Comparison of General Composition of Cooked Krill and Alcalase Optimization for Maximum Antioxidative Activity by Using Response Surface Methodology (자숙크릴의 일반성분 분석과 항산화 활성을 위한 반응표면법에 의한 알카라제 가수분해 최적화)

  • The objective of this study is to optimize enzymatic hydrolysis of cooked krill by using Alcalase. To optimize krill hydrolysis on such dependent variables as TCA, DPPH-scavenging, and Fe-chelating activities by using Alcalase, independent variables of hydrolysis pH and temperature were investigated Their formulas and three dimensional graphs were obtained by using SAS and Maple softwares, respectively. For comparison of general composition of raw krill, its contents of moisture, crude protein, crude fat, and ash were 17.48%, 53.74%, 15.66%, and 10.21%, respectively, and for cooked krill, its contents were 4.80%, 71.84%, 5.26%, and 15.09%, respectively. The composition of fatty acids for cooked krill was similar to that of raw krill. The most abundant fatty acid was palmitic acid(16:0) and the following order was oleic acid(18:1), eicosapentaenoic acid (20:5), palmitoleic acid(16:1), and docosahexaenoic acid(22:6). For DH optimization of hydrolysates from cooked krill, its result was pH 8.5 and $66.6^{\circ}C$ hydrolysis temperature for the maximum DH of 29.4% For DPPH-antioxidative optimization of hydrolysates from raw krill, its maximum result of 27.1% was obtained in the hydrolysis condition of pH 7.4 and $67.5^{\circ}C$. For Fe-chelating optimization of hydrolysates from cooked krill, its maximum result of 24.9% was in the condition of pH 8.7 and $65.5^{\circ}C$. These results can be used for basic data for using krill products and other fish products as bioactive ingredients.

Effects of Commelina communis L. on the Blood Glucose Level in Alloxan Induced Diabetic Rat and the Biochemical Properties of Glucose-6-Phosphate Dehydrogenase from the Rat Livers (당뇨유발쥐에서 닭의장풀의 혈당감소효과와 간조직내의 Glucose-6-Phosphate Dehydrogenase의 효소활성에 미치는 효과)

  • The hypoglycemic and metabolic effects of Commelina communis L. extract were investigated in alloxan induced diabetic rats. The increased blood glucose level in the diabetic rats was significantly reduced and the loss of body weight was recovered with the treatment of the plant protein fractions($30{\sim}70%$ ammonium sulfate precipitates). Administration of the plant protein fractions elicited the significant increase of glucose-6-phosphate dehydrogenase (G-6-P DH) activity and liver weight which were decreased in the diabetic rat liver. G-6-P DH was partially purified from extract- or insulin-treated diabetics, diabetic control, and normal rat liver and studied for the biochemical properties. The $K_m$ value(9.002 mM) of diabetic rat liver enzyme was greatly higher than that (0.033 mM) of normal enzyme indicating the affinity of enzyme for the substrate was significantly reduced in the diabetic rat liver. This reduced affinity of enzyme for the substrate in the diabetic rat was recovered in the extract- or insulin-treated rat liver enzyme having 0.164 or 0.208 mM of their $K_m$ values, respectively. Although there was no significant difference in the optimum pH(6.0) and optimum temperature($37^{\circ}C$) of enzyme among the experimental groups, the dependence of their activities on pH appeared to be slightly resistant in the extract- or insulin-treated group compared to the diabetic group. In order to investigate the antigenicity of rat liver enzyme among experimental groups, enzyme-linked immunosorbent assay was carried out by using anti-G-6-P DH anti-serum. Absorbance(0.102) shown in the normal rat liver was reduced even below zero in the alloxan-diabetic rat liver, but increased again in the extract- or insulin-treated rat liver(0.096 or 0.118, respectively). The result of this study suggested that G-6-P DH may be used as a marker enzyme to diagnose and to indicate the progress of the diabetics, and the hypoglycemic effect of the extracts of Commelina communis L. was certainly associated with action or mode of G-6-P DH on the rat liver.

