Myxococcus xanthus socD500에 의한 포자 형성 및 CsgA신호에 특이적 유전자의 발현에 관한 연구 (Myxococcus xanthus socD500 mutation causes Sporulation and Induction of two C-signal Specific Genes)

  • 군집을 이루어 생활을 하는 세균류의 하나인 M. xanthus의 csgA 유전자는 세포 표면 단백질을 암호화하는데 이 단백질은 M. xanthus의 자실체 및 포자 형성을 수반하는 발생 (development)에 필수적이다. csgA 돌연변이들은 정상적인 성장을 보이지만, 발생 과정에서 자실체 및 포자 형성이 불가능하다. 또한, CsgA 세포간 기인한 신호 전달체계에 의해서 유발되는 CsgA 특이적 유전자들의 발현이 없거나 감소한다. socD500돌연변이는, csgA 돌연변이체로부터 자실체 혹은 포자 형성을 회복하게 해주는 2차 돌연변이 중의 하나로서, csgA 돌연변이체의 포자 형성을 회복하도록 한다. socD500에 의한 포자 형성은 영양원의 고갈 및 자실체의 형성이 없이도, 단순히 성장 온도를 $32^{\circ}C$에서 $15^{\circ}C$로 낮추는 것이 의해서 이루어진다. 이런 과정을 거쳐 형성된 포자의 구조를 전자현미경으로 관찰한 결과 정상 포자와는 다르게 단지 얇은 여러 겹의 막들이 존재하였다. 또한 socD500 돌연변이체에서 10개의 CsgA 특이적 유전자들의 발현율을 측정한 결과, 자실체 형성을 수반하는 정상적인 발생시에만 발현하는 ΩDK4506 및 ΩDK4406 유전자들이 $15^{\circ}C$에서 높게 발현되는 것으로 나타났다. 이 결과는 socD500은 CsgA에 특이적인 신호 전달체계에 관련된 일부 유전자들의 발현을 조절하는 역할에 관련되었다는 것을 알 수가 있었다.

The Potential of Diffusion-Weighted Magnetic Resonance Imaging for Predicting the Outcomes of Chronic Subdural Hematomas

  • Objective : Diffusion-weighted magnetic resonance imaging (DW-MRI) has proven useful in the study of the natural history of ischemic stroke. However, the potential of DW-MRI for the evaluation of chronic subdural hematoma (CSDH) has not been established. In this study, we investigated DW-MRI findings of CSDH and evaluated the impact of the image findings on postoperative outcomes of CSDH. Methods : We studied 131 CSDH patients who had undergone single burr hole drainage surgery. The images of the subdural hematomas on preoperative DW-MRI and computed tomography (CT) were divided into three groups based on their signal intensity and density : 1) homogeneous (iso or low) density on CT and homogeneous low signal intensity on DW-MRI; 2) homogeneous (iso or low) density on CT and mixed signal intensity on DW-MRI; and 3) heterogeneous density on CT and mixed signal intensity on DW-MRI. On the basis of postoperative CT, we also divided the patients into 3 groups of surgical outcomes according to residual hematoma and mass effect. Results : Analysis showed statistically significant differences in surgical (A to B : p<0.001, A to C : p<0.001, B to C : p=0.129) and functional (A to B : p=0.039, A to C : p<0.001, B to C : p=0.108) outcomes and treatment failure rates (A to B : p=0.037, A to C : p=0.03, B to C : p=1) between the study groups. In particular, group B and group C showed worse outcomes and higher treatment failure rates than group A. Conclusion : CSDH with homogeneous density on CT was characterized by signal intensity on DW-MRI. In CSDH patients, performing DW-MRI as well as CT helps to predict postoperative treatment failure or complications.

Effects of Guanidination with Trypsin, Lys-C, or Glu-C Digestion on Mass Spectrometric Signal Intensity and Protein Sequence Coverage

  • The conventional peptide modification process of guanidination, in which the amino groups of lysine residues are converted to guanidino groups using O-methylisourea to create more basic homoarginine residues, is often used to improve the signal intensity of lysine-containing peptides in matrix-assisted laser desorption/ionization mass spectrometry (MALDI-MS). Here, we used three different protease enzymes (trypsin, Lys-C, and Glu-C) to evaluate the effects of guanidination on the MS signals of two enzymatically digested proteins. Horse heart myoglobin and bovine serum albumin were guanidinated either before or after digestion with trypsin, Lys-C, or Glu-C. The resulting peptides were subjected to MALDI-MS, and signal intensities and sequence coverage were systematically evaluated for each digest. Guanidination prior to Glu-C digestion improved sequence coverage for both proteins. For myoglobin, guanidination before enzymatic digestion with trypsin or Lys-C also enhanced sequence coverage, but guanidination after enzymatic digestion enhanced sequence coverage only with Lys-C. For albumin, guanidination either before or after Glu-C digestion increased sequence coverage, whereas pre- or post-digestion guanidination decreased sequence coverage with trypsin and Lys-C. The amino acid composition of a protein appears to be the major factor determining whether guanidination will enhance its MALDI-MS sequence coverage.

