Optimization of Fed-Batch Fermentation for Production of Poly-$\beta$-Hydroxybutyrate in Alcaligenes eutrophus

  • Production of poly-$\beta$-hydroxybutyrate (PHB) in fed-batch fermentation was studied. Utilization of carbon for PHB biosynthesis was investigated by using feeding solutions with different ratios of carbon to nitrogen (C/N). It was observed that at a high C/N ratio carbon source was more preferably utilized for PHB accumulation while its consumption for cellular metabolism appeared to be more favored at a low C/N value. A high cell concentration (184 g/l) was achieved when ammonium hydroxide solution was fed to control the pH, which was also utilized as the sole nitrogen source. For the mass production of PHB, two-stage fed-batch operations were carried out where PHB accumulation was observed to be stimulated by switching the ammonium feeding mode to the nitrogen limiting condition. A large amount of PHB (108 g/l) was obtained with cellular content of 80% within 50 hrs of operation.

  • 세정단계를 거치는 혐기성 소화시스템(ADEPT, Anaeorbic Digestion Elutriated Phase Treatment)에서 돈사폐수를 처리한 유출수를 혐기성 암모늄 산화공정(ANAMMOX)에 적용하기 위하여 부분질산화 반응조의 기질로 사용하였다. 중온($35^{\circ}C$)에서 아질산성 질소를 축적하는 조절인자는 용존산소보다는 오히려 HRT, pH, 유리 암모니아(FA)였다. 중탄산의 스트립핑과 완충에 사용된 양을 포함한 중탄산알칼리도의 소모량은 전환된 $NH_4-N$ g당 8.78 g이었다. HRT 1일, DO 농도가 $2.7{\sim}4.4mg/L$의 안정상태에서 부분질산화 반응 유출수의 $NO_2-N/NH_4-N$ 비는 약 $1{\sim}$으로서, 부분질산화-혐기성 암모늄산화 결합공정의 ANAMMOX 반응조의 유입수로서 적합하였다.

  • Plasma display panel(PDP) is a leading technology for large-area flat panel displays. A current issue in operating the PDP cell is that the efficiency of the PDP cell is very low. To increase the efficiency of the PDP cell, the visible light needs to be maximized and the power consumption minimized. Since the excited xenons are related to the production of the visible light, it is important to optimize the cell geometry and the gas composition that produce the excited xenons more efficiently. A 2D-fluid code (FL2P) is developed and used to simulate the plasma dynamics and the radiation transport in the PDP cell. The cell is optimized with the code for various operating conditions and cell dimensions such as the voltage pulse, electrode length, electrode spacing, gap size, dielectric constant, gas mixture ratio, pressure, and pulse duration.

흡연여대생의 지질섭취와 혈중지질 및 지방산 조성에 관한 연구 (Studies on Lipid Intake, Serum Lipid Profiles and Serum Fatty Acid Composition in College Women Who Smoke)

  • Cigarette smoking has long been recognized as a major risk factor in the development of coronary heart disease. Several investigators have reported the strong association between smoking and high serum cholesterol, triglyceride concentration, SFA and low HDL cholesterol, PUFA and $\omega$6 concentrations. Therefore, this study was done to investigate the effect of smoking on the serum lipid profile and fatty acid composition of college women. Sixty-one non-smokers and twenty-seven smokers were selected from college women students in the Seoul area. Their lipid intake, serum lipid concentration and fatty acid composition were examined. There were no differences in the general characteristics and anthropometric indices between the smokers and non-smokers. However, alcohol consumption was significantly higher in smokers than non-smokers (p<0.001). The daily caloric intake of smokers and non-smokers were 1875.84 kcal and 1915.53 kacl, respectively. On the other hand, the mean daily intake of lipids and cholesterol were significantly lower in smokers (p<0.05). In smokers, the mean concentrations of serum total cholesterol, LDL-cholesterol and the LDL-C/HDL-C ratio were higher, and the compositions of EPA and DHA were lowe than in non-smokers. There was a negative correlation between the serum triglyceride and PUFA levels in the two groups. Also, serum HDL-C correlated negatively with MUFA in smokers and non-smokers (p<0.01). These results suggest that smoking cause inadequate changes in serum lipid profile and serum fatty acid composition, thereby increasing the tendency for coronary heart disease.