Development of the breeding materials with diverse grain size and shape in japonica rice

  • We developed the breeding materials with diverse grain size and shape in japonica rice. Grain size and shape are important factors affecting consumer preference and choice. However, most of Korean japonica rice cultivars have small, short, and round grain. To diversify the grain size and shape of japonica rice, we conducted the breeding program using donor parents, Jizi1560 and Jizi1581. Jizi1560 and Jizi1581 are japonica germplasm with extremely large grain. Four crosses between the each donor parents and high yielding japonica rice cultivars, Deuraechan and Boramchan, were constructed and then anther culture method was applied. We obtained 290 doubled-haploid (DH) lines with appropriated morphological traits and selected 91 DH lines with diverse grain size and shape. The grain related-traits of the selected DH lines showed a higher diversity when compared with 319 cultivars developed by NICS (264 japonica, 13 black, and 32 Tongil type cultivars). We designated the selected DH lines, four parents, and Daeripbyeo 1, large grain japonica cultivar, as the breeding materials for further analysis. The breeding materials were classified into five groups, A to E, based on the grain-related traits. Group A (including Jizi1581) and Group B (including Daeripbyeo 1) showed similar grain width, whereas Group A exhibited longer grain length and heavier grain weight. Group C (including Deuraechan and Boramchan) showed shorter and rounder grain shape and smaller grain size than any other groups. Group D including solely Jizi1560 had extremely large grain, such as the longest grain length, width, and thickness and heaviest grain weight. Group E including only two DH lines had long and slender grain shape, so that showed the highest ratio of length to width. The grain size and shape of the breeding materials exhibited beyond the characteristics of previously developed Korean japonica cultivars. The breeding materials will be applied in the breeding programs to diversify the grain size and shape of japonica rice.

Development of Restriction Fragment Length Polymorphism(RELP) Markers in Silkworm, Bombyx mori (누에 RFLP(제한단편 다형현상)마커 개발)

  • A silkworm Bombyx mori genomic DNA library was constructed from polyphagous J111 strain and unpolyphagous $C_3$ strain to develop the genomic study by DNA makers. Genomic DNAs of two strains were digested with restriction enzyme EcoRI and ligated into pUC18. The ligated plasmids were transferred into E. coli host strain DH5$\alpha$. When the genomic DNAs were hybridized with insert DNAs from transformant, could be categorized from hybridization patterns to three groups as high repetitive sequence, moderately repetitive sequence, and low-copy number sequences. A total of 219 clones containing single or low-copy number sequence inserts were examined for any polymorphisms between two strains of J111 and $C_3$. Forty six clones showed RFLPs and 10 of these clones were used as a probe of analysis of $F_2$ population derived from crossing between J111 and $C_3$ strain. The genetic inheritance tested with each clones will be important tools to construct the genetic map of the silkworm, Bombyx mori.

Processing of Pen Shell By-product Hydrolysate Using Response Surface Methodology (반응표면분석법에 의한 키조개 부산물 단백질 가수분해물의 제조조건)

  • The hydrolysis of pen shell by-product by the APL $440^{TM}$, selected as the suitable alkaline protease on the basis of cost per unit enzyme activity, was optimized using response surface methodology(RSM). A model equation obtained from the results of RSM could be used for the prediction of degree of hydrolysis(DH) as follows: $%DH=51.126+2.419\;pH+2.415T-2.426S-2.846pH^2-4.211T^2-3.014t^2+2.419S^2$. From the ridge analysis, the conditions favoring the highest degree of hydrolysis were pH 10.2, $61.4^{\circ}C$, 2.58 hrs reaction time, 30.9% substrate concentration, and 0.32% enzyme/substrate ratio. The effect of autolysis affecting degree of hydrolysis in pen shell by-product was negligible. Hydrolysate produced under the optimal condition increased 3.5 times and 7.7 times in amino nitrogen and salinity, respectively, comparing with raw pen shell by-product.

