Development of PCR based Genetic Marker for Discrimination of Manchurian Clematis, Clematis terniflora var. mandshurica and Three-leaf clematis, Clematis apiifolia (으아리와 사위질빵 판별을 위한 PCR 기반의 마커 개발)

  • To distinguish manchurian clematis, Clematis terniflora var. mandshurica and three-leaf clematis, C. apiifolia, we collected 9 nuclear ITS sequences and two sequences of trnQ-trnH intergenic spacer and trnH region in GenBank database. Those sequences were aligned to find differences between those of C. terniflora var. mandshurica and C. apiifolia. Two primer pairs were newly designed base on the differences between two species and conducted multiplex PCR. The size of amplified fragments using generated primers were 380 base pairs (only three-leaf clematis) and 189 base pairs (both species). This genetic marker based on PCR is useful to discrimination of C. terniflora var. mandshurica and C. apiifolia

New Family of p-ary Sequences with Optimal Correlation Property and Large Linear Span (최적의 상관 특성과 큰 선형 복잡도를 갖는 새로운 p-진 수열군)

  • For an odd prime p and integer n, m and k such that n=(2m+1)ㆍk, a new family of p-ary sequences of period p$^{n}$ -1 with optimal correlation property is constructed using the p-ary Helleseth-Gong sequences with ideal autocorrelation, where the size of the sequence family is p$^{n}$ . That is, the maximum nontrivial correlation value R$_{max}$ of all pairs of distinct sequences in the family does not exceed p$^{n}$ 2/ +1, which means the optimal correlation property in terms of Welch's lower bound. It is also derived that the linear span of the sequences in the family is (m+2)ㆍn except for the m-sequence in the family.

  • In this paper, we propose a class of polyphase signature sequences, whose general odd correlation properties are useful for 4-phase signaling systems. The performance of the direct sequence code division multiple access (DS/CDMA) systems using the proposed sequences is compared with that using binary pseudo-noise sequences. The performance of the system using the proposed sequence is shown to be better than that using other conventional sequences both from the worst-case analysis and bit error rate simulations.

  • Let R be a commutative ring with identity. Let R be an integral domain and M a torsion-free R-module. We investigate the relation between the notion of e-exactness, recently introduced by Akray and Zebari [1], and generalized the concept of homology, and establish a relation between e-exact sequences and homology of modules. We modify some applications of e-exact sequences in homology and reprove some results of homology with e-exact sequences such as horseshoe lemma, long exact sequences, connecting homomorphisms and etc. Next, we generalize two special drived functor T or and Ext, and study some properties of them.

Construction of an Integrated Pepper Map Using RFLP, SSR, CAPS, AFLP, WRKY, rRAMP, and BAC End Sequences

  • Map-based cloning to find genes of interest, marker-assisted selection (MAS), and marker-assisted breeding (MAB) all require good genetic maps with high reproducible markers. For map construction as well as chromosome assignment, development of single copy PCR-based markers and map integration process are necessary. In this study, the 132 markers (57 STS from BAC-end sequences, 13 STS from RFLP, and 62 SSR) were newly developed as single copy type PCR-based markers. They were used together with 1830 markers previously developed in our lab to construct an integrated map with the Joinmap 3.0 program. This integrated map contained 169 SSR, 354 RFLP, 23 STS from BAC-end sequences, 6 STS from RFLP, 152 AFLP, 51 WRKY, and 99 rRAMP markers on 12 chromosomes. The integrated map contained four genetic maps of two interspecific (Capsicum annuum 'TF68' and C. chinense 'Habanero') and two intraspecific (C. annuum 'CM334' and C. annuum 'Chilsungcho') populations of peppers. This constructed integrated map consisted of 805 markers (map distance of 1858 cM) in interspecific populations and 745 markers (map distance of 1892 cM) in intraspecific populations. The used pepper STS were first developed from end sequences of BAC clones from Capsicum annuum 'CM334'. This integrated map will provide useful information for construction of future pepper genetic maps and for assignment of linkage groups to pepper chromosomes.

cDNA Sequence and mRNA Expression of a Putative Alcohol Dehydrogenase from the Mole Cricket, Gryllotalpa orientalis