하해혼성(河海混成) 논토양(土壤)의 양분수지(養分收支)에 관한 연구 (Study on Nutrient Balance in Paddy Field of Fluvio-Marine Deposit)

  • 벼 재배시 시용한 비료와 자재양분의 이동과 흡수에 미치는 영향을 구명하고자 전북통에서 동진벼를 공시하여 2개년간(1999~2000)시험한 결과는 다음과 같다. 시험후 토양의 pH는 생우분 시용구에서 높아졌고 유기물, 인산, 규산, 칼리, 석회, 전질소함량이 증가하였고, 토층내로의 침투수중 무기태질소함량은 $NH_4-N$보다 $NO_3-N$함량이 월등히 많았으며 그 정도는 생우분시용구에서 높았으며, $PO_4-P$함량은 생우분시용구에서 타처리 보다 높았으나 K함량은 표준시비구에서 높았다. 벼 생육기간중 물의 소비량은 477mm였으며, 양분의 공급량과 소비량 중 질소는 무질소구, LCU20%감비구, 무비구에서 공급량보다 소비량이 많았으며, 인산의 경우는 무인산, 무비구를 제외하고는 공급량보다 소비량이 적었다. 또한 칼리는 모든 처리에서 공급량보다 소비량이 많았다. 양분의 침투수 중 시비질소는 진단시비, 표준시비+생우분+규산시용구에서 많았고 수확물의 질소회수율은 LCU20%감비구에서 높았다. 시비인산의 침투율은 생우분시용구에서 많았고, 시비칼리는 LCU20%감비구에서 많았다. 수도체 질소이용율은 LCU20%감비, 인산과 칼리의 이용율은 표준시비+규산시용구에서 높았다. 쌀수량은 진단시비 대비 LCU20%감비>표준시비+규산=표준시비+생우분+규산>표준시비+생우분 순으로 높았다.

Alcohol과 쌀 곰팡이의 Toxicity가 간(肝)의 지방(脂肪) 축적(蓄積)에 미치는 영향(影響) (Effects of Alcohol Intoxication and Moldy Rice Consumption on the Liver Lipid Content in Rat)

  • For the elucidation of the metabolic effects due to alcoholic drink and moldy rice intake author designed two experiments as follows. In the first experiment, sixty male and female rats, divided into six groups, 10 rats each in both sexes, were given 7.5% sucrose-30% alcohol solution (Jinro-Soju) and diets varied of fat content (2%, 7%, 30% of diet) and protein quality (casein, bean, anchovy) for 6 weeks. In the Second experiment, six groups of rats, 10 male rats each, were fed the diets containing zero, 5%, 10%, 25%, 50%, and 100% moldy rice contaminated Penicillium Islandicum and the experimental periods were 4 weeks and 6 weeks. The results of these studies due to the toxicity of alcohol consumption and moldy rice ingestion of the fatty liver production and the other effects are as follows. 1. Food intake of alcohol consumed groups decreased to 50%-70% of that of standard group. In the second study, there was no significant differences on food intake due to the different contents of moldy rice and experimental period. 2. On the view paint of body weight gain, the body weight of alcohol consumed groups gained much less than that of standard group, because food consumption was decreased due to alcohol ingestion. 3. In comparison of F.E.R. and P.E.R. between standard and experimental groups, there was no significant differences in this study. 4. As a matter of fact, there was no significance on the nitrogen balance in both studies. 5. From the result of hematology, R.B.C. counts, W.B.C. counts, hemoglobin, and hematocrit showed normal values in all groups including in this study. So we might conclude that the toxicity of alcohol and moldy rice do not effect significantly on blood picture. 6. The larger organs shrank on the range of 20%-70% of that against standard group in this study. The major reason for the shrinkage of organs might be account of decreased food consumption due to alcoholic drink. There was no great changes on the organ weight due to the ingestion of moldy rice. 7. The nitrogen content in various organs in both experiments was revealed at the normal level for the worst condition in terms of the least food intake. In other word, it was noteworthy that the concentration of nitrogen in various organs was kept at the normal ratio as standard groups under the circumstances of this study. 8. The lipid content in the liver of rats fed alcohol and diets containing either various lipid contents or protein quality did not increase. Hepatic lipids accumulation due to the dietary fat content was observed, but there wvas no significances among the compared groups. In the second experiment, the difference of hepatic lipid content between the moldy rice groups and standard group was not showed. In addition to the result of total lipid, hepatic free cholesterol, free fatty acid, and triglyceride did not change in both studies, we might conclude that the toxicity of alcohol and moldy rice could not effect on the hepatic lipid contents. 9. There was no significant differences on the serum glucose level between alcohol groups and standard group. In the second experiment, serum glucose level increase in 6th week compared with in 4th week, but there was no significant differences.

