한우 씨수소의 고환 둘레와 정액 성상 및 농가 인공수정 수태율과의 관계 (Relationships of Scrotal Circumference of Hanwoo Sire to Semen Characteristics and Conception Rate of Artificial Insemination in Farm)

  • The relationships of scrotal circumference (SC) to semen characteristics and the conception rate (70 days-nonreturn rate) of artificial insemination in farm were studied with 137 heads of bull in Hanwoo. The average and range of SC were $38.27{\pm}3.90$ cm and 26.0~52.5 cm, respectively. Hanwoo bulls were classified with SC, divided into 34 cm below group, 34~39 cm group, and 39 cm over group. The 5,487 semen records of 43 heads of bull from July. 1. 2007 to June. 30. 2008. were used to determine the relationships between SC and semen characteristics. The semen concentration and total sperm number of each group were 11.18, 16.68, and $17.4{\times}10^8/ml$, and 69.83, 101.64 and $114.40{\times}10^8$/ejaculate. The bulls with 34 cm or more SC were higher than the bulls with 34 cm below in semen concentration and total sperm number (p<0.01). But between SC and semen volume have no significant relationship (p>0.05). The 9,862 mating records of 44 farm with 137 heads of bull were used to determine the relationships between SC and conception rate. The conception rate of 1st artificial insemination were 73.31, 74.16, and 77.33 % in each group. Also SC was positively correlated with pregnancy rate (r=0.12, p=0.17). These results indicate that SC correlates positively with semen characteristics, and maybe with pregnancy rate in Hanwoo.

精子 洗滌液의 pH와 洗滌頻度 및 수소個體가 精子洗滌液內 水素이온濃度와 소 精子의 尖帽反應에 미치는 影響 (Effects of pH of Washing Solution, Washing Frequency and Individual Bull on Proton Concentration in the Sperm Washed Solution and Sperm Acrosome Reaction)

  • 본 實驗은 精子 洗滌液의 pH와 洗滌頻度 및 수소개체가 精子洗滌液內 水素이온과 精子尖帽反應에 미치는 影響을 究明하기 위하여 실시하였는바 그 결과는 다음과 같다. 1. 精子를 SHP液으로 4차례 反復洗滌하여 培養하였을 때, 洗滌液의 吸光度差間 變化는 2차 精子洗滌液에서 가장 높았다. 2. 精子를 pH 5.99, 6.38, 6.78, 7.10, 7.40, 7.69, 8.15, 8.45 및 8.83 SHP液으로 3번 洗滌하였을 때, 精子洗滌液의 吸光度差間 變化는 pH 7.69~8.83에서 유의하게 증가하였으며, 1차 精子洗滌液이 2차와 3차 精子洗滌液보다 유의하게 높았다. 3. Holstein, KNC 및 Hereford 精子를 pH 5.99, 6.38, 6.78, 7.10, 7.40, 7.69, 8.15, 8.45 및 8.83의 SHP液으로 3번 洗滌하였을 때, 精子洗滌液의 光度差間 變化는 Holstein이 KNC와 Hereford보다 유의하게 높았으며, Holstein은 pH 7.69~8.83에서, KNC과 Hereford는 8.15에서 각각 유의하게 증가하였다. 4. 精子를 pH가 6.8, 7.1 및 7.4인 SHP液으로 3번 洗滌한 다음 pH 7.4인 mTALP液에서 15분간 培養하였을 때, 精子洗滌液의 吸光度差間 變化는 1차 및 2차 精子洗滌에서 pH 7.1과 7.4가 6.8보다 유의하게 높았으며, 精子尖帽反應率은 pH 7.1 및 7.4가 각각 68.8 및 72.9%로 pH 6.8의 49.1%보다 유의하게 높았다(P<0.05).

Testicular Cell Indices and Peripheral Blood Testosterone Concentrations in Relation to Age and Semen Quality in Crossbred (Holstein Friesian×Tharparkar) Bulls

