Isolation of a Promoter Element that is Functional in Bacillus subtilis for Heterologous Gene Expression

  • To construct an efficient Bacillus subtilis expression vector, strong promoters were isolated from the chromosomal DNA libraries of Clostridium acetobutylicum ATCC 4259, Thermoactinomyces sp. E79, and Bacillus thermoglucosidasius KCTC 3400. The $P_{C27}$ promoter cloned from the clostridial chromosmal DNA showed a 5-fold higher promoter strength than the $P_{SP02}$ promoter in the expression of the cat gene, and its sequence was estimated as an upstream region of the predicted hypothetical gene (tet-R family bacterial transcription regulator gene) in C. acetobutylicum. As a promoter element, $P_{C27}$ exhibited putative nucleotide sequences that can bind with bacterial RNAP and the 3'end of the 16S rRNA just upstream of the start codon. In addition, the promoter activity of $P_{C27}$ was distinctively repressed in the presence of glucose. Using $P_{C27}$ as the promoter element, a glucose controllable B. subtilis expression vector was constructed and the lipase gene from Staphylococcus haemolyticus KCTC 8957P was expressed in B. subtilis. When compared with the lipase expression by the T7 promoter induced by IPTG in E. coli, the $P_{C27}$ promoter showed about a 1.5-fold higher expression level in B. subtilis than that without induction.

  • This paper presents the investigations and the reduction methods for EMI in the Elevator(E/L) system. In the large capacity E/L. Power factor controlled IGBT converter is used, so that the conducted EMI level is mostly determined by a high frequency input current. We suggest the low cost EMI countermearsure for E/L system using by a LCL filter ed a low cost noise filter.

  • Residual resistance ratio ( RRR ) of Cu/Nb-Ti Superconductor wire was evaluated by fixed point method. Our measured value of RRR showed good consistency with the average value obtained from round robin test conducted by IEC/TC 90. Our test results showed that the coefficient of variation ( COV ) was kept within 2 %

  • MeOH extract obtained from the Kochia scoparia (KS) was observed to inhibit tumor necrosis factor-alpha (TNF-alpha), prostaglandins (PGs) and nitric oxide(NO) production in a lipopolysaccharide (LPS)-stimulated murine macrophage cell line, RAW 264.7. These effects of MeOH-KS were based on modulation of iNOS and COX-2 level. Western blot analysis showed that MeOH-KS reduced the iNOS and COX-2 level in LPS activated macrophages, in a dose dependent manner without cNOS and COX-1 protein level. (omitted)

  • Administration of Scopolamine can be considered a psychopharmacological model of Alzheimer's disease (AD). We made an animal model of Alzheimer's disease (AD) by administering Scopolamine to Blab/c mice. In this study, we investigated the effects of Resplex Alpha on memory impairment and cognitive function in mice in a mouse animal model of Scopolamine-induced memory impairment. Through Y-mazed and passive avoidance behavioral assays, we observed that Resplex Alpha recovered Scopolamine-induced short-term memory and cognitive functions. The results of our study imply that Resplex Alpha may be beneficial in the prevention of Alzheimer's disease (AD).

  • The improvement of luminance and luminous efficiency is the one of the most important parts in AC-PDPs. To achieve high luminance and luminous efficiency, high VUV emission efficiency is needed. We measured the emission spectra of the vacuum ultraviolet(VUV) rays in surface discharge AC-PDP with ternary gas mixture of He-Ne-Xe. The influence of He-Ne-Xe gas-mixture ratio on excited $Xe^{\ast}$ resonant atoms and $Xe_2\;^{\ast}$ dimers has been investigated. It is found that luminous efficiency of ternary gas mixture, He-Ne-Xe, is shown to be much higher than that of binary gas mixture of Ne-Xe. For improving discharge luminous efficiency, we have studied VUV emission characteristics of ternary gas mixture, He(50%)-Ne-Xe and He(70%)-Ne-Xe with Xe concentration and filling gas pressure.

Quality Characteristics of Shepherd′s Purse(Capsella bursa-pastoris) Kimchi during Fermentation (냉이 김치의 숙성시 품질 특성 및 변화)

  • This study was carried out to investigate the shelf life of Shepherd's purse(Capsella bursa-pastoris) Kimchi during fermentation at 1$0^{\circ}C$. Capsella bursa-pastoris was treated without or with blanching. The viable cells of lactic acid bacteria(LAB) of raw and blanched Kimchi after fermentation for 15 days at 1$0^{\circ}C$ were 7.91 log CFU/mL and 6.4 log CFU/mL, respectively. The viable cells of LAB of Capsella bursa-pastoris Kimchi at 1$0^{\circ}C$ were lower in the blanched one when compared to the raw one. The pH of raw Kimchi was lower than that of the blanched one during fermentation for 25 days at 1$0^{\circ}C$. The viable cells of total bacteria of the blanched Kimchi were lower than that of non-blanched one during fermentation at 1$0^{\circ}C$. The ascorbic acid and chlorophyll contents decreased more in the blanched Kimchi when compared to that treated without heat. The sensory quality of the blanched Kimchi was a little inferior to that treated without heat during fermentation.

  • Although plants have evolved to possess various defense mechanisms from local biotic and abiotic stressors, most of yield loss is caused by theses stressors. Recent studies have revealed that several different stress responsive reactions are inter-networking. Therefore, the identification and dissection of stress responsive genes is an essential and first step towards understanding of the global defense mechanism in response to various stressors. For this purpose, we applied cDNA microarray analysis, because it has powerful ability to monitor the global gene expression in a specific situation. To date, more than 10,000 non-redundant genes were identified from seven different cDNA libraries and deposited in our EST database (http://plant.pdrs.re.kr/ks200201/pepper.html). For this study, we have built 5K cDNA microarray containing 4,685 unigene clones from three different cDNA libraries. Monitoring of gene expression profiles of hot pepper interactions with biotic stress, abiotic stresses and chemical treatments will be presented. Although this work shows expression profiling at the sub-genomic level, this could be a good starting point to understand the complexity of global defense mechanism in hot pepper.