  • Alcohol dehydrogenases (AHDs) are enzymes responsible for the catalysis of the reversible conversion of various alcohols to their corresponding aldehydes and ketonesis. Until now cDNA sequences of ADH gene is informed exclusively from several diptean species. We describe here the cDNA sequence and mRNA expression of a putative ADH gene from the mole cricket, Gryllotalpa orientalis, and phylogenetic relationships among known insect ADHs. The G. orientalis ADH cDNA sequences comprised of 798 bp encoding 266 amino acid residues. The multiple sequence alignment of G. orientalis ADH gene and known dipteran ADHs shared 100% identity in the nine amino acid residues that are important for the enzymatic activity in Drosophila melanogaster. Percent sequence identity ranged from 25% to 32% among all insect ADHs including both types of ADHs. G. orientalis ADH gene showed no clear resemblance to any dipteran species and type. Phylogenetic analysis of the deduced amino acid sequences of G. orientalis ADH gene with available dipteran ADH genes including both types of ADHs further confirmed that the G. orientalis ADH gene is not clearly assigned to either type of ADHs. Northern blot analysis revealed a stronger signal in the fat body than midgut and epidermis, indicating that the fat body possibly is a main site for the synthesis of the G. orientalis ADH protein.

A New M-ary Sequence Family Constructed From Sidel'nikov Sequences (Sidel'nikov 수열로부터 생성한 새로운 M-진 수열군)

  • In this paper, for a positive integer M and a prime p such that $M|p^n-1$, families of M-ary sequences using the M-ary Sidel'nikov sequences with period $p^n-1$ are constructed. The family has its maximum magnitude of correlation values upper bounded by $3\sqrt{p^{n}}+6$ and the family size is $(M-1)^2(2^{n-1}-1)$+M-1 for p=2 or $(M-1)^2(p^n-3)/2+M(M-1)/2$ for an odd prime p.

  • In this paper we propose three methods to generate short hopping sequences for the frequency hopping system. First, we explain the one coincidence set of sequences and the polyphase power residue seqences which have been known previously, and we suggest a method by modifying the one coincidence sequence and two methods by using the power residue sequences. We verify that the optimal position deleted-power residue sequences have the best Hamming autocorrelation property and the first position deleted-power residue sequences and the modified one coincidence sequences follows with respect to Hamming autocorrelation. We also explain that these sequences have the good balance property and can be implemented with low complexity.

Isolation and Characterization of cDNA Encoding Pyridoxal Kinase from Ovine Liver

  • cDNA fragments of ovine liver pyridoxal kinase were amplified by PCR using degenerate oligonucleotide primers derived from partial amino acids sequences of the enzyme. Using PCR products as probes, several overlapping cDNA clones were isolated independently from an ovine liver and a human brain cDNA library. The largest cDNA clone for each was selected for sequence analysis. The ovine liver cDNA encodes a polypeptide of 297 amino acid residues with Mr of 32,925, whereas the human clone is comprised of an open reading frame encoding 312 amino acid residues with Mr of 35,102. The deduced sequence of the human brain enzyme is completely identical to that of human testes cDNA recently reported (Hanna et al., 1997). The ovine enzymes have approximately 77% sequence identity with the human enzyme although the two sequences are completely different in the N-terminus comprising 32 residues. This result suggests that pyridoxal kinase is highly homologous in mammalian species.

Morphological and molecular evidence of the hybrid origin of Crepidiastrum ×muratagenii in Korea (홍도고들빼기의 형태 다양성 및 잡종 기원의 분자 증거)

  • The plant "Hong-do-go-deul-ppae-gi" has been considered as Crepidiastrum × muratagenii, a hybrid between C. denticulatum and C. lanceolatum, based on its morphological traits and geographical distribution. To reveal the hybrid origin of Hong-do-go-deul-ppae-gi, we examined additional morphological traits of this plant and its putative parents (C. denticulatum, C. lanceolatum, C. platyphyllum) and analyzed one nuclear ribosomal internal transcribed spacer (ITS) region and four chloroplast regions (trnT-L, trnL-F, rpl16 intron, and rps16 intron). As a result of examining the morphological traits, putative hybrid individuals were classified into three types based on the habit, cauline leaf, outer phyllary, and achene beak traits. A molecular analysis found that the ITS sequences of Type 1 and Type 2 individuals showed additive species-specific sites of C. denticulatum and C. lanceolatum. Plastid sequences of Type 1 and Type 2 individuals showed C. denticulatum and C. lanceolatum sequences, respectively. However, Type 3 individuals had ITS and plastid sequences corresponding to C. denticulatum. Accordingly, Type 1 and Type 2 individuals not only share morphological traits with C. denticulatum and C. lanceolatum but also show additive species-specific sites for C. denticulatum and C. lanceolatum, and not C. platyphyllum, supporting its origin as a hybrid between C. denticulatum and C. lanceolatum. Type 3 had morphological traits similar to other hybrid types but was distinguished with respect to outer phyllaries and demonstrated some resemblance to C. denticulatum. In a molecular analysis, Type 3 was found to be identical with regard to the sequence of C. denticulatum and was judged to be an ecological variation of C. denticulatum.
