The Mechanism of t-Butylhydroperoxide-Induced Apoptosis in IMR-32 Human Neuroblastoma Cells

  • Apoptosis has been implicated in the pathophysiological mechanisms of various neurodegenerative diseases. In a variety of cell types, oxidative stress has been demonstrated to play an important role in the apoptotic cell death. However, the exact mechanism of oxidative stress-induced apoptosis in neuronal cells is not known. In this study, we induced oxidative stress in IMR-32 human neuroblastoma cells with tert- butylhydroperoxide (TBHP), which was confirmed by significantly reduced glutathione content and glutathione reductase activity, and increased glutathione peroxidase activity. TBHP induced decrease in cell viability and increase in DNA fragmentation, a hallmark of apoptosis, in a dose-dependent manner. TBHP also induced a sustained increase in intracellular $Ca^{2+}$ concentration, which was completely prevented either by EGTA, an extracellular $Ca^{2+}$ chelator or by flufenamic acid (FA), a non-selective cation channel (NSCC) blocker. These results indicate that the TBHP-induced intracellular $Ca^{2+}$ increase may be due to $Ca^{2+}$ influx through the activation of NSCCs. In addition, treatment with either an intracellular $Ca^{2+}$ chelator (BAPTA/AM) or FA significantly suppressed the TBHP-induced apoptosis. Moreover, TBHP increased the expression of p53 gene but decreased c-myc gene expression. Taken together, these results suggest that the oxidative stress-induced apoptosis in neuronal cells may be mediated through the activation of intracellular $Ca^{2+}$ signals and altered expression of p53 and c-myc.

  • 신경아세포종 세포 분화의 조절에 대한 연구는 아직 초기 단계에 불과하나 신경아세포종양의 임상적인 치료에 매우 중요한 기초가 된다. 본 연구는 신경아세포종 세포의 분화를 유도 할 수 있는 유용한 시약을 찾아내고자 하는 노력의 일환으로, 잘 알려진 DNA 합성 억제제가 신경 아세포종의 분화를 유도할 수 있는지를 살펴보고자 신경아세포종양 세포의 형태적, 생화학적 및 유전자 발현에 미치는 효과를 살펴보았다. 신경아세포종양으로부터 수립된 세포주, IMR-32 세포에 DNA 합성 억제제인 sodium butyrate, hydroxyurea, cytosine arabinoside를 각각 처리한 결과 처리하지 않은 대조군과는 달리 정상신경 세포 분화시 볼 수 있는 신경 돌기의 성장이 유도됨을 관찰할 수 있었으며 ,신경 전달 물질인 acetylcholine의 합성 효소인 choline acetyltransferase의 활성도가 현저히 증가되었다. 또한DNA합성 억제제를 처리하지 않은 IMR-32세포에서 탐지되지 않았던 c-myc 유전자의 발현이 시약 처리시 확연히 탐지됨을 관찰할 수 있었다. 이러한 실험 결과들은 DNA합성 억제제가 IMR-32세포의 성장을 억제하고 분화를 유도했음을 보여준 것이며, 어린 아이 시기에 많이 발병되는 신경아세포종양의 급속한 악성화나 전이의 억제기전을 제시해 줄 수 있으리라 생각한다.

청조구폐탕의 항암효과에 관한 연구 (Effect of Chungjokupye-tang on the Anti-cancer Property)

  • The purpose of this research was to investigate the effects of Chungjokupye-tang(CJKPT) on the anti-carcinogenic action. The cell viability of mouse spienocytes and thymocytes were enhanced by the addition of CJKPT. CJKPT were increased of splenic and thymic T lymphocytes, such as T/sub H/ cells were markedly increased by the treatment of CJKPT in vivo. CJKPT treatment induced the apoptotic cell death of Jurkat and HL60 leukemia cells. CJKPT reduced mitochondrial transmembrane potential and increased the expression of ICE, c-myc and p53 gene in Molt-4cells dose dependant manner. These results suggest that CJKPT have an anti-carcinogenic action via immunoregulatory mechanism.

식이 Capsaicin이 마우스의 주요 장기조직에서의 Proto-oncogenes Expression에 미치는 영향 (Effect of Dietary Capsaicin on Proto-oncogenes Expression in Various in Mice)

  • 매운맛 성분(capsaicin, CAP)이 암발생에 미치는 영향에 대한 분자적인 수준에서의 기초 정보를 확보하기 위해, 식이 CAP의 투여가 동물 조직 중 proto-oncogene 의 발현에 미치는 영향을 조사하였다. ICR mouse를 4 group으로 분류하여 각각 식이CAP 농도가 0, 5, 20, 100ppm이 되도록 조제한 먹이로 4주 동안 사육하였다. 사육기간 종료 후 동물들의 중요장기를 적출하여 total RNA를 분리하고, proto-oncogene(c-jun, c-myc, H-ras, erbB, p53)의 발현 수준을 slot blot hybridization assay를 통해 살펴 보았다. 이때, control probe로는 18SrRNA를 사용하였다. 그 결과, c-jun proto-oncogene의 발현은 각 주요 장기조직에 따라 다른 양상을 나타내었는데, 식이CAP 투여량이 증가함에 따라 간과 신장에서 그 발현이 증가하며, 위에서는 CAP 20ppm까지는 c-jun의 발현이 증가하다. 100ppm 투여시에는 감소하는 것으로 나타났으며, 비장에서는 식이CAP 투여량이 증가함에 따라 감소하는 경향을 보였다. 한편, tumor suppressor gene인 p53의 경우, 간에서만 CAP 20, 100ppm 처리시 약하게 발현되었다. 이들 결과로 보아, 식이 CAP에 의한 proto-oncogene의 발현은 CAP 투여량에 따라 그 정도를 달리하며, 그 발현 정도는 조직 특이성을 나타내는 것으로 평가된다.

