Studies on Discoloration of 16 Commercial wood Grown in Korea (한국산(韓國産) 유용목재(有用木材)의 변색(變色)에 관한 연구)

  • Change of color of woods is a trouble frequently found in decorative wood products. In this paper, studies were carried out on discoloation sensitivities of wood specimens to iron (0.1%, $FeCl_3{\cdot}6H_2O$), alkali (pH12.0, NaOH), acid (pH 1.0 $C_2H_2O_4$) and exposing to sunlight (40 hrs.). Four soft-woods and eleven hard-woods grown in Korea were used in this test. The results obtained were as follows: 1. In iron stain, strong discolored wood species were Larix leptolepis, Quercus mongolica. Zelkova serrata and Prunus yedoensis, and light discolored were Paulownia lomentosa and Pinus rigida. 2. In alkali stain. light discolored wood species were Abies holophylla, Kalopanax pictum, Populus ${\times}$ albaglandulosa, Acer mono, Pinus rigida, Pinus densiflora, Quercus mongolica, Betula platyphylla var. japonica, Prunus yedoensis, Zelkova serrata, Populus euramericana, Pinus koraiensis and Paulownia tomentosa, but there was no wood species of strong discolored compared with the color difference values of other reports. 3. In acid stain, light discolored wood species were Pinus rigida, Paulownia tomentosa. Pinus koraiensis, Quercus mongolica, Kalopanax piclum and Acer mone, but there was no strong discolored species compared with the color difference values of other reports. 4. In exposing to sunlight, strong discolored species were Larix leptolepis, Acer mono, Paulownia tomentosa, Populus euramericana, Pinus densijlora, Robinia pseudoacacia, Abies holophylla, Populus ${\times}$ albaglandulosa, Zelkova serrata, Prunus yedoensis and Pinus rigida, and light discolored was Alnus japonica. In general, it was shown that Korean wood species were susceptible to change of color by exposing to sunlight and iron stain compared with by alkali stain and acid stain.

The Influences of Vascular Endothlelial Growth Factor and Endothelin-1 on Speramtogenesis in Testis (정자형성 과정에서 Vascular Endothelial Growth Factor 및 Endothelin-1 발현의 면역조직화학적 연구)

  • Objective: The effects on spermatogenesis by expression of vascular endothelial growth factor (VEGF) and endothelin-1 (ET-1) were investigated. Materials and Methods: Testicular specimens were obtained from 40 infertile males due to primary testicular failure and from 10 fertile males with other urologic problems. The specimens of infertile males were devided into 4 groups according to histologic findings; Sertoli cell only syndrome (A), maturation arrest (B), hypospermatogenesis (C) and sloughing and disorganization (D). VEGF and ET-1 expression were detected with immunohistochemical stain. Results: VEGF expression on Leydig cell was detected in all cases. But, VEGF expression rates on germ cell were significantly higher in infertile group B, C, D compared to that of the control group (p<0.05). ET-1 expression rates on Leydig cell was significantly lower in all infertile group compared to that of the control group (p<0.05). But, ET-1 expression rates on Sertoli cell was significantly higher in all infertile group compared to that of the control group (p>0.05). In germ cell of infertile group, LH, FSH and prolactin were significantly decreased, and estradiol is increased in positive stain group on ET-1 immunohistochemical stain (p<0.05). VEGF and ET-1 expression were not correlated mean seminiferous tubule diameter (p>0.05). Conclusions: Abnormal spermatogenesis would be reflected in VEGF expression in germ cell.