Hospital Outpatients are Satisfactory for Case-control Studies on Cancer and Diet in China: A Comparison of Population Versus Hospital Controls

  • Background: To investigate the internal validity of a food-frequency questionnaire (FFQ) developed for use in Chinese women and to compare habitual dietary intakes between population and hospital controls measured by the FFQ. Materials and Methods: A quantitative FFQ and a short food habit questionnaire (SFHQ) were developed and adapted for cancer and nutritional studies. Habitual dietary intakes were assessed in 814 Chinese women aged 18-81 years (407 outpatients and 407 population controls) by face-to-face interview using the FFQ in Shenyang, Northeast China in 2009-2010. The Goldberg formula (ratio of energy intake to basal metabolic rate, EI/BMR) was used to assess the validity of the FFQ. Correlation analyses compared the SFHQ variables with those of the quantitative FFQ. Differences in dietary intakes between hospital and population controls were investigated. Odds ratios (ORs) and 95% confidence intervals (CIs) were obtained using conditional logistic regression analyses. Results: The partial correlation coefficients were moderate to high (0.42 to 0.80; all p<0.05) for preserved food intake, fat consumption and tea drinking variables between the SFHQ and the FFQ. The average EI/BMR was 1.93 with 88.5% of subjects exceeding the Goldberg cut-off value of 1.35. Hospital controls were comparable to population controls in consumption of 17 measured food groups and mean daily intakes of energy and selected nutrients. Conclusions: The FFQ had reasonable validity to measure habitual dietary intakes of Chinese women. Hospital outpatients provide a satisfactory control group for food consumption and intakes of energy and nutrients measured by the FFQ in a Chinese hospital setting.

조피볼락(Sebastes schlegelii)치어의 산소소비, 암모니아 질소배설 및 체조성에 미치는 사료내 에너지/단백질비의 영향 (The Effects of Dietary Energy/Protein Ratio on Oxygen Consumption, Ammonia Nitrogen Excretion and Body Composition in Juvenile Rockfish, Sebastes schlegeli)

  • 일정한 에너지를 함유한 사료내의 에너지 /단백질의 비를 11.5, 8.7, 7.0으로 각각 다르게 제조한 사료를 조피볼락, S. schlegeli이 치어에 공급시킨 후 24시간동안 2시간 간격으로 산소소비율과 암모니아 질소배설률을 측정하고 어체를 분석하여 이들에 미치는 사료내 에너지 /단백질비의 영향을 알아보았다. 사료 I, IIl, III을 각각 공급시킨 실험군의 평균산소소비율은 각각 $0,93\pm0.02^a,\;0.92\pm0.02^a\;and\;1.03\pm0.03^bml\;O_2/g/h$ml 이었다. 즉, 사료 III 공급군의 평균산소소비율이 다른 사료공급군에 비해 유의하게 높았다. 사료 공급 후 24시간동안 평균 암모니아 질소배설률은 사료내 에너지/단백질비가 증가함에 따라 증가하였으며, 사료 I, II, III의 공급군에서 평균 암모니아 배설률은 각각 $9.14\pm0.39^a,\;12.41\pm 0.53^a\;and\;14.89\pm0.55^c\;{\mu}g\;N/g/h$ 로 나타났고 각 실험군간의 평균암모니아 배설률값들은 모두 유의한 차를 보이고 있다 (P<0.05). 어체내 단백질 함량은 사료 II의 공급군에서 $62.2\%$로 가장 높게 나타났으며, 사료 I, II 공급군에 비해 유의하게 높았다 (P<0.05). 그리고 체내 지질 함량은 사료 II의 공급군에서 $16.0\%$로 가장 낮게 나타났고, 사료 I의 공급군에서는$23.2\%$로 가장 높게 나타났다. 회분함량은 에너지/단백질비가 낮아짐에 따라 약간씩 증가하며 탄수화물 함량은 사료 I에서 $5.51\%$로 가장 높게 나타났다. 그러나 지질, 회분 및 탄수화물의 함량은 사료 I, II, III의 공급군간에 유의한 차이는 없었다 (P>0.05). 따라서 본 실험에서는 사료 II가 생리학적인 측면에서 관찰한 결과 조피볼락 치어에 대해 가장 적합한 것으로 여겨진다.