Tumorigenicity of benzo(a)pyrene and benzo(a)pyrene diol epoxides in v-Ha-ras transgenic TG-AC mice

  • Tumorigenicity of benzo(a)pyrene (BP) and benzo(a)pyrene diol epoxides ((+)BPDE-1, (-)BPDE-1) was investigated in transgenic TG-AC mice carrying v-Ha-ras oncogene fused to the promoter of the mouse embryonic a-like, z-globin gene. Animals were topically treated twice per week for 25weeks with BPDE (10$\mu$g/mouse) and BP (10, 20, 40$\mu$g/mouse). In addition, animals were treated with BPDE or BP (initiated) followed by TPA (2$\times$2.5$\mu$g/week, for 4 weeks) for promotion study. In the continuous treatment of BPDE or BP, animals treated with 40$\mu$g BP showed $100\%$ tumor response after 20 weeks, $40\%$ of mice for 20$\mu$g BP, and $20\%$ for (+)BPDE-1, but (-)BPDE-1 and 10$\mu$g BP did not show any tumor response. After 25 weeks, most tumors turned out to be carcinomas in animals treated with 40$\mu$g BP. In BPDE or BP/TPA Initiation-promotion study, papilloma response occurred earlier (6 weeks after TPA treatment) than in continuously treated animals with BPDE or BP. RT-PCR assay for transgene expression showed that BP or BPOE was not transgene dependent in its tumorigenicity, but TPA was. Several Cytokine genes(TGF-a, TNF-a) and c-myc gene expressions were monitored in skin tissues during BP carcinogenesis. In early stage of BP treatment, the gene expressions were elevated(c-myc,TGF-a) or unchanged(TNF-a) compared to control, but the levels were gradually decreased during both middle and late stages of cacinogenesis, Gene expression levels of skin papillomas in acetone initiated-TPA promoted animals were close to those of middle stage or between middle and late stages. i-NOS was also highly expressed in carcinoma and papilloma, These data suggest that transgene expressions of TG-AC mice were not dependent on BP carcinogenesis and that TG-AC mice were more sensitive to TPA regardless of types of initiators. In addition, genes(TGF-a, c-myc, TNF-a, i-NOS) were modulated in the skin during BP cacinogenesis or TPA promotion.

Induction of Apoptosis and Cell Cycle Arrest by Dorema Glabrum Root Extracts in a Gastric Adenocarcinoma (AGS) Cell Line

  • Objective: Dorema glabrum Fisch. & C.A. Mey is a perennial plant that has several curative properties. Anti-proliferative activity of seeds of this plant has been demonstrated in a mouse fibrosarcoma cell line. The aim of the present study was to evaluate cytotoxicity of D. glabrum root extracts in a human gastric adenocarcinoma (AGS) cell line and explore mechanisms of apoptosis induction, cell cycle arrest and altered gene expression in cancer cells. Materials and Methods: The MTT assay was used to evaluate IC50 values, EB/AO staining to analyze the mode of cell death, and flow cytometry to assess the cell cycle. Quantitative real-time polymerase chain reaction (qRT-PCR) amplification was performed with apoptosis and cell cycle-related gene primers, for cyclin D1, c-myc, survivin, VEGF, Bcl-2, Bax, and caspase-3 to determine alteration of gene expression. Results: Our results showed that n-hexane and chloroform extracts had greatest toxic effects on gastric cancer cells with IC50 values of $6.4{\mu}g/ml$ and $4.6{\mu}g/ml$, respectively, after 72 h. Cell cycle analysis revealed that the population of treated cells in the G1 phase was increased in comparison to controls. Cellular morphological changes indicated induction of apoptosis. In addition, mRNA expression levels of Bax and caspase-3 were increased, and of bcl-2 survivin, VEGF, c-myc and cyclin D1 were decreased. Conclusion: Our study results suggest that D. glabrum has cytotoxic effects on AGS cells, characterized by enhanced apoptosis, reduced cell viability and arrest of cell cycling.