Biocontrol of Blue Stain in Pine Wood with Lyophilized Mycelium of Ophiostoma quercus Albino Strain

  • Mycelium of Ophiostoma quercus albino strain cultured in liquid culture media was harvested, lyophilized, and stored for examining biocontrol efficacy against wood discoloration by staining fungi in the laboratory and field conditions. Dry weight of mycelium grown in brown sugar yeast extract broth(BYB) showed 3.8 times higher than that grown in potato dextrose broth(PDB). The optimum culture period in BYB was 4 weeks. In vitality test of the albino strain, the lyophilized mycelium stored in liquid nitrogen($-196^{\circ}C$) or in a refrigerator($4^{\circ}C$) kept the vitality until 13 months after storage; however, the mycelium stored at room temperature lost the vitality completely after 13 months. The mycelium stored in liquid nitrogen or in a refrigerator protected wood chips from the discoloration by pretreating mycelial suspension on pine wood chips. The mycelium stored at room temperature for 7 months also showed complete protection. These results suggest that the lyophilized mycelium have a biocontrol efficacy only if it keeps the least vitality. In the field conditions, both albino strain and $Woodguard^{(R)}$(commercial chemical protectant) showed significant differences(p=0.05) in discoloration rate as compared to the non-treated control when these were treated on the wood logs of Pinus rigida. The albino strain showed better protection than $Woodguard^{(R)}$. Isolation frequency of blue stain fungi from the chips of wood logs treated with the albino strain was 0% at three months after treatment, while that treated with $Woodguard^{(R)}$ was 76.7%. In another experiment, pre-treatment of mycelial suspension on the cut surface of wood logs also showed significant protection from wood discoloration. Spraying of both albino strain on the cut surface and insecticides on the bark also showed relatively good control effects as compared to insecticide alone on the bark or nontreated control.

  • Gastrointestinal stromal tumors (GISTs) are the most common primary mesenchymal tumors of the digestive tract. They have been commonly observed in adults but have been rarely described in children. They arise typically from the intestinal wall and rarely in the mesentery, omentum, or retroperitoneum. GISTs originate from the interstitial cell of Cajal and are characterized by overexpression of the receptor tyrosine kinase c-kit. Up to 94% of these tumors express the CD117 on immunohistochemical stain. Surgery is the main modality of treatment for primary resectable GIST. Completely resectable GIST with low risk has excellent prognosis after primary surgical intervention, with over 90% of the 5-year survival. We report a case of 10-year-old girl presenting with an upper gastrointestinal bleeding caused by gastrointestinal stromal tumor.

  • The karyotypes of Korean Sciunis vulgaris coreas and Tamias sibiricus asiaticus were analyzed by conventional Giemsa staining and C-banding method. The diploid chromosome number (2n) of Sciunis vulgaris coreae 40 consisting of 6 metacentric, 8 submetacentric, 3 subtelocentric and 2 telocentric autosome pairs, submetacentric X and acrocentric or subtelocentric Y chromosome. The arm number (NF) of this species was obtained as 72, excluding the gonosomal arms. Tamias sibiricus asiaticus has a 2n of 38. The karyotype was represented by 3 metacentric, 4 submetacentric, 5 subtelocentric and 6 telocentric autosome paits and 2 sex chromosome. The X chromosome was submetacentric chromosome and the Y was the smallest chromosome with a median. The NF was 60. In S. vulgaris coreae constitutive heterochromatins were observed at the centromeres and telomeres. Constitutive heterochnomatins of T sibiricus asiaticus were primarily observed at the centromeres. These results suggested that non-Robensonian reanagenents and distribution of constitutive heterochromatin played an imporiant role in karyological differentiation of these species.

Optimization of Bleaching Conditions for Stain Removal in Japanese Hackberry (Celtis sinensis Persoon) Using Response Surface Methodology (반응표면분석법을 이용한 팽나무(Celtis sinensis Persoon)의 최적 변색제거조건 결정)

  • This research was performed to investigate the effect of hydrogen peroxide on the stain removal in japanese hackberry. Response surface method (RSM) was used to optimize the bleaching conditions such as reaction temperature, reaction time and the concentration of hydrogen peroxide. Fifteen different bleaching conditions were selected according to $2^3$ factorial central composite design (CCD). The bleaching effect were evaluated by lightness differences of wood surface before and after the bleaching. The RSM model was determined and its $R^2$ values were 0.93, showing it well represented the bleaching effect. The most affecting factor on the stain removal was the concentration of hydrogen peroxide, followed by reaction time and reaction temperature. Second degree of concentration was proved to have an effect on the bleaching. Bleaching rates above 3% concentrations of hydrogen peroxide were tended to be slightly decreased, and low bleaching effect was found at $20^{\circ}C$. The determined RSM model may offer very practical ways to obtain the desired levels of bleaching because it offers multiple solutions.