Smoking, Meat Intake and Exercise related to Alcohol Use

  • This study was conducted to investigate the relationship between drinking, smoking, meat intake, and exercise. The participants were 1,060 males and females who voluntarily responded to the survey by visiting the health promotion booth at the H Festival held in C city in 2014. Research data were collected in interview form using questionnaires on alcohol use, smoking, exercise, and meat intake. The data were analyzed by descriptive analysis, cross tabulation analysis, multinominal logistic regression after stratification of gender using the SPSS 24 program. The results of the study are as follows: Smokers had a significantly higher frequency of drinking ($x^2=163.33$, p<.001) than non-smokers. Meat intake was high when alcohol use was high ($x^2=35.13$, p<.001). The risk rates for smoking, meat consumption, and exercise related to alcohol use are as follows. The smoking was higher as the frequency of drinking increased. For men, smoking OR(odd ratio) was 6.26 (p=.001) and for women, smoking risk OR was 16.82 (p=.002). Meats intake showed a higher OR as the alcohol use increased. The OR of almost daily ingestion of meat at frequent drinking days was 4.40 (p=.002) for males and 4.52(p=.009) for females. As the frequency of drinking increased, the OR of days of exercise tended to decrease. In men, the OR was 0.36 (p=.003), the lowest in the probability of exercising more than 5 days when drinking high. In the case of women drinking less than once a week, the OR was 0.43 (p=.027), which was the lowest for exercise for 5 days or more. In conclusion, the higher the frequency of drinking, the higher the smoking and meat intake, and the less exercise. If drinking, smoking, and meat intake are high but there is a lack of exercise, a chronic disease can occur and cause premature death unless there is a change in the new health behavior. The results of this study suggest researches to understand the motives related to alcohol abstinence, smoking cessation, dietary control and exercise, and to develop programs.

농촌지역에 거주하는 노인의 생선 섭취량과 인지기능저하 위험도 간의 상관성 (Correlation between fish consumption and the risk of mild cognitive impairment in the elderly living in rural areas)

  • 본 연구는 2009-2010년 양평지역에서 수행된 양평 코호트 자료를 활용하여 60세 이상 노인을 대상으로 생선 섭취량과 인지기능저하와의 상관성을 확인하였다. 여자 노인의 경우 생선 섭취량이 높을수록 MMSE-KC 평균 점수가 높아지는 경향을 보였으나 (p for trend = 0.016), 남자 노인의 경우 유의한 차이가 나타나지 않았다. 또한 여자 노인의 경우 생선 섭취량이 높을수록 인지기능저하의 위험이 낮아지는 경향을 보였으나 (p for trend = 0.009), 남자 노인의 경우 통계적으로 유의한 결과가 나타나지 않았다. 여자 노인의 경우 생선 섭취량이 높을수록 항산화 비타민 (베타 카로틴, 비타민 A, 비타민 C, 비타민 E), 비타민 B군 (엽산, 비타민 B12), 지방산 (총 지방산, 포화지방산, 단일불포화지방산, 다불포화지방산, α-리놀렌산, EPA, DHA, 총 n-3 다불포화지방산) 섭취량이 높아지는 경향을 보였다. 남자 노인의 경우 생선 섭취량이 높을수록 항산화 비타민 (베타 카로틴, 비타민 A, 비타민 C, 비타민 E), 비타민 B군(엽산, 비타민 B12), 지방산 (총 지방산, 포화지방산, 단일불포화지방산, 다불포화지방산, α-리놀렌산, EPA, DHA, 총 n-3 다불포화지방산) 섭취량이 높아지는 경향을 보였다. 본 연구 결과 여자 노인에서 생선의 섭취가 많을 수록 인지기능저하 위험도가 감소함을 확인하였다. 향후 다양한 집단에서 생선 섭취량과 노인의 인지능력 간의 인과관계를 규명할 수 있는 종적연구가 필요하다.
