliferated on hormone-free medium but not in nontransformed roots. Hairy roots contained richer triterpenoids (lancemaside A, foetidissimoside A, and aster saponin Hb) than nontransformed roots. Transgenic plants were regenerated from the hairy roots via somatic embryogenesis. They showed phenotypic alterations such as shortened shoots and an increased number of axillary buds and adventitious roots. The transgenic plants also contained higher triterpenoid levels than wild-type plants. These results suggest that hairy roots and transgenic plants of C. lanceolata could be used as medicinal materials for the production of triterpene saponins.

더덕(Codonopsis lanceolata) 재배지 기생선충의 발생상황 (Studies on Plant Parasitic Nematodes in the Fields of Codonopsis lanceolata)

  • 더덕 (Codonopsis lanceolata) 재배지에서 발생 빈도와 서식 밀도가 높은 식물기생선충을 파악하기 위하여 3개도 34개 지역 더덕 포장에서 조사한 결과 검출된 식물기생선충은 Meloidogyne(뿌리 혹선충), Pratylenchus(뿌리썩이선충)등 10개 속(genus)이었는데, 그 중 뿌리혹선충은 97%의 포장에서 검출되었고, 검출된 평균 선충수도 1,700마리/토양 300$m\ell$ 이상으로 대단히 높았다. 또 논 토양보다는 밭 토양에서 검출율이 높았고, 평균 밀도도 2,450마리로 10배정도 많았다.