Conservation Treatment and Scientific Investigation of Daehye Bogakseonsaseo (Letters of Master Bogak) in the Goryeo Dynasty (고려시대 대혜보각선사서의 보존처리 및 과학적 조사)

  • Daehye Bogakseonsaseo (Letters of Master Bogak) in the possession of the National Museum of Korea is a Goryeo-era book produced in 1387, which was acquired in 2005 in a state of severe general damage. Subsequently, its production techniques and materials were examined using bibliographical characteristics and scientific analysis, based on which an overall conservation treatment was carried out. Janghwang(mounting) in bibliographical characteristics is a five-hole-stitch binding (五針眼線裝本), a single-lined rectangular box (四周單邊), and no column lines. The conservation treatment was examined in two aspects: the basic form and the conservation treatment process. The book is composed of the outer cover, original cover, end paper, and inner paper of 0.04-0.07mm thick, which is significantly thinner than general book paper. The conservation treatment was applied after the entire book was disassembled, based on the assessment that both the cover and the inner sheets of paper were in a dire condition due to deterioration. The conservation treatment comprised of the process of disassembly, cleaning, reinforcement of defective parts, scanning, and binding, with a paulownia box made for its storage. At the time of disassembly, samples were made for the inner sheets and the book string in order to conduct fiber identification. The fibers were identified using Safranin and C-stains. Safranin staining identified a thick fiber wall, in addition to cross-marking, dislocation, and transparent membranes. C-stain staining identified the fiber as mulberry, given the dark red residue. The book strap has a flat, circular cross-section, and was identified as cotton fiber, since a lumen was observed in the center.

The Neuroprotective Effects of Uncariae Ramulus et Uncus on focal cerebral ischemia in rats brains (백서(白鼠)의 국부(局部) 뇌경색(腦硬塞)에 대한 조구등(釣鉤藤)의 신경보호(神經保護) 효과(效果))

  • The goal of this study is to investigate whether Uncariae Ramulus et Uncus can protect nerve cells against ischemic neuronal damage is caused by middle cerebral artery occlusion (MCAO) in rats' brains and to investigate whether the neuroprotective effect of Uncariae Ramulus et Uncus is concerned with IEGs(immediate early genes) expression. Uncariae Ramulus et Uncus(l00mg/kg) was administered immediately after 2 hours of MCAO and maintained for 7 days. On 7th days after 2 hours of MCAO, the brains of rats were sliced through the hippocampus. c-Fos immunohistochemistry stain and Cresyl violet stain were done for microscopic examination. Each number of viable neurons and c-Fos immunoreactive cells in CA1 was counted. The density of neurons and c-Fos immunoreactive cells were significantly decreased in MCAO-operated ischemic rats compared to that sham-operated rats. Administration of Uncariae Ramulus et Uncus group significantly elevated MCAO-induced decrease in density of neurons, and elevated MCAO-induced decrease in c-Fos immunoreactive cells. These results suggest that the neuroprotective effect of Uncariae Ramulus et Uncus against focal cerebral ischemia. Also, we hypothesized that neuroprotective mechanism of Uncariae Ramulus et Uncus is related to IEGs expression.