  • Present study analyzed the changes in peripheral blood testosterone concentrations and testicular cytogram in relation to age and semen quality in crossbred males. Three different age groups of crossbred males viz. bull calves (6 months, n = 5), young bulls (15 months, n = 5) and adult bulls (4 to 6 years, n = 8) were utilized for the study. Testicular fine needle aspiration cytology technique was used to quantify testicular cytology and their indices. Peripheral blood testosterone concentrations were measured using enzyme-linked immunosorbent assay method. Semen samples collected from adult bulls were microscopically evaluated for quality parameters. Mean peripheral blood testosterone concentrations in bull calves, young bulls and adult bulls were $2.28{\pm}0.09ng/mL$, $1.42{\pm}0.22ng/mL$ and $5.66{\pm}1.08ng/mL$ respectively, and that in adult bulls were significantly different (p<0.01) from young bulls and bull calves. There was no significant difference between the proportion of different testicular cells in bull calves and young bulls. Between young and adult bulls, significant differences (p<0.01) were observed in the proportion of spermatocytes, spermatozoa, and sperm: Sertoli cell ratio. The proportions of Sertoli cells showed a significant difference (p<0.01) between the three age groups. The number of primary spermatocytes had a positive correlation with peripheral blood testosterone concentrations in bull calves (r = 0.719, p<0.01). Number of Sertoli cells per 100 germ cells was negatively correlated with blood testosterone concentration in young bulls (r = -0.713, p<0.01). Among different semen parameters in adult bulls, ejaculate volume (r = 0.790, p<0.05) had positive relationship, and sperm motility had significant negative correlation (r = -0.711, p<0.05) with testosterone concentrations. The number of Sertoli cells and Sertoli cell index had a positive correlation with various semen quality parameters (p<0.001). Results of the present study conclude that number of Sertoli cells and Sertoli cell index are good indicators of semen quality, but peripheral blood testosterone concentrations may not have a direct relationship with various seminal attributes in crossbred bulls.

제주흑우 동결정액 제조에 있어 Low Density Lipoproteins (LDL)과 항산화제로서 Taurine, Hypotaurine 그리고 Trehalose 조합이 동결 융해 후 정자의 성상에 미치는 영향 (Effect of LDL in Combination with Taurine, Hypotaurine and Trehalose as a Antioxidant on Freezing Thawed Semen Function in Korean Jeju Black Bull)

  • 본 연구에서는 제주흑우의 유전자원 보존과 증식에 있어 성공적인 인공수정을 위한 안정적인 동결정액 제조법을 수립하고 동결 융해 후 정자의 품질을 개선하기 위하여 제주흑우의 동결 정액 제조시 LDL, taurine, hypotaurine 그리고 trehalose를 첨가하여 이들이 동결 융해 후 정자의 성상에 미치는 영향에 대하여 알아보고자 수행하였다. 제주흑우의 정액 동결 시 LDL, LDL-taurine, LDL-hypotaurine 그리고 LDL-trehalose의 첨가는 ($63.40%{\pm}7.39$, $69.70%{\pm}6.12$, $67.25%{\pm}3.21$, $64.55%{\pm}2.43$) 대조구에($56.25%{\pm}6.42$) 비하여 모두 유의적인 생존율의 증가를 나타냈다 (p<0.05). 정자막 온전성 검사를 통한 꼬리막 팽창 정자의 비율은 LDL-taurine을 첨가 실험구에서 $70.55%{\pm}5.16$, LDL-hypotaurine 첨가 실험구에서 $64.45%{\pm}5.85$, LDL-trehalose 실험구에서 $64.50%{\pm}2.78$, LDL 단독 첨가 실험구에서 $61.65%{\pm}5.18$로 대조구 $51.90%{\pm}9.99$보다 모두 유의적으로 높은 결과를 나타냈다 (p<0.05). 동결 융해 후 정자의 첨체막 변화 양상에 있어서는 B pattern의 비율은 대조구와 실험구 모두 유의적 차이가 없었으며, F pattern은 LDL-taurine의 조합만이 대조구와 유의적인 차이를 나타내며 증가하였다 (p<0.05). 그러나 AR pattern의 비율은 LDL-taurine, LDL-trehalose, LDL-hypotaurine 그리고 LDL 처리의 순으로 대조구에 비하여 모두 유의적으로 낮은 수준의 AR pattern의 비율을 나타냈다(p<0.05). 정자의 수정능력 평가에 있어 LDL-taurine, LDL-hypotaurine 그리고 LDL-trehalose 모두 대조구에 대하여 유의적으로 높은 웅성전핵 형성율과 SFI를 나타냈으며 (p<0.05), 특히 LDL-taurine의 첨가는 LDL의 단독 처리에 비하여도 유의적으로 높은 수준의 웅성전핵 형성율과 SFI를 나타냈다 (p<0.05). 또한, decondenced sperm 비율에 있어 LDL, LDL-taurine, LDL-hypotaurine 그리고 LDL-trehalose 실험구 모두 대조구에 비하여 유의적으로 높은 결과를 나타냈다 (p<0.05). 본 연구의 결과는 제주흑우 정액의 동결정액 제조에 있어 안정적인 제조법을 공급할 수 있으며, 동결 융해 후 정자의 기능 개선 방법에 보다 많은 정보를 제공할 수 있을 것으로 사료된다.