Cloning and Expression of cDNA Encoding a Cysteine Protease Inhibitor from Clamworm and Its Possible Use in Managing Anoplophora glabripennis Motschulsky (Coleoptera: Cerambycidae)

  • A cDNA encoding a cysteine protease inhibitor (CPI) was isolated from the cDNA library of clamworm Perinereis aibuhitensis Grube. The deduced amino acid sequence analysis showed that the protein had 51%, 48%, and 48% identity with Zgc:153129 from Danio rerio, cystatin B from Theromyzon tessulatum, and the ChainA, stefin B tetramer from Homo sapiens, respectively. The gene was cloned into the intracellular expression vector pET-15b and expressed in Escherichia coli. The recombinant CPI (PA-CPI) was purified by affinity chromatography on Ni-charged resin and ion-exchange chromatography on DEAE-Sepharose FF. The relative molecular mass of PA-CPI was 16 kDa as deduced by SDS-PAGE. Activity analysis showed that the recombinant protein could inhibit the proteolytic activity of papain. A constitutive and secretive expression vector was also constructed, and the cDNA encoding CPI was subcloned into the vector for extracellular expression. Western blotting analysis results showed that the PA-CPI was secreted into the medium. Bioassay demonstrated that E. coli DH5${\alpha}$ harboring pUC18ompAcat-CPI showed a significant difference in mortality to the Asian longhorned beetle Anoplophora glabripennis compared with untransformed E. coli DH5${\alpha}$ and control.

A Method of Rating Curve Adjustment (수위유량곡선보정방법에 대하여)

  • With the use of many rivers increased nearly to the capacity, the need for information concerning daily quantities of water and the total annual or seasonal runoff has became increased. A systematic record of the flow of a river is commonly made in terms of the mean daily discharge Since. a single observation of stage is converted into discharge by means of rating curve, it is essential that the stage discharge relations shall be accurately established. All rating curves have the looping effect due chiefly to channel storage and variation in surface slope. Loop rating curves are most characteristic on streams with somewhat flatter gradients and more constricted channels. The great majority of gauge readings are taken by unskilled observers once a day without any indication of whether the stage is rising or falling. Therefore, normal rating curves shall show one discharge for one gauge height, regardless of falling or rising stage. The above reasons call for the correction of the discharge measurements taken on either side of flood waves to the theoretical steady-state condition. The correction of the discharge measurement is to consider channel storage and variation in surface slope. (1) Channel storage As the surface elevation of a river rises, water is temporarily stored in the river channel. There fore, the actual discharge at the control section can be attained by substracting the rate of change of storage from the measured discharge. (2) Variation in surface slope From the Manning equation, the steady state discharge Q in a channel of given roughness and cross-section, is given as {{{{Q PROPTO SQRT { 1} }}}} When the slope is not equal, the actual discharge will be {{{{ { Q}_{r CDOT f } PROPTO SQRT { 1 +- TRIANGLE I} CDOT TRIANGLE I }}}} may be expressed in the form of {{{{ TRIANGLE I= { dh/dt} over {c } }}}} and the celerity is approximately equal to 1.3 times the mean watrr velocity. Therefore, The steady-state discharge can be estimated from the following equation. {{{{Q= { { Q}_{r CDOT f } } over { SQRT { (1 +- { A CDOT dh/dt} over {1.3 { Q}_{r CDOT f }I } )} } }}}} If a sufficient number of observations are available, an alternative procedure can be applied. A rating curve may be drawn as a median line through the uncorrected values. The values of {{{{ { 1} over {cI } }}}} can be yielded from the measured quantities of Qr$.$f and dh/dt by use of Eq. (7) and (8). From the 1/cI v. stage relationship, new vlues of 1/cI are obtained and inserted in Eq. (7) and (8) to yield the steady-state discharge Q. The new values of Q are then plotted against stage as the corrected steadystate curve.