Mucosal Mast Cell Responses in the Small Intestine of C3H/HeN and BALB/c Mice Infected with Echinostoma hortense

  • In the intestinal mucosa, mast cells are thought to be responsible for the expulsion of parasites. We investigated the relationship of worm expulsion and mast cells in C3H/HeN and BALB/c mice infected with Echinostoma hortense. In addition, we examined whether the worm recovery rate was associated with the strain of mice, and whether a toluidine stain and immunohistochemistry using the c-kit antibody was effective in the detection of mast cells. In order to investigate the mucosal immune response of C3H/HeN and BALB/c mice, each mouse was infected orally with 30 E. hortense metacercariae. Then, the number of mucosal mast cells and worm recovery rates was observed in experimentally infected mouse strains between 1 week and 8 weeks post infection (PI). Mucosal mast cells were increased in 3 weeks P.I. in C3H/HeN and BALB/c mice. On the other hand, only mucosal goblet cells and worm recovery rates correlated in C3H/HeN mice (P=0.0482). Worm recoveries in C3H/HeN mice were 65.7$\pm$5.6, 53.3$\pm$5.4 and 6.7$\pm$0.6 in week 1, 2, and 3 P.I. and strongly decreased in week 3 P.I. Worm recoveries in BALB/c mice were 23.0$\pm$2.5, 10.0$\pm$1.0, and 6.7$\pm$0.6% in week 1, 2, and 3 P.I. and gradually decreased from week 1 P.I. to week 3 P.I. Worm recoveries in C3H/HeN mice were significantly higher than in BALB/c mice (P<0.00l). The number of mast cells in C3H/HeN and BALB/c mice using the anti-c-kit antibody reached to a peak in week 3 P.I. and recovered as normal level in week 5 P.I. and 6 P.I. The number in E. hortense-infected C3H/HeN mice (P=0.0015) was higher than in E. hortense-infected BALB/c mice (P=0.01) compared with the control group. There were significant differences in the number of mast cells among regions of the intestine in in C3H/HeN mice (P<0.05) but not in BALB/c mice (P>0.05). Immunohistochemistry using the anti-c-kit antibody was significant method as an examination of the number of mast cells (P=0.0002). In conclusion, the present study demonstrated that mast cells play an important role in worm recovery, and immunohistochemistry using the anti-c-kit antibody was superior to toluidine stain as an examination of mast cells.

Pulping Features of Blue-stained and Fungicide-treated Woods (청변균 및 살균제처리재의 펄프화특성)

  • This study was performed to understand the changes in wood extractives, mainly acetone extracts, in pine woods (Pinus densiflora and Pinus rigida) treated by three blue stain fungi (BSF) such as native BSF in Korea, Leptographium sp., screened Albino strain(BSFcs-1) and commercial Cartapip and fungicide, Wood guard. In addition their pulping and bleaching properties were investigated. BSF treatment has significantly reduced acetone extracts, $25.1{\sim}30.4%$ decreasing in red pine and $22.9{\sim}28.1%$ in pitch pine. Three week aging treatment showed about 20% decreasing in red pine and 19.3% in pitch pine. There were not so significant differences in extracts reduction among native BSF and Albino-type strains (Albino strain, BSFcs-1, and commercial Cartapip). But fungicide, Wood guard, treated wood showed relatively lower decreasing rates of extractives, 14% in red pine and 10.1% in pitch pine. Therefore it is understandable that the fungicide could protect the wood from blue stain fungi attack, but has no effect on its extractive reduction. Concerned to pulping properties of BSF and fungicide treated woods, red pine and pitch pine, optimum pulping condition was 20% active alkali, wood to liquor ratio 1 to 6, $170^{\circ}C$, and 2.5 hr. In the case of BSF woods, optimum pulping condition was same as the sound wood, $43.5{\sim}45%$ of pulp yields and $1.3{\sim}1.45%$ of rejects. Screened pulp yield of fungicide treated wood was lower than those of BSF treated woods. Rejects in pulps were higher in fungicide-treated wood than BSF treated woods. Bleaching pulp yields were ranged of 92 to 93.5%. BSF, Cartapip and fungicide treated woods resulted in lower brightness of $55{\sim}58%$, but Albino-type strain(BSFcs-1) $61.3{\sim}62.3%$, very similar to untreated one. Therefore bleaching chemicals could be saved in the processing of chemical pulping.