Construction and Validation of Human cDNA Microarray for Estimation of Endocrine Disrupting Chemicals (KISTCHIP-400 ver. 1.0)

  • Transcript profiling is a particularly valuable tool in the field of steroid receptor biology, as these receptors are ligand-activated transcription factors and therefore exert their initial effects through altering gene expression in responsive cells. Also, an awareness of endocrine disrupting chemicals (EDCs) and their potential screening methods to identify endocrine activity have been increased. Here we developed an in-house cDNA microarray, named KISTCHIP-400 ver. 1.0, with 416 clones, based on public database and research papers. These clones contained estrogen, androgen, thyroid hormone & receptors, sex hormone signal transduction & regulation, c-fos, c-myc, ps2 gene, metabolism related genes etc. Also, to validate the KISTCHIP-400 ver. 1.0, we investigated gene expression profiles with reference hormones, $10^{8}\;M\;17{\beta}-estradiol,\;10^{-7}\;M\;testosterone\;and\;10^{-7}\;M$ progesterone in MCF-7 cell line. As the results, gene expression profiles of three reference hormones were distinguished from each other with significant and identified 33 $17{\beta}-estradiol$ responsive genes. This study is in first step of validation for KISTCHIP-400 ver. 1.0, as following step transcriptional profile analysis on not only low concentrations of EDCs but suspected EDCs using KISTCHIP-400 ver. 1.0 is processing. Our results indicate that the developed microarray may be a useful laboratory tool for screening EDCs and elucidating endocrine disrupting mechanism.

Promoting Effects of Sanguinarine on Apoptotic Gene Expression in Human Neuroblastoma Cells

  • Neuroblastoma is the most common extracranial solid tumor in children. Approximately half of the affected patients are diagnosed with high-risk poor prognosis disease, and novel therapies are needed. Sanguinarine is a benzophenanthridine alkaloid which has anti-microbial, anti-oxidant and anti-inflammatory properties. The aim of this study is whether sanguinarine has in vitro apoptotic effects and which apoptotic genes might be affected in the human neuroblastoma cell lines SH-SY5Y (N-myc negative), Kelly (N-myc positive, ALK positive), and SK-N-BE(2). Cell viability was analysed with WST-1 and apoptotic cell death rates were determined using TUNEL. After RNA isolation and cDNA conversion, expression of 84 custom array genes of apoptosis was determined. Sanguinarine caused cell death in a dose dependent manner in all neuroblastoma cell lines except SK-N-BE(2) with rates of 18% in SH-SY5Y and 21% in Kelly human neuroblastoma cells. Cisplatin caused similar apoptotic cell death rates of 16% in SH-SY5Y and 23% in Kelly cells and sanguinarine-cisplatin combinations caused the same rates (18% and 20%). Sanguinarine treatment did not affect apoptototic gene expression but decreased levels of anti-apoptotic genes NOL3 and BCL2L2 in SH-SY5Y cells. Caspase and TNF related gene expression was affected by the sanguinarine-cisplatin combination in SH-SY5Y cells. The expression of regulation of apoptotic genes were increased with sanguinarine treatment in Kelly cells. From these results, we conclude that sanguinarine is a candidate agent against neuroblastoma.

인동등(忍冬藤)이 인간 유래 악성 흑색종 세포의 유전자 발현에 미치는 영향 (Effects of Lonicerae Caulis (LC) on Gene Expression of Human melanoma cells)

  • Objective : This study was designed to investigate anti-cancer and whitening activities (LC). So it was investigated the effects of LC on proliferation rates of melanoma genetic profile by LC. Methods : The genetic profile for the effect of LC on human derived melanoma cell, SK-MEL-2, was measured using microarray technique, and the functional analysis on these genes were conducted. Total 441 genes were up-regulated and 830 genes down-regulated in cells treated with LC. Genes induced or suppressed by LC were all mainly concerned with basic signalling pathways, which are involved in cell growth, differentiation and migration. Especially, many genes, which are related in apoptosis and cell cycle arrest were up-regulated by treatment with LC, and genes related in cell cycle were down-regulated. Result : The network of total protein interactions were identified by using cytoscape program, and some key molecules, such as BCL2L1, SIN3A, SMAD2 and c-myc that can be used for elucidation of therapeutical mechanism of medicine in the future. Conclusion : These results suggest possibility of LC as addition drug and whitening cosmetics. In addition, it was also suggested that related mechanisms are involved in BCL2L1, SIN3A, SMAD2 and c-myc related signalling pathways.

Development and Validation of the Custom Human cDNA Microarray (KISTCHIP-400) for Monitoring Expression of Genes involved in Hormone Disruption

Here we developed an in-house cDNA microarray, named KISTCHIP-400, with 401 clones, hormone related genes, factors, and ESTs, based on public database and research papers. Theses clones contained estrogen, androgen, thyroid hormone St receptors, sex hormone signal transduction & regulation, c-fos, c-myc, ps2 gene, metabolism related genes etc. And to validate the KISTCHIP-400, we investigated gene expression profiles with reference hormones, 10$\^$-8/ M 17be1a-estradiol, 10$\^$-7/ M testosterone, 10$\^$-7/ M progesterone, and thyroxin in MCF-7 cell line. Although it is in first step of validation, low doses and combinations of EDCs need to be tested. Our preliminary results that indicate the developed microarray may be a useful laboratory tool for screening EDCs and elucidating endocrine disrupting mechanism.