Seminal Plasma Heparin Binding Proteins Improve Semen Quality by Reducing Oxidative Stress during Cryopreservation of Cattle Bull Semen

  • Heparin binding proteins (HBPs) are produced by accessory glands. These are secreted into the seminal fluid, bind to the spermatozoa at the time of ejaculation, favour capacitation, acrosome reaction, and alter the immune system response toward the sperm. The present study was conducted with an objective to assess the effect of purified seminal plasma-HBPs (SP-HBPs) on cross bred cattle bull sperm attributes during two phases of cryopreservation: Pre freezing and freezing-thawing. SP-HBPs were purified from pooled seminal plasma by heparin affinity chromatography. Three doses of SP-HBPs i.e. 10, 20, $40{\mu}g/mLs$ semen were standardized to find out the optimum dose and $20{\mu}g/mLs$ was found to be an optimum dose. Semen as such and treated with SP-HBPs was diluted with sodium citrate-egg yolk diluter and cryopreserved as per the standard protocol. Sperm parameters i.e. motility, viability, Hypo-osmotic swelling test (HOST), acrosome damage, in vitro capacitation and lipid peroxidation were evaluated in SP-HBP treated and untreated (control) semen at both phases of cryopreservation. A considerable variation in percent sperm motility, viability, membrane integrity (HOST), acrosome damage, acrosome reaction and lipid peroxidation was observed at both phases among the bulls irrespective of the treatment. Incubation of neat semen with $20{\mu}g/mL$ SP-HBP before processing for cryopreservation enhanced the average motility, viability, membrane integrity by 7.2%, 1.5%, 7.9%, and 5.6%, 6.6%, 7.4% in pre-frozen and frozen-thawed semen in comparison to control. There was also an average increase of 4.1%/3.9% in in vitro capacitation and acrosome reaction in SP-HBPs-treated frozen-thawed semen as compared to control. However, binding of SP-HBPs to the sperm declined acrosome damage and lipid peroxidation by 1.3%/4.1% and 22.1/$32.7{\mu}M$/$10^9$ spermatozoa in SP-HBP treated pre-frozen/frozen-thawed semen as compared to control, respectively. Significant (p<0.05) effects were observed only in motility, HOST and in vitro acrosome reaction. It can be concluded that treatment of neat semen with SP-HBPs before cryopreservation minimized the cryoinjury by decreasing the generation of reactive oxygen species.

배양액중의 calcium 이온 농도 및 caffeine과 Ca-ionophore A23187 처리가 소정자의 수정능획득에 미치는 영향 (The effect of calcium ion concentrations in the medium and the treatment of caffeine and Ca-ionophore A23187 on in vitro capacitation of bull spermatozoa)

  • The present study was performed to investigate the effect of Ca ion concentration on sperm viability and acrosome reaction rate and to evaluate the effect of treatments using caffeine and Ca-ionophore A23187 on acrosome reaction rate in frozen-thawed bull spermatozoa. Viabilities of in vitro capacitated bull spermatozoa at 0, 2.25 and 4.5 mM Ca ion concenrations were 21.00, 26.00 and 22.59%, respectively and significantly higher in Ca ion 2.25mM added group than Ca ion free group (p<0.05) and acrosome reaction rates of in vitro capacitated bull spermatozoa were 17.09, 52.15 and 47.92%, respectively and significantly high in Ca ion added groups(p<0.05). Viabilities in vitro capacitation by caffeine and Ca-ionophore A23187 in control, caffeine treated group, Ca-ionophore A23187 treated group and caffeine+Ca-ionophore A23187 treated group were 37.91, 27.67, 22.33 and 25.59%, respectively and significantly higher in control than treated groups(p<0.05), there were no significant differences among the treated groups, and acrosome reaction rates were 10.33, 37.92, 48.09 and 57.17%, respectively and there were significant differences among the groups(p<0.05), especially higher in caffeine+Ca-ionophore A23187 treated group than others.

Effect of Antioxidant Preservative on Cold Protection Ability of Low Grade Riverine Buffalo (Bubalus bubalis) Bull Spermatozoa

  • An experiment was conducted to investigate the effect of Butylated Hydroxy Anisole (BHA), Butylated Hydroxy Toluene (BHT), Pentoxifylline (PTX), Theophylline (TPY) and Theobromine (TBR) on cold protection ability of Murrah buffalo semen at room ($22-25^{\circ}C$) and refrigerated temperature ($4-7^{\circ}C$). Each semen sample was divided into six parts of equal volume and sperm concentration; the first was kept as a control and the remaining five were treated with BHA, BHT, PTX, TPY or TBR. Sperm motility, abnormal spermatozoa, live-dead count, hypo-osmotic swelling and acrosomal integrity were studied at room and refrigerated temperature for various incubation periods viz.; 0, 4, 8, 12 and 24 h at room and 0, 12, 24, 36, 48, 60 and 72 h at refrigerated temperature. Significant improvement in sperm motility, live-dead count, hypo-osmotic swelling and acrosomal integrity were observed in BHT, PTX and TPY fortified extender at room and refrigerated temperature for various incubation periods. From the present study it could be concluded that cold protection ability of buffalo semen can be improved through the addition of BHT followed by PTX and TPY.