  • An automatic Q. C. tester for a production line of electric grinder is developed using vibration signals. The methods of measuring and analyzing the vibration signals are found through several experiments at laboratory and production line. The experiments show that checking a vibration signal at running condition only, without any sound signal, is enough to judge whether the product is good or not. The Q. C. tester is made of accelerometer and PC. Measured vibration signal using accelerometer is transmitted to PC through A/D board. Vibration level are calculated using FFT algorithm in PC for already selected five frequency bands, which can specify the cause of fault. The Judging criteria of vibration levels of each bands are decided through a lot of experiment with the comparison of manual judgement.

  • A clinically oriented EEG and EP mapping system was developed with user-friendly interface and easy interactive operations. The system was based on the graphical user interface developed with C/C++ and Software Development Kit (SDK) operated under Microsoft Windows 3.1. Continuous acquisition for the EEG signal and burst mode acquisition for EEG signal syncronized to the external stimuli arc implemented with real time display. A neural network based automatic artifact discrimation is developed and implemented with which examination time can be reduced by a factor of 3 or more. Several bands of spectral maps and spectrums arc displayed for EEG diagnosis. Amplitude maps of EP signal at specified times by operator are displayed together with cine mode of EP maps for dynamic study. Source localization and other statistical signal processing are also included.

A Brief Introduction to the Transduction of Neural Activity into Fos Signal

  • The immediate early gene c-fos has long been known as a molecular marker of neural activity. The neuron's activity is transformed into intracellular calcium influx through NMDA receptors and L-type voltage sensitive calcium channels. For the transcription of c-fos, neural activity should be strong enough to activate mitogen-activated protein kinase (MAPK) signaling pathway which shows low calcium sensitivity. Upon translation, the auto-inhibition by Fos protein regulates basal Fos expression. The pattern of external stimuli and the valence of the stimulus to the animal change Fos signal, thus the signal reflects learning and memory aspects. Understanding the features of multiple components regulating Fos signaling is necessary for the optimal generation and interpretation of Fos signal.

  • This paper concerns to the acquisition of Global Positioning System L1 C/A coded signals. It specifically addresses the issues of acquiring very low power signals which are attenuated due to special circumstances such as indoor environment or forest canopy etc. The proposed post-processing algorithm applies modified signal folding coherent integration scheme on weak signal record. It dynamically compensates the doppler effect on the length of C/A code before integrating the signal power. Experimental results show effectiveness of the algorithm on weak GPS signals recorded in a real environment.

Exclusion zones for GNSS signals when reconfiguring receiver hardware in the presence of narrowband RFI

  • Narrowband interference can severely degrade the performance of GPS receivers. Detecting the presence of interference and then characterizing it can lead to its removal. Receivers can be reconfigured to focus on other signals or satellites that are less vulnerable to that interference at that moment. Using hardware reconfigurability of FPGA receivers and characterizing the effect of narrowband interference on the GNSS signal quality lead us to a new RFI mitigation technique in which the highest quality and less vulnerable signal can be chosen at each moment. In the previous work [1], the post processing capability of a software GPS receiver, has been used to detect and characterize the CW interference. This is achieved by passing the GPS signal and the interference through the correlator. Then, using the conventional definition of C/No as the squared mean of the correlator output divided by its variance, the actual C/No for each satellite is calculated. In this work, first the 'Exclusion zone' for each satellite signal has been defined and then by using some experiments the effects of different parameters like signal power, jamming power and the environmental noise power on the Exclusion zone have been analyzed. By monitoring the Doppler frequency of each satellite and using the actual C/No of each satellite using the traditional definition of C/No and actual data from a software GPS receiver, the decision to reconfigure the receiver to other signal can be made.

  • Digital signal processor (DSP) is used to obtain the peak value and the time difference of ultrasonic signals, to make digital filter, and to derive mathematical transformation from analog circuit. In this study, C-scan system and control program have been developed to high speed data acquisition. This system consists of signal processing parts (DSP, oscilloscope, pulser/receiver, digitizer), scanner, and control program. The developed system has been applied to a practical ultrasonic testing in overlay weld, and demonstrated high speed with precision

톤버스트 응답 신호의 스펙트럼 추정에 의한 배터리의 SoC 측정에 관한 연구 (A Study on SoC Measurement of Battery by Spectrum Estimation of Tone Burst Response Signal)

  • 배터리의 열화는 SoC 및 SoH의 변화에 따라 결정된다. 배터리의 열화를 판정하기 위해 전달 함수에 의한 배터리 SoC를 측정 하는 방법을 연구 하였다. 배터리의 SoC는 DC 저항이 검출되는 1Hz 대역에서 가장 크게 변화 한다. 하지만 관측하기 협소한 대역이기 때문에 톤버스트 신호를 입력신호로 사용하여 배터리의 주파수 응답대역을 대역을 1kHz로 이동 시켰다. 신호의 주파수 영역 특성을 관측을 위해 Welch스펙트럼 추정 방법을 사용하였다. 스펙트럼 추정 결과 배터리의 응답 신호는 SoC가 40%, 60%, 80% 일 때 각각 1dB의 차이를 갖는 것을 확인 하였으며, 이를 통해 새로운 배터리 SoC의 측정 방식을 제안 하였다.
