Molecular Cloning and mRNA Expression of the Hanwoo CAT/enhancer-binding Protein α(C/EBPα) Gene (한우 CCAAT/enhancer-binding protein α(C/EBPα) 유전자의 동정과 mRNA의 발현)

  • CCAAT/enhancer binding proteins(C/EBP) are a group of transcription factors expressed during preadipocyte differentiation. In the C/EBPs, C/EBPa plays an important role in lipid deposition and adipocyte differentiation. In this studies, we report the identification, characterization, and expression of a Hanwoo CIEBP$\alpha$ The Hanwoo C/EBP$\alpha$DNA includes a 1059 bp open reading frame encoding a protein of 353 amino acids. The CIEBPa amino acid sequences of the Hanwoo show strong conservation with the corresponding sequences reported in other species. The distribution of C/EBP$\alpha$ mRNA in various tissues of Hanwoo aged 12 months were investigated using Northern blotting analysis. The highest expression was detected in adipose tissue and more lower expression was detected in colon and lung. We also identified expression of C/EBPa mRNA in Hanwoo sirloin and adipose tissue aged 12, 26, and 30 months by real-time RT-PCR. The higest expression were detected at 26 months in the sirloin and at 12 and 26 months in the adipose tissue.

A Sensorless and Versatile Temperature-Control System for MEMS Microheaters (온도센서를 사용하지 않는 MEMS 마이크로히터 온도제어시스템)

  • In this paper, we present a temperature-controlled system for MEMS electrical resistance heaters without a temperature sensor. To rapidly control the heater temperature, the microheater system developed consists of a power supply, power amplifier, digital ${\underline{P}}roportional-{\underline{I}}ntegral-{\underline{D}}ifferential$ (PID) controller, and a quarter bridge circuit with the microheater and three resistors are nominally balanced. The microheaters are calibrated inside a convection oven to obtain the temperature coefficient with a linear or quadratic fit. A voltage amplifier applies the supply voltage proportional to the control signal from the PID controller. Small changes in heater resistance generate a finite voltage across the quarter bridge circuit, which is fed back to the PID controller to compare with the set-point and to generate the control signal. Two MEMS microheaters are used for evaluating the developed control system - a NiCr serpentine microheater for a preconcentrator and a Nickel microheater for ${\underline{P}}olymerase\;{\underline{C}}hain\;{\underline{R}}eaction$ (PCR) chip.

  • The $Li_2O-MgO-MgF_2-SiO_2$glasses with addition of $B_2O_3$ were investigated in order to make glass-ceramics for low temperature firing substrate. Glasses were made by melting at $1450^{\circ}C$ in the electronic furnace and crystallized at $750^{\circ}C$. The crystal phases were polycrystalline of lithium boron fluorphlogopite and lithium fluorhectorite. The crystal shape was chanced to granule type from needle type with increasing $B_2O_3$ content. Average particle size of the glass-ceramics after water swelling was 3.77$\mu\textrm{m}$. The optimum sintering temperature and sintering shrinkage of the substrate were $900^{\circ}C$ and 13.4%, respectively.

Preliminary Studies on the Quality Changes of Eggplant as Influenced by Active Packaging

  • The effects of active packaging on the surface stiffness, mass, volume, density and weight changes of fresh as well as stored eggplant were studied at 11 and $25^{\circ}C$ for 10 days with active packaging material Type 1 and 2 and control. Mass, volume, and surface stiffness of eggplant decreased linearly throughout the storage period regardless of storage conditions; while the mass density showed a reverse trend in the ease of $11^{\circ}C$ storage. Reduction rate of mass, mass density and weight was observed minimum at $25^{\circ}C$ storage temperature with active packaging Type 1. The weight of eggplant decreased at a higher rate in the initial 4 days compared to that in the later period of storage regardless of storage temperature and type of packaging.

  • Considering ferrite grain size in the base metal, the prediction model for $A_{c3}$ temperature and prior austenite grain size at just above $A_{c3}$ temperature was proposed. In order to predict $A_{c3}$ temperature, the Avrami equation was modified with the variation of ferrite grain size, and its kinetic parameters were measured from non-isothermal data during continuous heating. From calculation using a proposed model, $A_{c3}$ temperatures increased with increasing ferrite grain size and heating rate. Meanwhile, by converting the phase transformation kinetic model that predicts the ferrite grain size from austenite grain size during cooling, a prediction model for prior austenite grain size at just above the $A_{c3}$ temperature during heating was developed.

  • We have developed a new heavy ion CT detector system for the use of a fan beam. The system consists of two sets of a position sensitive detector and an energy detector. The calibration runs were carried out using a $\^$12/C beam of 1mm in diameter with the energies of 290 MeV/u, 254.5 MeV/u, and 215.8 MeV/u. The spatial resolution of 1.1 mm and the energy resolution of about 1% were achieved.

Effects of growing degree days on growth and yield of maize depending on the sowing date

  • Total 15 different corn hybrids, Kwangpyeongok, Gangdaok, Yanganok, Singwangok, Jangdaok, Cheonganok, Cheongdaok, Andaok, Dapyeongok, Pyeongkangok, Pyeonganok, Daanok, Sunwon P3394, Gangilok, P3394, had been used to investigate the plant growth and yield of corn hybrids depends on the sowing date. Sowing dates were April 5th, June 25th, and July 5th and all experiments had three repeats. The growth of Gangdaok was the best. However, in the case of Kwangpyeongok, the growth was not the best to compare with Gangdaok, but the stem to ear height ratio was lower than Gangdaok, it may be better for the stable cultivation. Both of the growth and yield of Daanok was not good regardless of planting date, but the yield and ear shape of Pyeongkangok and Dapyeongok were good for fresh corn. The growth and yield of 15 different corn hybrids were variable depends on the planting date, however, the growth degree days (GDD) was the most important factor for the maturity of the corn. More than $1500^{\circ}C$ of GDD was enough to harvest mature corn hybrids in the central region of Korea. Besides the yield and growth, other characters such as sweetness and taste as a food should be investigated further for the fresh corn to be suggested.