Effect of Multiple Showering and Vitamin Supplementation on Sexual Behaviour, Quality and Freezability of Buffalo Bull Semen

  • In a summer study during May to July, involving 12 young Murrah buffalo bulls at forty months of average age, the effects of multiple shower vs single shower body cooling and vitamin A, D and E supplementation on the sexual behaviour, semen quality and freezability were investigated. The animals were divided into two groups (6 animals in each group) and housed in a half-walled shed with proper spacing, the feeding management being identical. The bulls in the control group were given a single shower at 1000 h, whereas the experimental bulls were given four showers at 10,12,14 and 16 h. In addition, the experimental bulls were given vitamin A, D and E injections at fifteen day intervals. The sexual behaviour of bulls was observed in terms of reaction time, sexual aggressiveness and ejaculatory thrust. Semen quality of all the bulls was assessed in terms of volume, mass activity, live-dead sperm and sperm concentration, sperm motility and morphology, and acrosomal abnormality. The sexual behaviour did not vary significantly between the groups, whereas semen quality differed significantly for volume, per cent live sperms, total sperms per ejaculate and total live sperm per ejaculate between groups. It can be concluded that sexual behaviour was not influenced by the thermal comfort treatment coupled with periodic vitamin A, D and E injections. But the treatments improved most of the seminal traits in the experimental group of bulls. However, benefit of treatment was not reflected in the freezability traits of the semen.

Effect of Caffeine, cAMP and Cattle Seminal Plasma on Freezability of Buffalo Bull Semen

  • An experiment was conducted to investigate the effect of caffeine, cAMP and cattle seminal plasma on preservation of semen at ultra low temperature ($-196{^{\circ}C}$). Each semen sample was divided into four parts equal in volume and sperm concentration; three were treated with caffeine, or cAMP, or cattle seminal plasma (CSP) and the fourth was kept as control. Sperm motility, abnormal spermatozoa, live-dead count and acrosomal damage were studied at different stages of freeze preservation viz.; just after dilution, at $5{^{\circ}C}$, at glycerolisation, before freezing, just after freezing, 24 hours of storage, and one week of storage. Sperm motility (58.39, 61.33, 52.00 and 50.39 per cent), non-eosinophilic spermatozoa (72.55, 69.98, 63.31 and 67.64 per cent), abnormal spermatozoa (5.71, 4.98, 8.04 and 5.66 per cent) and acrosomal damage (13.28, 13.33, 14.80 and 14.65 per cent) were observed in cAMP, caffeine, cattle seminal plasma and control, respectively, at every stage of freeze preservation. From this study it could be concluded that freezability of buffalo semen can be improved through the addition of caffeine followed by cAMP and cattle seminal plasma.

Correlation between Fluoromicroscipic Assessment of Mitochondria Function of Frozen-Thawed Hanwoo Spermatozoa and Blastocyst Development Following In Vitro Fertilization

  • This experiment was to investigate whether the mitochondria function assessment can be used for the prediction of sperm fertility through examining the correlation between mitochondria fluoromicroscopic frequency of frozen-thawed eight Hanwoo bull semen using rhodamine123 (R123) and in vitro embryo development following fertilization. Individual sperm were stained in 5 ${\mu}g/mL$ R123-added calcium-free Sp-TALP for 30 min at 0 h, 6 h, 12 h and 24 h after thawing and examined their mid-piece under an epifluorescence microscope using 495 nm excitation filter (x1,000). Three replications were taken, and at least 300 sperm per individual were examined. When semen samples were separated into two groups (good and poor) by sperm motility and fluorescent frequencies at just after thawing, average fluorescent frequencies were remarkably reduced as time going (0 h; $53.29{\~}72.94\%$, 6 h; $21.40{\~}58.90\%$, 12 h; $8.26{\~}25.93\%$, 24 h; $1.00{\~}13.78\%$, irrespective of selected group, and there were no differences at 6 h or 12 h after thawing between selected groups but indicated significant difference at 24 h after thawing (p<0.05). In vitro fertilization rates in good and poor groups ranging $70.8{\~}77.8\%$ and $52.1{\~}84.5\%$, respectively, were not significantly different. However, in vitro development rates of the same groups ranging $25.7{\~}40.0\%$ and $12.9{\~}1.8\%$, respectively, were significant different (p<0.05). These results demonstrate that mitochondria fluoromicroscopic assessment of frozen-thawed bovine sperm may be used as a criterion to select more fertile sperm.