Cloning and Characterization of a Cellulase Gene from a Plant Growth Promoting Rhizobacterium, Bacillus subtilis AH18 against Phytophthora Blight Disease in Red-Pepper (고추역병을 방제하는 PGPR균주 Bacillus subtilis AH18의 항진균성 Cellulase 유전자의 Cloning 및 효소 특성 조사)

  • Using PCR amplification, we cloned a cellulase gene (ce/H) from the Bacillus subtilis AH18 which has plant growth-promoting activity and antagonistic ability against pepper blight caused by Phytophthora capsici. The 1.6 kb PCR fragment contained the full sequence of the cellulase gene and the 1,582 bp gene deduced a 508 amino acid sequence. Similarity search in protein database revealed that the cellulase of B. subtilis AH18 was more than 98% homologous in the amino acid sequence to those of several major Bacillus spp. The ce/H was expressed in E. coli under an IPTG inducible lac promoter on the vector, had apparent molecular weight of about 55 kDa upon CMC-SDS-PAGE analysis. Partially purified cellulase had not only cellulolytic activity toward carboxymethyl-cellulose (CMC) but also insoluble cellulose, such as Avicel and filter paper (Whatman No. 1). In addition, the cellulase could degrade a fungal cell wall of Phytophthora capsici. The optimum pH and temperature of the ce/H coded cellulase were determined to be pH 5.0 and $50^{\circ}C$. The enzyme activity was activated by $AgNO_3$ or $CoCl_2$. However its activity was Inhibited by $HgC1_2$. The enzyme activity was activated by hydroxy urea or sodium azide and inhibited by CDTA or EDTA. The results indicate that the cellulase gene, ce/H is an antifungal mechanism of B. subtilis AH18 against phytophthora blight disease in red-pepper.

Effect of plasmid curing on the 2, 3-dihydroxybenzoic acid production and antibiotic resistance of Acinetobacter sp. B-W (Acinetobacter sp. B-W의 2, 3-dihydroxybenzoic acid 생산과 항생제 저항성에 미치는 플라스미드 제거 효과)

  • Acinetobacter sp. B-W producing siderophore, 2, 3-dihydroxybenzoic acid (DHB) was analyzed for plasmid content. Strain B-W harbored plasmid of 20 kb in size. Growth at $43^{\circ}C$ was effective in producing mutant cured of plasmid of strain B-W. This mutant lost the ability to produce 2, 3-DHB. Formation of siderophore halos on the chrome azurol S (CAS) agar medium was not detected by cured strain B-W. pHs of supernatants of wild type strain B-W and cured mutant grown in glucose and $MnSO_4$ containing medium at $28^{\circ}C$ for 3 days were 4.5 and 8.5, respectively. Antibiotic resistance against ampicillin, actinomycin D, bacitracin, lincomycin, and vancomycin was lost in cured mutant. Plasmid curing of strain B-W resulted in drastic reduction of minimal inhibitory concentration (MIC) of several antibiotics. E. coli $DH5{\alpha}$ was transformed with plasmid isolated from strain B-W. The transformant E. coli $DH5{\alpha}$ harbored a plasmid of the same molecular size as that of the donor plasmid. Transformant E. coli $DH5{\alpha}$ produced 2, 3-DHB and contained antibiotic resistant ability. Thus a single plasmid of 20 kb seemed to be involved in 2, 3-DHB production. Genes encoding resistance to antibiotics were also supposed to be located on this plasmid.

Properties of Zeolite A Synthesized by Natural Bentonite (천연 벤토나이트로부터 합성된 제올라이트 A의 특성)

  • Synthetic zeolite was prepared by using of natural bentonite from Kampo area and the application of detergent builder was investigated. The optimum synthetic condition was SiO$_2$/Al$_2$O$_3$= 2, Na$_2$O/A1$_2$O$_3$=1, H$_2$O/A1$_2$O$_3$=30 at 90$^{\circ}C$ for 3hr and it was found by XRD analysis that the zeolite synthesized under this condition was type A. When the zeolite A synthesized under the optimum condition was contacted with 40$^{\circ}$Dh CaCl$_2$solution at 30$^{\circ}C$ for 15min, the cation exchange capacity was 264.9mg CaO/g-zeolite. And the whiteness of the sample was 89% and the mean particle size was 9.95$\mu\textrm{m}$.
